Isolation and Structure Elucidation of Radical Scavengers from Chrysanthemum boreale Makino (산국(山菊)의 자유라디칼 소거 물질 분리 및 동정)

  • Two antioxidative compounds in Chrysanthemum boreale were isolated by a bioassay using a 2, 2-diphenyl-1-picrydrazyl(DPPH) free radical. They were identified as apigenin, linarin(acacetin-7-O-rutinoside) on the basis of $^1H\;and\;^{13}C-NMR$ and MS data. The DPPH radical scavenging activity$(RC_{50}:\;13.3{\mu}g)$ of apigenin was similar to that of ${\alpha}-tocopherol\;(12.1{\mu}g)$ and L-ascorbic acid$(13.1{\mu}g)$

Antioxidative and Free Radical Scavenging Effects of Rhus javanica Linne (오배자의 항산화 및 자유라디칼 소거효과)

  • This study was carried out to investigate the antioxidative activities of medicinal plants. Through the examination of methanol extracts from 30 species for radical scavenging effects using DPPH method, the extracts from Rhus javanica Linne, Smilax china Linne and Polygonum cuspidatum Siebold et Zucarinii showed strong antioxidative activity. Because of its highest antioxidative activity among 30 medicinal plants, radical scavenging effects of 4 different extract compartments (n- Hexane, EtOAc, BuOH and $H_2O$ extracts) from Galla Rhois MeOH extract of Rhus javanica Linne were examined by DPPH method and antioxidant effects on the 4 different extract compartments were tested by Ferric-Thiocyanate method. Antioxidative activities of n- Hexane, EtOAc and BuOH extracts were similar or even higher than that of natural (tocopherol) or synthetic antioxidants (BHA), suggesting that major fractions for the antioxidative activity of Rhus javanica Linne were the n- Hexane, EtOAc and BuOH extract compartments.

Anti-wrinkle Effects of Cervi Pantotrichum Cornu Pharmacopuncture Solution (녹용 약침액의 주름 개선 효과에 관한 연구)

  • 목적 : 본 연구는 녹용약침액의 주름 개선 효과에 대하여 여러 가지 in vitro 실험을 통해 확인해 보고자 계획되었다. 방법 : 녹용약침액의 항산화효과를 DPPH(1,1-diphenyl-2-picrylhydrazyl) 자유라디칼 소거법을 이용하여 측정하였고, 또, elastase 억제 효과를 측정하였다. 그리고 녹용약침액이 사람 정상 섬유아세포 HS 68에서 UVB 조사 후 type I procollagen의 생산량 회복에 미치는 효과를 ELISA법을 이용하여 측정하였다. 결과 : 녹용약침액에서 우수한 DPPH 자유라디칼 소거 효과가 관찰되었다. 또한 통계적으로 유의한 elastase 활성 억제 효과를 관찰할 수 있었다. 사람 섬유아세포 HS 68을 이용한 실험에서는 녹용약침액 처치군에서 UVB 조사로 감소된 type I procollagen이 유의하게 회복되는 것이 관찰되었다. 결론 : 본 연구 결과에 의하면, 녹용약침액에는 유효한 주름개선 효과가 있어, 미용약침 소재로 개발할 수 있을 것으로 사료된다.

Quality Characteristics of Tarts Made with Molecular Press Dehydrated Purple Sweet Potatoes during Storage (분자압축탈수된 자색고구마로 제조한 타르트의 저장 중 품질특성)

  • Purpose: Molecular press dehydration is one of the dehydration methods. The purpose of this study was to investigate the quality characteristics of tarts made with dehydrated purple sweet potatoes during the storage period. Methods: Quality characteristics of purple sweet potato tarts were evaluated by analyzing moisture content, water activity, total phenolics, anthocyanins, 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, color, and sensory evaluation for 45 days of storage. Results: Moisture content of tart crust made with molecular press dehydration treated purple sweet potatoes with concentrations of 20%, 40%, 60%, and 80% maltodextrin as a dehydrating agent for 45 days was 8.47%, 7.95%, 6.96%, and 6.24% respectively; however, the moisture content of non-treated tart crust was 11.99% (p<0.05). Total phenolics, anthocyanins, and DPPH free radical scavenging activity of dehydrated purple sweet potato tarts were lowered than those of non-treated tart (p<0.05). Conclusion: These results indicated that tarts made with molecular press dehydrated purple sweet potatoes had effectively controlled moisture content and water activity during storage although total phenolics, anthocyanins, DPPH free radical scavenging activity, color, and sensory evaluation were decreased.

Antioxidant Effect of Assai Palm Methanolic Extract (Assai 열매 메탄올 추출물의 항산화 효과)

  • The methanolic product of assai(Euterpe oleracea Mart) scavenged the intracellular reactive oxygen species (ROS) and 1,1-diphenyl-2- picrylhydrazyl (DPPH) radical, and prevented lipid peroxidation. The radical-scavenging activity of assai palm methanolic extract protected the viability of peritoneal macrophage cells exposed to hydrogen peroxide($H_2O_2$). Furthermore, the extract reduced the apoptotic-cell formation induced by $H_2O_2$, as demonstrated by the decrease in the number of hypodiploid cells and in the apoptotic-cell body formation. These results indicate that assai palm methanolic extract has radical-scavenging activity and ameliorates $H_2O_2$-induced cytotoxicity.

Antioxidant, Antimicrobial Acitivities and Cytotoxicity of Hot Water Extracts of Major Herbs in Korea (국내산 주요허브 열수 추출물의 항산화, 항균활성 및 세포독성)

  • In this study, the DPPH free radical scavenging activity, antimicrobial effects, growth inhibition and cytotoxicity of herb extracts were determined to screen alternative antibiotics. Hot water extracts of 10 species herbs (Origanum vulgare, Monarda didyma, Echinacea purpurea, Ocimum basilicum, Mentha piperita, Melissa officinalis, Thymus vulgaris, Stevia rebaudiana, Rosmarinus officinalis, Matricaria recutica) were used. DPPH free radical scavenging activity of all herb extracts was ranged from 31.4-49.9%, and significantly great activties were found at Rosmarinus officinalis, Origanum vulgare and Matricaria recutica (P<0.05). Hot water extracts of monarda didyma, origanum vulgare, thymus vulgaris and rosmarinus officinalis showed greater antimicrobial activities compared to others. Additionally, those four extracts represented relatively low cytotoxicity compared to others. As a result, it was found that Origanum vulgare and Rosmarinus officinalis which possessed great antioxidant and antimicrobial activity with less cytotoxicity. So these two herb extracts can be used as an alternative of antibiotics for organic farming.

Reactive Oxygen Species and Cytotoxicity of Bamboo (Phyllostachys pubescens) Sap (대나무수액의 활성산소 소거활성과 세포독성)

  • The antioxidant properties of bamboos sap isolated from Phyllostachys pubescens were investigated. This product scavenged intracellular reactive oxygen species (ROS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, and prevented lipid peroxidation. The radical scavenging activity of bamboo sap protected the viability of peritoneal macrophage cells exposed to hydrogen peroxide $(H_2O_2)$, Furthermore, bamboo sap reduced apoptotic cell formation induced by $H_2O_2$ as demonstrated by decreases in the number of hypo-diploid cells am apoptotic cell body formation. These results indicate that bamboo sap has radical scavenging activity and ameliorates $H_2O_2$ induced cytotoxicity.

Free radical scavenging and α-glucosidase inhibitory effects of a roots extract of Aruncus dioicus var. kamtschaticus (재배 삼나물 뿌리 추출물의 자유 라디칼소거 및 α-glucosidase 저해활성)

  • As part of our continuing search for bioactive natural products, the antioxidant and ${\alpha}$-glucosidase inhibitory activities of an 80% methanolic extract and organic solvent soluble-portions of Aruncus dioicus var. kamtschaticus roots were investigated by using a bioassay system. The antioxidant activity of A. dioicus var. kamtschaticus roots extract and organic solvent soluble-portions were assessed by examining with 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) ($ABTS^+$) radical scavenging. In addition, anti-diabetic effects of the A. dioicus var. kamtschaticus root extract and organic solvent soluble-portions were tested via ${\alpha}$-glucosidase inhibition assay. The total phenolic contents of the products were determined by applying UV-VIS spectrophotometry. All tested samples showed dose-dependent radical scavenging and ${\alpha}$-glucosidase inhibitory properties. In particular, the ${\alpha}$-glucosidase inhibitory and radical scavenging effects of the ethyl-acetate (EtOAc)-soluble portion from the roots of A. dioicus var. kamtschaticus were greater than those from other solvent-soluble portions. These results indicate that A. dioicus var. kamtschaticus could be considered a new effective source of natural antioxidants and anti-diabetic materials. More systematic research of the constituents of the roots of this A. dioicus variety will be conducted to further develop its antioxidative and anti-diabetic properties.

Comparison of the Antioxidant Activities of Organic Solvent Fractions of Leaf and Root Extracts of Peucedanum insolens Kitagawa (왕산방풍의 잎과 뿌리의 유기용매 분획물에서의 항산화 활성 비교)

  • This study compared and analyzed the antioxidant activities of various organic solvent fractions from the leaves and roots of Peucedanum insolens Kitagawa. For this study, the dried leaves and roots of P. insolens Kitagawa were first extracted using 70% ethanol. The extracts were sequentially sub-fractionated in the order of hexane, chloroform, ethyl acetate, n-butanol, and water. The results revealed that the distribution of total phenolic contents by organic solvent fractions showed the same pattern in both the leaves and roots, with the highest in the ethyl acetate fraction (101.1±1.0 mg vs 71.2±3.4 mg of GAE/mg), but the lowest content in the hexane fraction (9.5±0.2 mg vs 7.5±2.1 mg of GAE/mg). The distribution of total flavonoid content in the organic solvent fractions showed the same pattern as that of total phenolic content. The results of DPPH, ABTS, and FRAP assays showed that the leaf and root extracts exhibited free radical scavenging activity in the same pattern, particularly, the ethyl acetate fraction had the highest activity. These results indicate that not only the roots of P. insolens Kitagawa but also the leaves possess potential substances that exhibit strong antioxidant activity. Significant correlations (R=0.903, p<0.0001, DPPH radical; R=0.891, p<0.001, ABTS radical; R=0.745, p<0.05, FRAP radical) between total phenolics and radical scavenging activities, but also significant correlations (R=0.867, p<0.001, DPPH vs. ABTS radicals; R=0.882, p<0.0001, DPPH vs. FRAP radicals; R=0.973, p<0.0001, ABTS vs. FRAP radicals) between radical scavenging activities were found in the organic solvent fractions. Therefore, as in the roots of P. insolens Kitagawa, the leaves possess strong antioxidant capacity and can be used as the main antioxidant material.
