  • Dynamic parameters of biochemistry including the contents of soluble carbohydrate (SC) , crude fat (CF) , fatty arid (FA) and soluble protein (SP) in American send (Panax qufnquelolium L.) seed are reported in the present paper. When embryo ratio (ER) increased from 7.31 to 20.48%, the SC content decreased from 4.08 to 1.16%. After that, SC content changed little. The CF content decreased successively from 34.50 to 14.40% from the beginning to the end during the embryo afterripening. The changes of FA content did not correlate with that of ER and the changing range of SP content was not large during the seed stratification. All of these may provide some information for understanding the dormancy mechanisms of American seng seed.

The Effects of Environmental Conditions and Chemical Treatments on Seed Germination in Astilboides tabularis (Hemsl.) Engl.

  • Tabulis [Astilboides tabularis (Hemsl.) Engl.] has the potential for becoming a useful horticultural crop. This study was conducted to classify the seed dormancy types and to identify the germination conditions that improve the horticultural usefulness of this plant. We found that A. tabularis (Hemsl.) Engl. produced undifferentiated embryos and had a low germination rate of under 50%. In addition, water submersion led to moisture absorption. Therefore, A. tabularis seeds were designated as morphologically dormant. The germination rate was highest (86.3%) at $30^{\circ}C$ under light conditions. Higher temperatures generally led to an acceleration of the germination process, regardless of light condition. To improve the germination rate, seeds were submerged in various concentrations of growth regulators, such as $GA_3$ and kinetin, as well as minerals such as $KNO_3$ and KCl. An analysis of germination characteristics at $30^{\circ}C$ under light conditions revealed that treatment with 200 and $500mg{\cdot}L^{-1}GA_3$ led to an excellent germination rate of 97.0%. Treatment with $100mg{\cdot}L^{-1}GA_3$ led to a better germination rate than the control as well. However, treatment with kinetin, $KNO_3$, and KCl had no effect on germination, regardless of concentration.

Germination of West African Ebony (Diospyros mespiliformis Hochst) Seeds: Effects of Dehydration and Different Pre-sowing Treatments

  • Diospyros mespiliformis is a highly valued and threatened tree species within the Sahelo-Sudanian zone of Africa, but its seed germination requirements under cultivation are not well researched. In a first experiment which aimed at determining germination response of seeds to dehydration, fresh seeds were dried at room temperature for 26 days during which their moisture content, their germinability, and their viability were monitored at two-day intervals. In the second experiment, 14 pre-germination treatments were tested for their effect on the germination of dried seeds. Results showed that fresh seeds had 52.7% moisture and achieved 97.7% germination. As seeds were dried, percentage germination gradually decreased with decreasing moisture content and reached 0% when moisture content had dropped to 18%. Meanwhile, seed viability remained at 100% over drying duration. Seeds that were not germinated after air dry also recorded 100% viability. The most effective treatment for inducing germination of dried seeds was scarification using 98% sulfuric acid for 30 min which resulted in 96.6% germination. This study reports for the first time in D. mespiliformis seeds a desiccation-induced dormancy which can be efficiently alleviated by acid scarification. This study provides useful information that will contribute to efficient management of D. mespiliformis seed resources for propagation.

Germination of Two Rice Cultivars and Several Weed Species (벼와 수종(數種) 논잡초(雜草)의 휴면성(休眠性)과 발아성(發芽性))

  • An experiment was carried out at the International Rice Research Institute in 1987 to understand the seed dormancy and germination habit of rice and several weed species. The germinability of the weed seeds just after harvest was variable depending on the species and ranged from 0 to 72%. Two rice cultivars, IR64(lowland type) and UPLRi-5(upland type) had higher than 95% in germination ability throughout the experimental period due to the fact that the rice seeds came from the harvest of the previous season and dormancy had already been overcome. The length of the storage period needed to overcome dormancy at room temperature($25{\pm}2^{\circ}C$) was about 50 days for Echinochloa glabrescens Munro ex Hook, f., more than 60 days for E. crus-galli ssp. hispidula (Retz.) Honda and 20 days for Ludwigia octovalvis(Jacq.) Raven. Seeds of E. colona(L.) Link, Monochoria vaginalis(Burm. f.) Presl, Fimbristylis miliacea(L.) Vahl and Cyperus difformis L. appeared to have no dormancy. Among the nine species M. vaginalis had the lowest germination of less than 1% throughout the experimental period. However, its seed germinated easily when planted in soil. The low germinability of E. glabrescens, E. crus-galli ssp. hispidula and L. octovalvis just after harvest could be overcome through pretreatment of seeds either by soaking in nitric acid(0.1N) for 1 day or removal of the hull in the grass species, the nitric acid treatment being superior. The results imply that germination habit of weed species varied depending on the species through their differential dormancy period or differential germination strategy.

Four-year Survey on Transitions of the Life Form of Plants after Developing Human-made Wetlands along Boknaecheon of Juam Lake (주암호 복내천 인공습지 조성 후 식물의 생활형에 대한 4년간의 변화 연구)

  • Employing the Numata-type life form, the paper analyzed changes of plants for 4 years at the Human-made Wetlands along Boknaecheon of Juam Lake since its creation. The number of the species planted at the Human-made Wetlands along Boknaecheon of Juam Lake, which was completed in December 2002, were 15 in total including, 10 families, 13 genera, 12 species and 3 varieties. As for the three-featured life forms of the planted species, there were 6 perennial hydatophytes, recording the biggest number of species in dormancy form; species each of $R_5$, $R_3$, $R_{2-3}$ respectively in radicoid from; 20 species of geomantic dissenminule form ($D_1$) in disseminule form and erecred type(e) existed the most in growth form. With regard to the 3 features of life form identified during the final year of the monitoring that lasted 5 years after the completion of the Wetlands, the number of species and individuals was found to have increased but there was no significant change of tendency as against the composition ration(%) of life form. There were 43 species of therophytes (Th) that covered 24.29% in dormancy form, while $R_5$ was prevalent in radicoid form and $D_4$, $D_1$, and $D_{1,4}$ comprosed 77.39% of the whole disseminule form. Growth form was surveyed in the order of erected type (e), bunch type (t), temporal rosette type (pr), branch type (b) and straight rosette type (ps) and these species comprised 64.97% of the whole flora. Consequently, in case of the artificial wetlands along the Boknaecheon of Juam Lake, it turned out that the vegetation type in which pioneer species of succession, or gradually stabilized perennial vegetation favoring Wetlands because the higher dormancy form has its perennial plants' composition ratio getting, the more its succession is progressing. Even though single grained plants ($R_5$) belonging to radicoid in breeding form, succession is predicted to take place considering the fact that they actually belong to ~ plants like Phragmites japonica that form a connection on the surface of the earth. In addition, it is judged that geomantic disseminule form ($D_1$) conveyed by water and gravitational disseminule form favored by the development of waterside woody plants ($D_4$) seem to be better fit to this area in desseminule form. As for growth form, bunch type (t) is judged to become prevalent on the Wetlands while a good variety of phanerophytes will coexist on the earth due to artificial as well as natural disturbances.

Breaking Physical Dormancy with Sulfuric Acid in Seeds of Lespedeza tomentosa (Thunb.) Siebold ex Maxim (황산처리를 이용한 개싸리 종자의 물리적 휴면 타파)

  • Lespedeza species are mainly used for wildlife food and cover and for erosion control. The germination of these species can be enhanced after a fire occurrence in forest, which is known as fire-activated seeds to germinate. While the heat treatment could break seed dormancy of Lespedeza, its germination rate was quite low. We investigated that chemical scarification could promote germination of L. tomentosa. Seeds were soaked in 100% sulfuric acid (H2SO4) for 0, 1, 3, 6, 12, 24, 48, 96, 192, and 384 min, and then washed in distilled water for 24 h. Very few seeds were germinated in control (H2SO4 for 0 min). More than 90% of seeds were germinated in H2SO4 for 24, 48, and 92 min. However, some damage was observed in roots and cotyledons of seedling dipped in H2SO4 for a long time. To search the optimal soaking time in H2SO4 without defects, seeds scarified in H2SO4 for 30, 60, 90, 120, 150, 180, and 300 min were sown the commercial soil medium. Seeds treated with H2SO4 for 90 min and 150 min emerged by about 92% and 84%, respectively. Therefore, H2SO4 treatment could break the seed dormancy of Lespedeza species, and especially in case of L. tomentosa the optimal treatment time in sulfuric acid was one to two hours. Germination of L. tomentosa began promptly following the scarification and was completed within about one month, indicating that seeds has no physiological dormancy, just has physical dormancy.

Effects of Temperature and Gibberellin Treatment on Embryo Development and Germination of Sambucus racemosa subsp. pendula Seeds (온도 및 지베렐린 처리가 말오줌나무 종자의 배발달 및 발아에 미치는 영향)

  • This study was conducted to determine types of seed dormancy in Sambucus racemosa subsp. pendula (Nakai) H.I. Lim & C.S. Chang, an endemic tree species of Korea, whose seeds have been considered difficult to germinate. Seeds of S. racemosa subsp. pendula were stratified at 25/15 or $5^{\circ}C$ for 0, 6, or 12 weeks (wks) and incubated at 15/6, 20/10, 25/15, or $30/20^{\circ}C$ (12/12 h) under 14 h photoperiod. To determine the effect of $GA_3$ on seed germination of S. racemosa subsp. pendula, seeds were treated with 0, 500, or 1000 ppm $GA_3$ and then germinated at 25/15 or $5^{\circ}C$. The change in embryo length was investigated at 25/15 or $5^{\circ}C$. Seeds given 12 wks of cold stratification germinated to 33.4% at $15/6^{\circ}C$ and to 25.4% for seeds given 6 wks of warm stratification + 12 wks of cold stratification at $20/10^{\circ}C$. At $25/15^{\circ}C$, seeds given 12 wks of warm stratification + 6 wks of cold stratification germinated to 26.0%, and to 28.2% for seeds given 12 wks of warm stratification + 12 wks of cold stratification at $30/20^{\circ}C$. Warm stratification alone did not germinate seeds throughout the experiment, regardless of the thermoperiod. Linear embryos began to grow after 60 days of incubation at 25/15 or $5^{\circ}C$. The embryo length at day 69 increased from 1.4 mm to 1.50 or 1.62 mm at 25/15 or $5^{\circ}C$, respectively. Embryos of S. racemosa subsp. pendula seeds grew better at $5^{\circ}C$ than at $25/15^{\circ}C$. Gibberellin was effective to break seed dormancy of S. racemosa subsp. pendula. Seeds treated with 500 ppm $GA_3$ germinated up to 40.0% at $25/15^{\circ}C$ and to 62.7% for those treated with 100 ppm $GA_3$ at $5^{\circ}C$. With these results, seeds of S. racemosa subsp. pendula have both nondeep complex and intermediate complex morphophysiological dormancy.

Studies on Germination Characteristics and Differential Responses to Herbicides in Setaria Spp. (강아지풀 종류별 종자발아특성과 제초반응 차이)

  • To supply seeds with a good quality as plant materials for herbicide screening and to know whether the germination characteristics could be associated with a differential response to herbicides, germination characteristics and differential responses to herbicides were investigated with 3 species of a genus Setaria ; Yellow foxtail(Setaria glauca P. Beauv, SETGL), Giant foxtail(Setaria faberi Hetrm, SETFA), and Green foxtail(Setaria viridis P. Beauv, SETVI). Degree of dormancy was high in the order of SETGL, SETVI and SETFA. The dormancy of SETGL seed was relatively well removed by room temperature and drying storage, but SETFA and SETVI by low temperature and wetting storage(stratification). For breaking dormancy of SETGL, SETVI and SETFA, it was necessary for being kept under the above storage conditions for at least 2, 4 and 4-5 months, respectively. When the dormancy-breaked seeds were transfered to low temperature($4^{\circ}C$) and drying condition, SETGL showed germination rate of 96% even after 2 month storage. However, SETVI and SETFA showed a decreased germination of 54% and 69%, respectively, with a decreased velocity of germination, indicating that secondary dormancy might be induced. On the other hand, a significant change in germination rate was not observed as the seeds were transfered to room temperature($25^{\circ}C$) and drying condition. The germinability of SETGL seed began to decline from 6th year after storage in room temperature and drying condition. All of 3 species showed relatively high germination rate at alternating temperature of $30^{\circ}C$/$20^{\circ}C$(14hr/10hr) and their germination were not increased by light. All of 3 species exhibited similar responses to cycloxydim, sethoxydim and primisulfuron in greenhouse experiment. In contrast, SETVI and SETFA were relatively susceptible to fenoxapropethyl, SETFA to fluazifop-butyl, SETGL and SETFA to clorimuron-ethyl, and SETGL to EK-2612. The difference in herbicidal response among 3 species was the highest in the treatment of EK-2612. These results suggest that there is no a consistent tendency in responses of 3 species to herbicides which have the same target site. And the relationship between germination characteristics and differential responses to herbicides was not found.

Effect of Chemical Treatments on Seed Germination of Saururus chinensis (Lour.) Baill., an Endangered Species in Korea (멸종위기 야생식물 삼백초의 종자발아에 미치는 화학적 처리의 영향)

  • This research was performed to develop seminal propagation method of Saururus chinensis (Lour.) Baill. by conducting a rigorous germination study. Well-selected seeds were dry-stored at 4 ± 1.0℃ during the experiment. To study dormancy type, non-stored seeds were analyzed by embryo observation, germination test and detecting for any difficulties in seed coat to absorb moisture. Then to improve germination, seeds were submerged for 24 hours in a solution of varying concentrations containing one of plant growth regulators and minerals. According to research, fleshly matured seeds had an undifferentiated embryo and had a low germination rate below 5%. In addition, water submersion led to moisture absorption, embryo in the seeds grew and germinated so it was deemed morphophysiological dormant seeds. Percent germination (PG) and germination energy (GE) was greatly improved by soaking in plant growth regulators and minerals for 24 hours. Especially, 500 ㎎/L GA3 treatment resulted in the highest GE as 46.1%. KNO3 meaningfully improved PG (54.3∼57.7%) at 10∼20 mM but effect of minerals on germination acceleration as GE were negatively impacted in all concentrations.

Effects of Gibberellic Acid and Alternating Temperature on Breaking Seed Dormancy of Panax ginseng C. A. Meyer (개갑된 인삼종자 휴면 조기타파에 미치는 GA3 및 변온처리 효과)

  • Background: Developing new ginseng cultivars is a significant time-consuming process owing to the three years of growth required for ginseng to flower. To shorten the ginseng breeding process, it is necessary to establish rapid progression through each generation. In this study, we examined it was possible to rapidly break ginseng seed dormancy using gibberellic acid ($GA_3$) treatment and alternating temperature. Methods and Results: Seeds were obtained from local variety. Seeds were treated with either $GA_3$ at a concentration of $100mg/{\ell} $, constant temperature ($-2^{\circ}C$ and $2^{\circ}C$), alternating temperature ($2^{\circ}C$ followed by $-2^{\circ}C$, followed by $2^{\circ}C$) or a combination $GA_3$ and temperature treatment. Following experimental treatment, seeds were sown into trays and placed in a greenhouse. Low germination rates were observed in seeds that did not receive $GA_3$ treatment, which were similar following $2^{\circ}C$ and $-2^{\circ}C$ constant temperature treatment. Germination rates increased in proportion to $GA_3$ and more so when combined with alternating temperature treatment. In additon, stem and leaf lengths of the resulting ginseng plants were increased following $GA_3$ treatment, although no synergistic effect was observed with alternating temperature treatment. Conclusions: These results suggest that a combination $GA_3$ and alternating temperature treatment enhances ginseng seed germination, which can contribute to shortening the time required to progress through a single ginseng generation for breeding.
