To investigate the effects of carboxyethylgermanium sesquioxide (Ge-132) on the cyclophosphamide (CY)-induced immunotoxicity, hemagglutinin titer (HA-titer), splenic IgM plaque forming cells (PFC) and contact-delayed type hypersensitivity (CDTH) were assessed in mice. Ge-132 was orally administered alone (single dose of 300, 600, 900 mg/kg b.w.) or with CY (10 or 50 mg/kg, i.p.) to mice on the 2nd day before, simultaneously with, the 2nd day after immunization. Within Ge-132 alone-treated groups, HA-titer and PFC to SRBC were significantly and dose-dependently enhanced when compared with control group. HA-titer and PFC numbers suppressed by the treatment of CY alone were significantly restored by the concomitant treatment of CY and Ge-132. Also, Ge-132 significantly decreased DNFB-induced CDTH and inhibited the CY-enhanced CDTH. These results indicate that Ge-132 may be able to increase humoral immunity and inhibit the immunotoxicity by CY.
The purpose of this research was to investigate the effect of Kamidangkwieumja(KDEJ) water extract on the allergy reaction in mice. The results were obtained as follows: 1. The passive cutaneous anaphylaxis induced by egg albumin in fat was not affected. 2. The lethal anaphylaxis induced by platelet activating factor in mice. was inhibited. 3. The degranulation of peritoneal mast cells induced by compound 48/80 was not affected. 4. The acute hind paw edema was inhibited after 2hours later when it was induced by histamine. 5. The permeability of evans blue into peritoneal cavity induced by acetic acid was not affected. 6. Arthus reaction in mice was not affected. 7. The delayed type hypersensitivity induced by SRBC was inhibited. 8. The contact dermatitis induced by DNFB was not affected. 9. The hemagglutination titer induced by SRBC was inhibited. 10. The writhing syndrome induced by acetic acid was inhibited. 11. The population of heper T cells in mice thymus was enhanced. 12. The phagocytic activity of peritoneal macrophages was enhanced. 13. The production of nitric oxide from peritoneal macrophages was not affected. These results suggest that the anti-allergy effect of KDEJ is caused by steroidlike and enhanced immune action. The steroidlike action of KDEJ correspond with steroid-applying-method that frequently used in clinic, so it is used io treatment of psoriasis. The research on anti-allergy of KDEJ might has to be continued.
Atopic dermatitis (AD) is an allergic and inflammatory skin. Recently, the limitations and side effects of drug therapy, and possibility of alternative therapies, such as music therapy are emerging in the treatment of AD. Thus, the present study determined whether traditional Korean music, Daegeum Sanjo, regulates AD symptoms by comparing the rhythm, Jinyangjo-jangdan and Jungmori-jangdan in an AD-like murine model. Jinyangjo-jangdan and Jungmori-jangdan of Daegeum Sanjo reduced the duration of scratching behavior increased by DNFB challenge. Jinyangjo-jangdan and Jungmori-jangdan of Daegeum Sanjo attenuated clinical symptoms. However, Jinyangjo-jangdan and Jungmori-jangdan of Daegeum Sanjo did not inhibit IgE, histamine, interleukin (IL)-4, IL-6, or thymic stromal lymphopoietin levels in serum or AD-like skin lesions. In conclusion, the present study suggests that it is possible for Jinyangjo-jangdan and Jungmori-jangdan of Daegeum Sanjo to ameliorate AD symptoms. However, further study is needed to clarify significant mechanisms of Jinyangjo-jangdan and Jungmori-jangdan of Daegeum Sanjo therapy for AD symptoms.
The purpose of this research was to investigate the effects of Kwakhyangjeonggisan (KJS) on the anti-allergic action. In the present study, we examined the effect of KJS on type I and type IV allergic reaction. KJS inhibited the systemic anaphylaxis induced by compound 48/80 and platelet activating factor (PAF), and inhibited the passive cutaneous anaphylaxis (PCA) induced by anti-dinitrophenyl (DNP)-IgE and DNP-human serum albumin (HSA) in vivo. In addition, KJS dose-dependently inhibited the release of histamine from peritoneal mast cells in rat. Also, KJS inhibited the delayed type hypersensitivity (DTH) induced by SRBC and the contact dermatitis induced by dinitrofluorobenzene (DNFB). KJS inhibited the proliferation of splenocytes, the subpopulation of B220+ cells and CD4+CD8-(Th) cells in splenocytes and the production of γ-interferon in serum and splenocytes. These findings suggest that KJS prevented the type I allergy by the inhibition of histamine release from mast cells and the type IV allergy by the inhibition of γ-interferon production and B lymphocytes subpopulation. These results indicate that KJS may be useful for the prevention and treatment of type I and type IV allergy related disease.
사람 장내세균은 그 개체가 100조 이상으로 알려져 있으며, 그 종류도 수 천 종에 이르러 건강에 지대한 영향을 미치고 있다. 이에 많은 연구를 통해 장내세균의 구성과 각종 질환과의 연관성이 밝혀지고 있으며, 장내세균 구성을 변화시켜 질환을 치료하려는 시도도 이어지고 있다. 본 연구에서는 천연식의약소재의 활성 검토를 위해 피부염 동물모델을 구축하고, 장내세균과의 상관관계를 정립하기 위해 실험을 수행하였다. 실험동물의 등 부위를 제모하고 DNFB로 피부염을 유발시켰으며, 유도 전에 대변을 채취하였다. 피부염의 유발을 확인하기 위해 비장의 무게를 측정하였다. Arctigenin과 dexamethasone을 실험군으로 설정하여 동물에 투여하여 그 효과를 검증함과 동시에 대변을 채취하여 장내세균의 구성을 T-RFLP법으로 분석하였다. 또한, 실험동물의 장내세균을 납품회사 별로 분석하여 향후 연구에 활용하고자 하였다. 그 결과, 장내세균의 구성이 피부염의 유발에 직접적으로 영향을 주는 것이 확인되지 않았다. 하지만, 투여 성분의 채내 동태 등에 대한 연구를 실시하여 약물대사를 포함한 장내세균과의 상호작용 연구를 통해 질환과 약물, 그리고 장내세균과의 관계를 정립할 수 있는 기반을 마련하였다.
Objective : Active treatment for a skin condition has produced a substantial change in the history of allergic contact or wound induced dermatitis. The frequency of opportunistic contacts and allergen-related dermatitis has increased, though new skin treatments have developed. Especially, there has been a interest in the korean herbal bathing. This study was performed to investigate the effects of the korean herbal bathing candidates on skin lesions in mice model. These candidates are Korea's representative herbs on skin lesions. Method : ICR mice were divided into five groups : naive, control, experimental group A and B and C. Control and experimental groups induced skin lesions by DNFB and TMA. Each experimental group was pasted with corresponding korean herbal bathing candidates on skin lesions. Result : Significant improvements in skin wound index, epidermal thickness associated with skin lesions were seen with the korean herbal bathing candidates. Conclusion : These results suggest that the korean herbal bathing candidates could be at least as effective as steroid, anti-histamine for the treatment of skin lesions.
본 연구에서는 초액 4종인 2차 정제초액 (2nd refined liquid smoke, 2 CV), 3차 정제초액 (3'rd refined chaff liquid smoke, 3 CV), 쑥 정제초액(Artemisia plus refined chaff liquid smoke, A CV), 자소엽 정제초액 (Perillae folium plus refined chaff liquid smoke, Pf CV)의 항산화 능력과 항아토피 효과를 연구하였다. 항산화능 측정결과로 DPPH 활성은 3차 정제초액을 제외한 3종에서 높은 항산화 활성을 확인할 수 있었고, FRAP 실험 역시 DPPH와 마찬가지로 3종의 초액에서 높은 활성을 보였다. DNFB를 이용한 C57BL/6 마우스의 귀에 염증 유발을 통한 실험에서는 4가지 초액 모두 높은 활성을 보였다. 특히 육안, 귀의 두께 및 귀의 상피 두께를 확인한 결과 자소엽 초액이 염증을 유도한 군에 비해 두께가 현저히 감소하여 가장 높은 활성을 보였다.
This study investigated the effects of mycelium and culture supernatant of Cordyceps ochraceostromat(Co) on air way hyper-responsiveness, pulmonary immune cell infiltration, and Th2 cytokine expression in animal models of atopy and asthma. After ConA(+/-) activation of mouse primary spleen cells, decreased IL-4 and IL-13 cytokine production were seen in the presence of Co mycelium extracts and culture supernatant. The asthma model involved mice sensitized to ovalbumin by i.p. injection treatment; Co mycelium extract was also injected. The atopy model was the dinitrofenylbenzene-treated mouse ear. Ear thicken ing induced by DNFB was decreased by Co mycelium extract, and the extract also inhibited lung cell infiltration in ovalbumin-induced asthmatic mice. The results thus indicated that the Co mycelial extract reduced the undesirable immune responses seen in asthma and atopy.
Objectives Hataedock is a Korean herbal medical oral treatment that removes fetal toxic heat and meconium from new born babies. The purpose of this study is to evaluate whether Hataedock treatment of Duchi extracts has anti-inflammation effects on AD (Atopic Dermatitis)-induced NC/Nga mice. Methods After Hataedock treatment of Duchi extracts on days 0, 3-week-old NC/Nga mice were sensitized on days 28, 35, 42 by exposure of DNFB (dinitrochlorobenzene) and were induced to have AD. Immunohistochemistry of NF-${\kappa}B$ p65, iNOS, COX-2 and TUNEL assay of apoptotic body was used to identify changes of skin damages and anti-inflammation effects. Results The alleviate effect of the skin damage and angiogenesis was observed in DT group. The damage of stratum corneum, hyperplasia, edema, infiltration of lymphocytes and distribution of capillary were decreased in DT group. Also, the study results suggested that Hataedock treatment of Duchi extracts in DT group remarkably downregulated levels of NF-${\kappa}B$ p65 by 70% (p < 0.001), as well as COX-2 by 51%, iNOS by 62% (p < 0.001). Additionally, Hataedock treatment of Duchi extracts in DT group up-regulated apoptosis of inflammatory cells by 68% in atopic dermatitis-like skin lesion. Conclusions From the study results, we observed that Hataedock treatment of Duchi extracts alleviates AD through diminishing various inflammatory cytokines in the skin lesions, which are involved in the initial steps of AD development. It is anticipated to have potential applications for prevention and treatment of atopic dermatitis.
Allergic contact or wound induced dermatitis are common environmental health issue and seriously affect the patient's quality of life. In now, people are interested in the korean herbal bathing that get better the skin conditions. This study was performed to investigate the effect of korean herbal bathing candidates extract on allergic contact or wound induced dermatitis. ICR mice were divided into four groups: naive, control, experimental group A and B. Control and experimental groups induced allergic contact or wound induced dermatitis by DNFB and TMA. Since then, each experimental group was pasted with corresponding korean herbal bathing extract into the skin. All groups were observed after 5 days. When the korean herbal bathing extract was applied, arbitrary skin wound index and epidermal thickness were significantly decreased compared to that of control. These data suggest that these korean herbal bathing candidates may be an effective therapeutic supplements for the treatment of skin.
