Galectins are a group of animal lectins consisting of galectin-type carbohydrate recognition domains (CRD) with relatively minor domains. The biological properties of galectins include the regulation of inflammation, intercellular adhesion, cell differentiation and cell death. The diverse kinds of galectin suggest variety in their biological roles. Galectin-1 is released during adipocyte differentiation and is associated with fat which is one of the important factors for meat quality. To verify expression level, a 0.5 kb clone of galectin-1 was obtained from cDNA prepared from back fat tissue of a Sancheong Berkshire pig with good quality meat, and the galectin-1 gene identified. The deduced amino acid sequence of the galectin-1 gene was compared with those obtained from other species. By using RT-PCR and Real time-PCR, an attempt was made to determine the expression level of galectin-1 and to compare with various tissues (tenderloin and back fat) taken from pigs in different groups. Grouping of pigs was based on growth-stage (weighing 60, 80, and 110 kg) and the sub-speciation (Yorkshire and Sancheong Berkshire pigs). We attempted to determine influences of pig species, growth stages and tissue variations on the expression level of the galectin-l gene and it was revealed that the expression pattern of the galectin-1 gene was significantly different (p<0.01 or p<0.05). Galectin-1 genes were expressed more highly in the back fat tissues of pigs weighing 110 kg than in those weighing 60 kg or 80 kg. However, the lowest expression was seen in the tenderloin tissues of pigs weighing 110 kg. Sancheong Berkshire pigs showed higher expression of the galectin-1 gene compared to Yorkshire pigs. Accordingly, it is considered that the expression pattern of the galectin-1 gene influences the growth of back fat tissues and the pig speciation relationship. Previous studies suggested that different expression of galectin-1 genes represents variety among the breeds and is closely related to fat tissue growth, conjugation and catabolism. Further, this study suggests that the expression of galectin-1 at a specific growth stage and tissue contributes significantly to the overall meat quality of Sancheong Berkshire pigs.
Toll-like receptors (TLRs) are expressed in immune and tumor cells and recognize pathogen-associated molecular patterns. Cervical cancer (CC) is directly linked to a persistent infection with high risk human papillomaviruses (HR-HPVs) and could be associated with alteration of TLRs expression. TLR9 plays a key role in the recognition of DNA viruses and better understanding of this signaling pathway in CC could lead to the development of novel immunotherapeutic approaches. The present study was undertaken to determine the level of TLR9 expression in cervical neoplasias from Tunisian women with 53 formalin-fixed and paraffin-embedded specimens, including 22 samples of invasive cervical carcinoma (ICC), 18 of cervical intraepithelial neoplasia (CIN), 7 of condyloma and 6 normal cervical tissues as control cases. Quantification of TLR9 expression was based on scoring four degrees of extent and intensity of immunostaining in squamous epithelial cells. TLR9 expression gradually increased from CIN1 (80% weak intensity) to CIN2 (83.3% moderate), CIN3 (57.1% strong) and ICC (100% very strong). It was absent in normal cervical tissue and weak in 71.4% of condyloma. The mean scores of TLR9 expression were compared using the Kruskall-Wallis test and there was a statistical significance between normal tissue and condyloma as well as between condyloma, CINs and ICC. These results suggest that TLR9 may play a role in progression of cervical neoplasia in Tunisian patients and could represent a useful biomarker for malignant transformation of cervical squamous cells.
오늘상 국민소득이 증가하고 웰빙시대를 맞아 국민 주식(主食)인 쌀 브랜드에 관한 관심이 고조되고 있는 배경하에 쌀 공동브랜드의 브랜드 자산, 신뢰, 애착이 충성도에 미치는 영향의 관계를 분석한다. 연구방법은 선행연구의 검토하에 설문을 개발하고 연구가설을 설정하였다. 연구대상은 충남 ○○의 쌀 전업농가 163명에게 설문을 배포 및 회수하여 통계·분석하였다. 실증분석 결과는 첫째, 가설 1의 '소비자의 쌀 공동브랜드의 자산은 브랜드 신뢰에 통계적으로 유의미한(+) 영향을 미칠 것이다'는 쌀 브랜드의 지각된 품질과 원산지효과를 제외하고 인지도 및 이미지는 지지하였다. 특히 통계값의 크기로 볼 때, 인지도> 이미지보다 소비자의 신뢰에 대한 영향의 차이를 보였다. 둘째, 가설 2의 '소비자의 쌀 공동브랜드의 신뢰는 애착 및 충성도에 유의미한(+) 영향을 미칠 것이다'는 통계적으로 채택되었다. 여기서 브랜드 신뢰는 애착보다 충성도의 값이 높았다. 셋째, 가설 3의 '쌀 공동브랜드의 애착은 충성도에 유의미한(+) 영향을 미칠것이다'도 통계적 값에 따라 지지되었다. 전체 변수의 값은 브랜드 애착→충성도 영향>신뢰→충성도>신뢰→애착>인지도→신뢰>이미지→신뢰의 순으로 나타났다. 본 연구의 전략적 함의는 첫째, 시중에 유통되는 쌀 공동브랜드를 바라볼 때, 소비자들은 유통 쌀의 지각된 품질에 불신을 갖고 있음에 따라 생산농가는 소비자들에게 신뢰의 단서(GAP 등 국가인증마크의 획득, 팸투어)를 제공해야 한다. 둘째, 쌀 생산 원산지효과도 의문을 제기하므로 생산이력제/쌀 DNA 검증의 도입으로 쌀 품종의 혼입(단일미 유지)을 예방하는 조치가 요구된다.
대장균에서 6개의 Leu 코돈중 가장 흔한 코돈은 CUG이다. 이 코돈을 인식하는 tRNA는 4개의 유전자에 의해 합성되는데, leuPQV와 leuT 2개의 locus로 나누어져 있다. 이 CUG를 인식하는 모든 tRNA가 결핍된 균주를 만들기 위해, 우선 leuPQV가 삭제된 균주(${\Delta}leuPQV$)와, leuT의 anticodon CAG를 GAG로 돌연변이시킨 균주[$Km^R$, $leuT^*$(GAG)]를 각각 만들었다. 이 두 돌연변이 유전자를 모으기 위해 ${\Delta}leuPQV$ 균주를 recipient로, $leuT^*$(GAG) 균주를 donor로하는 transduction을 수행한 결과, 콜로니 크기가 큰 것과 작은 것 두 종류의 transductant를 얻었다. PCR 후 염기서열 분석 결과 큰 콜로니는 예측한 recombinant로 판명됐으나, 작은 콜로니는 donor와 recipient 염색체 간의 상호교환재조합(reciprocal recombination)으로는 설명이 되지 않는, 돌연변이 유전자[$leuT^*$(GAG)]와 야생형 유전자(leuT(CAG)]를 모두 가진 균주로 밝혀졌다. 이 heterozygous diploid는 광학현미경으로 관찰시 세포의 형태와 크기에서 특이점이 발견되지 않았으나, 영양배지에서 야생형에 비해 생장이 한참 느리면서, 선형생장곡선(linear growth curve)이라는 예측하지 못한 생장특성을 보였다. 이 2배체 균주는 선택배지에서는 항상 작은 균일한 콜로니를 형성하였으나, 배지에 선택항생제 없을 경우, $leuT^*$(GAG) 유전자형 세포와 leuT(CAG) 유전자형 세포로 분리가 일어났다. 우리의 결과를 종합해볼 때, 이 2배체 균주는, $leuT^*$(GAG)와 leuT(CAG) 부분만 2배체로 갖는 부분이배체(merodiploid)라기 보다는, $leuT^*$(GAG)와 leuT(CAG)가 서로 다른 염색체에 있는 완전이배체라는 모델을 지지했다. 우리는 이러한 2배체가 어떻게 생성되었으며, 어떻게 분리되는지, 또 이 균주는 왜 선형생장곡선을 보이는지 등에 대한 모델을 토론하였다.
glysosyl hydrolase의 하나인 CCF 유사 유전자를 지렁이 Eisenia anderi의 중장으로부터 분리하여 클로닝하였다. 이 유전자의 전체 염기서열 크기는 1,152bp로 나타났으며 개시코돈을 포함하여 384개의 아미노산을 인코딩한다. N-말단 지역의 17개 잔기들은 signal peptide이다. CCF 관련 유전자의 아미노산 서열을 분석한 결과 본 연구에서 분리한 CCF 유사 유전자는 glycosyl hydrolase family 16 (GHF16)에 속하며, 다른 종의 지렁이에서 밝혀진 CCF 및 CCF-유사 단백질과 79~99%의 높은 상동성을 보였다. 여러 지렁이 종에서 분리된 CCF 및 CCF-유사 단백질들은 가수분해활성에 중요한 polysacchride-binding motif와 glucanase motif가 100% 상동성을 나타냈다. 본 연구의 대상종과 유사종인 E. fetida에서 분리된 CCF는 이 종의 서식지가 미생물 활성이 높은 부패 유기질층이기 때문에 다른 종의 CCF에 비해 높은 기질인식 특이성을 갖고 있는 것으로 알려져 있다. 이러한 사실은 본 연구에서 분리한 CCF도 넓은 기질 특이성을 갖고 있을 가능성을 제시해 주고 있으며 이는 산업적 응용 측면에서 유용한 특징 중의 하나라고 생각된다. BLASTX를 이용한 계통수 분석 결과 GHF16 효소는 크게 후생동물군, 녹색식물군, 진정세균군 등의 세 그룹으로 나눌 수 있었으며, 후생동물군의 GHF16은 다시 촉수담륜동물형와 탈피동물형(후생동물형 포함)으로 나뉘어졌다. 본 연구에 의해 E. andrei로부터 분리된 CCF-유사 단백질의 더 많은 생물학적 특성 연구를 통해 ${\beta}$-D-글루칸의 분해 및 생산을 조절하는 산업적 활용이 가능할 것으로 사료된다.
