본 연구는 태령 19일된 백서 태자 두 개관에서 분리한 골모세포유사세포에 화학발암물질인 7,12-Dimethylbenz(a)anthracene (DMBA: 0.5 ㎍/ml) 및 tumor promotor인 12-O-tetradecanoyl-phorbol-13-acetate (TPA; 1.0 ㎍/ml)를 단독 혹은 복합 처리하여 PTRCC-DMBA, RCC-DMBA 및 RCC-DMBA-TPA 세포주를 확립시키고, 각 세포의 세포형태, 세포성장곡선, alkaline phosphatase와 acid phosphatase 활성 및 in vitro tumorigenicity를 연구하였다. 또한 c-myc, c-랜, c-jun, p53 및 Rb 유전자의 발현변화와 항암단백질인 p53 및 pRb 단백질의 발현변화를 관찰하여 골모세포유사세포가 악성형질전환되는 분자기전의 일단을 연구하고자 시행하였다. 본 실험에 사용한 모든 세포군에서 높은 aikaline phosphatase 활성과 낮은 acid phosphatase/alkaline phosphatase ratio를 보여 골모세포의 특성을 나타내었다. RCC-DMBA와 RCC-DMBA-TPA 세포는 정상세포나 PTRCC-DMBA에 비해 빠른 성장속도를 보였으며, 또한 SOFT AGAR상에서 colony를 형성하여 anchorage-independent growth를 나타내었다. 화학발암 물질로 악성변형된 세포들은 정상세포나 PTRCC-DMBA 세포에 비해 c-myc 유전자의 과발현이 관찰되었다. 정상세포에서 p53 유전자의 발현은 1.9 kb의 message만이 발현되었다. 그러나 화학발암물질로 형질전환된 세포에서는 1.9 kb message외에도 1.6 kb의 message가 더 발현되었으며, message의 양도 현저히 증가되었다. p53 단백질의 발현은 RCC-DMBA-TPA 세포에서 정상세포에 비해 현저히 감소하였으나, RCC-DMBA 세포에서는 유사한 경향을 보였다. Rb 유전자의 발현은 RCC-DMBA-TPA 세포에서만 현저히 감소하였으나, Rb 단백질의 발현은 정상세포에 비해 형질전환된 세포들에서 모두 현저히 감소되었고, 특히 RCC-DMBA-TPA 세포에서는 거의 발현되지 않았다. 이상의 결과에서 백서 태자 두 개관에서 분리한 골모세포유사세포는 화학발암물질인 DMBA에 의해 악성형질전환이 유도되었으며, c-myc의 과발현 및 p53과 Rb 단백질의 발현감소가 정상 골모세포유사세포의 악성변형과정에 밀접히 연관되어 있음을 시사한다.
This study was carried out to investigate the immunopathological effects of 7, 12-Dimethylbenz[a]anthracene(DMBA) on mouse thymus. DMBA was administered subcutaneously to BALB/C mice by interscapular single injection of 50 or 100 ng/g of body weight. Each DMBA treatment group and additional corn oil control group of mice were studied on day 1, 3, 7, 14 and 21 following the injection of DMBA. DMBA treatment resulted in marked decreases in weights and cellularity of thymus. Thymus weights were decreased by 50.6 and 66.0% at 21 days, respectively, after treatment of 50ng/g and 100 ng/g DMBA.
Our previous finding that pre-initiation treatment of indole-3-carbinol (I3C) represents a chemopreventive effect in dimethylbenz[a]anthracene (DMBA)-induced mammary carcinogenesis has prompted us to test the global expression of genes at an early stage. Rats were continuously fed 300 ppm I3C in their diet at 6 weeks of age and were injected with DMBA at 7 weeks of age, and were sacrificed at 8 weeks of age. Global gene expression analysis using oligonucleotide microarrays was conducted to detect altered genes in DMBA- or DMBA plus I3C-treated mammary glands. Altered genes were identified by fold changes of 1.2 and by t-test (P<0.05) from the log ratios of the hybridization intensity of samples between control (Group 1) and DMBA (Group 2), and from those of samples between DMBA (Group 2) and DMBA plus I3C (Group 3). From these genes, we chose altered genes that were up- or down-regulated by DMBA treatment and recovered to the control level by I3C treatment. For early stage of carcinogenesis, I3C treatment induced the recovery to normal levels of several genes including cell cycle pathway (cyclin B2, cell division cycle 2 homolog A), MAP signaling pathway (fibroblast growth factor receptor 1, platelet derived growth factor receptor, beta polypeptide), and insulin signaling (protein phosphatase 1, regulatory (inhibitor) subunit 3B and flotillin 2), which were up-regulated by DMBA treatment. In addition, I3C treatment induced the recovery to normal levels of several genes including those of MAPK signaling (transforming growth factor, beta receptor 1 and protein phosphatase 3, catalytic subunit, beta isoform), which were down-regulated by DMBA treatment. These results suggest that the targeting of these genes presents a possible approach for chemoprevention in DMBA-induced mammary carcinogenesis.

Carcinogenesis is a multi-stage process that generally consists of at least three steps; initiation, promotion, and progression. If one of these carcinogenic steps were suppressed or delayed, the cancer could be prevented. Cancer chemoprevention is defined to be inhibition or reversal of the carcinogenic process by the specific chemical agents and is a novel approach to cancer management alternative to conventional chemotherapy. Chlorophylln(CHL), a water-soluble derivative of chlorophyll, containing sodium and copper, has been known to be strong antimutagen in several test systems, but its mechanism of antimutagenic action is unknown. In the present experiment, the possibility of CHL as chemopreventive drugs on 7,12-dimethylbenz[a]anthracene(DMBA)-induced hamster buccal pouch carcinogenesis was investigated by mutagenicity test, carcinogenicity test, and frequency or spectrum of H-ras mutations in the both of DMBA-induced and chlorophylln-pretreated-DMBA induced tumor by polymerase chain reaction and non-isotopic restriction fragment length polymorphism. The treatment of CHL reduced the yields and multiplicity of the 0.5% DMBA-induced tumor, 86% to 62.5% and $3.7{\pm}0.6$ to $1.4{\pm}0.3$, respectively. The occurrence of histidine revertant by $20{\mu}mole$ DMBA was inhibited 25.6 to 81.7% by 1 to $5{\mu}M$ CHL in a dose-dependent manner. The mutation rates of H-ras gene in DMBA-induced and CHL-pretreated-DMBA induced tumor were 96%, 94% of which the most mutations were in codon 12/13. These results suggest that CHL inhibits the carcinogenic action of DMBA by the formation of complex between CHL and DMBA or the inhibition of the activation of DMBA in vivo. But CHL did not affect the mutation rates or its spectrum in already formed tumor.

DMBA treatment resulted in marked decrease in weights and cellularity of spleen. Spleen weights showed the greatest decrease at 14days after 50${\mu}g/g$ DMBA treatment, and at 21days after 100${\mu}g/g$ DMBA treatment. Spleen cellularity was similarly deceased in comparison with spleen weights. Spleen showed morphologically no typical changes throughout the experiment after 50${\mu}g/g$ DMBA treatment. Following the treatment of 100${\mu}g/g$ DMBA the spleen showed severe fibrosis, hemosiderin precipitation, and megakaryocytes decrease in red pulp at 14 days, while hemopoietic function was partly restored in addition to the appearance of a few megakaryocytes at 21 days. In spleen sections treated with antibodies to IgM or Thy1.2, lymphocytes strongly stained with IgM antibody were infiltrated around the central artery within the white pulp, and T-lymphocytes of periarterial lymphatic sheath (PALS) were diminished and destructed in sections treated with Thy1.2 antibody, at 14 days after the treatment of 100${\mu}g/g$ DMBA. By the electron microscopy phagocytic epithelial cells or macrophages were remarkably increased in spleen at 14and 21days following the treatment of 100${\mu}g/g$ DMBA.
Background: The search for naturally occurring agents in routinely consumed foods that may inhibit cancer development is of high priority. [6]-Paradol is a pungent phenolic bioactive component from ginger with welldocumented health promoting antioxidant, antimutagenic, antigenotoxic and anti-inflammatory properties. However, anticarcinogenic effects have yet to be fully explored. The objectives of the present study were therefore to assess protective effects against 7,12-dimethylbenz(a)anthracene (DMBA) induced buccal pouch carcinogenesis in male golden Syrian hamsters. Methods: Oral squamous cell carcinomas developed in the left buccal pouch of hamsters on painting with 0.5% of DMBA, three times in a week. To assess the apoptotic associated gene expressing potential of [6]-paradol, it was orally administered to DMBA treated hamsters on alternate days from DMBA painting for 14 weeks. Results: We observed 100% tumor formation with marked levels of neoplastic changes and altered the expression of apoptotic associated gene (p53, bcl-2, caspase-3 and TNF-${\alpha}$) was observed in the DMBA alone painted hamsters as compared to control hamsters. Oral administration of [6]-paradol at a dose of 30 mg/kg b.wt to DMBA treated animals on alternative days for 14 weeks significantly reduced the neoplastic changes and improved the status of apoptosis associated gene expression. Conclusion: These observations confirmed that [6]-paradol acts as a tumor suppressing agent against DMBA induced oral carcinogenesis. We also conclude that [6]-paradol also effectively enhances apoptosis- associated gene expression in DMBA treated animals.
In attempts to evaluate carcinogenic chemical effect on $Na^+,K^+$-ATPase activity in the rat submandibular gland tumor induced by DMBA, pellet from DMBA powder was inserted into the right submandibular gland. Both right and left submandibular gland were excised and weighed following the period of experiment. Excised glands were observed microscopically and estimated biochemically. The results were obtained as follows. 1. Swelling and nodular mass in the right submandibular gland region ould be found at 11th week post-implantation with DMBA. 2. The weight and size of the right submandibular gland was markedly increased following the period of the experiment. 3. Epithelial dysplasia and invasive epidermoid carcinoma could be observed at 7th and 11th week after implantation of DMBA, respectively. 4. The rate of tumor induction in the right submandibular gland was about 76% at 17th week following implantation of DMBA. 5. DMBA caused markedly depressed $Na^+,K^+$-ATPase activity as well as the activity ratio in the rgiht submandibular gland following the period of experiment.
Ever since the expression of new tumor-specific antigens was reported during malignant transformation, studies on separation, purification and characterization of these proteins have been so activated recently. Following experiment was performed to observe tumor-specific antigens by implanting DMBA pellet into submaxillary gland of rat for inducing salivary gland tumor. After dividing 280 rats into 2 groups, in control group, sham operation was performed on right submaxillary gland and, in experimental group, DMBA pellet (5mg) was implanted into right submaxillary gland. Then proteins from excised submaxillary gland by killing 10 rats every two weeks for 28 weeks were extracted with 3M KCl, and SDS-PAGE and PAS-staining were carried out for biochemical examination. The obtained results were summarized as follows; 1) At 12th week since implantation of DMBA pellet, tumor mass formation was inspected. And dysplasia at 6th week and invasive epidermoid carcinoma at 10th week were observed by microscope. 2) In control group, the weight ratio of both submaxillary glands had no any change, however, in experimental group, the ratio was increased remarkably. And at 28th week after DMBA implantation, there was more than 15 times of differences in weight between control and experimental group. 3) There was no DMBA remnant after 22nd experimental week. 4) In the SDS-PAGE, high molecular protein bands (more than 100 kd) were appeared much, and new prominent protein bands (66, 48, 41.5, 39, 37, 37.5 kd) were appeared after 4th week since DMBA implantation. However, 38, 27, 22kd protein bands were disappeared. 5) In PAS-staining, high molecular proteins were proteins were all glycoproteins and 37.5kd protein was proved as to be glycoprotein. And 38kd glycoprotein was disappeared after 4th week since DMBA implantation.
This study was examined on the effect of ex-vivo immunotherapy in 7, 12-dimethylbenz[a]anthracene (DMBA)-induced rat mammary carcinogenesis. Sprague-Dawley female 40 rats were divided into Jour groups. As a positive control, Group I was intubated with DMBA, 5 mg /100 g body weight and single dose, at experimental onset. Group II was treated ex-vivo immunotherapy with polyinosinic-polycytidylic acid (Poly I : C) and Group III was treated with Interleukin-2 (IL-2). Group IV was negative control. All rats were sacrificed at 16 weeks after DMBA intubation. Mammary gland wet weight, dry fat free tissue weight, incidence of tumor, and the number of lobules, alveolar buds, terminal end buds, and terminal ducts were examined. Morphological changes of the mammary gland after treated with DMBA were analyzed by whole mount and histopathological method. As results, the induced mammary tumors of Group I, II and III were 60%, 33% and 0%, respectively. Histopathological types of induced-mammary tumors were adenoma, adenocarcinoma and carcinosarcoma. In analysis of the whole mount method, the number of the terminal end buds, terminal ducts and lobules were significantly lower in Group II (p<0.01) and III (p<0.01) than DMBA alone treated Group I. In microscopic observation, hyperplastic alveolar nodules were significantly lower in Group III than Group I (p<0.01). In conclusion, IL-2 had strong inhibitory effect on the mammary gland tumorigenesis induced by DMBA in rats. Whole mount method may be a useful technique to assess the mammary carcinogenesis. Moreover, hyperplastic alveolar nodules were very sensitive parameter to assess the mammary carcinogenesis.
This study was undertaken to evaluate the immune responses to sheep red blood cell (SRBC) and potential anti-tumor effect of Bacillus Calmette-Guerin (BCG) in the mice bearing rumor induced by DMBA. The frequencies of tumor appearances were 62% in DMBA-treated mice and 14% in DMBA and BCG-treated group, respectively. Cellular immune response such as delayed-type hypersensitivity (DTH) to SRBCs, natural killer (NK) cell activity and antigen-binding cell (ABC) assay were decreased apparently in the tumor bearing mice compared to the normal controls. Humoral immune responses such as hemagglutinin (HA) and hemolysin (HE) were noted to be reduced in the tumor bearing mice, but the spleen index increased in tumor bearing mice. All the immunological parameters in the DMBA and BCG-group appeared to be higher than those of only DMBA-treated group. These results indicated that DMBA-induced tumor suppressed host immune responses. Also, they imply the idea that BCG enhanced the immune responses of tumor-bearing host and antitumor effects.
