IN SITU RUMINAL DEGRADATION KINETICS OF FORAGES AND FEED BYPRODUCTS IN MALE NILI-RAVI BUFFALO CALVES

  • The rate and extent of digestion of dietary carbohydrates has a tremendous impact on ruminal fermentation and the productivity of the animals. The objective of the study was to determine the dry matter (DM) and neutral detergent fiber (NDF) degradabilities and rate and extent of feed byproducts (cotton seed cake, wheat bran), legumes [berseem (Egyptian clover), lucern (Medicago sativa), cowpeas (Vigna sinensis)], grasses [maize (Zea mays), millet (Panicum miliaceum), sorghum (Sorghum vulgare)] and wheat straw in ruminally fistulated male buffalo calves. By using nylon bags, 10 grams sample was exposed to the ruminal fermentation for 0, 1, 2, 4, 6, 10, 16, 24, 36, 48 and 96 hours. Dry matter and NDF degradability was measured at 48 hours. Extent of DM and NDF disappearance was determined at each time point. Rates of disappearance of DM and NDF were determined by regressing the natural logarithm of the percentage of original DM and NDF remaining in the bags between 1 and 96 hours. The dry matter digestibility (DMD) of the feed byproducts (FBP) and legume forages when incubated in the rumen of male buffalo calves were greater (p < 0.05) than grasses. Extent of digestion followed similar pattern as DMD. Rate of DMD was higher in FBP than in legumes and was the lowest in the wheat straw. The NDF degradability (NDFD) of FBP, legumes and grasses did not differ, however, wheat straw had the lowest NDFD from all the feeds tested. The lowest NDFD of wheat straw may have been due to the depressing effect of lignin on fiber digestion. The FBP and legumes had higher (p < 0.05) rates and lower extents of NDF digestion than grasses.

In Situ Ruminal Digestion Kinetics of Forages and Feed Byproducts in Cattle and Buffalo

  • The relative disappearance and rate of degradation of dry matter (DM) and neutral detergent fiber (NDF) of nine different feedstuffs were determined by simultaneously suspending groups of substrates, using the nylon bags, in the rumen of males of Sahiwal cattle and Nili-Ravi buffalo. The digestion kinetics of leguminous forages (Lucerne, berseem and cowpeas) and feed byproducts (cotton seed cake, wheat bran and wheat straw) did not differ between the two species. However, the DM and NDF digestibilities and rates of digestion of grasses and wheat straw were greater in buffalo than in cow bulls, indicating that buffaloes are better converters of poor quality roughages than are Sahiwal. The lag time for DM of grasses did not differ between these two species but the NDF lag time was lower in buffalo than in cows, indicating that both the rate and lag time of digestion may be reliable indicators for assessing the NDF quality.

볏짚사일리지 제조를 위한 유기감귤 첨가효과 (Effects of Supplementing Organic Citrus in Making Rice Straw Silage)

  • The effects of supplementation of organic citrus on the quality of rice straw silage was investigated in this study. The aim of the study was to improve the quality of rice straw silage using agricultural by-products and therefore to secure the good quality forage sources. Firstly, looking at chemical composition of citrus added rice straw silage, crude protein content appeared improved with the addition of citrus to rice straw silage compared to control while NDF and ADF contents decreased. pH of the silages were different between control(4.39) and treatments$(3.47\sim3.53)$. Lactic acid content was higher in citrus added silage(4.87mM) than in control (3.65mM). External quality of the silages was also better in citrus added silage when citrus added at the equal ratio with rice straw(1 : 1). This trend was the same as with pH and lactic acid content. Propionate was significantly higher in citrus addd silage but butyrate was significantly lower. DM and NDF disappearance rates in the rumen were highest when citrus added at the equal ratio with rice straw(1:1). In conclusion, supplementation of citrus to rice straw silage with equal ratio contributed the best to the improvement of the quality of silage and it may have been due mainly to abundant content of non-structural carbohydrate in citrus.

EFFECT OF ALKALINE HYDROGEN PEROXIDE TREATMENT OF RICE STRAW ON IN SACCO RUMINAL DIGESTIBILITY

  • The objective of this experiment was to compare the effect of pH-regulated alkaline hydrogen peroxide (AHP) treatment of rice straw with those of sodium hydroxide (NaOH) and anhydrous ammonia ($NH_3$) treatments on in sacco digestivility. Three non-lactating ruminally cannulated Holstein cows were fed a diet containing 90% forage and 10% concentrate on a dry matter (DM) basis. The AHP treatment significantly (p<0.05) reduced acid detergent lignin content of the straw, resulting in significant (p<0.05) increase of neutral detergent fiber (NDF), acid detergent fiber (ADF) and cellulose concentrations. Disappearance rates of DM and NDF of the straw significantly (p<0.05) increased at the incubation time of 24 h. On the other hand, those of ADF and cellulose were significantly (p<0.05) higher at the incubation time of 12 h than those of the others. The effective degradability of DM(EDDM), NDF(EDNDF), ADF(EDADF) and cellulose (EDCE) were determined using in sacco nylon bag technique on the basis of 0.05/h solid outflow rate. The greater differences (p<0.05) of EDDM, EDNDF, EDADF and EDCE were found between AHP treated straw and the others. In general, AHP treatment of the straw recorded higher digestion coefficients than untreated straw as well as NaOH and $NH_3$ treated straws. The results of this study demonstrate that AHP treatment can be used as a effective method for improving the nutritive value of rice straw for ruminants.

사과박 첨가가 볏짚 사일리지의 품질과 in situ 소실율에 미치는 영향 (The Effect of Addition of Apple Pomace on Quality and in situ Degradability of Rice Straw Silage)

  • The quality of the rice straw silage added with apple pomace was investigated in this study and the amount of apple pomace added in different treatments were 0, 20, 40 and 60%, respectively. Crude protein contents (6.4-7.5%) of rice straw silage added with apple pomace were significantly (P<0.05) higher than that of 100%. rice straw silage (5.3%), however, crude ash contents were lower (P<0.05) in supplementation of apple pomace. The trends of changing chemical composition between raw materials and end products of silages particularly in the contents of crude protein and crude ash were more apparent in the silages added with apple pomace by 40-60%. Values of pH and the contents of lactic acid and total acid in 40-60% apple pomace added silages were 3.9-4.1, 1.0- 1.5% and 2.7-4.5%, respectively which were significantly (P<0.05) higher than those of 4.6, 0.02% and 0.34% in 100% rice straw silage, respectively. In situ dry matter (DM) and neutral detergent fiber (NDF) disappearance rates in the rumen in the treatments of 40- 60% apple pomace added silages were significantly (P<0.05) higher than those of 100% rice straw silage particularly since after 3 and 24 hour incubation on DM and NDF disappearance, respectively. Although quickly degraded fraction (a) among the treatments were not significantly different, 28.4-28.5% of slowly degraded fraction (b) and 27.2-27.4% of effective degradability (ED, k=0.08) for DM were significantly (P<0.05) higher than those of 100% rice straw silage (12.5 and 24.6% respectively). NDF was in the same trend as in DM. 31.6-63.2% of NDF for b fraction and 18.7- 19.4% for ED in 40-60% apple pomace added silages were significantly (P<0.05) higher than those of 100% rice straw silage (12.4 and 17.6% respectively). Effect of Sodium Hydroxide plus Hydrogen Peroxide Treated Mustard (Brassica campestris) Straw Based Diets on Rumen Degradation Kinetics (In sacco), Fermentation Pattern and Nutrient Utilization in Sheep

  • Two experiments were conducted to determine the effect of alkaline hydrogen peroxide (AHP) treatment (1% NaOH+1.5% $H_2O_2$; 1 AHPMS, 2% NaOH+1.5% $H_2O_2$; 2AHPMS) on rate and extent of degradation of mustard straw (MS) in sacco in sheep, and its in vivo digestion and ruminal fermentation characteristics when fed to sheep with concentrate (200 g per sheep daily). The treatment of straw with 1 and 2% AHP increased its sodium content by 148 and 296% to that of untreated straw (UMS). There was significant decrease in NDF and hemicellulose contents of AHP treated straw and increase in cellulose and lignin contents. Phenolic acids like ferrulic, $\rho$-coumaric and o-coumaric significantly (p<0.001) reduced by AHP treatment of mustard straw. In first experiment the in sacco degradation of DM, OM and NDF was significantly (p<0.01) greater for 2 AHPMS than for UMS at all incubation periods. The disappearance of nutrient from 1 AHPMS and 2 AHPMS treated straws continue to increase up to 96 h whereas in UMS the peak disappearance was found at 48 h. By using the equation {(y=a+b) ($1-e^{-ct}$)} the degradation rates (c) for DM, OM, and NDF were significantly higher for UMS than AHP treated straws. Level of alkali (1 and 2%) had significant effect on degradation characteristics (a, b, c and $P_{0.05}$) of DM and NDF fraction of MS. However, the effect was not pronounced on OM fraction of MS. In feeding experiment, the intake of nutrients for DM, OM, cell wall constituents and energy was higher on 2 AHPMS, whereas no effect on the digestibility of these nutrients was observed. The apparent nitrogen retention was higher (p<0.05) both in 1 and 2 AHPMS groups. Water intake by animals was significantly increased due to AHP treated mustard straw feeding. Rumen liquor pH was higher in 2 AHPMS fed animals. The $NH_3-N$ of rumen liquor was not affected by feeding of AHP treated MS based diets. Total VFA concentration was significantly (p<0.01) higher in UMS fed group. The fractional out flow rate of DM was higher (p<0.05) in animals fed on 2 AHPMS diets compared to UMS and lAHPMS fed groups. The population of large holotrichs was higher (p<0.05) on AHP treated MS fed diets compared to UMS. The study indicated that treatment of mustard straw with AHP changed its chemical composition towards a better feed. The nutritive value of 2% AHP treated mustard straw was better in terms of dry matter intake and apparent nitrogen retention. The higher in sacco DM, OM and NDF disappearance however, was not confirmed by in vivo data in this study.

EFFECT OF ALKALINE HYDROGEN PEROXIDE AND PERACETIC ACID ON IN SACCO RUMINAL DIGESTIBILITY OF ASPEN SAWDUST

  • The influence of alkaline hydrogen peroxide (AHP) and peracetic acid treatment on in sacco digestion of aspen was evaluated in three non-lactating ruminally cannulated Holstein cows fed a diet containing 90% forage and 10% concentrate on a DM basis. AHP treatment decreased lignin concentration by 40 to 60% resulting in increased concentrations of neutral detergent fiber (NDF), acid detergent fiber (ADF) and cellulose. Lignin concentrations in peracetic acid treated samples were less than 10% of values for control samples. In sacco disappearance rates of aspen DM, NDF, ADF and cellulose increased (p<.05) with AHP and peracetic acid treatment. Effective degradability of DM, NDF, ADF and cellulose were determined at a ruminal outflow rate of $.05h^{-1}$. Effective degradabilities of AHP treated aspen were approximately three-fold greater and peracetic acid treated samples five-fold greater than untreated control samples. For all parameters measured, peracetic acid treatment resulted in higher (p<.05) digestion coefficients than AHP treated aspen. Results demonstrate that peracetic acid or AHP treatment can enhance the nutritive value of aspen sawdust for ruminants to a level comparable to that reported for many forages.

식물성유 첨가가 In vitro 발효성상, NDF 소실율 및 지방산염 형성에 미치는 영향 (Effects of Added Vegetable Oils on In vitro Formation of Fatty Acid Soaps and Fermentation Characteristics and NDF Disappearance Rate)

  • 식물성유 첨가시 배양시간에 따른 지방산염의 형성 정도와 발효성상 및 NDF 소실율에 미치는 영향을 규명하고자 in vitro 시험을 실시 하였다. 기질은 알팔파 건초로 하고 1) oil 무첨가구, 2) 대두유 10%첨가구 및 3) 옥수수유 10%첨가구로 하였으며, oil 첨가 수준은 기질중량 건물 기준으로 첨가하였다. 발효성상 및 NDF 소실율을 위한 시험에서는 120ml serum bottle을 사용하여 3반복 실시하여 pH, $NH_3-N$, VFA농도 및 건물과 NDF 소실율을 분석하엿다. 지방산 분획을 위한 시험에서는 60ml serum bottle을 사용하요 3반복 실시하여 NEFA, EFA 및 FAS의 형성 정도를 분석하였다. pH는 배양시간이 증가함에 따라 유읭적으로 감소하였고, $NH_3-N$ 농도는 유의하게 증가하였다(P<0.0001). 건물 및 NDF 소실율은 처리구, 배양시간 및 oil간 유의하게 차이를 보여주었다(P<0.05). 총 휘발성 지방산 농도는 배양시간이 증가함에 따라 유의하게 증가하였다(P<0.0001). Acetate는 시험구 모두 배양시간이 증가함에 따라 유의적으로 감소하였고(P<0.0001), 대조구에 비해 oil 첨가구가 유의적으로 낮아졌다(P<0.05). Propionate는 시험구 모두 배양시간이 증가함에 따라 유의적으로 증가하였지만(P<0.0001), 처리구 및 oil간 효과는 없었다. Oil 첨가구에서 A/P비율이 유의하게 낮아졌다(P<0.05). EFA는 배양시간이 증가함에 따라 유의하게 감소하였고(P<0.0001), NEFA는 EFA의 가수분해로 인하여 증가하였다. 배양시간 48시간에서는 NEFA가 양이온과 결합하여 FAS의 형성 비율이 유의적으로 증가하였다(P<0.0001). 특히 $C_{18:0)$의 FAS 형성은 배양시간 48시간에서 대두유 및 옥수수유 첨가구에서 각각 12.53 및 15.17mg/g DM으로 0시간에 비해 각각 27.5 및 32.5배 증가하였다(P<0.0001). 배양시간이 증가할수록 칼슘을 포함한 양이온 함량이 높은 알팔파가 분해되면서 양이온이 용해되어 유리된 지방산과 결합하여 지방산염을 형성하게 된다. 이때 불포화지방산보다는 포화지방산이 양이온과 반응하여 지방산염을 형성하기 쉽다. 또한 $C_{16:0}$ 이상의 장쇄지방산이 주로 지방산염을 형성하였다. 이러한 특성 때문에 배양액내에 유리된 지방산이 염을 형성하기 쉽고 oil 첨가시 발생하는 발효성상 및 NDF소실율에 미치는 부의 영향을 최소화 한 것으로 사료된다.

Effects of Different Levels of Concentrate in Complete Rations on Nutrient Digestibilities and Ruminal Metabolites in Sheep and Growth Performance in Korean Native Bulls

  • Objective of the present study were to investigate effects of different levels of concentrate in complete rations on nutrient digestibilities and ruminal fermentation in sheep and growth performance in Korean native bulls. Increasing levels of concentrate (35, 50, 65, and 80% of complete rations) improved digestibilities of dry matter (DM), crude protein (CP) and ether extract (EE) without affecting digestibility of neutral detergent (NDF) and acid detergent fiber (ADF). Increasing levels of concentrate decreased ruminal fluid pH but increased concentrations of $NH_3-N$, propionic acid, and total volatile fatty acids (VFA). Both the disappearance rates of DM and nitrogen (N) in an in sacco study were linearly increased as the levels of concentrate in complete rations increased. Meanwhile, increasing levels of concentrate in complete rations improved growth rate and feed conversion ratio in Korean native bulls. In conclusion, the complete rations containing 80% concentrate showed better digestibility and energy supply than those of the lower levels (35, 50 and 65%) of concentrate of the rations, resulting in improved growth performance of Korean native bulls.

혐기성 미생물로 제조한 synbiotics 혼합배양물의 첨가가 발효 TMR의 발효특성과 소실률에 미치는 영향 (Effects of Supplementation of Synbiotic Co-cultures Manufactured with Anaerobic Microbes on In Vitro Fermentation Characteristics and In Situ Degradability of Fermented TMR)

  • 본 연구는 혐기성 박테리아, 곰팡이 및 효모로 제조한 synbiotics 혼합 배양물을 TMR 제조 시에 접종하여 발효기간에 따른 반추위 in vitro 발효특성과 in situ 분해율에 미치는 영향을 조사하고자 수행되었다. 처리구는 무처리구(Control), 곰팡이와 박테리아로 제조한 synbiotic 첨가구(T1), 곰팡이와 효모로 제조한 synbiotic첨가구(T2), 그리고 박테리아와 효모로 제조한 synbiotic 첨가구(T3)로 나누어, 6회의 발효기간(1, 3, 5, 7, 14 및 21일)에 따라 처리당 3반복으로 총 72개의 F-TMR (4처리 $\times$ 6회 $\times$ 3반복)을 제조하였다. In situ시험은 반추위 누관이 장착된 Holstein (평균체중 550 kg) 젖소 2두를 사용하여 4처리의 F-TMR을 반추위내에서 현수시간별(1, 3, 6, 9, 18, 24, 48 및 72 시간)로 각각 3반복하여 두당 96개의 nylon bag (4 처리 $\times$ 8 발효시간 $\times$ 3반복)을 제조하였다. 발효기간에 따른 TMR의 온도변화는 발효기간이 진행됨에 따라 높아지는 경향 이었으며, pH는 4.39~4.98범위로서 발효기간이 진행됨에 따라 감소되는 경향이었으며, 각 발효시간대별로는 대조구에서 높았고, synbiotics 처리구에서 낮게 나타났다(p<0.05). F-TMR의 암모니아 농도는 발효 7일째까지는 처리간 차이가 없었으나 14일째 이후에는 대조구에 비해 박테리아와 효모를 첨가한 synbiotics인 T3구에서 가장 낮았다(p<0.05). Lactic acid 함량은 발효 1일째에 T3구에서 가장 낮았으나 다른 발효시간대에서는 처리간 차이가 없었다. 발효기간별 F-TMR의 미생물 성장률(OD값)은 각 발효시간대별로 처리간에 차이가 없었으며, 발효기간 7일째에 정점을 나타낸 이후로 점차 감소하는 경향이었다. In situ 건물 소실률은 발효초기인 1~3시간대에 곰팡이와 박테리아 synbiotics 첨가구인 T1구가 대조구에 비해 건물소실율이 높았으나, 발효 48시간대에는 오히려 대조구에서 건물소실율이 가장 높았다(p<0.05). 다른 발효시간대의 건물 소실율과 유효분해도는 처리간 차이가 없었으며, NDF와 ADF소실율은 건물 소실율의 결과와 비슷하였다. 결론적으로 발효의 척도가 되는 pH와 lactic acid 함량은 synbiotics 첨가구가 대조구에 비해 좋은 결과를 나타내었으며, in situ 시험에서 발효초기에 건물과 섬유소 소실율이 곰팡이와 박테리아를 조합한 synbiotics구에서 높게 나타났으나 유효 분해도에서는 차이가 거의 나타나지 않아 처리효과가 미흡한 것으로 조사되었다.
