Development of Elite Lines with Improved Eating Quality Using RIL Population Derived from the Korean Weedy Rice, Wandoaengmi6 (국내 잡초벼(완도앵미6) 유래 RILs 집단의 식미 관련 특성분석 및 우량계통 선발)

  • As the main objective of rice breeding programs, rice eating quality is one of critical factors directly determining the market price and the consumer preference. However, the genetic complexity of eating quality and the difficulty in accurate evaluation often constrain improvement of the eating quality in rice breeding programs. In addition, given that the rice eating quality of current cultivars has already reached some high-level, diversifying of genetic resources are demanded more than ever to improve the rice eating quality. In this study, we developed a recombinant inbred lines (RILs) population derive from Wandoaengmi6, a japonica-type Korean weedy rice with high eating quality and a high degree of glossiness of cooked rice. Year-to-year correlations between the traits in three years were shown normal distribution for major agronomic traits and physicochemical characteristics. After evaluating tested traits related to eating quality procedure, a total of ten lines were ultimately selected from the population. Increasement of the taste of cooked rice (TA) and the overall eating quality (OE) were confirmed in the selected lines, which are caused by alleles derived from Wandoaengmi6 without any linkage drag. These results indicate that the utility of wide genomic resources in Korean landrace could be of application in various rice breeding programs and countermeasure to contribute to properly response to climate change.

Rumen Fermentation was Changed by Feed Inoculation Method in In Vitro (사료 접종 방법에 의한 in vitro 반추위 발효 상성 변화)

  • The objective of this study was to investigate the effect of different feed inoculation method on rumen fermentation in an in vitro. Three experimental treatments were used: control (CON, direct dispersion of feed (2 g) in rumen fluid), combinations of direct dispersion (1 g) and nylon bag (DNB, pore size: 50 ㎛, 1 g), and nylon bag (NB, 2 g). An in vitro fermentation experiment was carried out using strained rumen fluid for 48 h incubation time and timothy was used as a substrate. At the end of the incubation, in vitro dry matter digestibility (IVDMD), in vitro neutral detergent fiber digestibility (IVNDFD), pH, volatile fatty acids (VFA), ammonia nitrogen (NH3-N), and microbial community were evaluated and gas production was estimated at 3, 6, 12, 24, 48 h incubation periods. Gas production was higher in CON than DNB and NB at 6 and 12 h incubation time (p<0.01). There were no differences in final gas production, pH, NH3-N concentration, total VFA production, and VFA profiles among treatments. The IVDMD was lowest in CON (p<0.01) but the IVNDFD was not differed by feed distribution methods. There were no significant differences in general bacteria and fungi. Protozoa count was highest in NB treatment among treatments (p<0.01). The abundance of cellulolytic bacteria, Ruminococcus flavefaciens and Fibrobacter succinogenes, was highest in the CON among treatments (p<0.01).

Discount Presentation Framing & Bundle Evaluation: The Effects of Consumption Benefit and Perceived Uncertainty of Quality (묶음제품 가격 할인 제시 프레이밍 효과: 지각된 소비 혜택과 품질 불확실성의 영향을 중심으로)

  • Constructing attractive bundle offers depends on more than an understanding of the distribution of consumer preferences. Consumers are also sensitive to the framing of price information in a bundle offer. In classical economic theory, consumers' utility should not change as long as the total price paid stays same. However, even when total prices are identical, consumers' preferences toward a bundle product could be different depending on the format of price presentation and the locus of price discount. A weighted additive model predicts that the impact of a price discount on the overall evaluation of the bundle will be greater when the discount is assigned to the more important product in the bundle(Yadav 1995). Meanwhile, a reference dependent model asserts that it is better to assign a price discount to a tie-in component that has a negative valuation at its current offer price than to a focal product that has a positive valuation at its current offer price(Janiszewski and Cunha 2004). This paper has expanded previous research regarding price discount presentation format, investigating the reasons for mixed results of prior research and presenting new mechanisms for price discount framing effect. Prior research has hypothesized that bundling is used to sell a tie-in component with an offer price above the consumer's reference price plus a focal product of the same offer price with reference price(e.g., Janiszewski and Cunha 2004). However, this study suggests that bundling strategy can be used for increasing product's attractiveness through the synergy between components even when offer prices of bundle components are the same with reference prices. In this context, this study employed various realistic bundle sets with same price between offer price and reference price in the experiment. Hamilton and Srivastava(2008) demonstrated that when evaluating different partitions of the same total price, consumers prefer partitions in which the price of the high-benefit component is higher. This study determined that their mechanism can be applied to price discount presentation formats. This study hypothesized that price discount framing effect depends not on the negative perception of tie-in component with offer price above reference price but rather on the consumers' perceived consumption benefit in bundle product. This research also hypothesized that preference for low-benefit discount mechanism is that perceived consumption benefit reduces price sensitivity. Furthermore, this study investigated how consumers' concern for quality in a price discount--a factor not considered in previous research--influences price discount framing. Yadav(1995)'s experiment used only one magazine bundle of relatively low quality uncertainty and could not show the influence of perceived uncertainty of quality. This study assumed that as perceived uncertainty of quality increases, the price sensitivity mechanism for assigning the discount to low-benefit will increase. Further, this research investigated the moderating effect of uncertainty of quality in price discount framing. The results of the experiment showed that when evaluating different partitions of the same total price and the same amount of discounts, the partition that discounts in the price of low benefit component is preferred to the partition that decreases the price of high benefit component. This implies that price discount framing effect depends on the perceived consumption benefit. The results also demonstrated that consumers are more price sensitive to low benefit component and less price sensitive to high benefit component. Furthermore, the results showed that the influence of price discount presentation format on the evaluation of bundle product varies with the perceived uncertainty of quality in high consumption benefit. As perceived uncertainty of quality gradually increases, the preference for discounts in the price of low consumption benefit decreases. Besides, the results demonstrate that as perceived uncertainty of quality gradually increases, the effect of price sensitivity in consumption benefit also increases. This paper integrated prior research by using a new mechanism of perceived consumption benefit and moderating effect of perceived quality uncertainty, thus providing a clearer explanation for price discount framing effect.

Text Mining of Successful Casebook of Agricultural Settlement in Graduates of Korea National College of Agriculture and Fisheries - Frequency Analysis and Word Cloud of Key Words - (한국농수산대학 졸업생 영농정착 성공 사례집의 Text Mining - 주요단어의 빈도 분석 및 word cloud -)

  • In order to extract meaningful information from the excellent farming settlement cases of young farmers published by KNCAF, we studied the key words with text mining and created a word cloud for visualization. First, in the text mining results for the entire sample, the words 'CEO', 'corporate executive', 'think', 'self', 'start', 'mind', and 'effort' are the words with high frequency among the top 50 core words. Their ability to think, judge and push ahead with themselves is a result of showing that they have ability of to be managers or managers. And it is a expression of how they manages to achieve their dream without giving up their dream. The high frequency of words such as "father" and "parent" is due to the high ratio of parents' cooperation and succession. Also 'KNCAF', 'university', 'graduation' and 'study' are the results of their high educational awareness, and 'organic farming' and 'eco-friendly' are the result of the interest in eco-friendly agriculture. In addition, words related to the 6th industry such as 'sales' and 'experience' represent their efforts to revitalize farming and fishing villages. Meanwhile, 'internet', 'blog', 'online', 'SNS', 'ICT', 'composite' and 'smart' were not included in the top 50. However, the fact that these words were extracted without omission shows that young farmers are increasingly interested in the scientificization and high-tech of agriculture and fisheries Next, as a result of grouping the top 50 key words by crop, the words 'facilities' in livestock, vegetables and aquatic crops, the words 'equipment' and 'machine' in food crops were extracted as main words. 'Eco-friendly' and 'organic' appeared in vegetable crops and food crops, and 'organic' appeared in fruit crops. The 'worm' of eco-friendly farming method appeared in the food crops, and the 'certification', which means excellent agricultural and marine products, appeared only in the fishery crops. 'Production', which is related to '6th industry', appeared in all crops, 'processing' and 'distribution' appeared in the fruit crops, and 'experience' appeared in the vegetable crops, food crops and fruit crops. To visualize the extracted words by text mining, we created a word cloud with the entire samples and each crop sample. As a result, we were able to judge the meaning of excellent practices, which are unstructured text, by character size.

Functional Components and Antioxidant Activities of Perilla Leaf Genetic Resource (잎들깨 유전자원의 기능성분 및 항산화 활성)

  • The objective of this study was to investigate the distribution of functional compounds in perilla leaves of various genetic resources and their antioxidant activities. A comprehensive analysis of functional compounds was conducted for 90 genetic resources, focusing on total polyphenol content (TPC), total flavonoid content (TFC), individual phenolic content (IPC), and lutein. Their antioxidant activities were then analyzed based on their radical scavenging capacity using ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) and DPPH (2,2-diphenyl-1-picrylhydrazyl). The TPC content exhibited a range of 13.19 to 35.85 mg gallic acid equivalent/g, whereas the TFC content varied from 11.74 to 46.51 mg catechin equivalent/g. Total IPC was detected in a range of 6,310.98 to 40,491.82 ㎍/g. Lutein was detected at levels between 70.97 and 597.97 ㎍/g. ABTS and DPPH radical scavenging activities of perilla leaves ranged from 30.39 to 58.58 mg trolox equivalent (TE)/g and from 7.74 to 46.56 mg TE/g, respectively. Furthermore, correlation analysis demonstrated that rosmarinic acid, a phenolic acid, exhibited a significantly positive correlation with antioxidant activity. These findings suggest that various genetic resource of perilla leaves could effectively mediate antioxidant capacity. Results of this study provide valuable information for use of perilla leaves in Korea as functional food materials.

A Study on Method of Citizen Science and Improvement of Performance as a Ecosystem Conservation and Management Tool of Wetland Protected Areas (Inland Wetland) - Focused on the Target of Conservation·Management·Utilization in Wetland Protected Area Conservation Plan - (내륙 습지보호지역의 생태계 보전·관리 도구로서 시민과학연구 방법론 및 성과 제고 방안 - 습지보호지역 보전계획의 보전·관리·이용 목표를 중심으로 -)

  • This study suggested methodology of Citizen Science as a tool of ecosystem conservation and management to achieve Wetland Protected Area (WPA) Conservation Plan and examined whose applicability in 3 WPAs (Jangrok of Gwangju metropolitan city, Madongho of Goseong in South Gyeongsang Province, and Incheongang estuary of Gochang in North Jeolla Province). It consists of a) figuring out main interests and stakeholder or beneficiaries of WPA and their information demand based on conservation, utilization, and management target in the WPA Conservation Plan, b) conducting research activities to gain outcome to address stakeholder's demand, and c) returning the research outcome to citizen scientists and making diffusion to the society. Based on the suggested method and process, citizen scientists conducted ecosystem monitoring (plants including Invasive Alien Plants, terrestrial insects, traces of mammals, discovering unknown wetland). As a result, citizen scientists contributed to collecting species information of 16 plans, 43 species of terrestrial insects, 5 mammals including Lutra lutra (Endangered Species I) and Prionailurus bengalensis (Endangered Species II). The authors constructed and provided distribution map of Invasive Alien Plants, which included information of location and density which citizen scientists registered, for Environment Agencies and local governments who manage 3 WPAs to aid data-based ecosystem policy, In further studies, not only accumulating research data and outcomes acquired from citizen science to suffice the policy demands but also deliberate reviewing policy applicability and social·economic ripple effect should be processed for the suggested Citizen Science in WPA to be settled down as a tool of ecosystem conservation and management.

The Heading Response and Characterization of the Adaptable Heading Ecotypes of Korean Rice Varieties in Dandong, China (중국 단동 지역에서 국내 벼 품종의 출수 반응과 적응 출수생태 특성)

  • We examined the heading responses of rice varieties over two years in Dandong, China to select the varieties that are adaptable to the bordering northwestern plains of North Korea. Dandong had slightly lower mean temperature than Sinuiju, North Korea but slightly higher than Supung, but it had almost the same day-length as the two regions in North Korea. In the two experimental years and two transplanting-date treatments, eight varieties (Jinbuol, Baegilmi, Joun, Jinok, Jopyeong, Jinbu, Sanhomi, and Odae) from South Korea and five varieties (Olbyeo2, Sonbong9, Onpo1, Gilju1, and Pyongdo5) from North Korea reached the heading stage not later than the latest heading dates of Dandong domestic varieties and the safe marginal heading dates of Dandong. We examined the basic vegetative phase (BVP), photoperiod-sensitivity (PS), and thermo-sensitivity (TS) of rice heading in a phytotron study to characterize the heading ecotypes of rice varieties adaptable to Dandong. For the rice varieties grown in Dandong experimental field, number of days to the heading stage was highly positively correlated with PS and, except middle and mid-late maturing three varieties, it was significantly positively correlated with BVP. Two-dimensional distribution plotting revealed that both 35 days or less BVP and 25 days or less PS were the characteristic heading ecotypes of the rice varieties adaptable to Dandong.

Derivation of Inherent Optical Properties Based on Deep Neural Network (심층신경망 기반의 해수 고유광특성 도출)

  • In coastal waters, phytoplankton,suspended particulate matter, and dissolved organic matter intricately and nonlinearly alter the reflectivity of seawater. Neural network technology, which has been rapidly advancing recently, offers the advantage of effectively representing complex nonlinear relationships. In previous studies, a three-stage neural network was constructed to extract the inherent optical properties of each component. However, this study proposes an algorithm that directly employs a deep neural network. The dataset used in this study consists of synthetic data provided by the International Ocean Color Coordination Group, with the input data comprising above-surface remote-sensing reflectance at nine different wavelengths. We derived inherent optical properties using this dataset based on a deep neural network. To evaluate performance, we compared it with a quasi-analytical algorithm and analyzed the impact of log transformation on the performance of the deep neural network algorithm in relation to data distribution. As a result, we found that the deep neural network algorithm accurately estimated the inherent optical properties except for the absorption coefficient of suspended particulate matter (R2 greater than or equal to 0.9) and successfully separated the sum of the absorption coefficient of suspended particulate matter and dissolved organic matter into the absorption coefficient of suspended particulate matter and dissolved organic matter, respectively. We also observed that the algorithm, when directly applied without log transformation of the data, showed little difference in performance. To effectively apply the findings of this study to ocean color data processing, further research is needed to perform learning using field data and additional datasets from various marine regions, compare and analyze empirical and semi-analytical methods, and appropriately assess the strengths and weaknesses of each algorithm.

Anti-tumor and Anti-inflammatory Effects of Ecklonia cava in CT26 Tumor-bearing BALB/cKorl Syngeneic Mice (CT26 고형암을 내포하는 BALB/cKorl Syngeneic 마우스에서 Ecklonia cava의 항암효과 및 항염증효과)

  • The inflammatory response have been considered as one of important targets for cancer treatment because they play a key role during all steps of tumor development including initiation, promotion, malignant conversion and progression. To investigate the anti-inflammatory response during anti-tumor activity of an aqueous extracts of Ecklonia cava (AEC), alterations on the distribution of mast cells and the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), nuclear factor (NF)-κB, inflammasome compositional protein and inflammatory cytokines were examined in CT26 colon tumor-bearing BALB/cKorl syngeneic mice after administrating AEC for five weeks. After treatment of AEC, total weight of tumor and necrotic region of tumor section were significantly decreased compared to vehicle treated group. The number of infiltered mast cells was higher in AEC treated group than vehicle treated group, while the expression levels of COX-2 and iNOS were decreased in AEC treated group. Also, similar decrease pattern were detected in the expression levels of NF-κB, NLR family pyrin domain containing 3 (NLRP3), apoptosis-associated speck-like protein containing a caspase recruitment domain (ASC) and caspase-1 (Cas-1) after AEC treatment although the decrease rate was varied. Furthermore, the mRNA expressions of three inflammatory cytokines including tumor necrosis factor-α (TNF-α), interleukin-1α (IL-1α) and interleukin-6 (IL-6) were remarkably decreased in AEC treated group compared to vehicle treated group. These results suggest that inhibition of inflammatory response may be tightly associated with anti-tumor activity of AEC in CT26 colon tumor-bearing BALB/cKorl syngeneic mice.

A study of analytical method for Benzo[a]pyrene in edible oils (식용유지 중 벤조피렌 분석법 비교 연구)

  • The benzo[a]pyrene in edible oils is extracted using methods such as Liquid-liquid, soxhlet and ultrasound-assisted extraction. However these extraction methods have significant drawbacks, such as long extraction time and large amount of solvent usage. To overcome these drawbacks, this study attempted to improve the current complex benzo[a]pyrene analysis method by applying the QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) method that can be analyzed in a simple and short time. The QuEChERS method applied in this study includes extraction of benzo[a]pyrene into n-hexane saturated acetonitrile and n-hexane. After extraction and distribution using magnesium sulfate and sodium chloride, benzo[a]pyrene is analyzed by liquid chromatography with fluorescence detector (LC/FLR). As a result of method validation of the new method, the limit of detection (LOD) and quantification (LOQ) were 0.02 ㎍/kg and 0.05 ㎍/kg, respectively. The calibration curves were constructed using five levels (0.1~10 ㎍/kg) and coefficient (R2) was above 0.99. Mean recovery ratio was ranged from 74.5 to 79.3 % with a relative standard deviation (RSD) between 0.52 to 1.58 %. The accuracy and precision were 72.6~79.4 % and 0.14~7.20 %, respectively. All results satisfied the criteria ranges requested in the Food Safety Evaluation Department guidelines (2016) and AOAC official method of analysis (2023). Therefore, the analysis method presented in this study was a relatively simple pretreatment method compared to the existing analysis method, which reduced the analysis time and solvent use to 92 % and 96 %, respectively.
