4갈락토오즈 측정을 위한 전기화학적 바이오센서 개발 (Development of electrochemical biosensor for determination of galactose)

  • 본 연구에서는 갈락토오즈의 현장검사(point-of-care testing, POCT)를 위한 일회용 갈락토오즈 바이오센서 개발에 관해 논하고자 한다. Galactose oxidase(GAO)와 horseradish peroxidase(HRP) 두 효소를 0.05 M phosphate 완충용액에 용해시킨 후 스크린 프린팅(screen printing) 방법으로 제작한 전극위에 고정화하여 센서를 제작하였다. 이렇게 제작된 센서를 이용하여 $100{\mu}L$ 이하의 시료를 이용하여 갈락토오즈를 90초 이내에 측정하였다. 전극에서의 반응을 최적화하기 위하여 GAO 효소가 가장 우수한 활성을 나타내는 pH 7.0 완충용액을 이용하여 GAO와 HRP 효소를 1:1로 고정화하고, 1mM 1,1'-ferrocene-dimethanol 전자전달매개체를 도입하여 센서를 제작하였다. 유리 탄소전극의 경우 100 mV (vs Ag/AgCl), 스크린 프린트된 전극의 경우 -100 mV(vs Ag/AgCl)의 인가전압을 적용하였다. 본 연구에 의해 개발된 센서는 $0{\sim}400{\mu}M$의 갈락토오즈 농도에 대하여 우수한 직선성($r^2$ = 0.997, S/N = 3)을 나타내었고 낮은 인가전압을 적용하여 갈락토오즈를 측정하므로, ascorbic acid, uric acid 그리고 acetaminophen과 같은 방해물질의 영향을 최소화 할 수 있었다. 또한 갈락토오즈 표준 용액에 대하여 만족할 만한 재현성을 나타내었다(CV = 5%).

이동형 무대장치 활용을 통한 무대 공간성 창출 방안 연구 - 뮤지컬 <레미제라블>을 중심으로 - (A Study on Creating Stage Spatial Effects through the Utilization of Mobile Stage Sets- Focusing on the Musical <Les Misérables> -)

  • 본 연구는 대규모 뮤지컬 작품에서 필요로 하는 물리적 공간성과 극장 시설의 제약 없이 소극장에서 이동형 무대장치를 적용하여 공간성 창출 가능성을 중심으로 진행되었다. 국내 뮤지컬 시장에서는 대규모 뮤지컬의 성장세가 지속되고 있고, 이러한 배경을 바탕으로 본 연구에서는 이동형 무대장치를 기획하고 설계하여 대규모 뮤지컬 작품의 실습을 통해 소극장에서의 대규모 뮤지컬 작품을 공연 가능성을 분석하였다. 이를 위해 국내 뮤지컬 시장의 작품 분포를 분석하고, 대학 뮤지컬 교육에서 대규모 뮤지컬 작품 진출을 위한 환경 조성을 위해 공간성 창출 방법을 제시하였다. 이동형 무대장치는 고대 연극의 무대장치인 페리아크토이의 원리를 중심으로 옥외광고에 활용되는 페리아크토이의 정보전달의 특성과 뮤지컬 <노트르담 드 파리>의 이동형 기둥, 뮤지컬 <빨래>의 이동형 무대장치의 특성을 바탕으로 4면으로 구성된 기둥 형태의 이동형 무대장치로 제작되었으며, 이를 뮤지컬 <레미제라블>에 적용하여 공간형성과 인물의 내면 상태, 극의 분위기 형성을 위한 적용 가능성을 도출하였다. 본 연구는 대학 뮤지컬 교육에서 대규모 뮤지컬 작품의 공연 기회를 제공하는 것이 뮤지컬 산업과 교육의 발전을 위한 기반이 될 것으로 기대하며, 이동형 무대장치의 활용을 통해 소극장에서의 대규모 뮤지컬 작품을 공연의 적용 가능성을 제시하였다.

혁신 및 제조 허브의 미국으로 이동에 관한 연구 (Study on Shift of Innovation and Manufacturing Hubs to the United States)

  • 본 연구는 허브(혁신,제조)의 타국 이동에 따른 자국 산업과 허브화된 미국의 산업(배터리,반도체,전기차)에 관한 것이다. 미국과 중국 사이의 지속적인 무역 긴장은 기업들이 그들의 사업을 미국으로 옮기는 데 또한 역할을 했을 수있다. 이러한 움직임의 결과는 잠재적으로 미국의 일자리 창출과 미국 제조업 분야에 대한 투자 증가를 포함할 수 있다. 다만, 이전 과정에서 소비자들의 가격이 상승하거나 공급망에 차질이 빚어지는 등 부정적인 결과가 초래될 수도 있다. 그러나, 중국을 포함한 각 나라의 주요 첨단 산업(배터리, 반도체, 전기차)들이 미국으로 이동한다면, 그 문제는 달라질수 있다. 미국 경제는 성장하고 첨단 산업 공장을 미국에 설립한 기업이나 국가는 곤경에 처할 수 있다. 그러나 이러한 IRA, Chips Act 시나리오는 일자리와 투자를 잃고 결과적으로 경제적 어려움에 직면할 수 있기 때문에 산업이 미국으로 이동한 국가에도 부정적인 결과(자국 물가상승)를 초래할 가능성이 높다. 한국과 미국의 수출규모에 대한 실증분석결과, 글로벌 공급 허브의 이동 변화가 지정학적 가격상승과 소비감소와 같은 요소들과 연관이 된다. 본 논문은 이러한 변화에 대응하기 위해 규모의 생산지를 이동시켜 탈 첨단화되는 결과를 방지해야 함을 강조하고자 한다.

  • 영화를 본다는 것은 극장처럼 대형화된 스크린 혹은 대형화를 추구하는 TV를 통해 시청하는 것이 일반적이었지만 현재에는 모바일 디바이스 같은 소형 플렛폼을 이용하여 시청하는 빈도수가 급속도로 증가하고 있다. 이러한 변화는 인터넷 기반 동영상 스트리밍 서비스 플랫폼인 OTT(Over The Top)의 등장과 깊은 연관이 있다. OTT의 개발은 케이블 TV 시청의 필수 디바이스인 세톱박스를 이용하지 않고 시간과 공간의 제약에서 벗어난 자유로운 영상 시청을 제공하게 되었고 이 시장의 선두엔 인터넷 기반 DVD대여 서비스로 사업을 시작한 넷플릭스(NETFLIX)가1) 있다. 모바일 시장과 비례하여 동반 성장 중인 넷플릭스는 2018년 말 기준 1억3926만명의2) 회원을 보유하고 있으며 그 외 OTT 참여 회사로 콘텐츠 기반의 Pooq, TVing, 플랫폼 기반의 올레TV모바일, 옥수수, LTE비디오포털 등이 있다. 이와 같은 신 성장 사업의 규모는 점차적으로 증가하여 현재 스마트폰 전체 이용자의 25.4%가 스마트폰 같은 소형 모바일 디바이스를 이용 하여 영상콘텐츠를 시청하고 있다. 따라서 탈 대형화 중심 즉, OTT 서비스가 가능한 소형 모바일 디바이스 시청을 위해 차별화된 영상 문법이 대두되어야 판단된다. 이를 위해 본 논문은 대중적 영상콘텐츠를 소형 모바일 디바이스로 시청함에 있어 그 문제점을 파악하고 시청자에게 미치는 영향을 설문을 이용하여 조사 연구하겠다.

Diversification Strategy through Market Creation: The Case of CJ Group

  • The purpose of this paper is to investigate upon a diversification strategy through market creation of CJ Group, which has contributed in positioning of the firm as one of the leading conglomerates in South Korea. With such objective, the background of CJ Group, followed by its business diversification strategies were explored, with reference to several case studies. The history of CJ Group began with establishment of CheilJedang Industrial Corporation in 1953, as the first domestic sugar producer and exporter of South Korea. The corporation gradually expanded its business ever since at both national and global level, to include the fields of food production, pharmaceutical, biotechnology, and life chemicals. Later, CheilJedang (CJ) Group was established as an affiliate of CheilJedang Industrial Corporation. With such independence, extension of business has been witnessed across the industries of media, entertainment, finance, information technology and distribution. Thus, the current CJ Group pursues to define itself as a progressive global living culture company with four major business categories from food and food service, biotechnology, entertainment and media, and logistics. Despite its success in today's market, CJ Group underwent hardships in its business diversification in 1990s due to indiscreet management, along with the Asian financial crisis. Here, many firms overcame the financial difficulties by taking advantage of the exchange rate for overseas expansion. Though, CJ Group tried to differentiate itself by focusing on the domestic market by creating something out of nothing. Hence, CJ Group takes a unique position among many cases of business diversification and their categorization. In an effort to identify and classify the types of growth experienced by the top 30 companies in South Korea, the firms were categorized into four groups according to their diversification strategies adapted after the Asian financial crisis. Based on the mode and time of entry, corporations were identified either as the 'Explorer', 'Invader', 'Venture Capitalist', or 'Assimilator'. Here, the majority of the firms showed the qualities of Invader, entering mature markets through large-scaled mergers and acquisitions. However, CJ Group was the only firm that was categorized as an Explorer, for its focus on the newly emerging service sector in culture-contents industry. This diversification strategy through market creation is worth examining, due to its contribution in generating simultaneous growth between the market and the company itself. Diverse brands of CJ Group have been referred to as case studies in this regard, from 'Hatban', 'Cine de Chef', 'VIPS' to 'CJ GLS'. These four businesses, each to represent processed food, film, restaurant service, and logistics industries respectively, show CJ Group's effectiveness in creating a whole new category of goods and services that are innovative. In fact, such businesses not only contributed in advancement of consumers' wellbeing, but toward generating additional value and employment. It is true that the diversification strategy of CJ Group requires long-term capital investment with high risk, compared to the other strategies mentioned in the paper. However, this model does create high employment and additional values that are positive to both the society and the firm itself. Therefore, the paper comes to a conclusion that the diversification strategy through market creation conveys the most positive impact relative to the others.

Effects of zinc oxide and calcium-doped zinc oxide nanocrystals on cytotoxicity and reactive oxygen species production in different cell culture models

  • Objectives: This study aimed to synthesize nanocrystals (NCs) of zinc oxide (ZnO) and calcium ion (Ca2+)-doped ZnO with different percentages of calcium oxide (CaO), to evaluate cytotoxicity and to assess the effects of the most promising NCs on cytotoxicity depending on lipopolysaccharide (LPS) stimulation. Materials and Methods: Nanomaterials were synthesized (ZnO and ZnO:xCa, x = 0.7; 1.0; 5.0; 9.0) and characterized using X-ray diffractometry, scanning electron microscopy, and methylene blue degradation. SAOS-2 and RAW 264.7 were treated with NCs, and evaluated for viability using the MTT assay. NCs with lower cytotoxicity were maintained in contact with LPS-stimulated (+LPS) and nonstimulated (-LPS) human dental pulp cells (hDPCs). Cell viability, nitric oxide (NO), and reactive oxygen species (ROS) production were evaluated. Cells kept in culture medium or LPS served as negative and positive controls, respectively. One-way analysis of variance and the Dunnett test (α = 0.05) were used for statistical testing. Results: ZnO:0.7Ca and ZnO:1.0Ca at 10 ㎍/mL were not cytotoxic to SAOS-2 and RAW 264.7. +LPS and -LPS hDPCs treated with ZnO, ZnO:0.7Ca, and ZnO:1.0Ca presented similar NO production to negative control (p > 0.05) and lower production compared to positive control (p < 0.05). All NCs showed reduced ROS production compared with the positive control group both in +LPS and -LPS cells (p < 0.05). Conclusions: NCs were successfully synthesized. ZnO, ZnO:0.7Ca and ZnO:1.0Ca presented the highest percentages of cell viability, decreased ROS and NO production in +LPS cells, and maintenance of NO production at basal levels.

Parameter estimation and assessment of bias in genetic evaluation of carcass traits in Hanwoo cattle using real and simulated data

  • Most carcass and meat quality traits are moderate to highly heritable, indicating that they can be improved through selection. Genetic evaluation for these types of traits is performed using performance data obtained from commercial and progeny testing evaluation. The performance data from commercial farms are available in large volume, however, some drawbacks have been observed. The drawback of the commercial data is mainly due to sorting of animals based on live weight prior to slaughter, and this could lead to bias in the genetic evaluation of later measured traits such as carcass traits. The current study has two components to address the drawback of the commercial data. The first component of the study aimed to estimate genetic parameters for carcass and meat quality traits in Korean Hanwoo cattle using a large sample size of industry-based carcass performance records (n = 469,002). The second component of the study aimed to describe the impact of sorting animals into different contemporary groups based on an early measured trait and then examine the effect on the genetic evaluation of subsequently measured traits. To demonstrate our objectives, we used real performance data to estimate genetic parameters and simulated data was used to assess the bias in genetic evaluation. The results of our first study showed that commercial data obtained from slaughterhouses is a potential source of carcass performance data and useful for genetic evaluation of carcass traits to improve beef cattle performance. However, we observed some harvesting effect which leads to bias in genetic evaluation of carcass traits. This is mainly due to the selection of animal based on their body weight before arrival to slaughterhouse. Overall, the non-random allocation of animals into a contemporary group leads to a biased estimated breeding value in genetic evaluation, the severity of which increases when the evaluation traits are highly correlated.

Assessment of Phenolic Content, Saponin Content, and Antioxidant Activities in Gray, Red, and White Adzuki Bean Germplasm: A Multivariate Analysis

  • Seed color is controlled by several genes and is a key trait in determining the metabolite content and biological activities of legume genotypes. In this study, 296 adzuki bean accessions, including 159 grey, 99 red, and 38 white adzuki beans, were grown in Korea. Variations in total phenolic content (TPC), total saponin content (TSC), DPPH scavenging activity, ABTS•+ scavenging activity, and ferric reducing antioxidant power (FRAP) were assessed and were reported to be in the ranges of 1.52-8.24 mg GAE/g, 14.36-114.22 mg DE/g, 0.23-12.84 mg AAE/g, 1.05-17.66 mg TE/g, and 0.59-13.14 mg AAE/g, respectively, with a wide variation across adzuki beans. Except for DPPH scavenging activity, the average values declined in the order gray > red > white adzuki beans, each demonstrating a significant variation (p < 0.05). White adzuki beans, which showed low metabolite content and antioxidant activity, were clearly separated from the gray and red genotypes using principal component and hierarchical cluster analyses. Moreover, TPC, TSC, and antioxidant activities were strongly correlated, regardless of seed color. Overall, the diversity of the TPC, TSC, and antioxidant activity in a broad population of adzuki bean genotypes was determined. Furthermore, this study found that seed color variation in adzuki beans had a significant effect on the metabolite content and antioxidant activity. Superior accessions with high levels of TPC, TSC, and antioxidant activity were also discovered and could be used for functional plant breeding and human consumption. The findings of this study may be useful for understanding the relationship between seed coat color and metabolite concentration in adzuki beans, paving the way for molecular-level analyses.

Dimensional Stability and Mechanical Properties of Citric Acid Impregnated Samama Wood (Anthocephalus macrophyllus (Roxb) Havil) at High Curing Temperatures

  • Samama wood (Anthocephalus macrophyllus (Roxb) Havil) is a fast-growing and lesser-utilized wood species that has inferior properties; therefore, its quality needs to be improved. This research aimed to determine the effect of citric acid impregnation at high curing temperatures on the dimensional stability and mechanical properties of wood. Citric acid solution with 10% concentration (w/w) was impregnated into wood samples by vacuum-pressure method (-0.5 cmHg, 30 min; 0.7 MPa, 3 h), followed by curing process at 140℃, 160℃, and 180℃ of temperature for 1 h. In comparison, the other wood samples were heat treated at the same temperatures and time. The results showed that the increase in curing and heat temperatures for both treatments were directly proportional to the dimensional stability, but inversely proportional to the mechanical properties. Citric acid impregnated had higher density, dimensional stability, and mechanical properties, except for modulus of rupture, than that of heat treatment. The optimum temperature is suggested at 160℃ in both treatments.

Opto-Mechanical Detailed Design of the G-CLEF Flexure Control Camera

  • The GMT-Consortium Large Earth Finder (G-CLEF) is the first instrument for the Giant Magellan Telescope (GMT). G-CLEF is a fiber feed, optical band echelle spectrograph that is capable of extremely precise radial velocity measurement. G-CLEF Flexure Control Camera (FCC) is included as a part in G-CLEF Front End Assembly (GCFEA), which monitors the field images focused on a fiber mirror to control the flexure and the focus errors within GCFEA. FCC consists of an optical bench on which five optical components are installed. The order of the optical train is: a collimator, neutral density filters, a focus analyzer, a reimager and a detector (Andor iKon-L 936 CCD camera). The collimator consists of a triplet lens and receives the beam reflected by a fiber mirror. The neutral density filters make it possible a broad range star brightness as a target or a guide. The focus analyzer is used to measure a focus offset. The reimager focuses the beam from the collimator onto the CCD detector focal plane. The detector module includes a linear translator and a field de-rotator. We performed thermoelastic stress analysis for lenses and their mounts to confirm the physical safety of the lens materials. We also conducted the global structure analysis for various gravitational orientations to verify the image stability requirement during the operation of the telescope and the instrument. In this article, we present the opto-mechanical detailed design of G-CLEF FCC and describe the consequence of the numerical finite element analyses for the design.
