Time course monitoring of FCW/DCW ratio, production of intra and extracellular protein, and peroxidase activity were performed during suspension culture of Taxus chinensis cells. The observed FCW/DCW ratio was 12 at day 14, which was the lowest value during cultivation, and the specific protein production, based on dry cell weight, was also the lowest at day 14, which showed 4.3 mg/g DCW. The pattern of POD activity was similar to that of protein production. The results in this report were obtained using actively growing cells in flasks, therefore it is possible to use those results to control the process and indicate the stresses imposed on cells during large-scale cultivation.
In the fed-batch fermentation of penicillin specific uptake rates of carbon source and ammonia nitrogen, and specific production rate of penicillin as the most important process variables were evaluated over the fermentation course and their effects on the productivity studied. As the results, glucose and lactose each as a major carbon source showed the following values, respectively ; the specific uptake rates of 47-93 mg hexose per gm-DCW per he and 37-44 mg hexose per gm-DCW per hr, the specific uptake rates of 4.6-6.8 mg $NH_3-N$ per gm-DCW per hr, and 1.2 mg $NH_3-N$ per gm-DCW per he and the specific production rates of 32-42 arbitrary unit per gm-DCW per hr and 46-50 arbitrary unit per gm-DCW per hr. The productivity of penicillin could be improved by controlling the feed rates of glucose and ammonia nitrogen to meet the uptake rates.
The morphology of filamentous fungi closely correlates with the productivity in submerged culture. Using itaconic acid (IA) production by Aspergillus terreus as a research model, the quantitative relationship between the growth form of A. terreus and IA production was investigated. IA fermentation was scaled up from shake flasks to a 7 L stirred tank bioreactor based on the quantitative relationship. Our results demonstrated the following: (1) Three morphologies of A. terreus were formed by changing the inoculum level and shape of the flask. (2) Investigation of the effects of the three morphologies on broth rheology and IA production revealed the higher yield of IA on dry cell weight (DCW, IA/DCW) and yield of glucose on DCW (consumed glucose/DCW) were achieved during clump growth of A. terreus. (3) By varying the $KH_2PO_4$ concentration and culture temperature, the relationships between clump diameter and IA production were established, demonstrating that the yield of IA on DCW ($R^2$ = 0.9809) and yield of glucose on DCW ($R^2$ = 0.9421) were closely correlated with clump diameter. The optimum clump diameter range for higher IA production was 0.40-0.50 mm. (4) When the clump diameter was controlled at 0.45 mm by manipulating the mechanical stress in a 7 L fermentor, the yield of IA on DCW and yield of glucose on DCW were increased by 25.1% and 16.3%, respectively. The results presented in this study provide a potential approach for further enhancement of metabolite production by filamentous fungi.
Effects of various environmental factors on cell size index(FCW/DCW) in Thalictrum rugosum. Lithospermum erythrorhizon and Taxus cuspidata plant cell suspension cultures were investigated. Time course change of cell size index were also observed. In batch cultures, FCW/DCW increased according to the decrease of sugar concentration. For short-term experiment within 24 hr, FCW/DCW value could be reduced significantly by increasing sugar concentration. When an osmoticum such as mannitol was added, FCW/DCW converged to a low value. Therefore, it was confirmed that osmolality of the medium was important in determining cell size or water content of the cells. Inorganic salts or treatment with organic solvent also exhibited some effect on the cell size index. However, pH and centrifugal force did not show any influences. On the other hand, it was found that the addition of Pluronic F-68 reduced FCW/DCW. By combining these results effectively, it may be possible to increase the cell concentration in high density culture to a higher extent.
To obtain a yeast mutant with high RNA content and high growth rate, Saccharomyces cerevisiae MTY62 was mutated with ethylmethane sulfonate. Among the selected mutants that were sensitive to the high concentration of KCl, M40-10 strain was finally selected due to its rapid cell growth and high RNA content in the tube and baffled-flask cultures. In the batch culture of M40-10 mutant, the maximum specific growth rate ($\mu_{max}$) of $0.38 h^{-1}$ , RNA concentration of 3210 mg-RNA/1, and RNA content of 183 mg-RNA/g-DCW were obtained, which were 23%, 15%, and 12% increased levels, respectively, compared to those of MTY62 parent strain. The intermittent fed-batch culture of M40-10 strain resulted in the maximum cell concentration of 35.6 g-DCW/1, RNA concentration of 5677 mg/1, and RNA content of 160 mg-RNA/g-DCW. Through the constant fed-batch culture, the maximum cell concentration of 46.4 g-DCW/1, RNA concentration of 6270 mg-RNA/1, and RNA content of 135 mg-RNA/g-DCW were obtained. At the 20 h culture time in the fed-batch cultures of M40-10 strain, the cell and RNA concentrations were increased by 30% and 10%, respectively, over the parent strain MTY62. In addition, it was also found that the accumulated RNA within the mutant cell was not degraded until the end of fed-batch cultivation, indicating that the M40-10 cell is a mutant with weak acidic RNase activity.y.
Methyl ethyl ketone (MEK) and methyl isobutyl ketone (MIBK) have been widely used as solvents in various industries. Biodegradation of MEK and MIBK by Pseudomonas putida KT-3, which could utilize MEK or MIBK as a sole carbon source, was characterized, and the cosubstrate interaction in MEK/MIBK mixture was also studied. Within the range of initial MEK concentration (from 0.5 to 5.5 mM), an increased substrate concentration increased the specific degradation rate of MEK by P putida KT-3 (from 3.15 to 10.58 mmol/g DCW$\cdot$h), but the rate sightly increased at 11.0 mM of initial MEK concentation (11.28 mmol/g DCW$\cdot$h). The similar degradation rates of MIBK (4.69-4.92 mmol/g DCW$\cdot$h) were obtained at more than 3.0 mM of initial MIBK concentation. Kinetic analysis on the degradation of MEK/MIBK mixture by P. putida KT-3 showed that MEK or MIBK acted as a competitive inhibitor. Maximum degradation rate ($V_{max}$), saturation constant ($K_{m}$) and inhibition constant ($K_{1}$) were as follows: $V_{max,MEK}$=12.94 mmol/g DCW$\cdot$h; $K_{m,MEK}$=1.72 mmol/L; $K_{l,MEK}$=1.30 mmol/L; $V_{max,MIBK}$=5.00 mmol/g-DCW$\cdot$h; $K_{m,MIBK}$=0.42 mmol/L; $K_{l,MEK}$=0.77 mmol/L.
To increase the efficiency of esterase production by Bacillus, high cell-density culture of recombinant Escherichia coli through fed batch fermentation was tested. Cells were cultured to $OD_{600}$ of 76 (35.8 g/l DCW) with dissolved oxygen level controlled to least above 30% air saturation by supplying pure oxygen. Cells were cultured to an $OD_{600}$ of 90 (42.4 g/l DCW) with glucose feeding controlled to at least 1 g/l. However, the cells reached stationary phase at the late stage of culture, despite glucose being supplied. Cells were cultured to an $OD_{600}$ of 185 (87.3 g/l DCW) by supplying additional medium with fortified yeast extract. To increase the productivity of the recombinant protein, cell growth and esterase productivity based on induction time were evaluated. Late exponential phase induction for esterase production in fed batch fermentation resulted in maximum optical density $OD_{600}$ of 190 (89 g/l DCW) and maximum esterase activity of 1745 U/l, corresponding to a 5.8-fold enhancement in esterase production, compared to the early exponential phase induction. In this study, we established fermentation methods for achieving maximum production of Bacillus-derived esterase by optimizing IPTG induction time in high-cell density culture by supplying pure oxygen and a nitrogen source.
We investigated the effects of dissolved oxygen (D.O.) and fructose (C-source) on cell growth and biosynthesis of cyclosporin A (CyA) produced as a secondary metabolite by a wild-type filamentous fungus, Tolypocladium inflatum. This was performed by controlling the level of D.O. and the residual C-source, as required, through adjustment of medium flow rate, medium concentration and agitation rate in fed-batch cultures. CyA production was furned out to be maximal, when D.O. level was controlled around 10% saturated D.O. and concentration of the C-source was maintained sufficiently low (below 2 g/L) not to cause carbon catabolite repression. Under this culture condition, we obtained the highest values of CyA concentration (507.14 mg/L), Qp (2.11 mg CyA/L/hr), $Y_x/s$ (0.49 g DCW/g fructose), $Y_p/s$<(22.56 mg CyA/g fructose), and YTEX>$_p/x$ (48.31 mg CyA/g DCW), but relatively lower values of cell concentration (11.98 g DCW/L) and cell productivity (0.043 g DCW/L/hr), in comparison with other parallel fed-batch fermentation conditions. These results implied that, in the carbon-limited culture with 10% saturated D.O. level, the producer microorganism utilized the C-source more efficiently for secondary metabolism.
The usage of ethanol (EtOH)-blended gasoline (gasohol), has been increasing in recent years. EtOH has influence on the distribution and biodegradation of aromatic compounds such as BTEX (benzene (B), toluene (T), ethylbenzene (B), and xylene (X)) that are gasoline compositions. In this study, the effect of EtOH on the aerobic biodegradation of B, T and E was investigated using a BTEX and EtOH-degrading bacterium, Rhodococcus sp. EH831. The degradation rates of B in the conditions of 1:1, 1:4, and 1:0.25 mixtures with EtOH (B:EtOH, mol:mol) were ranged from $3.82{\pm}0.20$ to $5.00{\pm}0.37{\mu}mol{\cdot}g-dry$ cell wight $(DCW)^{-1}{\cdot}h^{-1}$. The degradation rate of T was the fastest in the 1:0.25 mixture ($6.63{\pm}0.06{\mu}mol{\cdot}g-DCW^{-1}{\cdot}h^{-1}$), and it was the lowest in the 1:4 mixture ($4.41{\pm}0.04{\mu}mol{\cdot}DCW^{-1}{\cdot}h^{-1}$). The degradation rates of E were increased with increasing the addition amount of EtOH: The degradation rate of E was the highest in the 1:4 mixture ($1.60{\pm}0.03{\mu}mol{\cdot}g-DCW^{-1}{\cdot}h^{-1}$), and the rates were $1.42{\pm}0.06$, $1.30{\pm}0.01$, and $1.01{\pm}0.30{\mu}mol{\cdot}g-DCW^{-1}{\cdot}h^{-1}$ in the 1:1, 1:0.25, 1.0 mixtures, respectively. In conclusion, the biodegradation of B, T, E by Rhodococcus sp. EH831 was not significantly inhibited by the co-existence of EtOH.
In order to product the furofuranoid lignans, (+)-eudesmin which is one of the secondary products from Magnolia sieboldii. through cell suspension cultures; various culture media, initial sucrose concentration, elicitations, shaking speeds, and inoculum sizes. Among the culture media tested, MS medium had a pronounced effect on suspension cell growth and (+)-eudesmin contents. The maximum dry cell weight (DCW) of 3.71 g per flask was obtained at inoculum size of 0.5 g and in MS medium supplemented with $3\%$ sucrose plus 0.5 mg/L 2,4-D after 8 weeks. (+)-Eudesmin biosynthesis was stimulated with high initial sucrose concentration ,and the maximum (+)-eudesmin production of $3.2{\mu}g/g$ DCW was achieved at 200mg/L chitosan and $5\%$ initial medium sucrose. The optimal shaking speeds for dry biomass accumulation and (+)-eudesmin contents was 130 rpm. This work is considered to be helpful for large-scale bioprocessing of Magnolia sieboldii suspension cell cultures in bioreactor.
