Expedments were perfonned to examine the role of cAMP-dependent protein kinase (PK-A) and diacylglycerol-dependent protein kinase (PK-C) during the meiodc resumption and the first mitotic cell cycle of mouse embryogenesis. Mejoric GV oocytes and one-cell embryos derived from in vitro fertilization were cultured in vitro, and morphological changes in response to activators of PK-A and PK-C were examined. Treatments with a membrane-permeable cAMP analog, dbcAMP (0.1 mg/mi), phosphodiesterase inhibitor, IBMX (0.1 mM), biologically active phorbol ester, WA (10 nglmi), or a synthetic diacylglycerol, sn-diC8 inhibited resumption of melosis. Combination of PK-A and PK-C activator brought about furiher inhibition. On the contrary, dbcAMP (0.1 mg/mi), IBMX (0.2 mM), WA (10 nglml), and sn-diC8 (0.5 mM) did not inhibit pronucleus membrane breakdown (PNBD) when added S or G2 phase of cell cycle. However, activators of PK-C inhibited cleavage of one-cefl embryos. This result indicates that the action mechanism of PK-A and PK-C on dissolution of nuclear membrane in primary meiotic arrest oocytes may be different from that of mitotic one-cell embryos.
Human polymorphonuclear leukocytes(PMN) are the most numerous host cell in periodontal pockets and their presumed role is to form a protective barrier between the bacteria and periodontal tissues. Microbial component LPS activates macrophages to produce $IL-1{\beta}$, $MIP-1{\alpha}$, $-1{\beta}$, $TNF-{\alpha}$ and IL-6, etc. These cytokines have autocrine function to the macrophages, and paracrine function to other cell such as PMN and affect them to produce some biological functions. In the present study, human PMN were tested for the expression of $IL-1{\beta}$ and $MIP-1{\alpha}$ mRNA. Also we performed the receptor binding assay and in vitro assay for the antimicrobial action of HL-60 cell to determine whether HL-60 can replace the peripheral PMN in analyzing the biological functions. PMN were stimulated with $IL-1{\beta}$, TPA, $MIP-1{\alpha}$, LPS, IL-2 and total cytoplasmic RNA were extracted for the northern blot analysis. In order to determine the induction kinetics of $IL-1{\beta}$ or $MIP-1{\alpha}$ mRNA expression, cells were stimulated for 0,1,2,3 hours. We found peak expression of $IL-1{\beta}$ mRNA after 1hr of induction with $IL-1{\beta}$, LPS and after 2hr of induction with TPA. $MIP-l{\alpha}$ also induced but a scarce $IL-l{\beta}$ message from PMN. In contrast to the $IL-l{\beta}$ mRNA expression, $MIP-1{\alpha}$ were not induced from PMN in any culture conditions. Receptors for $MIP-1{\alpha}$ were identified on dibutyryl cyclic AMP(dbcAMP)-treated HL-60 as well as peripheral PMN. dbcAMP treatment significantly enhanced antimicrobial action of undifferentiated HL-60 cell. MIP-1 further increased enhancing effect of dbcAMP. $IL-1{\beta}$, to a lesser extent, also increased dbcAMP-induced enhancing effect of antimicrobial action of HL-60 cell.
Cyclic-AMP-dependent protein kinase (PKA) seems to function as a negative regulator of the c-Jun $NH_2-terminal$ kinase (JNK) signaling pathway. We demonstrate here that the activity of the PKA catalytic subunit (PKAc) is reduced in apoptotic PC12 pheochromocytoma cells. Apoptotic progress was inhibited by dibutyryl cyclic AMP (dbcAMP), an analog of cAMP. The rescue by dbcAMP was attributable to inhibition of the JNK but not of the p38 signaling pathway, due to the induction of PKA activity. JNK was present in immunocomplexes of PKAc, and PKAc phosphorylated JNK in vitro. Presence of p38 kinase, however, was not prominent in immunocomplexes of PKAc. Our data suggest that JNK is a target point of negative regulation by PKAc in the JNK signaling pathway.
We evaluated the effects of the substrate and inhibitors related to phosphatidylinositol metabolism on in vitro maturation and fertilization of porcine oocytes. Cumulus-oocyte complexes were cultured in mTLP-PVA medium supplemented with or without inositol (250 mM) fur 46h. Subsequently, these oocytes were inseminated with fresh boar semen in mTALP-PVA medium for 6h. At 6h after insemination, oocytes were cultured for further 12 h in TCM-199 supplemented with 10% FBS (fetal bovine serum). The higher percentage of oocytes in inositol-supplemented medium reached metaphase of the second meiotic division compared to those in control (81.4% vs. 67.3%; P<0.()5). following 18 h of insemination, more number of male pronuclei were formed in the oocytes matured in inositol-supplemented medium than in those of control experiment (42.0% vs. 27.3%; P<0.05). When oocytes were cultured in medium with 10mM LiCl (chloride lithium) or 0.5mM dbcAMP (dibutyryl cyclic adenosine monophosphate) to determine the role of inositol on the maturation of oocytes, these two drugs inhibited the meiotic division of oocytes (P<0.05). However, addition of inositol to the culture medium did overcome the inhibitory effect of these drugs on the oocyte maturation. DbcAMP and verapamil supplemented synergistically arrested the meiotic division of oocytes. Addition of verapamil did not inhibit germinal vesicle breakdown, but it severly inhibited the second meiotic division of oocytes. These results suggest that inositol exert its improving effects on maturation, by activating the PI (phosphatidylinositol) cycle and causing beneficial changes in both cytoplasm and membrane of oocytes.
It has been found that the rat oocytes maintain germinal vesicle (GV) in general in the follicles either untreated or punctured, or in the foreign follicles for 17 hours culture unless they are cultured in the medium supplemented with human chorionic gonadotropin (hCG). That is, the proportion of oocytes with GV was in range of 88.8% and 95.2% in the plain medium, and on the other hand, only 11.1% to 19.4% of the oocytes were intact with GV when the follicles were exposed to hCG. The experiments with the oocytes which had once been cultured in the presence of dbcAMP or IBMX, and returned to the follicles for the additional culture showed almost the same results as above. That is, when the oocytes exposed to dbcAMP or IBMX for a certain length of period had been returned to the follicles, and set the additional culture, their maturation continuously suppressed even in the cultivation in the plain medium in which most of the oocytes usually resume meiosis. That is, despite of the cultivation in the plain medium, the oocytes transferred into the follicles failed to start maturation division, while the oocytes once exposed to the inhibitors immediately resume their maturation process in the inhibitor-free medium. Thus, it is apparent that the follicles provide inhibitory environment to the oocytes, and the inhibitory function is nullified by the presence of hCG.
The aim of this study was to investigate what protein(s) of porcine epididymal fluid (pEF) are able to enhance the nuclear maturation of porcine germinal vesicle (GV) oocytes in vitro. Proteins of pEF were fractionated by affinity, ion exchange, and gel filtration chromatography. Porcine cumulus-oocytes complexes (COC) from follicles were cultured in tissue culture medium (TCM 199) containing various fractions obtained by chromatography. Porcine COCs were also cultured in TCM 199 containing various meiosis inhibitors and pEF. After 24 or 48 h culture, oocytes were examined for evidence of GV breakdown, metaphase I, anaphase-telophase I, and metaphase II. When porcine COCs were cultured in the medium with meiosis inhibitor such as, dibutyryl cAMP (dbcAMP) and forskolin (Fo), more than 80% of oocytes were unable to resume meiosis. However, porcine COCs supplemented with pEF were able to overcome the inhibitory effect of dbcAMP and Fo. Maturation rate of oocytes was significantly (p<0.05) increased in the media supplemented with cationic protein(s) during in vitro maturation than in those with anionic protein(s) (44.1% vs 20.0%). When oocytes were cultured in the TCM 199 with fractions obtained by gel filtration, the maturation rate of oocytes was significantly (p<0.05) higher in fraction 11 containing 18 kDa than other fractions. The present study suggests that 1) dbcAMP and Fo prevent the spontaneous maturation of oocyte after isolation from follicles, and that pEF contain a substance(s) that improves meiosis resumption in vitro of porcine COCs, 2) cationic 18 kDa protein(s) are responsible for promotion of Mil stage.

This paper reports an experimental result on optimizing dissimilarity-based classification(DBC) by simultaneously using a dynamic time warping(DTW) and a multiple fusion strategy(MFS). DBC is a way of defining classifiers among classes; they are not based on the feature measurements of individual samples, but rather on a suitable dissimilarity measure among the samples. In DTW, the dissimilarity is measured in two steps: first, we adjust the object samples by finding the best warping path with a correlation coefficient-based DTW technique. We then compute the dissimilarity distance between the adjusted objects with conventional measures. In MFS, fusion strategies are repeatedly used in generating dissimilarity matrices as well as in designing classifiers: we first combine the dissimilarity matrices obtained with the DTW technique to a new matrix. After training some base classifiers in the new matrix, we again combine the results of the base classifiers. Our experimental results for well-known benchmark databases demonstrate that the proposed mechanism achieves further improved results in terms of classification accuracy compared with the previous approaches. From this consideration, the method could also be applied to other high-dimensional tasks, such as multimedia information retrieval.

Objective : Chronic subdural hematomas (cSDHs) are generally known to result from traumatic tears of bridging veins. However, the causes of repeat spontaneous cSDHs are still unclear. We investigated the changes in vasculature in the human dura mater and outer membrane (OM) of cSDHs to elucidate the cause of their spontaneous repetition. Methods : The dura mater was obtained from a normal control participant and a patient with repeat spontaneous cSDHs. The pathological samples from the patient included the dura mater and OM tightly adhered to the inner dura. The samples were analyzed with a particular focus on blood and lymphatic vessels by immunohistochemistry, 3-dimensional imaging using a transparent tissue clearing technique, and electron microscopy. Results : The dural border cell (DBC) layer of the dura mater and OM were histologically indistinguishable. There were 5.9 times more blood vessels per unit volume of tissue in the DBC layer and OM in the patient than in the normal control. The DBC layer and OM contained pathological sinusoidal capillaries not observed in the normal tissue; these capillaries were connected to the middle meningeal arteries via penetrating arteries. In addition, marked lymphangiogenesis in the periosteal and meningeal layers was observed in the patient with cSDHs. Conclusion : Neovascularization in the OM seemed to originate from the DBC layer; this is a potential cause of repeat spontaneous cSDHs. Embolization of the meningeal arteries to interrupt the blood supply to pathological capillaries via penetrating arteries may be an effective treatment option.
The minimum concentrations of nonionic and anionic surfactants inducing phytotoxicity were investigated after spraying or flooding surfactant solutions to annual plants. Of the sufactants tested, LE, NP, SPSS, LN, PAAS and DBC induced phytotoxicity at the lowest concentrations through all treatments and Tween, Span, SP, SC, STPP and CLIS induced the least phytotoxicity even at high concentration. At flooded paddy field tests, anionic surfactants induced phytotoxicity at the lower concentration than nonionic, but showed similar tendencies with other treatments. In pre-emergence treatments of upland and dry paddy field tests, there was little phytotoxicity induced at over 10 percent, while phytotoxicity was induced at the lowest concentration among 6 treatments in flooded paddy field tests.
The pepsinogen secretion was stimulated by the cholecystokinin, caerulein, isoproterenol, and carbachol, respectively. But it was increased slightly and returned to control level by the combiantions of total saponin with each above the agents. Even though the atropine had the inhibition effect, the pepsinogen secretion was recovered to normal level from depressed condition by the combination of the atropine with total saponin. Propranolol showed the same pattern as atropine, too. On the other hand, the pepsinogen secretion was stimulated by the DBcAMP alone, but decreased to control level by the combination with the total saponin. In the case of DBcGMP, the pepsinogen secretion was decreased by itself, but stimulated the above control level by the combination with total saponin. Histamine alone had little effect on the pepsinogen secretion, but when combinated with total saponin, the pepsinogen secretion was increased. Serotonin alone and with total saponin, had no effect respectively, From the above results, the total saponin may have the normalization action stimulating or decreasing the pepsinogen secretion to the control level.
