Performance of a portable GC that can be utilized for the real time determination of volatile organic compounds in air was evaluated. It employs purified/compressed ambient air as the carrier gas eliminating the need for high pressure gas tanks. The compact system with dimensions of 35 × 26 × 15 ㎤ and weight of 5 kg is powered by either a 24 V DC external adapter or battery pack. Chromatograms of the mixture sample including benzene, toluene, ethylbenzene, and oxylene at concentrations of 1 ppmv and 20 ppmv represent a good reproducibility: 3.79% and 0.48% relative standard deviations (RSDs) for peak area variations; 0.40% and 0.08% RSDs for retention times. The method detection limit was 0.09 ppmv. A 30 m long, 0.28 mm I.D. column operated at an optimal condition yielded a peak capacity of 61 with good resolution for a 10 min isothermal analysis. The relative standard deviations (RSD) of the peak area variations and retention times during consecutive measurements over 27 h were less than 2.4%RSD and 0.5%RSD, respectively. Thus, this instrument makes it suitable for continuous and field analysis of low-concentration VOC mixtures in the indoor/outdoor environment as well as the spillage accident of hazardous chemicals.
To investigate the current pest management status in Korea, a survey was conducted from 151 students and graduates in the Korea National College of Agriculture and Fisheries (KNCAF) by on-line. The questionnaire consists of two divisions, basic questions and pest control questions. The basic questions were including the respondent's age, academic status, cultivating crops and cultivating area. The pest control questions were including pest control methods, pesticide selection rationale, and pest forecasting methods. As a summary of basic questions, the respondents in their 20s accounted for 91.2%. Moreover, 34.5% of the respondents had over 3 hectares of cultivating area. The cultivating methods were differed by cultivating crops. As a summary of pest control questions, major control methods were using the conventional chemicals (>66%). To understand the pesticide selection rationale, farmers/respondents made their own decisions based on existing control techniques (30%) or depended on the decisions of pesticide vendors (29%). As for the pest forecasting method, it was mainly conducted by the Rural Development Administration affiliated organization (29%) and the National Crop Pest Management System (27%). Regarding the reliability of the pest diagnosis and pesticide prescription of pesticide vendors, 97% of the respondents marked above average. However, there was no choice on strong reliability. Interestingly, 79% of the respondents agreed to train experts for pest diagnosis and pesticide prescription with high necessity and, in particular, 47% of respondents were very strongly supported. These results suggest that the farmers might be need more qualified experts in pest diagnosis and pesticide prescriptions. Taken together, these survey results would provide important information to understand the current status of pest management by farmers' point of view and useful to set the direction of pest control.
The machine transplanted rice has highly extended in the Republic of Korea due to a wage rise since 1976 and thus rice seedling raising nursery method has shifted from field into polyethylene film house. The rice seedling grown under a polyethylene film house was weak and tall because of limited sunlight and wind and so there was so complained among rice growing farmers in terms of missing hill and broken seedling after machine transplanting. In recent iron-coated seedling has reported on healthier and stronger than that of polyethylene film house. These seedlings were more stiff and thus more easy to handle during transplanting and less missing hills. The Iron-coated seed was proven to more stiff like seedling height, root growth and weight of shoot and root growth. This was more convenient during transfer of seedling raising box im machine transplanting. Thus, this study was carried out to grow a stiff and healthy seedling growth and development for machine transplanting. There were a normal seed germination and sprouting after iron-coated seeds treatment and this seedling was smaller and stiffer than that of untreated control. The leaf number and root growth of seedling which was iron-coated seeds would be better and total seedling fresh weight was greater than that of untreated control.
This research was carried as a part of the basic study, in which the aptitude of theKorean oak varieties as barrels for aging apple fine spirits was investigated, and thefollowing results were obtained. 1. Following was the result of the chemical analysis of the fruits which are now mass-produced and can be used as a substitute for raw materials for wine production. Apple (Malus pumila Miller var. domestica Schneider) : Total sugar. total acid, volatile acid and pectin of Jonathan (Hong-og) were 13.95%, 0.46%, 0.012%, 0.20% respectively. Total sugar, total acid, volatile acid and pectin of Ralls (Koog-kwang) were 13.35%, 0.43%, 0.011%, 0.45% respectively. 2. Because of low yield of apple juice due to cellulose, pectin, hemicellulose which are present besides sugars, acids in apples, the apple juice were treated with xylanase of Aspergillus niger SUAFM-430, cellulase and pectinase of Aspergillus niger SUAFM-6. This treatment increased the yield of apple juice. And the apple juice was sterilized by adding potassium metabisulfite
The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The current study was conducted to determine the effect of suspension method on satisfaction level of Korean consumers and objective meat quality traits in Hanwoo longissimus dorsi(LD), triceps brachii(TB) and semimembranosus(SM) muscles. Eighteen Hanwoo steers were slaughtered and alternative sides were hung either by pelvic bone(TS) or Achilles tendon(Al). Sensory characteristics, WB-shear force, sarcomere length, water-holding capacity, saroomere length and cooking loss were determined after a 7-d chiller ageing. Higher carcass quality grade received significantly(p < 0.05) greater eating quality for LD, but the grade did not affect eating quality for both TB and SM. TS did not influence objective and subjective meat quality for TB, but that significantly(P < 0.05) improved eating quality for LD and SM. The most noticeable result was that when SM was tenderstretehed, eating quality was equivalent to that of nonna1ly hung LD. In relationship between objective and subjective meat quality assessments, eating qualty for LD had a significant(P < 0.05) relationship with intramuscular fat content, while that for SM was greatly(P < 0.05) related to saroomere length. The current study indicated that pelvic hanging was an effective way to improve eating quality both LD and SM, and carcass quality grades did not greatly reflect eating quality of SM and TB. The data also implied that instnunental measurements poorly estimated the satisfaction level of Korean conswners.
It is very important in forestry to study the shear strength of weathered granitic soil, because the soil covers 66% of our country, and because the majority of land slides have been occured in the soil. In general, the causes of land slide can be classified both the external and internal factors. The external factors are known as vegetations, geography and climate, but internal factors are known as engineering properties originated from parent rocks and weathering. Soil engineering properties are controlled by the skeleton structure, texture, consistency, cohesion, permeability, water content, mineral components, porosity and density etc. of soils. And the effects of these internal factors on sliding down summarize as resistance, shear strength, against silding of soil mass. Shear strength basically depends upon effective stress, kinds of soils, density (void ratio), water content, the structure and arrangement of soil particles, among the properties. Background : The p53 gene codes for a DNA-binding nuclear phosphoprotein that appears to inhibit the progression of cells from the G1 to the S phase of the cell cycle. Mutations of the p53 gene are common in a wide variety of human cancers, including lung cancer. In lung cancers, point mutations of the p53 gene have been found in all histological types including approximately 45% of resected NSCLC and even more frequently in SCLC specimens. Mutant forms of the p53 protein have transforming activity and interfere with the cell-cycle regulatory function of the wild-type protein. The majority of p53 gene mutations produce proteins with altered conformation and prolonged half life; these mutant proteins accumulate in the cell nucleus and can be detected by immunohistochemical staining. But protein overexpression has been reported in the absence of mutation. p53 protein overexpression or gene mutation is reported poor prognostic factor in breast cancer, but in lung cancer, its prognostic significance is controversial. Method : We investigated the p53 abnormalities by nucleotide sequencing, polymerase chain reaction-single strand conformation polymorphism(PCR-SSCP), and immunohistochemical staining. We correlated these results with each other and survival in 75 patients with NSCLC resected with curative intent. Overexpression of the p53 protein was studied immunohistochemically in archival paraffin- embedded tumor samples using the D07(Novocastra, U.K.) antibody. Overexpression of p53 protein was defined by the nuclear staining of greater than 25% immunopositive cells in tumors. Detection of p53 gene mutation was done by PCR-SSCP and nucleotide sequencing from the exon 5-9 of p53 gene. Result: 1) Of the 75 patients, 36%(27/75) showed p53 overexpression by immunohistochemical stain. There was no survival difference between positive and negative p53 immunostaining(overall median survival of 26 months, disease free median survival of 13 months in both groups). 2) By PCR-SSCP, 27.6%(16/58) of the patients showed mobility shift. There was no significant difference in survival according to mobility shift(overall median survival of 27 in patients without mobility shift vs 20 months in patients with mobility shift, disease free median survival of 8 months vs 10 months respectively). 3) Nucleotide sequence was analysed from 29 patients, and 34.5%(10/29) had mutant p53 sequence. Patients with the presence of gene mutations showed tendency to shortened survival compared with the patients with no mutation(overall median survival of 22 vs 27 months, disease free median survival of 10 vs 20 months), but there was no statistical significance. 4) The sensitivity and specificity of immunostain based on PCR-SSCP was 67.0%, 74.0%, and that of the PCR-SSCP based on the nucleotide sequencing was 91.8%, 96.2% respectively. The concordance rate between the immunostain and PCR-SSCP was 62.5%, and the rate between the PCR-SSCP and nucleotide sequencing was 95.3%. Conclusion : In terms of detection of p53 gene mutation, PCR-SSCP was superior to immunostaining. p53 gene abnormalities either overexpression or mutation were not a significant prognostic factor in NSCLC patients resected with curative intent. However, patients with the mutated p53 gene showed the trends of early relapse.
