Effect of Male-Sterile Cytoplasm on the Genetic Performance of Agronomic Traits in $F_1$ Hybrid Rices

  • Three different male-sterile cytoplasmic lines and their common maintainer 'Zhenshan 97B' and two elite restorer lines were used to study cytoplasmic effects on agronomic trait manifestation per se under different nitrogen supply levels. The result showed that cytoplasmic effects could be modified by nitrogen environments. The cytoplasmic effect on grain yield under 150 kg N/ha varied depending on crosses, while it was significantly negative in most crosses under both 60 and 330 kg N/ha. The correlation and path-coefficient analyses suggested that it was expected to improve cytoplasmic effects through reducing maximum tillers and increasing the percentage of productive tillers, leading to increased productive tillers and higher yield in hybrid rice by the aid of cultural practice and genetic transformation. This study also revealed that the same cytoplasm in different combinations had differential effect under the same nitrogen environment, indicating that cytoplasmic effect was produced by interaction of nuclear genes with cytoplasm rather than cytoplasm per se. These results indicated the usefulness of evaluating diverse cytoplasmic sources in various nuclear genotypes bred for hybrid rice breeding program. The finding also suggested that negative cytoplasmic effect could be effectively overcome by elite restorer lines through the interaction of nuclear genes with female cytoplasm.

  • We characterized the resistances such as to waterlogging, Phytophythora and viruses in hybrids between Italian and Korean mother lines and screened them for complex resistances and agronomic traits to select elite multi-resistant lines for hybrid breeding. Resistance to waterlogging was selectable due to diversity of the resistance. Phytophythora resistance introduced from Italian lines could also be combined with resistance to other diseases and restoration abilities from cytoplasmic male sterility that has been maintained in Korean varieties.

  • The glucose transporters found in the plasma membrane of all animal cells are known to have 12 putative transmembrane domains. Among 7 cytoplasmic loops, the fourth loop is the largest one. Since previous studies showed that cofilin, an actin-modulating protein, was found to interact with the largest cytoplasmic loop of (Na, K)ATPase, we tested if cofilin interacts with the largest cytoplasmic loop of Glut4. We demonstrated by the two-hybrid system that the largest cytoplasmic loop of Glut4 did not show any interaction with cofilin, suggesting that cofilin is not required for the membrane targeting process of other membrane proteins but only for a P-type ATPase.

세포질전환 기법에 의한 신품종 느타리 '천화심'의 육성 및 자실체 특성 (Characteristics and breeding of a new oyster mushroom (Pleurotus ostreatus) variety 'Chunhwashim' by using cytoplasmic hybrid)

  • 느타리버섯(Pleurotus ostreatus)의 품종 육성을 위해서 는 많은 노력과 시간을 필요로 하고 있어, 최근에는 분자 육종방법으로 마커를 이용하여 빠르고 정확하게 교잡주를 선발하고 있다. 본 실험에서는 육종효율을 증진하기 위해 교잡효율이 높은 Di-Mon 교배방법으로 부친의 핵과 모친의 미토콘드리아를 가진 세포질전환 교잡주를 선발하였다. 느타리버섯 수한3호와 흑변이체의 교잡종인 '다굴'의 이핵균주에 춘추2호의 단핵균주와 교잡하여 120여개의 교잡주를 얻고 여기에 1차로 미토콘드리아 microsatellite DNA 마커 (MtPO1)를 이용하여 단핵균주 춘추2호의 미토콘드리아 밴드를 형성하는 교잡주 80개를 1차 선발하였다. 선발된 80균주중에서 URP 프라이머를 이용하여 '다굴'의 2핵균주 핵 DNA 패턴을 가진 26교잡주를 선발하였다. 이 중에서 수량성이 우수한 3개 교잡주의 자실체 특성을 조사한 결과 초발이일수가 2일이나 빠르면서 수량성이 좋은 품종을 선발하여 '천화심'이라 명명하였다. 천화심은 수량이 높으면서 여름에도 재배가 가능해 춘추2호의 약점을 보완함으로써 춘추대체품종으로도 재배가 가능하리라 본다.

Possible Implication for an Indirect Interaction between Basic Fibroblast Growth Factor and (Na,K)ATPase

  • The (Na,K)ATPase is responsible for generating the ionic gradients and membrane potentials by the exchange of intracellular $Na^+$ for $K^+$. It has been recentl y shown that (Na,K)ATPase is involved in the exocytic pathway of basic fibroblast growth factor (bFGF), although it is not known that bFGF is secreted to the outside of cell through direct interaction with (Na,K) ATPase. To understand the role for (Na,K)ATPase in the secretary pathway of bFGF, we have sought to identify the cytoplasmic domains of the alpha1 isoform of (Na,K)ATPase interacting with bFGF by yeast two-hybrid system. We have also investigated the interaction between the alpha2 isoform of (Na,K)ATPase and bFGF to find out whether the interaction is isoform-specific. We found that none of the cytoplasmic domains of (Na,K)ATPase isoforms interacted with bFGF. The result suggests that the interaction between bFGF and (Na,K)ATPase might be indirect, thus requiring other proteins which are involved in the formation of protein complexes for the interaction, although we cannot exclude the possibility that the interaction requires the element of the whole alpha subunit structure that was not present in the isolated alpha subunit cytoplasmic domains.

  • 효모에는 여러 가지 종류의 thiol peroxid se 동위 효소들인 cytoplasmic TPx I, cTPx II, cTPx III, mitochodrial TPx (mTPx), 및 nuclear TPx (nTPx)가 존재하고 있다. 특히 cTPx II는 다른 효모TPx와 비교해 볼 때 매우 낮은 peroxidase 활성을 보이나, cTPx II를 제거한 cTPx II mutant균주는 심하게 성장이 저해되는 특징을 보인다. 본 연구에서는 효모에서의 cTPx II의 생리학적 기능을 밝히는 연구의 첫 과정으로 cTPx II와 상호 작용하는 단백질을 탐색하였다. Sacchromyces cerevisiae genomic DNA library에서 yeast two-hybrid system을 이용하여 cTPx II와 상호 결합하는 단백질을 탐색하여 그 단백질들의 기능을 연구하여, 궁극적으로 cTPx II의 생리기능을 밝히는데 이 연구의 목적을 두었다.

  • 우리나라 수도 품종을 배경으로 한 세포질적 유전적 웅성불임계통들에 우리나라 품종의 임성회복친을 교잡시킨 HR1619A/남풍벼, HR1619A/가야벼, HR1619A/계통234, 통일A/남풍벼, 통일A/청청벼, 수원296A/계통209, 수원296A/계통237, 수원296A/계통252 및 계통201A/계통234 등 9개 Hybrid를 육성하고 이들의 수량 및 수량 구성요소, 흰빛잎마름병 저항성 및 미질을 조사하여 다음과 같은 결과를 얻었다. 1 공시된 9개 Hybrid들 중 계통201A/계통234, 통일A/남풍벼 및 HR 1619A/남풍벼 등 3개 Hybrid의 수량성이 높았는데 각각 우수친에 비해 36%, 17% 및 10%의 증수(Heterobeltiosis)를, 표준품종인 청청벼에 비해 각각 19%, 17% 및 14%의 증수(Standard Heterosis)를 보였다. 2. 공시된 Hybrid중 HR1619A/가야벼와 수원296A/계통237의 수량은 양친보다 낮았다. 3. 수량구성요소 중 수당입수의 잡종강세가 현저하였고 주당수수, 천립중 및 임실율의 잡종강세는 인정되지 않았다. 4. Hybrid의 흰빛잎마름병 저항성은 양친 중 강한친과 같은 반응을 보였다. 5. Hybrid들의 Amylose 함량은 20.8%∼24.7%였고 딘백질함량은 9.3%∼10.0%, 알칼리 붕괴도는 3.7∼7.0으로서 각각 양친품종과 비슷하였다.

Sources of Resistance to Bacterial Wilt Found in Vietnam Collections of Pepper (Capsicum annuum) and Their Nuclear Fertility Restorer Genotypes for Cytoplasmic Male Sterility

  • Eleven hot pepper accessions collected in Vietnam showed stable resistance to bacterial wilt as well-known resistance sources, MC4 and MC5, in repeated inoculation tests with different Ralstonia solanacearum isolates conducted from 2004 to 2010. Seven of these accessions (specifically KC981, KC1006, KC1021, KC1027, KC1045, KC1050, and KC1055) resulted in stable male sterile F1 plants in the crosses with a cytoplasmically male sterile (CMS) Chilseong (CMS-A, Srfrf ), and therefore, they were maintainers (CMS-B) with a genotype of Nrfrf. The rest (KC980, KC995, KC999, and KC1009) produced stable male fertile F1 plants in the crosses, and therefore, were restorers (CMS-C) with a genotype of N(S)RfRf. Therefore, the maintainer and restorer sources of resistance may be used in preference in breeding maternal (CMS and their maintainers) and paternal parents (restorers) for resistance to bacterial wilt, respectively, in the hybrid breeding system utilizing cytoplasmic male sterility.

  • A $F_1$ hybrid out of 143 crosses of rape (Brassica napus) using cytoplasmic-genetic male sterility yielded 5.18 tons per hectare. European and native varieties used, were found out to have 78% and 58% of genes restoring fertility, respectively. The $F_1$ hybrids of three way cross using non-isogenic maintainer showed higher heteroses than those of double cross. To produce F 1 hybrids of three way cross, three stages of crossing operations are needed and seeds produced from about 60 hectares can be covered to 60, 000 hectares.
