Stability of Partial Nitrification and Microbial Population Dynamics in a Bioaugmented Membrane Bioreactor

  • Bioaugmentation of bioreactors focuses on the removal of numerous organics, with little attention typically paid to the maintenance of high and stable nitrite accumulation in partial nitrification. In this study, a bioaugmented membrane bioreactor (MBR) inoculated with enriched ammonia-oxidizing bacteria (AOB) was developed, and the effects of dissolved oxygen (DO) and temperature on the stability of partial nitrification and microbial community structure, in particular on the nitrifying community, were evaluated. The results showed that DO and temperature played the most important roles in the stability of partial nitrification in the bioaugmented MBR. The optimal operation conditions were found at 2-3 mgDO/l and $30^{\circ}C$, achieving 95% ammonia oxidization efficiency and nitrite ratio ($NO_2^-/{NO_x}^-$) of 0.95. High DO (5-6 mg/l) and low temperature ($20^{\circ}C$) had negative impacts on nitrite accumulation, leading to nitrite ratio drop to 0.6. However, the nitrite ratio achieved in the bioaugmented MBR was higher than that in most previous literatures. Denaturing gradient gel electrophoresis (DGGE) and fluorescence in situ hybridization (FISH) were used to provide an insight into the microbial community. It showed that Nitrosomonas-like species as the only detected AOB remained predominant in the bioaugmented MBR all the time, and coexisted with numerous heterotrophic bacteria. The heterotrophic bacteria responsible for mineralizing soluble microbial products (SMP) produced by nitrifiers belonged to the Cytophaga-Flavobacterium-Bacteroides (CFB) group, and $\alpha$-, $\beta$-, and $\gamma$- Proteobacteria. The fraction of AOB ranging from 77% to 54% was much higher than that of nitrite-oxidizing bacteria (0.4-0.9%), which might be the primary cause for the high and stable nitrite accumulation in the bioaugmented MBR.

Seasonal Variation of Eubacterial Community Structure and Their Structure Affecting Environmental Parameters in Reservoir (남매지에서 Eubacteria 군집구조의 계절적 변화와 그에 영향을 미치는 환경요인)

  • For elucidating the correlation between the eubacterial community structure and environmental parameters in Nammae Reservoir located in Kyungsan, Kyungbuk, the bacterial community structure and their structure affecting environmental parameters were analyzed using Fluorescent In Situ Hybridization (FISH) monthly over year. $\alpha$ . $\beta$ . $\gamma$-subclasses of Proteobacteria and Cytophaga-Flavobacterium (CF) group known as dominant bacterial group in freshwater were detected in 3 stations over year. The ratio of each subclass to total bacteria was determined; $\alpha$.$\beta$ . $\gamma$-subclasses and CF group varied in the range of 4.0~29.2%, 1.7~25.8%, 1.8~12.8%, 4.9~36.3%, respectively and there was no substantial differences between stations. In terms of the correlation between each group specific bacteria and environmental parameters such as temperature, SS, pH, DOC, NH$_4$-N, NO$_3$-N, PO$_4$-P, standing crops of algae, the results were as follows: 1) total bacterial numbers correlated positively with temperature, SS and DOC, 2) Eubacteria positively with DOC and Chl-$\alpha$, 3)${\gamma}$-subclass positively with DOC, and 4) CF group positively with standing crops of chlorophyceae, 5) whereas $\beta$-subclass bacteria correlated negatively with standing crop of cyanobacteria and that of total algae.

Changes in the Microflora of Marine Fishes during Storage by Partial Freezing (해산어(海産魚)의 부분동결(部分凍結)에 의한 Microflora의 변화(變化))

  • Marine fishes, sardine(Sardinops melanosticta), scad kingfish(Caranx equula), horse mackerel(Trachurus japonicus) and file fish(Navodon modestus), were stored for fifty days with partial freezing at$-3.5^{\circ}C$. During the storage, the changes in microflora and volatile nitrogen content was investigated. The fishes exhibited $10^4\;to\;10^6$ of bacterial cells per square centimeter of their skin just before they were submitted to the storage. The bacterial cell number was increased as $10^6\;to\;10^8$ cells as the storage time passed over twenty-two days. Offensive odor which is typical in the spoilage of fishes became strong as increase the bacterial cell numbe. The major isolates among the three hundred strains of bacteria isolated from the fish skins were identified as Pseudomonas I/II, III/IV-H, Vibrio and Moraxella. The same was found in the spoiled fishes, however, Pseudomonas I/II, was predominant on contrast to that of fresh fishes. Pseudomonas III/IV-NH, Flavobacterium, Cytophaga and Micrococcus were also found in early period of storage, but they disappeared as the progress of storage. Nine per cent of isolates were unidentified.

Seasonal Variations of Water Quality in the Lower Part of the Nagdong River (낙동강 하류수질의 계절적 변화)

  • The Nagdong is one of the biggest rivers in Korea, which is very important water source not only for tap water of Pusan city but also for the industrial water. Therefore, authors tried to check the water quality year by year. In this experiment one hundred and twenty water samples collected from August 1983 to July 1984 were analyzed bacteriologically and physiologically. Fifteen sampling stations were established between near Samrangjin and estuary of the river. To evaluate the water quality, temperature, pH, chloride ion, salinity, chemical oxygen demand (COD), electrical conductivity, nutrients, total coliform, fecal coliform, fecal streptococcus, viable cell count and bacterial flora were observed. The variation of water temperature was ranged $-1.5{\sim}29.0^{\circ}C$ (Mean value $13.9{\sim}16.5^{\circ}C$), it in spring was higher as $10{\sim}15^{\circ}C$ about $10^{\circ}C$ than in winter and it in autumm was very stabilized as about $20^{\circ}C$ at each station. The pH variation of the samples was ranged $6.68{\sim}9.15$. The range of concentration of chloride ion and salinity varied $7.4{\sim}l,020.5$ mg/l and $1.05{\sim}33.0\%0$, respectively. Especially, salinity of the 3rd water war was the higher than others as $25.76{\sim}31.58\%0$. COD was ranged $1.45{\sim}14.94$ mg/l and the lower part of the Nagdong River was heavily contaminated by domesitc sewage and waste water from the adjacent factor area. The range of electrical conductivity was $1.360{\times}10^2{\sim}5.650{\times}10^4{\mu}{\mho}/cm$ and that was by far higher the estuary than the upper. Concentration of nutrients were $0.008{\sim}0.040$ mg/l (Mean value $0.019{\sim}0.068$ mg/l) for $NO_2-N,\;0.038{\sim}5.253$ mg/l ($0.351{\sim}2.347$ mg/l) for $NO_3-N,\;0.100{\sim}2.685$ mg/l($0.117{\sim}1.380$ mg/l) for $NH_4-N,\;0.003{\sim}0.084$ mg/l($0.014{\sim}0.065$ mg/l) for $PO_4-P$ and $0.154{\sim}6.123$ mg/l ($1.165{\sim}3.972$ mg/l) for $SiO_2-Si$, respectively. Usually nutrients contents of the water in the upper part(included station 1 to 5) were higher than those of the estuarine area. The bacterial density of the samples ranged 7.3 to 460,000/100 ml for total coliforms, 3.6 to 460,000/100 ml for fecal coliform, $0{\sim}46,000/100ml$ for fecal streptococcus and $<30{\sim}1.2{\times}10^5/ml$ for viable cell count. Composition of coliform was $28\%$ Escherichia coli group, $18\%$ Citrobacter freundii group, $31\%$ Enterobacter aerogenes group and $22\%$ others. Predominant species among the 659 strains isolated from the samples were Pseudomonas spp. ($42\%$), Flavobacterium spp. ($20\%$) and Moraxella spp. ($12\%$).
