Ankyrins are a ubiquitously expressed family of intracellular adaptor proteins involved in targeting diverse proteins to specialized membrane domains in both the plasma membrane and the endoplasmic reticulum. Recently, the studies with C-terminus of ankyrins have identified that ankyrin-B is capable of interacting with Hsp40 and sAnkl is capable of interacting with obscurin and titin, but the function of C-terminal domain of ankyrin-G remains unknown. To identify proteins interacting C-terminus of ankyrin-G, we used the C-terminus of ankyrin-G as a bait for a yeast two-hybrid screen of brain cDNA library. Approximately 1.33$\times$l0$^6$ transformants were screened, of which 13 positive clones were obtained as determined by activation of HIS3, ADE2 and MELl reporter genes. Sequence analyses of these 13 plasmids revealed that cDNA inserts of 13 colonies showed highly homologous to 11 genes, including 5 known (i.e., Na$^+$/K$^+$ ATPase $\beta$1, SERBPl, UTF2, cytochrome C oxidase and collagen IV $\alpha$2) and 6 unknown genes. The evaluation of the proteins that emerge from these experiments provides a rational approach to investigate the those proteins significant in interaction with ankyrin-G.
버섯파리는 국내 양송이 균상 재배지에서 발생하여 버섯 균사와 자실체를 가해하고 간접적으로는 병을 매개하여 피해는 해마다 증가하고 있다. 부여군 세도면, 보령시 성주면, 용인시 남사면, 칠곡군 약목면 등 총 4개 지역의 버섯 재배사에서 끈끈이트랩을 이용하여 채집한 버섯파리 성충을 DNA barcoding 방법으로 Cytochrome Oxidase I (COI)의 염기서열 분석을 통해 종 동정을 실시하였다. 그 결과 4개 조사지역의 양송이에서 발생하는 버섯파리는 모두 긴수염버섯파리(Lycoriella ingenua)로 동정되었다.
Gammarus gageoensis n. sp., a new species of freshwater gammarid, was collected from Gageodo Island, South Korea. This new species is characterized in terms of its morphology by the flagellum of antenna 2 with calceoli, the pereopods 3 and 4 with numerous long curled setae on the posterior margins, the terminal article of the outer ramus in uropod 3, which is longer than the adjacent spines, and the presence of both rami with long simple setae. The morphological characters of the new species that are different to related species are presented in the text. This paper also compares the new species to related species by the partial sequence of the mitochondrial cytochrome c oxidase subunit 1 (CO1) gene. The sequence difference, consisting of a 39-44% difference relative to closely related species, is also large enough to designate this gammarid as a different species from other freshwater gammarid species. This is the first finding of a new species of Korean freshwater gammarid since 1992. Including this new species, a total of 10 species are known to Korea.
Interspecific hybrids between tiger grouper Epinephelus fuscoguttatus and giant grouper E. lanceolatus were genetically identified based on the partial sequence analysis of mitochondrial cytochrome c oxidase I (COX I) gene and nuclear recombination activating gene 2 (RAG 2) gene. Out of 585 base positions of RAG 2, a total of five nucleotide substitutions were detected between the two parental species (E. fuscoguttatus and E. lanceolatus). The hybrids had two distinct types of RAG 2 sequences corresponding to those of both parental species. Mitochondrial COX I gene sequencing showed that hybrids had sequences identical to E. fuscoguttatus. Molecular data clearly demonstrate that hybridization does occur between E. fuscoguttatus and E. lanceolatus, but with E. fuscoguttatus as the maternal parent.
Diphyllobothrium latum and Diphyllobothrium nihonkaiense are the 2 reported main causes of human diphyllobothriasis in the Republic of Korea. However, the differentiation of these 2 species based on morphologic features alone is difficult. The authors used nucleotide sequencing of the mitochondrial cytochrome c oxidase subunit 1 (cox1) gene to diagnose Diphyllobothrium spp. Two patients visited the emergency room at Kyungpook National University Hospital on 3 April and 12 April 2013, respectively, with fragments of parasites found while defecating. The parasites were identified as Diphyllobothrium spp. based on morphologic characteristics, and subsequent cox1 gene sequencing showed 99.9% similarity (1,478/1,480 bp) with D. nihonkaiense. Our findings support the hypothesis that D. nihonkaiense is a dominant species in Korea.
Objectives : Parkinson's disease, a progressive neurodegenerative disease, is caused by the loss of dopaminergic neurons in the substantia nigra. There is no clear treatment or remedy for Parkinson's disease; therefore, the development of novel therapies related to anti-inflammatory and antioxidant effects is required. This study was performed to evaluate the neuroprotective effect of water extracts from Cervi Cornu (CC) in dopaminergic cells. Methods : We studied effects of CC on apoptosis, cell death and inflammation in SH-SY5Y neuroblastoma cells treated by methylpyridinium ion (MPP+). SH-SY5Y cell line was treated with CC for 24 hours and then 500 μM MPP+ for 18 hours. Results : Cervi Cornu treatment inhibited the decrease in tyrosine hydroxylase (TH) expression and decreased the activation of inflammatory factors mitochondrial cytochrome C oxidase (COX2) and inducible NO synthase (iNOS) against MPP+ neurotoxicity. Apoptosis factors BCL2 associated X, apoptosis regulator (BAX) levels were decreased and B-Cell CLL/Lymphoma 2 (BCL2) levels were increased. Conclusions : These results suggest that CC treatment had neuroprotective effects in the SH-SY5Y neuroblastoma cells against toxicity induced by MPP+. The results suggest new possibilities of CC for the treatment of Parkinson's disease.
In the northwestern Pacific region, the pen shell (Atrina pectinata) is a widely distributed bivalve and economically important in fisheries. Recently, stock of this species has been greatly reduced due to overexploitation and marine pollution, which arouses interest in conservation. Studies on genetic and taxonomic entities of pen shells have not been tried in Korea, which makes difficult to take measures for effective conservation of this marine resource. In this study, we investigated mitochondrial genetic polymorphism of pen shells collected from 4 locations in Korea and Japan using cytochrome c oxidase I (COI) gene sequences. A total of 39 haplotypes were identified among 86 individuals of pen shell. Although only 5 haplotypes were shared, no significant genetic differentiation was observed between Korean and Japanese populations. These results suggest that pen shell populations of these regions share an ancestral population which might have experienced expansion during the Pleistocene, but gene flow must have been highly restricted after expansion.

Alaria is the second largest genus of the Laminariales, which is distributed far into the northern Pacific and Atlantic oceans. Due to its high morphological plasticity, over 100 specific and sub-specific names have been used in Alaria, this has been tailored down to the present 17 species through morphological revision and molecular phylogenetic analysis. Endemic species of Alaria from Russian Far East have not been thoroughly revised since their original description, and few of them were confirmed using molecular data until recently. We carried out morphological and molecular studies on A. paradisea which is an endemic species distributed on the Kurile Islands, first described by Miyabe and Nagai in 1932 as Pleuropterum paradiseum. The range of morphological variability and its distribution was re-evaluated using the type specimen as well as other specimens. Analyses of partial mitochondrial cytochrome c oxidase subunit 1 and nuclear-encoded internal transcribed spacer sequences showed that A. paradisea nested within the genus Alaria, but differs morphologically from any other Alaria species in having additional sporophylls with a central midrib (${\beta}-sporophylls$). Our results showed that A. paradisea clearly belongs to the genus Alaria based on DNA data, although the key morphological character that was used to include this species to the genus Pleuropterum, ${\beta}-sporophylls$, is stable and distinguishes it from other Alaria species.
A mutant library of Cordyceps militaris was constructed by improved Agrobacterium tumefaciens-mediated transformation and screened for degradation features. Six mutants with altered characters in in vitro and in vivo fruiting body production, and cordycepin formation were found to contain a single copy T-DNA. T-DNA flanking sequences of these mutants were identified by thermal asymmetric interlaced-PCR approach. ATP-dependent helicase, cytochrome oxidase subunit I and ubiquitin-like activating enzyme were involved in in vitro fruiting body production, serine/threonine phosphatase involved in in vivo fruiting body production, while glucose-methanol-choline oxidoreductase and telomerase reverse transcriptase involved in cordycepin formation. These genes were analyzed by bioinformatics methods, and their molecular function and biology process were speculated by Gene Ontology (GO) analysis. The results provided useful information for the control of culture degeneration in commercial production of C. militaris.
Formic acid is a representative carboxylic acid that inhibits bacterial cell growth, and thus it is generally considered to constitute an obstacle to the reuse of renewable biomass. In this study, Saccharomyces cerevisiae was used to elucidate changes in protein levels in response to formic acid. Fifty-seven differentially expressed proteins in response to formic acid toxicity in S. cerevisiae were identified by 1D-PAGE and nano-liquid chromatography-tandem mass spectrometry (nano-LC-MS/MS) analyses. Among the 28 proteins increased in expression, four were involved in the MAP kinase signal transduction pathway and one in the oxidative stress-induced pathway. A dramatic increase was observed in the number of ion transporters related to maintenance of acid-base balance. Regarding the 29 proteins decreased in expression, they were found to participate in transcription during cell division. Heat shock protein 70, glutathione reductase, and cytochrome c oxidase were measured by LC-MS/MS analysis. Taken together, the inhibitory action of formic acid on S. cerevisiae cells might disrupt the acidbase balance across the cell membrane and generate oxidative stress, leading to repressed cell division and death. S. cerevisiae also induced expression of ion transporters, which may be required to maintain the acid-base balance when yeast cells are exposed to high concentrations of formic acid in growth medium.
