Physiological gradient of IAA-oxidizing enzyme activities was investigated in order to elucidate the mechanism of shoot differentiation in Cymbidium sp. (‘Jungfrau’) protocorms by using phenolic compounds (2, 4-dichlorophenol, catechol), auxin-inhibitors (PCIB, TIBA), and hormones (GA3, ABA, BA). The activity of IAA oxidase was decreased in protocorms treated with catechol decreased the catalytic activity of IAA oxidase or TIBA but this enzyme activity was increased after a temporary decrease at initial stages in the presence of 2, 4-dichlorophenol or PCIB. The activity of IAA oxidase in BA-treated protocorms (white and crown gall-like) was the highest of all. However, the catalytic activity of peroxidase increased after a temporary decrease at initial period. These results suggest that shoot differentiation and growth may be influenced by effective IAA levels in the protocorms causing IAA-oxidizing enzyme and phenolic compounds.
Changes in starch content and activities of ADPG- and UDPG-starch synthase and $\alpha$- and, $\beta$-amylase were studied in order to investigate effects of gibberellic aicd and abscisic acid on endogenous starch content during shoot differentiation and protocorm propagation in Cymbidium spp. (Jungfrau) protocorm. Shoot differentiation was promoted during the degradation of endogenous starch and protocorn propagation was promoted during starch accumulation in protocorm. The activities of ADPG- and, UDPG-starch synthase and $\alpha$- and $\beta$-amylase seemed to be related with starch content. Shoot differentiation and protocorm propagation were slightly inhibited in protocorm explants treated with 100$\mu$M gibberellic acid. The explants treated with 10$\mu$M abscisic acid lost the capacity for shoot differentiation and protocorm propagation, and that could not be overcome by 100$\mu$M gibberellic acid added to culture medium. Starch content fluctuated as the control even after 10$\mu$M abscisic acid. None the less, the treatment completely inhibited shoot differentiation and protocorm propagation.
This study was carried out to detect cymbidium mosaic potexvirus (CymMV) and odontoglossum ringspot tobamovirus (ORSV) in cultivated orchid plants in Korea. The standard double antibody sandwich enzyme-linked immunosorbent assay (ELISA) and reverse transcription polymerase chain reaction (RT-PCR) were carried out for detection of the viruses in the collected orchid samples. ELISA was suitable for massive-scale diagnostic method for virus detection in orchids. RT-PCR was rapid, time-saving and reliable detective method, and detection limit data showed that RT-PCR was 103 times more sensitive than ELISA. Of the 321 individual orchids representing 5 orchids genera tested by the ELISA, CymMV and ORSV were detected in 15.6% and 22.4%, and mixed infection of the both viruses with 4.9%, respectively. Of the Cymbidium plants tested, cultivated plants showed 52.5% virus infection rate with either CymMV or ORSV and both viruses.
Changes in polamine titers during shoot differentiation in Cymbidium sp. (Jungfrau) protocorms were studied in order to investigate the mechanism of shoot differentiation by using auxin-inhibitors(PCIB, TIBA), hormones(GA3, ABA, BA), and phenolic compounds (2,4-dichlorophenol, catechol). The shoot differentiation and propagation of protocorms were promoted by PCIB or 2,4-dichlorophenol, and the growth of differentiated shoot were promoted by TIBA or catechol. In BA-treated protocorms, white or brown protocorms were observed. Putrescine was the most abundant polyamine during the propagation and differentiation processes. As compared with putrescine, spermidine did not show significant changes and spermine was not detected at all. Putrescine titers decreased after a temporary increase, and then again increased in the presence of GA3, ABA, 2,4-dichlorophenol, and then again increased in the presence of GA3, ABA, 2,4-dichlorophenol, catechol, or PCIB. But, in BA-treated protocorms, putrescine level was much lower than spermidine.
Orchids have been propagated vegetatively for a long time without adequate control measures against virus diseases in Korea. As a result, it is presumed that most of the orchid varieties in Korea may have been degenerated. Nevertheless there has been little work on the virus diseases of orchids in Korea. Therefore studies were initiated to isolate an4 characterize the orchid viruses occurring in Korea. The results obtained are summerized as follows. 1. Symptoms of virus diseases on orchid varieties can be grouped 1) mosaic, 2) necrotic streak with mosaic, 3) ring necrosis, 4) chlorotkc ring and 5) necrotic spot. 2. A total of 102 orchid plants representing 4 genera were investigated on the occurrence of Cymbidium mosaic virus and tobacco mosaic virus by serological agar-gel double diffusion test. The test revealed that approximately $45\%$ of the orchids were infected with Cymbidium mosaic virus. None of the plants were found to be infected with tobacco mosaic virus. 3. Local lesions appeared on the inoculated leaves of Chenopodium amaranticolor Cassia occidentalis and Datura stramonium 7-12 days after mechanical inoculation with Cymbidium mosaic virus. 4. Physical properties of the Cymbidium mosaic virus determined by inoculation on Chenopodium amaranticolor were as follows: Thermal inactivation Point; $75-80^{\circ}C$, dilution end Point; $10^{-5}-10^{-6}\%$ aging in vitro; 8 days. 5. Three different buffers at pH 7.0 and pH 9.0 were compared for the efficiency of agar-gel double diffusion test with Cymbidium mosaic virus. Phosphate, imidazol and tris buffer at pH 7.0 gave equally satisfactory results. 6. Electron microscopic examination of the Cymbidium mosaic virus revealed rod shaped particles measuring 460-580mu.
This text was analyzed and investigated the vegetation and floristic composition by ordination and classification of phytosociological method, to evaluate the species composition and community structure of Cymbidium goeringii group in Korean islets. In habitat of 33 plots, the mean altitude was 65.9m, the direction was the southeast slope, the mean slope was 7.9%. The coverage of Cymbidium goeringii was 4.5%. The appearing plants with the Cymbidium goeringii was the total 102 taxa, and it was the kind of trees 68 taxa (66.7%), herbs 34 taxa (33.3%), evergreen plants 36 taxa (35.3%) and deciduous plants 66 taxa (64.7 %) respectively. The frequency of appearing plant was the highest in the Eurya japonica (48.5%), followed by Pinus thunbergii (45.5%), Smilax china (36.4%), Carex lanceolata (33.3%), Hedera rhombea (33.3%), Machilus thunbergii (30.0%), Styrax japonicus (30.3%) and Pinus densiflora (27.3%), respectively. The vegetation of tree layer in Cymbidium goeringii group was classified into Pinus thunbergii community, Pinus densiflora community, Castanopsis sieboldii community and Quercus variabilis community. Pinus densiflora community showed a strong combination with Cymbidium goeringii group in Korean islets. Pinus thunbergii community among communities was combined with Castanopsis sieboldii community, and Pinus densiflora community and Quercus variabilis community were combined.
In ultrathin section of the tissue of odontoglossum ringspot virus (ORSV)-infected Cymbidium goeringii Reichenbach, ORSV particles appeared as bundles of irregular aggregates of various length which were called stacked plates or rounded plates. Virus particles were found in the cytoplasm in electron clear zones and they also found between cell wall and plasma membrane. They mainly clustered in parallel aggregates and sometimes oriented randomly. The X-bodies and paramural bodies were observed near the cell membrane and these contained vacuole-like cavities. The cell wall of infected tissue expanded largely. Some chloroplast in ORSV infected cell was irregular. No virus particle was present in mitochondria, nuclei, vacuoles, vesicles or other organelles. The plasmodesmata slightly enlarged, and virus-associated granules were present around it.

Orchid fleck virus (OFV) is an unassigned plant virus in the family Rhabdoviridae. OFV was isolated from Cymbidium sp. showing oval necrotic lesions on their leaves in Korea, and designated as OFV-NHHS1. The complete nucleotide sequence of the RNA1 (6,413 nt) (GenBank accession no. AB516442) and RNA2 (6,001 nt) (GenBank accession no. AB516441) was determined in this study. RNA1 and RNA2 contained five and one ORF respectively. RNA1 encodes nucleocapsid (N) of 49 kDa, ORF2 of 26 kDa, ORF3 of 38 kDa, ORF4 of 20 kDa and glycoprotein (G) of 61 kDa proteins, whereas RNA2 encodes a single polymerase of 212 kDa. OFV-NHHS1 shared extremely high similarity of 98.6-100% and 98.9-99.6% in nucleotidle and amino acid sequences with a Japanese isolate, OFV-so, respectively. However, the N, G and L of OFV-NHHS1 revealed 6.9-19.3%, 7.3-12.0%, and 13.4-26.6% identities to those of 29 Rhabdoviruses, respectively. To survey the infection rate of OFV in commercial orchids in Korea, 51 Cymbidium sp., 10 Phalaenopsis sp., 22 Oncidium sp. and 21 Dendrobium sp. plants that showed typical viral symptoms were collected. RT-PCR with specific primers for detection of Cymbidium mosaic virus (CymMV), ORSV and OFV showed high infection rate by ORSV alone and double infection by ORSV and CymMV. One of the orchids tested was infected with OFV. This is the first report of the complete nucleotide sequences of OFV isolated in Korea.
Cymbidium goeringii REICHB. fil was extracted with 80% MeOH and solvent-fractionated with EtOAc, n-BuOH and $H_2O$, successively. From EtOAc fraction, three sterols were isolated on repeated silica gel and ODS column chromatographies. The chemical structures were determined as ${\beta}-sitosterol$ (1), daucosterol (2) and ergosterol peroxide (3) by NMR, MS and IR, which is the first to be isolated from Cymbidium goeringii REICHB. fil.
Effects of plant growth regulators and anti-oxidants for rapid multiplication of Cymbidium kanran were investigated. The best gelling agent was 2.5 g/1 gelrite which needed less quantity (about 28%) and half price than 9 g/1 chemi-cal agar. Undefined edible agar was only a little bit worse than chemical agar in growth, but the price was half as much as the latter. The higher concentration of BA and NAA, the deeper browning of medium that prevented from performing its functions of plant growth regulators. Polyvinylpyrrolidone (M.W. 40,000) was the most effective anti-oxidant other than ascorbic acid, aspartic acid, and rutin in protecting the browning of medium, enhancing the effect of plant growth regulators, and thus prolonging the subculture cycle. Vigorous seedlings were obtained by 0.1∼1.0 mg/1 BA,0.1 mg/1 NAA and 1 g/1 polyvinylpyrrolidone treatments. Therefore, the best result for growth and econo-mic aspects in rhizome culture of Cymbidium kanran were obtained by using MS basal medium with 2.5 g/1 gelrite, 1 g/1 polyvinylpyrrolidone, 0.1∼1.0 mg/1 BA and 0.1 mg/1 NAA.
