The triclosan was shown to have anti-microbial and anti-inflammatory effect with inhibition of inflammatory mediators such as prostaglandin $E_2(PGE_2)$. The purpose of this study was to elucidate whether and how $PGE_2$ could be inhibited by triclosan in human gingival fibroblast. Human gingival fibroblast-1 cells (ATCC CRL2014) were pre-treated for 1 hour with triclosan (0.001 ${\mu}/ml{\sim}10$${\mu}/ml$) and then stimulated with $TNF-{\alpha}$ (1.0 ng/ml). $PGE_2$ synthesis was evaluated by ELISA and gene expression of COX-1 and COX-2 was evaluated by RT-PCR after $TNF-{\alpha}$, triclosan, and NS-398 (COX-2 inhibitor, 5, ${\mu}M$) and/ or cycloheximide (protein synthesis inhibitor, 2 ${\mu}g/ml$). Triclosan was cytotoxic to human gingival fibroblasts in the concentration higher than 1.0 ${\mu}g/ml$ for longer than 24 hours in tissue culture. The $PGE_2$ synthesis was inhibited by triclosan in dose-dependent manner. Greater COX-2 mRNA suppression was observed with triclosan (0.1 ${\mu}g/ml$) than with $TNF-{\alpha}$ alone, without change in COX-1 gene expression. Inhibitory effects of triclosan on $PGE_2$ synthesis disappeared in presence of cycloheximide. This study suggests that triclosan inhibit prostaglandin $E_2$ at the level of COX-2 gene regulation and require de novo protein synthesis.

As a step toward a more complete understanding of the molecular actions of TNF, we prepared a cDNA library from TNF-treated human FS-4 fibroblasts and used differential hybridization to identify cDNA clones corresponding to mRNAs enriched in TNF-treated eells. In Quiescent FS-4 cells n induces an increase in the level of some mRNAs within 20 to 30 min. Some of these immediate-early response mRNAs are elevated only transiently for about 30 to 120 min, e. g., c-fos and c-myc (Lin and Vilcek,1987) or the transcription factor IRF-1 (Fujita et al.1989). Such immediate-early gene products may be important for the activation of other genes, but their transient induction suggests that they are not the actual effector molecules responsible for the phenotypic changes induced by TNF. We chose a 3-h incubation with W because we were seeking cDNAs corresponding to messages that are more stably elevated after TNF treatment. Indeed, the results shown in Figure 8 and 9 indicate that all of the mRNAs corresponding to the eight TSG cDNAs isolated remained significantly elevated after 16h of continuous treatment with TNF, and their kinetics of induction were clearly different from those of the immediate-early response mRNAs such as c-fos, c-myc or IRF-1. Nevertheless, only the induction of TSG-21 (collagenase) and TSG-27 (stromelysin) nNAs was completely inhibited by cycloheximide and the induction of TSG-37 (metallothionein-II) was reduced in the presence of this inhibitor of protein synthesis. Induction of the other five TSG mRNAs by TNF was completelyresistant to cycloheximide, suggest ins that no protein intermediate is needed for the upregulation of these mRNAs.
Isolates of the different species groups of Trichoderma from the mushroom culture beds were identified according to Rifai's classification and influence of antibiotics produced by them against the oyster mushroom was examined. Trichoderma islolates were identified as Trichoderma hamatum, Trichoderma viride and Trichoderma koningii. Among the Trichoderma isolates, fungistatic action of Trichoderma viride was found to be most remarkable. Pleurotus ostreatus and Pleurotus sajor-caju were the most susceptable of the edible mushrooms tested, followed by Lentinus edodes, Flammulina velutipes and Auricularia auricula. A needle-shaped crystal gained from the chloroform extract of the culture filtrate of Trichoderma viride repressed distinctively the mycelial growth of the oyster mushroom. The grade of repression of the crystal at 500ppm and 1/10 aequous solution of the chloroform extract against the oyster mushroom, seemed equal to that of cycloheximide at $100{\sim}200ppm$.
The apoptosis-inducing activity of ursolic acid (UA) was examined in mouse F9 teratocarcinoma cells on the bases of biochemical and morphological characteeristics. UA, pentacyclic trierpene acid, exhibits antitumor activities including inhibition of skin tumorigenesis, induction of tumor cell differentiation and antitumor promotion. Treatment with UA showed that the decrease of cell viability was dose-dependent. UA also induced genomic DNA fragmetation, a hallmark of apoptosis, indicating that the mechanism of UA-induced F9 cell death was through apoptosis. When the morphology of the F9 cells was examined by electron microscopy, the cells treated with UA showed the charcteristic morphological features of apoptosis such as chromatin condensation and nuclear fragmentation. DNA fragmentations by UA were inhibired by cycloheximide, which suggest that de novo protein synthesis was required for DNA fragmentation by UA. Inaddition, the expression of c-jun was increased, but those of c-myc and laminin B1 were decreased during apoptosis induced by UA in F9 cells. These results suggest that UA causes an apoptosis in F9 cells. Further, the increased expression of c-jun may be involved in the UA-induced apoptosis of f9 cells.

Auxotrophs and drug resistant mutants from the mycelia of Lentinus edodes were obtained by UV irradiation at survival rates of $0.024{\sim}2.45%$ and ethidium bromide (EtBr) enrichment after UV irradiation. The mutation rate was 0.40%, and back mutation rate was $4.81{\times}10^{-4}{\sim}8.46{\times}10^{-4}$. Various amino acid-, nucleic acid-, and vitamin-requiring auxotrophs were isolated. The concentrations of several fungicides, antibiotics and amino acid analogues inhibiting the growth of L. edodes were determined. The MIC values for cycloheximide, benomyl, and p-fluorophenylalanine were 2, 2000, and 1000 ug/ml respectively. Five p-fluorophenylalanine-resistant mutants and eight benomyl-resistant mutants were selected by UV irradiation.
In rat hippocampus, kainic acid (KA; 10 mg/kg; i.p.) increased the phosphorylated forms of ERK1/2 (p-ERK1/2) and Jun kinase1 (p-JNK1), but not p-JNK2 and p38 (p-p38). The preadministration with cycloheximide (CHX; 5 mg/kg; i.p.) inhibited KA-induced increase of p-JNK1, but not p-ERK1/2. Surprisingly, the phosphorylated upstream MAP kinase kinases (p-MKKs) were not correlated with their downstream MAP kinases. The basal p-MKK1/2 levels were completely abolished by KA, which were reversed by CHX. In addition, p-MKK4 and p-MKK3/6 levels were enhanced by CHX alone, but were attenuated by KA. Thus, our results showed that KA increased the p-ERK and p-JNK levels in rat hippocampus, which were not parallel with their classical upstreamal kinases.
Aspergillus niger van Tieghem was cultured by the method of synchronous and submerged culture. Throughout the culture, sporulation was occured. Highly phosphorylated nucleotides in sporulating mycelia were detected to assure whether the eucaryotic Aspergillus niger produce these substances or not during the differentiation. Phosphorylated nucleotides were extracted from the conidiophore, bearing mycelia and spore forming body, these nucleotides were identified by TLC with P.E.I. cellulose. Guanosine tetraphosphate was found in both phialide forming mycelia and spore forming body. The contents of free amino acids were assayed and its level was found to increase at early stage of sporulation. The effects of 8-azaguanine examined, it was found to prevent spore formation and to made abnormal structure. The effects of inosinic monophosphate and guanosine monophosphate on spore formation were examined, spore formation was enhanced by these nucleotides.
This study was to evaluate the in vitro survival of bovine enucleated MII (eMII) oocytes according to minimum volume cooling (MVC) freezing method and activation timing, and their in vitro development after somatic cell nuclear transfer (SONT). in vitro matured bovine oocytes for 20 h were stained with 5 $\mu\textrm{g}$/$m\ell$ Hoechst, and their 1st polar body and MII plate were removed by enucleation micropipette under UV filter. Also, eMII oocytes were subjected to activation after (group II) and before (group III) vitrification in 5 ${\mu}{\textrm}{m}$ ionomycin added CRlaa medium for 5 min. For vitrification, eMll oocytes were pretreated with EG10 for 5 min, exposed to EG30 for 30 sec and then directly plunged into L$N_2$. Thawing was taken by 4-step procedures at 37$^{\circ}C$. Survived eMII oocytes were subjected to SONT with cultured adult bovine ear cells. Reconstructed oocytes were cultured in 10 $\mu\textrm{g}$/$m\ell$ of cycloheximide and 2.5 $\mu\textrm{g}$/$m\ell$ of cytochalasin D added CRlaa medium for 1 h, and then in 10 $\mu\textrm{g}$/$m\ell$ of cycloheximide added CRlaa medium for 4 h. Subsequently, the reconstructed oocytes were incubated for 2 days and cleaved embryos were further cultured on cumulus-cell monolayer drop in CRlaa medium for 6 days. Survival rates of bovine vitrified-thawed eMII oocytes in group II (activation after vitrification and thawing) and III (activation before vitrification) were 81.0% and 84.9%, respectively. Fusion rates of cytoplasts and oocytes in group II and III were 69.0% and 70.0%, respectively, and their results were not different with non-frozen NT group (control, 75.2%). Although their cleaved rates (53.4% and 58.4%) were not different, cytoplasmic fragment rate in group II (32.8%) was significantly higher than that in group III (15.6%)(P<0.05). Also, subsequent development rate into >morula in group II (8.6%) was low than that in group III(15.6%). However, in vitro development rate in group III was not different with that in control (24.8%). This result suggested that MVC method was appropriate freezing method for the bovine eMII oocytes and vitrified eMII oocytes after pre-activation could support in vitro embryonic development after SONT as equally well as fresh oocytes.
The roles of RNA- and protein-synthesis and $H^{+}$ excretion in 1AA ($10\;\mu\textrm{M}$)-induced elongation were investigated using abraded hypocotyl segments of sunflower (Helianthus annuus L.). The response of elongation initiated about 13 min after IAA treatment. Removal of cuticle, acting as diffusion barrier for inhibitors, by mechanical abrasion of hypocotyl segments enhanced the effect of inhibitors markedly, but the degree of abrasion for the saturated effect of inhibition was different among inhibitors. The elongation induced by 1M was completely inhibited when cycloheximide ($10\;\mu\textrm{M}$) was applied to abraded hypocotyl segments as shortly as 4 min before the onset of the growth response (= 10 min after administration of IAA). Cordycepin ($200\;\mu\textrm{M}$) prevented completely 1AA-induced elongation when applied as shortly as 19 min before the onset of the growth response (=5 min before administration of 1AA). Vanadate (1 mM) inhibited both lAA-induced elongation and medium acidification via lAA-induced $H^{+}$ excretion to apoplast. Cycloheximide and cordycepin also prevented lAA-induced $H^{+}$ excretion strongly. However, inhibition by cycloheximide of lAA-induced elongation was not alleviated by acidifying the cell wall to pH 4.5. The results indicate that, a few minutes before the initiation of growih, protein synthesis is demanded for the initiation of 1AA-induced elongation and the $H^{+}$ excretion to cell wall, and that the H+ excretion, even though it may be necessary for elongation, does not seem to bring about acid growth simply through acidifying cell wall.l wall.
Background : Tumor necrosis factor(TNF) has been considered as an important candidate for cancer gene therapy based on its potent anti-tumor activity. However, since the efficiency of current techniques of gene transfer is not satisfactory, the majorities of current protocols is aiming the in vitro gene transfer to cancer cells and re-introducing genetically modified cancer cells to hoot. In previous study, it was shown that TNF-sensitive cancer cells transfected with TNF-$\alpha$ cDNA would become highly resistant to TNF. Understanding the mechanisms of TNF-resistance in TNF-$\alpha$ gene transfected cancer cells would be an important step for improving the efficacy of cancer gene therapy as well as for better understandings of tumor biology. This study was designed to evaluate the role of new protective protein synthesis in the acquired resistance to TNF of TNF-$\alpha$ gene transfected cancer cells. Method : We transfected TNF-$\alpha$ c-DNA to WEHI164, a murine fibrosarcoma cell line, using retroviral vector(pLT12SN(TNF)) and confirm the expression of TNF with PCR, ELISA, MIT assay. Then we determined the TNF resistance of TNF gene transfected cells(WEHI164-TNF) and the changes of TNF sensitivities after treatments with actinomycin D(transcription inhibitor) and cycloheximide ( translation inhibitor). Results : WEHI164 which was sensitive to TNF became resistant to TNF after being transfected with TNF-$\alpha$ gene and the resistance to TNF was partially reversed after treatment with actinomycin D, but not with cycloheximide. Conclusion : The acquired resistance to TNF after TNF-$\alpha$ gene transfection may be associated with synthesis of some protective proteins.
