Dynamics of Bacterial Communities Analyzed by DGGE during Cyanobacterial Bloom in Daechung Reservoir, Korea (대청호 수화발생시기의 미생물 다양성 및 계통분류학적 분석)

  • The change of microbial communities during cyanobacterial bloom was comparatively analyzed by 16S rDNA PCR-DGGE in Daechung Reservoir during 2003~2005. Morphological analysis showed that Cyanophyceae dominated algal community in the bloom. Dominant cyanobacteria were Microcystis, Planktothrix (Oscillatoria), Phormidium and Anabaena. We used 16S rDNA-denaturing gradient gel electrophoresis (DGGE) profiles and phylogenetic affiliations of the DGGE bands to analyze the community structure and diversity of the predominant microbial community. The DGGE band patterns demonstrated that the most frequent bands were identified as Microcystis during the monitoring periods, Planktothrix also dominated on September 2003 and 2004, whereas Anabaena was showed a peak on September 2005 and Aphanizomenon on August 2003. DGGE and phylogenetic analysis provided us new information that could not be obtained by traditional, morphological analysis. The relationship between cyanobacteria and other aquatic bacteria can be traced and their genetic diversity also identified in detail.

Characteristics of Algae Occurrence in Lake Paldang (팔당호의 조류 발생 특성)

  • This study was performed to evaluate characteristics of algal occurrence in Lake Paldang. Chlorophyll a concentration was highest during spring, based on weekly measurements from 2006 to 2008, which revealed a different trend compared to the data before 2000 that showed the highest level during summer. Total algae cell quantity was also highest during spring, based on monthly data. Overall, Bacillariophyceae was dominant and represented 87% of total algae cells and occupation was more significant during spring and winter. Cyanophyceae was dominant during summer. 2-MIB and geosmin originating in algae which caused odor problems was high in July 2008, and more research on the relationship between algae and production of odor causing material is needed.

Phytoplankton Community, Pollution Sources and Water Quality of Drinking Water Lakes in Ulsan (울산지역 상수원 호수 환경에 따른 식물플랑크톤 분포)

  • This study presented the phytoplankton communities of the three lakes (Sayeon, Daeam, Hoeya ) using for drinking water in the Ulsan reservoir. The water storage of the Lake Sayeon, Daeam and Hoeya were 25, 13, 21 million ton respectively and most of which were being utilized for industrial and residential purposes. The total precipitation of the Ulsan region in 2010 was 1,162 mm, decreasing 10 % from 1,275 mm of the annual. As for pollutant loads, BOD and TN discharge loads of Daeam was the highest with 3,277 kg/day, 1,931 kg/day and 90 % of them were came from non-point pollutant sources. TP discharge loads showed the highest in the lake Hoeya with 643 kg/day and 97 % of them were came from point sources as household, industry and livestock. We assessed water quality of the lake Sayeon, Daeam and Hoeya using 17 variables. The water quality assessment found that the lake Daeam met the fourth to fifth grade because of high concentration of COD, SS and chlorophyll-a. Eutrophication assessment was conducted by revised Carlson's Index (TSIm, Aizaki) and found that Lake Daeam was more eutrophicated than the other two lakes all the year through as for chlorophyll-a, transparency and the total phosphorus (TP). A total of 95~111 phytoplankton species were identified from the three lake samples. Among them, the largest number of species were Chlorophyceae with 35~51, followed by Bacillariophyceae with 36~45, Cyanophyceae with 9~11, and Cryptophyceae with 6~9 species. The total cell number of phytoplankton was the highest in February (15,254 cells/mL) with Bacillariophyceae in the lake Daeam and the seasonal succession shows that Bacillariophyceae (Stephanodiscus spp.) in the spring, Cyanophyceae (Anabaena spp.) in the summer and the autumn, Bacillariophyceae (Stephanodiscus spp.) in the winter.

Monthly Variation of Phytoplankton Community in Asan Bay, Korea (아산만 식물플랑크톤 군집의 월 변동)

  • Phytoplankton community composition was investigated in the Asan Bay (South) Korea. Water samples were collected monthly from five stations during January to December 2008. Phytoplankton blooms were observed in February and in August but the location of the bloom varied with the sampling time. A total of 134 phytoplankton species from seven classes (Bacillariophyceae, Chlorophyceae, Chrysophyceae, Cryptophyceae, Cyanophyceae, Dinophyceae, Euglenophyceae) were observed. Diatoms were dominant in all stations through-out the year except in summer when dinoflagellates were numerous and highly distributed within the outer bay rather than in the river mouth. Dominant and subdominant species were composed mostly of diatoms but were replaced by dinoflagellates in June and July. Results of ecological index showed that the diversity indices were increasing toward outer bay but the dominance indices showed a reverse trend.

Response of Growth and Toxigenicity to Varying Temperature and Nutrient Conditions in Aphanizomenon flos-aquae (Cyanophyceae) (환경조건에 따른 Aphanizomenon flos-aquae (Cyanophyceae) 균주의 성장 반응 및 독소 생성)

  • The purpose of this study is to investigate growth response and toxigenicity under various temperature and nutritional conditions, in order to understand the physioecological characteristics of Aphanizomenon flos-aquae, which is a bloom-forming cyanobacterium in the Nakdong River. The strain was inoculated into media under combinations of four temperatures (4, 12, 21, $30^{\circ}C$) and three nutrients (modified CB medium, P-depleted CB medium, N-depleted CB medium) for 28 days. The algae-inhibition tests were performed to assess the potential allelopathic effects of the strains' filtrates on the growth of four algae strains (Microcystis aeruginosa, Aulacoseria ambigua f. spiralis, Aphanizomenon flos-aquae, Scenedesmus obliquus). Toxin production of a strain was measured by Enzyme-Linked ImmunoSolbent Assay (ELISA). The optimal growth temperature (Topt) of strains was $19.9^{\circ}C$ ($18.3-21.2^{\circ}C$), and the temperature range for growth was from $-0.3^{\circ}C$ to $34.3^{\circ}C$. Specific growth rate (${\mu}$) in modified CB medium varied from 0.10 to $0.16day^{-1}$, and the maximum growth rate (${\mu}_{max}$) was $0.17day^{-1}$. Although growth curves under N-existed and N-depleted conditions were almost the same, growth under N-depleted condition was relatively slowed (${\mu}=0.09$ to $0.14day^{-1}$), with a decreased maximum cell density. However, growth under the P-depleted condition was restricted for all temperatures, Two stains of Aphanizomenon flos-aquae were confirmed as not producing toxins, because saxitoxin and cylindrospermopsin were not detected by ELISA. The exudates or filtrates from the Aphanizomenon flos-aquae (DGUC003) resulted in significant inhibition of algal growth on the Aulacoseira ambigua f. spiralis (DGUD001) and Aphanizomenon flos-aquae (DGUC001) (p < 0.01).

Succession of Cyanobacterial Species and Taxonomical Characteristics of Dolichospermum spp. (Nostocales, Cyanophyceae) in the Weir Regions of the Nakdong River (낙동강 보 구간에서 남조류의 천이 및 Dolichospermum 속(Nostocales, Cyanophyceae)의 분류학적 고찰)

  • Freshwater cyanobacterial genus Dolichospermum is one of the most commonly spotted types of phytoplankton, whereas a limited number of studies on morphology of Dolichospermum spp. have been performed in South Korea. The purpose of this study is to investigate the succession pattern of cyanobacteria after weir construction, as well as morphological characteristics of Dolichospermum spp. from natural samples collected in the weir regions of Nakdong River. A total of 31 cyanobacterial taxa observed in this study were classified as belonging to 15 genera, 5 families, and 3 orders. Among them, morphological characteristics in the four species were classified into genus Dolichospermum, for most of the planktic former members of the genus Anabaena, were observed through light microscopy. Water bloom frequently occurred in the middle region of Nakdong River, the maximum number of cyanobacterial species appeared in the lower region of Nakdong River. The appearance of order Chrooccocales was only observed during summer when population density of Microcystis aeruginosa reached an annual peak. In contrast, filamentous cyanobacteria was observed throughout the whole year, even if when water temperature was lower than $5^{\circ}C$. It implied that the low-temperature-adapted filamentous cyanobacteria can grow in a range of water temperatures. Coil diameter of D. crassum from natural samples was $75{\sim}140{\mu}m$ ($ave.=91.3{\mu}m$; n = 94), slightly larger than those reported by previous studies. Dolichospemum smithii ($Kom{\grave{a}}rek$) Wacklin et al. 2009, was described for the first time in Nakdong River.

A study of newly recorded genera and species of filamentous blue-green algae (Cyanophyceae, cyanobacteria) in Korea

  • Cyanobacteria were sampled at five sites in the Han River, Nakdong River, and Geum River watershed from June 2014 to May 2015 and then cultivated. Two genera and five species of the cyanobacteria were newly recorded in Korea. The newly recorded species were Limnothrix redekei, Pseudanabaena galeata, Pseudanabaena amphigranulata, Sphaerospermopsis aphanizomenoides, and Calothrix parietina. As a result, the Korean flora of the cyanobacteria now include four orders, 22 families with 73 genera, 143 species, and two varieties, giving a total of 146 taxa.

Notes on Six Unrecorded Indigenous Species of Filamentous cyanobacteria (Cyanophyceae, Cyanophyta) in Korea

  • Samples were collected between August 2016 and May 2017 at Sangju-si, Gyeongsangbuk-do, Jeju-do, and Uljin-gun, Gangwon-do. As a result, one genus and six species were newly recorded in Korea. The unrecorded indigenous genus was Borzia, and the six species were Anagnostidinema acutissimum, Komvophoron bourrellyi, Hydrocoleum stankovicii, Borzia trilocularis, Phormidium tinctorium and Pseudanabaena lohchoides. Phormidium tinctorium and Pseudanabaena lohchoides had been reported to inhabit freshwater, but in this study, these were found in brackish water. It had also been reported that B. trilocularis trichomes are composed of up to 8 cells, but in this study, a maximum of 10 cells was observed.

낙동강 중류의 식물플랑크톤상과 군집구조

  • 낙동강 중류에서 조사된 식물플랑크톤상은 7강 42속 77종류였고 Bacillariophyceae가 43종류(55.8%), Chlorophyceae 24종류(31.2%), Cyanophyceae 4종류(5.2%), Dinophyceae와 Euglenophyceae이 각각 2종류(2.6%), Chrysophyceae와 Cryptophyceae가 각각 1종류 (1.3%)였다. 주요종은 Stephanodiscus hantzschii를 비롯한 6속 7종류였고 모두 규조류였다. 현존량은 최저 79 cells/$m{\ell}$(3월, 고령)에서 최고 33,023 cells/$m{\ell}$(2월, 물금)로 나타났다. 우점도지수는 최저 0.434(8월, 남지), 최고 0.999(1월, 2월, 물금)였다. 종다양성지수는 최저 0.026(2월, 물금), 최고 3.073(9월, 남지)로 조사되었다.

Studies on the Water Pollution in Gum Ho River (금호강의 수질오염에 관한 연구)

  • A study on the water pollution of Gumho river by the relationship between physio-chemical conditions and water quality level by phytoplankton was examined at 6 sampling positions during the period from April 21 to August 11, 1982. Examination of physio-chemical water analysis such as temperature, pH, DO, BOD and biological analysis has shown 1. The average for Gumho river was 6.2 - 7.2 of pH, 2.6 - 9.4 mg/l of DO, 28.8 - 122.4 mg/l of BOD. 2. The phytoplankton identification in this survey period showed, Cyanophyceae is 7 genera 13 species, Bacillariophyceae 11 genera 32 species and Chlorophyceae 17 genera 27 species. 3. The results of biological water analysis were as follows: Banyawol was from oligosaprobic to $\beta$-mesosaprobic. Dongchon and Gumdan was from $\alpha$-mesosaprobic to $\beta$-mesosaprobic, 3rd gongdan was from oemesosaprobic to $\beta$-polysaprobic, Paldal was $\beta$-polysaprobic and Gangchang was repolysaprobic. 4. To appear dominant algae there were Microcycstis, Oscillatoria, Anabaena, Melosira, Nitzschia and Synedra.
