Nutrition Contents and Health Status, Blood Glucose Response Effect for NIDDM Patients of Inonotus Obliquus in Culturing Method by Using Unpolished Rice (현미에서 배양된 차가버섯 분말의 영양성분과 제2형 당뇨환자의 건강지표 및 혈당관련인자에 미치는 영향)

  • The purpose of this study was to investigate the possibility of Inonotus obliquus as an functional resource. To assess the effects of Inonotus obliquus in culturing method by using unpolished rice in 15non-insulin dependent diabetes mellitus persons, we examined changes of hematological and chemical variables, DM indices content in serum during the Inonotus obliquus supplementation. GOT, GPT concentrations were significantly decreased. But TIBC content was significantly increased. Fasting glucose level during Inonotus obliquus supplementation was decreased respectively. These results show that modest dose of Inonotus obliquus in culturing method by using unpolished rice supplementation can decrease fasting blood glucose level without any changes in health indices and nutrition status of the non-insulin dependent diabetes mellitus person. These results indicated that Inonotus obliquus in culturing method by using unpolished rice diet is effective therapeutic regimen for the control of metabolic derangements in diabetes mellitus. Also, the results imply that Inonotus obliquus in culturing method by using unpolished rice can be used as possible functional food materials. However, large amounts of Inonotus obliquus in culturing method by using unpolished rice showed be used cautiously application.

  • Epifluorescence microscopy and direct viable counting methods have shown that only 0.01-0.1% of all the microbial cells from marine environments form colonies on standard agar plates. To culture novel marine microorganisms, high throughput culturing (HTC) techniques were developed to isolate cells in very low nutrient media. This approaches was designed to address microbial metabolic precesses that occur at natural substrate concentrations and cell densities, which are typically about three orders of magnitude less than in common laboratory media. Approximately 5000 cultures of pelagic marine bacteria were examined over the course of 3 years. Up to 14% of cells from coastal seawater were cultured using this method, a number that is 1400 to 140-fold higher than obtained by traditional microbiological culturing techniques. Among the cultured organisms are many unique phylogenetic lineages that have been named as new phyla (7), orders (2, 5, 12), families (3), and genera (1, 4, 6). Over 90% of the cells recovered by this method do not replicate in standard agar plating, the most common method of microbial cell cultivation.

Culturing Method and Dietary Value of Benthic Copepod, Tigriopus japonicus (부착성 요각류, Tigriopus japonicus의 배양방법 및 먹이효율)

  • The harpacticoid copepod, Tigriopus japonicus is one of the important zooplankton as a live food the production of marine fish fry. This reserch was carried out to investigate the culturing method and dietary value of T. japonicus. The reproduction of this copepod was analysed in the culturing systems (tne opened and the closed systems) and the surface area of the substate plates for the mass culture. Survival rate of the mature female and the number of nauplii produced per female for two day was higher in the open culturing system than in the close system. However, the result of survival rate of the nauplius was reversed. Larger surface area of the culturing vessel accommodated higher density of this bentic copepod. In polyculture of T. japonicus and rotifer (B. plicatilis), growth of T. japonicus seems to be dependent on the rotifer density. But rotifer seems to be independent on the copepod. With regard to dietary value, this copepod is better than Artemia nauplius for the larvae of red sea bream and tiger puffer which feed by picking. However, it is inadequate for bottom fish larvae as flounder.

Optimal Design of Mooring Steel Pile for Submersible Fish Cage (부침식 가두리 계류용 말뚝의 최적설계)

  • To develop a new fish cage which is required for offshore or moving cage culturing system has been gradually increased against being closely dense of fish cage in shallow water. Though submersible fish cage culturing system is essential technology for converting from shallow water into the offshore, it was pointed out the serious problem about stability of which are sinking and floating state. This study is presented conceptual design of submersible fish cage centered with a mooring steel pile to acquire stability and faculty. Design of mooring steel pile for submersible fish cage culturing system needs to carry out optimal design of mooring steel pile for which much efforts are required. Formulation and optimal design process of submersible fish cage are organized into using Sequential Quadratic Programming method of numerical optimization. For submersible fish cage system centered with a mooring steel pile, process of the optimal design is proposed and the optimal solutions are obtained.

In Vitro Germination and Propagation by Embryo Culture of Taxus cuspidata for the Taxol Production

  • To develop an efficient propagation method for yew tree, zygotic embryos were cultured under various conditions. When dissected embryos were cultured on GA$_3$ containing media, the highest germination frequency was observed on WPM medium contaning 1.0 mg/L GA$_3$. For germination of the embryos, two different conditions were compared; culturing embryos with endosperm (Method I), and 2) culturing embryos only (Method II). Maximum germination was achieved in 0.5 mg/L GA$_3$ when embryos with endosperm were cultured on the media. Of the media tested, White and WPM medium were the most suitable on germination of embryos. The abnormality of yew embryos found was observed when it cultured on GA$_3$ or culture media. About 40% of the precociously germinated embryos could be developed into full seedlings. Seedlings contained taxol in high quantity (535 $\mu\textrm{g}$/g dry weight). In vitro techniques will be sewed as a useful tool for the development of transformed root cultures and biosynthesis studies.

  • The purpose of this study was to establish a basic principal procedure for the processing of cultured meat. The first stage involved isolating satellite cells from the desired muscle of an animal using enzymatic digestion (i.e., by using proteases, collagenases, and pronases). The second stage involved culturing the isolated muscle satellite cells in a growth medium containing fetal bovine serum and penicillin/streptomycin with growth factors for an optimal period of time. The second stage involved a basic method for the isolated muscle cells to proliferate while sub-culturing to further induce differentiation in gelatin-coated culture dishes with the general culture medium. The third stage involved the induction of differentiation of muscle satellite cells or formation of myotubes using myogenic medium. Lastly, the fourth stage involved the identification of cell differentiation or myotube formation (myogenesis) using fluorescent dyes. Moreover, the principle of these protocols can be applied to perform primary culture of animal cells. This study will assist beginners with the technical aspects of culturing meat (isolation, cultivation, and differentiation of muscle satellite cells as well as identification of myotube formation for myogenesis).

  • To develop the optimal design method for the longline type scallop culturing facilities in the open sea numerical calculations and hydraulic model experiments are carried out for the stability and function optimization. Using the results for the motion and tension of the facilities, stable design concepts and effects of motion control system by vertical anchor and resistance discs art discussed. The results of this study that can be applied to the design are as follows: 1) Total external forces by design wave $(H_{1/3}\;=\;6,7\;m,\;T_{1/3}\;=\;12sec)$ at the coastal waters of Jumunjin for unit facility (one main line) are estimated to 5-20 tons, and required anchor weights are 10-40 tons in the case of 2-point mooring system. Though the present facilities are stable to steady currents, but is unstable to the extreme wave condition of return period of 10 years. 2) The dimensions and depth of array systems must be designed considering the ecological environments as well as the physical characteristics including the mooring and holding forces that are proportional to the length and relative depth of main line to wave length, and the number of buoys and nets. 3) Oscillation of the facility is influenced by water particle motion and the weight of hanging net, and is excited at both edge, especially at the lee side. To reduce the motion of the nets, the vertical anchoring system and the resistence disc method are recommended by the experimental results, 4) The damage of rope near the anchor by abrasion should be prevented using the ring-type connection parts or anchor chains.

Development of in vitro 3D hair growth model using tissue engineering technology

  • The recent development of methods for culturing hair follicles in vitro has proved an important tool to investigate many aspects of drug screening. Hair follicles develop as a result of epithelial-mesenchymal interactions between epidermal keratinocytes and dermal cells. We isolated some follicle cells using explantation and enzymatic digestion method from human scalp hair follicles. So we could culture some follicular cells, such as outer root sheath (ORS) cells, dermal papilla (DP) cells, dermal sheath (DS) cells, matrix cells and melanocytes. To induce hair morphogenesis in vitro the cells were 3-D cultured as skin structures. Moreover, to develop hair follicel organ culture model, we applied dermal equivalent (DE) to culturing hair follicles to expand hair growth period.

Studies on Sexing of Bisected Mouse Embryos by Rat H-Y Antibody (Rat H-Y 항체에 의한 생쥐 분할란의 성 조절에 관한 연구)

  • This experiment was carried out to develop a new technique of identifying XX of XY-bearing bisected embryos prior to implantation by immunological method. H-Y antiserum prepared in inbred Wastar female rats by repeated immunization with spleen cells from males of the same strain. The reactivity of H-Y antibody was confirmed by culturing mouse embryos in the medium containing H-Y antiserum and complement obtained from the guinea pig. The optimal condition for the activity of H-Y antibody was also investigated by culturing embryos under the concentraton or affected H-Y antibody was also investigated by culturing embryos under the concentration or affected H-Y antibody and culture rate. However, production of live young or sex rates of male and female from embryos transferred with psudopregnant. The biological test with the morula stage embryos showed that H-Y antibody was formed in all female rats immunized with spleen cell, but it was formed only in 80% female rats immunized with the antigen. When the bisected mouse embryos were cultured in vitro for 5~6 hours in morula stage, of 457 bisected embryos 81.4% of then were developed to the blastocyst stage. When the concentration rate of complement to H-Y antiserum varied from 1.0~5.0${mu}ell$, the lysis-rate of embryo was 19.5 to 67.3%. The concentration rate of complement did not influence the lysis-rate of embryos(P<0.05). The morphology embryos of bisected, zona-free and intact embryos showed the embryos lysis rate of 58.6, 42.7 and 48.5% respectively(P<0.05). Pregnancy rate were 50.0, 45.5 and 57.1% in psudopregnant recipient transferred with bisected, zona-free and intact blastocyst embryos. However, production of live youngs, sexual rate of male or female was 24(50.0:50.0), 22(45.5:55.5) and 36(58.3:41.7)mice, but affected and non affected half embryos with H-Y antiserum treatment was 23.1 and 26.7%. Also production of live youngs and sexual rate was 14(92.9:7.1) and 17(17.6:82.4)mice in affected and non affected half embryos in H-Y antiserum treatment(P<0.05).

A Study of Technical Development of Mariculture in the Coastal Water (천해양식어업발달과정에 관한 연구 - 기술개발활동을 중심으로 -)

  • Mariculture is contrasted with inland aqua-culturing fisheries. It is defind as the Industry of rearing Aquaorganism in limited coastal area relatively shallow in depth. Then, It's coming into being realization of Mariculture in it is long in history that Mariculture was realized in Korea. But it is from the early part of 1960s, that this industry has normally developed. Owing to 200 miles economy-zone problems of coastal countries, the development of deep sea fishing was limited, so the Korean Government has now appreciated the importance of cultured industries in the field of coastal fisheries. And the Korean Mariculture the output of which was only 18, 000 M/T in '60s attained 540, 000M/T in 1980s, has now occupied its relative importance in Korean Fisheries Industry. So the purpose of this report is to suggest the prospect of technical development of mariculture in the future of Korea, through the analysis of the various problems that affect upon the individual management '||'&'||' fishing ground utilization, along with the appreciation of "how to extend of those technical innovation" and "how the fishermen's technique level is extended at this stage. According to this study, the result is summarized as follows. First, Maricultural technique is classified into 8 sub-techniques as follows, as shown in fig. 1.Fig. 1. The Formation structure of mariculture technique Second, the change of technical method of mariculture in coastal area of Korea has made as 5 stages; 1) Scattering of culturing organism 2) Culturing by putting stone and installing bamboo 3) Culturing by installing rope and seeding 4) Culturing of putting objectives in cages 5) Culturing fish by feed Third, the maricultural fisheries of Korea has about 70 years long in history. It began from 1910s. But at that time there was no special technique in aquaculture and its technique was confined in searching out the object of species. The species was laver, oyster ect.Forth, although realization of mariculture in Korea has been long time, it is of late from 1960s that this has been industrial with normal development, and its technique of mariculture has mainly has developed from 1970s. Its result not only contributed to the high growth in Korean ecconomy along with the well balanced development between industires, but also it played a great role for the resolution of nation's food problem. Especially maricultural production has shown its sustained annual increase of 13.8% during the last 20 years. So the portion of mariculture among total fisheries stucture was extended from 4.1% in the early 1960s to 22.4% in 1980s.Fifth, it could be safely said that such development in maricultural field is resulted from the activity of aquacultural institutes such as Fisheries Reseach '||'&'||' Development production of major kinds such as Oyster, Sea-mustard, and Laver etc. As well as in the innovation of aquaculturing method with synthetic fiber utilization. FRDA has played important role in the efficient propargation of new aquacultural technique.Sixth, as for the change in aquaculture structure and its during period between 1970s and 1980s, the private management participation shown 25% increase from household number of 45, 173 to 56, 268 in total number. And in the respect of the management scale, of their management decreased, while it showed an increase in relative large scale management, the increase over 3 employees compared with other fisheries field between '70s and 80s. This must be an major trait to be recorded, Now the data above mentioned are shown as in table 1 and 2.Table 1. The maricultural fishing ground development situation in 982.Table 2.
