Quality Characteristics of Muffins with added Dukeum (pan-fired) Ramie Leaf (Boehmeria nivea) Powder using Response Surface Methodology (반응표면분석에 의한 덖음 모시풀잎 가루 첨가 머핀의 품질특성)

  • The objective of this study was to produce muffins with added dukeum ramie leaf powder and to compare them to a control. We used two different optimal composition ratios to produce muffins. The physical and sensory characters were varied by changing the ratio of dukeum ramie leaf powder, butter, and sugar to identify a critical value using response surface regression analysis. The ranges for the added dukeum ramie leaf powder, sugar, and butter were 15-35, 60-100, and 60-100 g, respectively. Muffins with added dukeum ramie leaves had comparable mineral and antioxidant vitamin compositions to those of dried ramie leaves. Muffins with added dukeum ramie leaf powder satisfied the suitability of the model, showing a p-value within 5% of significance level (p<0.05) for every aspect Volume and height differed without any association to ingredients. Lightness increased when less dukeum ramie leaf powder was added with more butter and sugar, redness increased when more dukeum ramie leaf powder and sugar were added with 70-80 g of butter, whereas yellowness increased when less dukeum ramie powder was added. Texture and toughness increased when less dukeum ramie leaf powder was added. The degree of gumminess and chewiness depended hugely on the amount of sugar. Cohesiveness, decreased with more dukeum ramie leaf powder and butter, whereas hardness increased with a medium range of ingredients. Sensory character increased with less dukeum ramie powder, and overall quality increased with more butter and sugar, but depended on the amount of added dukeum ramie power. The critical values from the sensory characteristics showed that 27 g of dukeum ramie powder, 72 g of sugar, and 86 g of butter was optimum.

Varietal and Culture-Seasonal Variation in Physicochemical Properties of Rice Grain and Their Interrelationships (쌀의 식미 관련 이화학 성분의 품종 및 작기간 변이와 상관 관계)

  • This study was carried out to understand the varietal variation in physicochemical properties of rice grain and those environmental changes by different transplanting time, and to elucidate the interrelationships among the factors related with eating quality of cooked rice. Fifty three rice samples, among which fifty samples were harvested at ordinary or late transplanted plots of the Crop Experiment Station in Suwon and three samples were harvested orpurchased from Niigata prefecture in Japan, were tested for various physicochemical components of rice grain and some physical factors of cooked rice. All of twenty seven rice cultivars tested were the recent-bred Korean japonica rice showing the wide range of maturity from early to medium-late heading and considerable difference in palatability of cooked rice. Amylose content, taste value by Nireco palatability tester (TVN), iodine blue color of cooking extracts(IB), and the ratio of IB /extracted solid amounts (ES) increased significantly by late transplanting, while viscosity (VN) and Mg / K. N value by Nireco tester, hot-water absorption of milled rice (HA), loss tangent of cooked rice by Rheolograph-Micro(LT), and most viscogram characteristics except setback viscosity (C-P) decreased drastically by late transplanting as compared with ordinary transplanting. Most of physicochemical properties of milled rice revealed narrower varietal variation in lately transplanted plot than in ordinary transplanted one. Protein content (PRO), volume expansion rate of cooked rice(VE), C-P and all physical factors of cooked rice by Rheolograph-Micro showed almost negligible seasonal variation, while amylose content (AM), VN, HA, IB/ES, peak viscosity(P), hot viscosity(H) and breakdown(P-H) viscosity exhibited considerably large seasonal variation. The early-headed varieties revealed lower amylose content and smaller seasonal variation of IB/ES compared with medium or medium-late headed rice varieties. AM was closely associatied with IB and IB / ES and VN was highly correlated with Mg/K. N and TVN in both ordinary and late transplanted plots. VN also was highly negatively correlated with cooking characteristics and highly positively correlated with viscogram properties in ordinary culture. PRO was closely connected with moisture content of milled rice and L T in ordinary transplanted plot. IB, which was closely connected with ES, was also singificantly associated LT, P and P-H in ordinary seasonal culture. IB/ES was highly negatively correlated with P, P-Hand P-H / C-P in ordinary culture but with LT and dynamic viscosity of cooked rice in late seasonal culture. The thirty rice cultivars were largely classified into two varietal groups by cluster analysis with physicochemical properties related with eating quality of cooked rice. Korean and Japanese high-quality rice cultivars were separately distributed in two respective varietal group.

Isolation and Mycelial Cultivation Submerged of Phellinus sp. (Phellinus sp.의 분리 및 균사체의 액체배양)

  • Fruit bodies similar to the Phellinus sp. residing on the mulberry were collected at Yang-yang in Kang-won-do province and one strain of Phellinus sp. was isolated from the fruit bodies. For mass production of the isolated mycelia in a submerged culture, the culture conditions, medium composition, and the effect of various culture systems on the mycelial growth, were investigated. The morphological characteristics of the fruit body were as follows: covered with blackish to black and rough, lower surface with yellowish-brown to dull-brown and smooth, 5-7 cm thick and hard woody. Also, the pure cultured mycelia showed yellowish-brown color, capability of purplish-brown pigment production on the PDA plate media, no-formation of clamp-connection, much binding branch, and enzyme activities such as laccase, tyrosinase and peroxidase. Therefore, pure cultured strain was identified to be Phellinus sp. In the flask culture, the optimum culture conditions for the mycelial production were obtained after cultivation of 8 days at inoculum level of 5%(v/v), media volume of 70 mL, 150 rpm, initial pH 6, and temperature of $30^{\circ}C$. Optimum medium composition from the response surface analysis were determined to be glucose 12.12 g/L, sucrose 12.12 g/L, yeast extract 11.15 g/L, malt extract 11.15 g/L, $KH_2PO_4$ 0.855 g/L and $CaCl_2$ 0.855 g/L. The production of the mycelia after 4 and 8 days of cultivation was 1.95 and 9.89 g/L, respectively. The maximum specific growth rate and productivity were $0.020\;hr^{-1}$ and 1.25 g/L/day, respectively. Among the three different culture systems for the growth of mycelia, the maximum mycelial dry weight of 7.5 g/L was obtained after cultivation of 4 days in the air-lift fermentor under aeration rate of 2.5 vvm. The maximum specific growth rate and productivity were $0.033\;hr^{-1}$ and 1.9 g/L/day, respectively, which were about 1.7 and 4.2 times higher than those of flask culture.

Effects on yield of Pleurotus ostreatus(Suhanneutari 1ho) according to afterripening conditions with bottle cultivation (후숙 조건에 따른 느타리(수한느타리버섯 1호)의 생육특성)

  • This study of afterripening conditions during oyster mushroom (Suhanneutari 1ho) cultivation in bottles was investigated. Medium materials were used poplar-sawdust (68%), cotton seed peel (16%), Beet pulp (8%) and cotton seed cake (8%). Mix of materials was used as a percentage of the volume and to adjust moisture content (67%). Autoclaved mediums were placed in low temperature storage ($20^{\circ}C$) and then moved in inoculation room and conducted mechanical inoculation. Mycelial culture temperature was maintained at $20{\sim}21^{\circ}C$ and cultured during 18 days. The afterripening period were 6days, 9days, 12days and 15 days at $25{\sim}27^{\circ}C$. The yield of fruit body was higher for 9 days (163.1g/bottle) and 12 days (160.7g/bottle) than that of other afterripening period. Second, in the changes in moisture content of the medium according to the afterripening period, no significant changes were observed during mycelial grwoth. The longer afterripening period was showed slightly lower weight of media. The moisture content of media after harvest at afterripening for 9 days had the biggest reduction than any other treatments. In addition, weight of media and yield of afterripening for 9 days were the lowest and highest, respectively.

Nutrient Absorption by Citrus unshiu Marc. Grown in Out-Door Solution Culture (양액재배에서 감귤나무의 무기양분 흡수)

  • Nutrients and water absorption by Citrus unshiu Marc. cv. Miyagawa Wase were examined using spray type of out-door hydroponics system. Three different concentration levels of nutrient solution were treated to citrus trees to examine nutrient uptake. To do so, concentration and the volume change of nutrient solution was measured for each treatment. By weighing the volume of solution and citrus trees, amounts of water uptake and fresh weight increase were observed periodically. Water uptake by trees increased as tree grew and time elapsed, but was lower at high level of nutrient concentration than the rest, due partly to the reduction in hydraulic conductance and to the sustained high salt concentration. Rapid increase in fresh weight and nutrient uptake occurred from May to July and from August to September. The amounts of nutrient uptake were significantly different among nutrient levels: the higher concentration, the greater uptake by citrus tree. The absorbed amounts of $NO_3$, K and Ca were much higher than those of $NH_4$, S, P, Mg, and Fe. Most mineral contents in leaves were proportional to the concentration of supplied nutrient solutions. From the results of nutrient absorption and contents in leaves at different levels of nutrient solutions, the composition of major elements for citrus nutrient solution can be modified as follows: 27.1, 16.5, 66.0, 80.0, and $24.0mg\;L^{-1}$ for N, P, K, Ca, and Mg, respectively.

Development of a Functional Mortar for Algae Growth Restraining by Using Soluble Glass (수용성 유리를 이용한 조류 생장 억제형 기능성 모르타르의 개발)

  • This study focuses on the algae growth restraining. Many researches on a critical damage from algae growth are published, but it is hard to find how th restrain. Abnormal algae increasing is a problem, because it makes red tides, biodeterioration, etc. Therefore this study aims to decrease the damage from algae growth. Some metal ions have been used microorganism killing materials from old times. Especially, Cu ions are highly effective. Based on these uses of the metal ions, a functional mortar which restrains algae growth is developed. The mortar contains soluble glass which dissolve in water. The soluble glass was made of Cu ions and phosphates. When the soluble glass is dissolved, Cu ions are soaked out stably from the soluble glass. Culture mediums which incubate algae were made to evaluate the developed mortar specimens. Culture mediums were filled with fresh water and sea water. Algae were incubated for fourteen days in culture mediums. The evaluating methods are measuring volume of the dissolved organic carbon and the chlorophyll. Using these two measurements, the mortar specimens are judged that can restrain algae or not. According to the result, the functional mortars of culture medium filled with fresh and sea water shows similar trend. The functional mortar for restraining algae growth performs that's role well.

Antimicrobial Activity of Hwangryunheadok-tang and Extracts of Scutellariae Radix, Phellodendri Cortex, Coptis Rhizome, and Gardenia Jasminoides against Salmonella Typhimurium (黃連解毒湯, 黃連, 黃芩, 黃柏, 梔子의 Salmonella Typhimurium에 대한 항균효과)

  • Objectives: This experiment investigated the antimicrobial activity of Hwangryunheadok-tang and extracts of Scutellariae radix , Phellodendri cortex , Coptis rhizome , and Gardenia jasminoides against Salmonella typhimurium . Methods: After spreading S. typhimurium on a bacterial culture medium plate, antimicrobial activity was tested by dripping diluted Hwangryunheadok-tang or extracts of Scutellariae radix , Phellodendri cortex , Coptis rhizome , or Gardenia jasminoides (80 μl, diluted 100, 50, 10, and 1%) onto the plate, followed by culture for 16 to 72 hours. The minimal inhibitory concentration (MIC) was tested by dripping the minimum dilution that showed antimicrobial activity (80, 60, 40, and 20 μl) and measuring the density. The antimicrobial activity of Hwangryunheadok-tang and four extracts showed continuous antibacterial activity against S. typhimurium throughout the experiment. Result: 1. S. typhimurium . (Standard Microorganism, ATCC) 1) The Hwangryunheadok-tang and extracts of Scutellariae radix , Phellodendri cortex , and Coptis rhizome showed antibacterial activity in the undiluted solutions and in 50% dilutions. Gardenia jasminoides extract showed potency only in the undiluted solution. The antimicrobial potency of the undiluted solution was increased when the volume of inoculation was increased, but no difference was noted when the culture time was extended. All the extracts showed antimicrobial potency against S. typhimurium . 2. S. typhimurium isolated from diarrhea patients 1) When compared to the standard microorganism experiment on S. typhimurium , the MICs of the five extracts were increased. However, whereas the antimicrobial potency of doxycycline was lost entirely against bacteria isolated from patients with diarrhea, the antimicrobial potency of all the extracts was diminished but did not disappear. 2) The antimicrobial activity of Hwangryunheadok-tang and four extracts was continuous even when the culture time was extended to 16, 24, and 72 hours. Conclusions: The Hwangryunheadok-tang and four kinds of extracts have antimicrobial activity against S. typhimurium . Comparison of a standard microorganism with S. typhimurium isolated from diarrhea patients showed that the antimicrobial activity of all the extracts was better than that of antibiotics. Further studies should focus on the value and benefits of Hwangryunheadok-tang , and the four kinds of extracts as clinical treatments.

Influence of Physico.Chemical Properties of Root Substrates on the Growth of 'Maehyang' Strawberry Daughter Plants Produced by Bag Culture of Stock Plants (포트 충전용 상토의 물리.화학성이 플라스틱 백 재배를 통해 발생한 '매향' 딸기의 자묘 생육에 미치는 영향)

  • This research was conducted to determine the influence of physiological and chemical properties of root substrates on the growth of the daughter plants of 'Maehyang' strawberry produced by bag culture. The daughter plants produced by stock plants during bag culture were individually separated and grown in 10-cm diameter plastic pots containing six different formulations of root substrates: a) 50% peatmoss plus 50% vermiculite (5:5 by volume, A), b) 70% peatmoss plus 30% perlite (7:3, B), c) 70% coir dust plus 30% perlite (7:3, C), d) mixture of 35% coir dust, 35% peatmoss, and 30% perlite (3.5:3.5:3.0, D), e) mixture of 20% rice hull, 70% coir dust, and 10% perlite (2:7:1, E), and f) 30% rice hull plus 70% coir dust (30:70, F). The container capacity and air filled porosity of the growing medium varied greatly among the six substrate formulations evaluated. The substrates E and F had less container capacity and higher air-holding spaces than the rest of the formulations. Therefore, these two formulations (E and F) may cause a problem in water management during the production of healthy daughter plants. The substrate formulations A, B, and D retained higher nitrogen (N) concentrations than other formulations containing coir dust or rice hull. The substrate formulations E and F which contained rice hull had lower N, phosphorus (P), and potassium(K) concentrations than other substrate formulations containing coir. The quality of the daughter plants grown in all six different substrate formulations was good with the crown diameters at around 10 mm. Fresh weights of the daughter plants grown in substrate formulations A, C, and D were higher than those obtained from B, E, and F. Dry weights of the daughter plants showed a similar trend. The daughter plants having high fresh and dry weights and increased crown diameter are in demand by the industry. For this reason, the substrate formulations A, C and D can readily be used as potting mixes during the production of 'Maehyang' strawberry transplants utilizing the bag culture system.

Cell Growth in Suspension-Culture of Populus nigra var. italica and the Efficiency of Micro-Callus Formation according to Cell Plating Method (Populus nigra var. italica현탁배양(懸濁培養) 세포(細胞)의 생장(生長) 및 Cell Plating방법(方法)에 따른 Micro-Callus형성능력(形成能力))

  • In order to know the growth of suspended cells by explant sources, the change of nitrogen contents of cultured cells following the growth periods, capability of micro-callus formation according to cell plating methods, growth of suspended cells on various media, and efficiency of micro-callus formation by using growth regulators and different N strengths were investigated. 1. When suspension culture was tried by using the callus induced from internode and petiole, cell fresh weight and packed cell volume increased with similar way and the growth reached at stationary phase after 12 culture days. 2. N-contents of cultured cells increased upto 3 days and decreased around 6days. But the values increased again upto 9 days, after that they showed gradual decreases. 3. Of cell plating methods, embedding method was the best for micro-callus formation. 4. Growth of suspened cells showed the rest performanoes, when they were cultured on LM medium with 1/2N strengths and BAP 0.01.2.4-D 0.1, and NAA $1.0mg/{\ell}$, after 15 cultured days(upto 76.9 folds). LM medium was better than MS or GD. The combination of auxin and cytokinin was better for cell growing than auxin-treatment only. 5. Micro-callus from single cell and small cell aggregates was formed only on MS and LM media with 2,4-D $1.0mg/{\ell}$.

Development of Saccharomyces cerevisiae Strains with High RNA Content (리보핵산을 다량으로 함유하는 Saccharomyces cerevisiae 균주의 개발)

  • RNase activity of Saccharomyces cerevisiae ATCC 7754 was investigated to obtain strains with high ribonucleic acid (RNA) content. The yeast strain contained two RNase activities; an acidic RNase with a optima of pH $3{\sim}4$ and an alkaline RNase with a optima pH 9. The acidic RNase activity was inhibited by $0.08\;M\;HgCl_{2}$ most drastically. The alkaline RNase activity was inhibited by 2.0 M NaCl or KCl, while enhanced by addition of $0.05\;M\;CaCl_{2},\;0.02\;M\;ZnSO_{4},\;or\;0.008\;M\;HgCl_{2}$. Various mutants of Saccharomyces cerevisiae ATCC 7754 were isolated by ethylmethane sulfonate (EMS) treatment or $\gamma$-ray/ultra violet irradiation. Among the mutants that were sensitive to high concentration of KCl which inhibits alkaline RNase, B24 was selected for high RNA content per culture volume. Growth characteristics of the mutant were comparable to those of the mother strain with optimum growth at pH $4.5{\sim}5.5$. The mutant accumulated higher content of RNA than the mother strain when glucose was used as the carbon source. However, both growth rate and total RNA content of the mutant were higher in molasses medium than in glucose medium. RNA content of the mutant increased rapidly during the early stage of growth, and then decreased gradually until the culture reached stationary phase by a fed-batch culture in a 5 L jar fermenter. Maximal cell harvest and the final RNA content using the mutant B24 were 69.6 g/L culture broth and 19.8 g/100 g of the dry cell while those using the mother strain were 68 g/L culture broth and 16.1 g/100 g of dry cell, respectively.
