  • Amoebic gill disease of flounder, Paralichthys olivaceus was diagnosed at commerical culture facility in South Korea. The amoeba was identified as a species of the genus Neoparamoeba based on the morpholgical characteristics of trophozoites. Transmisson electron microscopy revealed the presence of a symbiotic organism, parasome in the cytoplasm and dense glycocalyx on the surface of the trophozoites. They lacked the boat-shaped microscales on the surface and contained numerous vacuoles and channels, mitochondria in the cytoplasm. Colonization of amoebae on gill tissue elicited extensive fusion and hyperplasia of gill lamella.

  • This paper dealt with the light microscopical and electron microscopical findings on the morphological changes of the liver of Korean native goat injected with toxin (culture filtrate) of Clostridium perfringens which was isolated from Korean native cattle died of acute Clostridium perfringens enterotoxemia. The results observed are summarized as follows. In the microscopical findings, hyperemia and minute hemorrhage of the liver parenchyma, dilatation of hepatic central vein and centrilobular necrosis of liver, cloudy swelling and hydropic degeneration of hepatic cells, and appearance of light eosinophilic granular bodies in the vacuoles were recognized. In the electron microscopical findings, appearance of pinocytotic vesicle (coated vesicle), fusion of these vesicles, formation of vacuole and accumulation of minute granular proteinous materials in the vacuole were observed in the hepatic cells. Decreased number of glycogen granules, swelling and destruction of mitochondria, proliferation of smooth-surfaced endoplasmic reticulum, enlargement of rough-surfaced endoplasmic reticulum, dispersal of thready agranular membranous structure and appearance of secondary lysosome were recognized in the hepatic cell cytoplasm.

  • Mesophyll protoplasts derived from young leaves of Nicotiana rustica and N. tabacum cv Burley 21 were fused with the aid of polyethylene glycol(PEG). Cytological examination of protoplasts after PEG treatment revealed 12.8 % heterokaryocytes. After 7 weeks culture, the hybrid calli showing greenish white with a compact appearance were selected in contrast to parental type calli tinged with white or green color. The somatic hybrid plants were verified by morphological, biochemical and cyclological analysis. A heterosis effect for plant vigor and height was observed but the shape of leaves and flower characteristics were intermediate between N. tabacum and N. rutstica. The isozyme banding patterns for peroxidase of somatic hybrid lines were compared with the parent species. A number of isozyme bands derived from both parental species were found in the hybrids. Somatic hybrid plants have been successfully backcrossed to the parental N. tabacum particularly with somatic hybrid plants as female parents. These hybrid plants yielded small seeds, only few which were germinable.

  • Rapamycin, known as an inhibitor of Target of Rapamycin (TOR), is an immunosuppressant drug used to prevent rejection in organ transplantation. Despite the close association of the TOR signaling cascade with various scopes of metabolism, it has not yet been thoroughly investigated at the metabolome level. In our current study, we applied mass spectrometric analysis for profiling primary metabolism in order to capture the responsive dynamics of the Chlamydomonas metabolome to the inhibition of TOR by rapamycin. Accordingly, we identified the impact of the rapamycin treatment at the level of metabolomic phenotypes that were clearly distinguished by multivariate statistical analysis. Pathway analysis pinpointed that inactivation of the TCA cycle was accompanied by the inhibition of cellular growth. Relative to the constant suppression of the TCA cycle, most amino acids were significantly increased in a time-dependent manner by longer exposure to rapamycin treatment, after an initial down-regulation at the early stage of exposure. Finally, we explored the isolation of the responsive metabolic factors into the rapamycin treatment and the culture duration, respectively.

  • Many age-dependent neurodegenerative diseases are associated with the accumulation of abnormally folded proteins within neurons. One of the major proteolytic pathways in the cell is the autophagy pathway, which targets cytoplasmic contents and organelles to the lysosomes for bulk degradation under various physiological and stressful conditions. Although the importance of autophagy in cellular physiology is well appreciated, its precise roles in neurodegeneration remain largely unclear. Recent studies indicate that components of the endosomal sorting complex required for transport (ESCRT) are important in the autophagy pathway. Reduced activity of some ESCRT subunits leads to the accumulation of autophagosomes and failure to clear intracellular protein aggregates. Interestingly, rare mutations in CHMP2B, an ESCRT-III subunit, are associated with frontotemporal dementia linked to chromosome 3 (FTD3). Mutant CHMP2B proteins seem to disrupt the fusion of autophagosomes and lysosomes in cell culture models. These findings suggest a potential mechanism for the pathogenesis of FTD3 and possibly other neurodegenerative diseases as well.

  • The remedial junction is found in the network of single walled carbon nanotubes after the irradiation of protons not only for the better mechanical strength but also for the higher property of electrical conductivity. The irradiated proton formed a beam transferred sufficient energy to change the sp2 structure of atomic carbon as much as damage of crystalline formation, however it is shown the cross bonding while recovery of structure. This improved network in 2-D atomic chain of carbon is expected to use in a critical part in space energy harvesting system related with the solar radiation.
