Cultivation of Nostoc flagelliforme on Solid Medium

  • In order to construct an artificial cultivation of Nostoc flagelliforme on solid medium, we attempted to assess the viability of approaches, which utilized either BG-11 agar or sand medium using both sterile and non-sterile algal segments. In the trial in which the BG -11 agar medium was inoculated with the non-sterile algal segments, the algae exhibited the rapid growth in the initial 4 days of cultivation. However, after 4 days of cultivation, the growth rate of the algae slowed, and the algal growth was completely stopped by 7 days of cultivation. When the BG -11 medium was inoculated with the sterile algal segments, the algae exhibited the rapid growth for a longer period of 8 days, reaching a length of 24.9 mm. The growth rate during this period was measured to be $24.5\%$. After the 8 days of cultivation, the algal growth rate began to slow and had almost stopped by the 13 days of cultivation. On the other hand, when the sterile algal segments were inoculated onto a sand plate, the algal segments decomposed, reaching total decomposition after 11 days of cultivation. By way of contrast, the desiccation treatment samples continued to grow for 14 days of cultivation. After 14 days of cultivation, the algae achieved a length of 26.1 mm, with a growth rate of $30.6\%$. Our results indicate that periodic desiccation may constitute an effective strategy for the prevention of algal decomposition.

Effect of Cultivation Methods and Harvesting Time on Yield and Quality of Safflower(Carthamus tinctorius L.) Seeds (홍화 재배유형 및 수확시기가 종실 수량 및 품질에 미치는 영향)

  • This study was conducted to investigate the plant growth and seed yield between rain-shelter plastic house and out-door cultivation, and analysed the effect of harvesting time on seed quality. In rain-shelter plastic house cultivation, emergence date was 3 days and flowering date was 8 days earlier than out-door cultivation. Seed yield of rain-shelter plastic house cultivation was increased $25\%$ compared to out-door cultivation and it's more beneficial than out door cultivation. The crude fat content was higher and seed was whiter and better quality in rain-shelter plastic house cultivation than out-door cultivation. In out-door cultivation, the crude fat content was steadily increased until 40 days after full bloom, but the seed color was getting dark which was harvested at 30 days after full bloom and the color was turn to the dark brown when the seed was harvested at 40 days after full bloom. Harvesting of safflower seed's quality is best between 20 days and 30 days after full bloom.

Changes of Growth Characteristics and Yield according to the Cultivation Types of Waxy Corn, Chalok 1

  • The appearances of growth and yield according to the cultivation types were investigated in chalok 1. The days to harvest under double vinyl house, open mulching and open cultivation were 87.7, 95.8, and 101.8 days, respectively and significantly different in each cultivation types. The branched ears of open mulching, double vinyl house and open cultivation were started at 7,8, and 13 days after silking, respectively. The frequency of branched ear per total plants to double vinyl house, open cultivation and open mulching were 10%, 13%, and 19%, respectively. The ear weight of open mulching and open cultivation was superior to branched ear weight. The yields (kg/10a) of double vinyl house, open mulching and open cultivation were 755.7kg, 740.7kg, and 530.0kg, respectively. The yields(kg/10a) of double vinyl house and open mulching were significantly different to that of open cultivation and LSD(5%) was 133.42. Thus, early cultivations in double vinyl house were more beneficial than other cultivation types because of the highest yield and the lowest branched ear.

The Growth-Curve Analysis of Tobacco in Various Cultivation Types (잎담배의 재배방법에 따른 생장 분석에 대하여)

  • The growth of flue-cured tobacco was analyzed with the mathematical treatment. The results are summarized as followings: 1. The growth curve was fitted to the quadratic polynomial equation in improved-mulching cultivation, blot to the Gompertz equation in the other cultivations. 2. The initial point of the maximum growth phase for dry weight was about 50 days after transplanting in improved-mulching cultivation, but about 40 days in the other cultivations, and the maximum growth period was for 25 days in all cultivations. 3. The growth rate of the maximum growth period in dry weight decreased in the order of improved-mulching cultivation, mulching cultivation, and non-mulching cultivation. 4. A relative growth amount in the maximum growth period was higher in later sowing. 5. The length of maximum growth was 5 days shorter in leaf area than in dry weight. The maximum growth phase was 7 days earlier in leaf area than in dry weight.

Changes of Antioxidant Activity as affected by cultivation period in Buckwheat (Fagopyrum species) Sprouts (재배기간에 따른 쓴메밀(Fagopyrum tataricum Gaertner)싹의 항산화 활성 및 생리활성 평가)

  • Buckwheat (Fagopyrum species) has long been an excellent functional food. Besides, buckwheat sprouts contain various functional substances. In this study, we investigated the antioxidant activity of buckwheat sprouts in the context of cultivars harvested after different cultivation periods (0, 3, 5, 7, 9, 13, and 15 days after planting). Buckwheat sprouts were cultivated at 25℃ for up to 15 days and then extracted with ethanol. Antioxidant components were then extracted from sprouts and leaves using a freeze dryer. The total polyphenolic content, flavonoid content, and antioxidant activity were then analyzed. The total polyphenol content increased from 32.26 mg GA eq/100 g for raw buckwheat to 114.75 mg GA eq/100 g after 7 days of cultivation. Also, the flavonoid content increased from 20.61 mg catechin eq/100 g (0 days) to 56.54 mg/g after 9 days of cultivation. The DPPH radical scavenging activity (concentration of extract at 0.25 mg/mL) increased from 7.89% at day 0 to 53.48% after 9 days of cultivation. Additionally, the ABTS radical scavenging activity increased from 10.26% at day 0 to 32.89% after 7 days of cultivation; of note, the activity decreased afterward. These results suggest that the best buckwheat sprouts with higher biological activities are those cultivated for 7-9 days. For a complete understanding of the potential of buckwheat sprouts as functional foods, we plan to further analyze their antioxidant activity in the future.

Characteristics of Immobilized Culture of Mentha piperita Cells for Oil Production

  • To investigate the characteristics of immobilized peppermint (Mentha piperita) cells, dry cell weight (DCW), change of cell viability, and oil productivity of the immobilized cells were determined. Peppermint cells were immobilized in polyurethane (PU) foams of $5{\times}5{\times}5$ mm and cultured in a shaking flask. The maximum DCW was 2.1 mg per foam piece after 20 days of cultivation and the cell density was approximately 420 mg per flask containing 200 foams in 200 ml medium. For the first five days of cultivation, the cell viability was about 80$%$ and decreased to 70$%$ during 5 to 20 days of cultivation. The maximum oil productivity, 148 mg/l was achieved after 40 days of cultivation. The immobilized cells were also cultivated in a bioreactor, equipped with a round spiral type impeller, containing 2, 400 PU foams. The cell viability after 30 days of cultivation with chitosan as an elicitor in the bioreactor was 67$%$ and DCW was 2.0 mg per foam piece. Though the cell viability was relatively high in the bioreactor system, the oil productivity was relatively lower than that of the flask system.

The ptimum temperatures during cultivation period of Gastrodia elata according to growth stages (천마 생육단계별 변온에 의한 최적온도 및 재배기간)

  • This study was carried out to investigate the optimum temperature and the cultivation period according to the different growth stages of Gastrodia elata (G. elata). The growth period for the indoor cultivation of G. elata is divided into four stages that require specific temperatures during the enlargement of the tuber. The optimum temperatures and cultivation periods during the growth stages of G. elata were observed to be $20^{\circ}C$ for 30 days during the mycelial growth stage (MGS), $25^{\circ}C$ for 120 days during the tuber formation stage (TFS), $6-24^{\circ}C$ for 60 days during the tuber enlargement stage (TES), and $5^{\circ}C$ for 30 days during the dormant stage (DS). The total cultivation period was shortened by 120 days in the indoor cultivation facilities by reduction of 30 days from the mycelial growth stage, addition of 30 days to the tuber formation stage, and reduction of 120 days from the dormancy stage as compared to the outdoor field cultivation. These results provide a basis for a growth model that permits year-round cultivation of G. elata.

Evaluation of Methane Emissions with Water Regime before the Cultivation Period in Paddy Fields

  • Anaerobic decomposition of organic material in flooded rice fields produces methane, which escapes to the atmosphere primarily by transport through the rice plants. The annual amount of $CH_4$ emitted from a given area of rice is a function of the number and duration of crops grown, water regimes before and during cultivation period, and organic and inorganic soil amendments. Soil type, temperature, and rice cultivar also affect $CH_4$ emissions. The field experiment was conducted for three years to develop methane emission factor for water regime before the cultivation period from the rice fields. It was treated with three different water regimes prior to rice cultivation, namely: non-flooded pre-season < 180 days, non-flooded pre-season > 180 days, flooded per-season in which the minimum flooding interval is set to 30 days. Methane emission increased with days after transplanting and soil redox potential (Eh) decreased rapidly after flooding during the rice cultivation. The average methane emission fluxes were $5.47kg\;CH_4\;ha^{-1}day^{-1}$in flooded pre-season > 30 days, 5.04 in non-flooded pre-season < 180 days and 4.62 in non-flooded pre-season > 180. Methane emission flux was highly correlated with soil temperature and soil Eh. Rice yields showed no difference among treatments with water regime before the cultivation period.

Evaluation of the Antioxidant Activity of Wheat Sprouts Produced by Different Cultivation Methods and Harvest Times (새싹밀의 재배방법과 수확시기에 따른 항산화 성분 및 활성 평가)

  • In this study, the antioxidant activity of methanol extracts of wheat sprouts grown in wild fields and cultivated fields and harvested at various times were analyzed to compare the quality characteristics of the wheat sprouts as a function of their cultivation and harvest time, as well as for the development of functional materials. The total polyphenolic and flavonoid content, as well as antioxidant activity of the wheat extracts, were subsequently analyzed. The ABTS radical scavenging activity of the wheat extract increased from 16.97 mg TEAC/g sample on cultivation day three in cultivated field wheat sprouts to 25.99 mg TEAC/g sample after seven days of wild field cultivation. The total polyphenol content increased from 17.08 mg GA eq/100 g in cultivated field wheat sprouts grown for three days to 28.70 mg GA eq/100 g after seven days of wild field cultivation. In addition, the flavonoid content increased from 7.02 mg catechin eq/100 g (7 days) to 8.47 mg/g after 12 days of wild field cultivation. Notably, the activity subsequently decreased. These results suggest that the wheat sprouts with higher biological activity were those produced from the wild field after 20 days.

Comparison of optimal culture and growth characteristics of the Lentinula edodes fruit body 'Sanjo701ho', 'Nongjingo' (표고 '산조701호', '농진고'의 최적 배양 및 자실체 특성 비교)

  • This study investigated the characteristics of different cultivars of Lentinula edodes in sawdust culture at different cultivation days. Between cultivation days 30-120, the color of 'Sanjo701ho' displayed a reduction in L value (brightness) from $83.8{\pm}2.5$ to $45.7{\pm}2.3$, values a and b increased, but hardness decreased from $9.4{\pm}0.9g/mm$ to $2.6{\pm}0.2g/mm$. Between cultivation days 30-120, 'Nongjingo' displayed a reduction in L value from $86.2{\pm}2.1$ to $53.4{\pm}1.3$. Values a and b increased with longer cultivation; however, hardness decreased from $4.8{\pm}0.7g/mm$ on day 30 to $3.8{\pm}1.0g/mm$ on day 120. 'Sanjo701ho' was first harvested at 46 days after a 30-, 89 days after a 60-, 8 days after a 90- (the shortest), and 9 days after a 120-day cultivation. The average fruit body weight was the highest on day 90 of cultivation, at 48.3 g, when the diameter and thickness of the mushroom cap also appeared highest. However, the colorimetric results showed that fruiting bodies produced in the culture medium for 120 cultivation days showed the most excellent commercial properties. 'Nongjingo' was first harvested at 22 days after a 30-, 18 days after a 60-, 8 days after a 90- (the shortest), and 9 days after a 120-day cultivation. Therefore, this study determined that a stable quantity of mushrooms with high commerciality can be produced with 120 cultivation days, considering the shiitake culture and the characteristics of the fruit body.
