The baking performance of Cudrania tricuspidata leaf powder as a value-added food ingredient was investigated in a model system of sponge cakes. Cudrania tricuspidata leaf powder was incorporated into cake batter at 5 levels (0, 5, 10, 15, and 20%, w/w) by adding equivalent amount based on total weight of wheat flour. The specific volume, height, moisture content, volume index, and symmetry index of sponge cakes decreased significantly with the increase in Cudrania tricuspidata leaf powder content (p < 0.05). Sponge cakes became darker and firmer with increase in Cudrania tricuspidata leaf powder content (p < 0.05), as indicated by decrease in the $L^*$ and increase in the firmness measured by the texture measuring instrument. Total polyphenol content also increased gradually as the Cudrania tricuspidata leaf powder content increased. Finally, the consumer acceptance test indicated that addition of Cudrania tricuspidata leaf powder up to 10% in the formulation of sponge cakes did not significantly influence the consumers' acceptability with respect to taste and flavor. In addition, the overall acceptability was not different from each other for control and 5% sample (p > 0.05).
The purpose of this study was to develop hydrolysate from silkworm (Cudrania Silkworm Fibroin Peptide; CSFP), a food containing components for improving blood vessel health. In general, Cudrania tricuspidata leaves contain about 5 times more rutin and 25 times more dihydroquercetin than mulberry leaves. 1-Deoxynojirimycine (1-DNJ), dihydroquercetin and rutin inhibit the activity of carbohydrate-digesting enzymes, inhibit blood lipid peroxidation, and regulate insulin secretion, which helps blood vessels to be healthy. When the diet-controlled silkworm was enzymatically hydrolyzed, it was confirmed that rutin content was about 8 times higher than that of the in general silkworm as a control. In the silkworm hydrolysate, CSFP, developed as a final food material, the active ingredients were 65 mg/kg for rutin, 3,328 mg/kg for DNJ, 0.43 mg/kg for dihydroquercetin, and 82,624 mg/kg for total polyphenol, which was confirmed through LC-MS/MS analysis. In conclusion, it was found that silkworms fed with C. tricuspidata leaves as a diet had more active components that can help control blood sugar and improve blood vessel health than silkworms fed with mulberry leaves.

Objectives : Cudrania tricuspidata (C. tricuspidata) is well-known traditional herbal remedy and its root, leaf and fruit were used for treatment of inflammation, tumor and painkilling. However, effect of C. tricuspidata fruit on tight junction is still unknown. The aim of this research was to determine the effect of C. tricuspidata fruit extract on human keratinocyte HaCaT cells. Methods : The antioxidant effects of water extract of C. tricuspidata (WECT) and ethanol extract of C. tricuspidata (EECT) were analyzed by using an ABTS assay. To confirm the cytotoxicity of WECT and EECT, MTS assay was performed. The mRNA expression levels of tight junction related genes were analyzed using quantitative RT-PCR analysis. Furthermore, dispase assay was used to investigate the alteration of cell-cell adhesion strength of EECT treated HaCaT cells. Results : WECT and EECT showed strong antioxidant activity. No obvious cytotoxicity was observed in both WECT and EECT until $2.0mg/m{\ell}$ concentration. The mRNA expression level of Claudin 6 were significantly increased by EECT treatment, whereas the WECT did not affect the expression of Claudin 6. Furthermore, EECT treatment enhances cell-cell adhesion strength. Conclusions : In this study, we investigated the physiological activities of the extracts of Cudrania tricuspidata fruit extracts on human keratinocytes by two different extraction methods. EECT might have an anti-aging activity on the skin by reducing oxidative stress. Moreover, it may be a useful ingredient in atopic dermatitis and skin-moisturizing, given its effects of altering Claudin 6 gene expression and enhancing cell-cell adhesion strength.

The antioxidative activities of water-soluble extracts from leaves and stem bark of Morus alba and Cudrania tricuspidata were compared in vitro experimental models. Antioxidative activities were measured by inhibition activity against lipid peroxidation of mouse liver microsome, and they were showed in the following order; stem bark of C. tricuspidata(53%)>stem bark of M. alba(43%)>leaves of C. tricuspidata(38%)>leaves of M. alba(43%). In antioxidative activities determined by thiocyanate method and TBA method, the water-soluble extract of stem bark of C. tricuspidata showed the highest antioxidative activity. The water-soluble extracts of leaves were slightly stronger than other extracts in DPPH$({\alpha},{\alpha}'-diphenyl-{\beta}-picrylhydrazyl)$ method. The concentrations of total polyphenolic compound from water-soluble extracts of leaves and stem bark of M. alba and C. tricuspidata were 1.32%, 1.28%, 1.34% and 1.30% respectively. In these results, the water-soluble extract of stem bark from Cudrania tricuspidata showed the highest antioxidative activity.
Water extracts of root, leaf, branch and fruit of Cudrania tricuspidata were tested to use as natural antiwrinkle and whitening cosmetic ingredient. Root extract indicated 86.5 mg/g polyphenol concentration and 55.5 mg/g flavonoid concentration. From MTT test, root, branch and fruit extract did not show cell toxicity up to $1,000{\mu}g/mL$, however, leaf extract displayed significant cell toxicity in concentrations higher than $200{\mu}g/mL$. Cudrania tricuspidata root extracts displayed excellent 81% tyrosinase inhibition (whitening effect), 58% elastase inhibition (antiwrinkle effect) at $500{\mu}g/mL$ and showed minor UVB absorbance. When solution and lotion formulation were made including Cudrania tricuspidata root extract, they are stable at $25^{\circ}C$ for 21 days, but showed significant deviation in viscosity and particle size at $47^{\circ}C$. From the study, Cudrania tricuspidata root extract demonstrated good potential for natural whitening and antiwrinkle cosmetic agent.
High-performance liquid chromatography coupled to an online $ABTS^+$-based assay (online HPLC-$ABTS^+$) system was used to determine the principal antioxidants in Cudrania tricuspidata fruits. Six prenylated isoflavonoids (1 - 6) were isolated from C. tricuspidata fruits according to the online HPLC-$ABTS^+$ system. The structures of isolated compounds, alpiniumisoflavone (1), 6,8-diprenylorobol (2), 6,8-diprenylgenistein (3), pomiferin (4), 4'-methylalpiniumisoflavone (5), and osajin (6) were identified by their retention time, UV spectra, ESI-MS, and NMR data. Among these compounds, 6,8-diprenylorobol (2) and pomiferin (4) reduced A2E photooxidation in a dose dependent manner.
Cudrania tricuspidata Bureau (Moraceae) is a traditional oriental medicine that has been widely used as anti-oxidant, anti-inflammatory and immunomodulatory in Korea. This study was performed that the 70% ethanol extract of the roots of C. tricuspidata (CTE) suppressed receptor activator of NF-${\kappa}B$ ligand (RANKL)-induced osteoclastogenesis, actin ring formation in RAW 264.7 cell lines. CTE significantly inhibited the JNK/mitogen-activated protein kinase (MAPK) signaling pathway without affecting ERK and p38 signaling in RANKL-stimulated RAW 264.7 cells. Also, CTE inhibited RANKL-induced expression of c-Fos, an upstream activator of NFATc1. Consequently, CTE suppresses osteoclast differentiation by inhibiting RANKL induced MAPK signaling pathways and disrupts the actin rings in mature osteoclasts. Thus, CTE can be used for the development of osteoporosis treatment drug with a natural material.
In this study, silkworms fed Cudrania tricuspidata leaves were powdered, and hydrolytic enzymes, including viscozyme, papain, and flavourzyme, were added to verify the functionality of the different mixtures. In the general component analysis, the C. tricuspidata silkworm (CS) group exhibited higher crude protein and ash content than did the other groups, and the enzyme-treated groups exhibited higher carbohydrate content than the CS group. The DPPH and ABTS radical scavenging activities were significantly higher in the CS treated with viscozyme (CSV) and the CS treated with viscozyme/flavourzyme (CSVF) than in the other groups, with the CSV group showing the highest reducing power. ACE inhibitory activity was significantly higher in the CS treated with visocozyme/papain (CSVP) than in the CS group. In conclusion, rather than using powdered silkworms fed C. tricuspidata leaves, it would be more effective to use hydrolysates from C. tricuspidata silkworms as raw materials for functional foods.
The purpose of this paper is to describe the differences in mineral, ascorbic acid, and total polyphenol content, and also the relationship between polyphenol content and antioxidant activity(DPPH radical scavenging, FTC, and TBA methods) in three sizes of Cudrania tricuspidata leaves. For the mineral contents of the leaves, Ca was highest(17.26 mg/g.d.w.), followed by K(15.53 mg), P(2.61 mg), and Mg(1.99 mg). Ascorbic acid was found to be slightly higher(43.4 mg/100g.f.w.) in the basal parts of the medium size leaves and lowest in the whole parts(9.9 mg) of the small size leaves. Total polyphenol content was highest($1,384{\sim}1,258mg/100g.f.w.$) in the leaves, for all sizes, when extracted with 80% ethanol, followed by the stem bark(543 mg%), roots(369 mg%), stems(243 mg%), and thorns(223 g%). By determining DPPH activity, we found that antioxidant activity was higher in the ethanol extracts of the leaves than in the ethanol extracts of the thorns, stems, roots, and stem bark. While in the tea, no differences were found between the ethanol and water extracts.

뽕나무 (Morus alba) 및 꾸지뽕나무(Cudrania tricuspidata) 잎으로부터 추출한 수용성 물질을 0.5% 콜레스테롤 투여 식이에 1%(w/w) 수준으로 첨가하여 각 조직에서의 지질 과산화물 생성정도를 나타내는 TBARS 측정법으로 항산화 활성을 검토하였다. 흰쥐의 체중 및 식이 섭취량은 실험군 간의 유의적인 차이는 없었다. 간장 및 뇌 조직 중량은 뽕잎 추출물군에서 증가하였다. 신장의 지질 과산화물 함량은 대조군에 비해서 뽕잎 추출물군에서 유의적으로 감소하였다. 뇌 및 심장의 지질 과산화에서 유의적으로 감소하였다. 뇌 및 심장의 지질 과산화물 함량은 대조군에 비교해서 꾸지뽕잎 추출물군에서 유의적으로 증가하였다. 간장 및 비장의 경우에는 각 실험군 간에 유의적인 차이가 없었다. 각 조직의 microsomes 분획으 지질 과산화물은 심장과 비장에서 뽕잎 및 꾸지뽕잎 추출물군에서 감소하였으나, 다른 조직에서는 현저한 차이가 없었다. 한편, 각 조직의 microsomes 분획을 얻어서 Fe2+/ascorbate로 지질 과산화물을 유도하기 위하여 37$^{\circ}C$에서 한시간 반응을 시킨 결과, 뽕잎 및 꾸지뽕잎 추출물군에서 약 절반 정도 감소하였으며, 상대적 증가량은 뇌조직 microsomes에서 현저하였다. 본 실험의 결과 신장조직 및 간장 microsome막 지질 과산화물 함량은 뽕잎 및 꾸지뽕잎 수용성 추출물에 의해 감소되었으며, 뇌 및 심장조직의 지질 과산화물 함량은 꾸지뽕잎 수용성 추출물에 의해 증가됨으로써 봉잎 추출물간의 차이를 나타내었다.
