Viruses and Symptoms on Peppers, and Their Infection Types in Korea (우리나라 고추 바이러스 종류, 병징 및 발생 형태)

  • The incidence of virus disease on peppers was investigated at the 52 areas in the whole country in 2002, 2004 to 2006. Among the six viruses, Cucumber mosaic virus (CMV), Pepper mottle virus (PepMoV), Pepper mild mottle virus (PMMoV), Broad bean wilt virus 2 (BBWV2), Tobacco mild green mosaic virus (TMGMV) and Tomato spotted wilt virus (TSWV), infecting peppers in Korea, the incidence of CMV, PepMoV, PMMoV and BBWV2 was 29.4%, 13.6%, 14.3%, 25.6%, orderly. TMGMV and TSWV had the same low infection rate of 1.0%. The infection rate of CMV was higher as 53.3% and 34.2% in 2002 and 2004, but it was decreased to 18.2% and 11.9% in 2005 and 2006, respectively. The infection rate of BBWV2 was lower as 1.3% in 2002 and 1.8% in 2004, but it was increased abruptly to 41.3% in 2005 and 58.2% in 2006. For the types of mixed infections of pepper viruses, the incidence of CMV+PepMoV was 62.6% in 2002 and 50.0% in 2004, and that of CMV+BBWV2 was increased suddenly from 33.3% in 2005 to 83.2% in 2006. The triplex infection of CMV+BBWV2+PepMoV was 6.4% in average. CMV caused severe mosaic and BBWV2 induced ring spots, and the two mixed virions caused chlorosis on the leaves of red peppers. TSWV induced the typical symptoms of multiple ring spots on the leaves and fruits of red peppers.

  • The genetic diagnostic method of virion capture (VC)/RT-PCR was developed for the simultaneous detection of three rod shaped viruses of Cucumber green mottle mosaic virus (CGMMV), Kyuri green mottle mosaic virus(KGMMV) and Zucchini green mottle mosaic virus (ZGMMV) transmitted by seed in Cucurbit. Out of 12 primer combinations for the three tobamoviruses, a primer set of CGMMV-C724, KGMMV-K513 and ZGMMV-Z407A was useful for mono and triplex VC/RT-PCR. The triplex VC/RT-PCR for the three tobamovirus in Cucurbit could detect specifically without interference among primers and/or plant species of watermelon, gourd, cucumber, melon, pumpkin, squash and Nicotiana benthamiana.

Inhibitory Effect of Cheese Whey on Cucumber Mosaic Virus and Pepper Mottle Virus in Capsicum annuum (치즈 유청의 오이모자이크바이러스와 고추모틀바이러스 감염 억제 효과)

  • Evaluations were made for the effects of cheese whey treatment on infection of pepper plants by cucumber mosaic virus-Vch (CMV-Vch) and pepper mottle virus-Kr (PepMoV-Kr). In a greenhouse, pepper plants sprayed with whey, prior to inoculation by CMV-Vch using aphids, showed a viral infection rate significantly lower (6.6%) than for the control (23.3%). In an open field experiment, in which CMV infection relied on natural transmission by aphids, pepper plants were sprayed with undiluted whey once a week, starting on the transplanting date (May 2) to the end of June. On June 5, these whey-sprayed plants showed a CMV infection rate reduced by 18.9% and 16.7%, compared to untreated and pesticide-treated plants, respectively. In the greenhouse, pepper plants inoculated with PepMoV-Kr mixed with whey showed a viral infection rate decreased by 60% compared to the control. The accumulated amount of PepMoV-Kr coat protein was less than that for the virus-only control at 6 days post inoculation (dpi), but increased up to a similar level as the control at 9 dpi. This study showed that cheese whey is effective in reducing infection of both CMV and PepMoV in pepper plants.

Occurrence of Virus Diseases on Cucumber in Gyeongbuk Province (경북지역 오이에 발생하는 주요 바이러스 종류 및 발생실태)

  • Cucumber is high valued cash crop, for it is grown during the winter season in plastic house. Recently, virus disease spread widely in cucumber growing area and cause severe income loss. Therefore, occurrence of virus disease on cucumber were surveyed from 2004 to 2006 in Sangju and Gunwi area, Gyeongbuk province. The rate of plastic house which has infected plants was $55.0{\sim}88.6%$. Infection rate was the highest at Sangju in 2006 than others and ranged from 15 to 90.0% per plastic house. The 217 samples showing virus symptom were analyzed by RT-PCR using appropriate detection primer. Zucchini yellow mosaic virus(ZYMV) has the highest infection rate(detected over 85%) and followed by Cucumber green mottle mosaic virus(CGMMV). But Watermelon mosaic virus-2(WMV-2) was not detected in our survey. Therefore, we conclude that ZYMV is major pathogene of virus disease on cucumber. ZYMV induced chlorosis and severe mosaic on the leaves and distortion on the surface of fruits.

An Unusual Potyvirus from Pepper in Taiwan (대만에서 고추에 발생한 미보고 Potyvirus에 관한 연구)

  • A virus which induced yellowing, vein banding and ruffling on pepper in the field was investigated. The virus reacted strongly with PVY - antiserum in ELISA, but not with antisera of cucumber mosaic virus, tobacco mosaic virus, tomato black ring virus, alfalfa mosaic virus, tomato spotted wilt virus, tobacco etch virus, pepper mottle virus, and tobacco ringspot virus. Electron micrographs revealed that the virus was a flexuous rod of 750-760nm in length. The virus was transmitted mechanically and by Myzus persicae in a nonpersistent manner. The host range was similar to that of PVY, except that Chenopodium amaranticolor and C. quinoa were infected systemiclly.

Detection of Virus in Fruit and Seed of Vegetables Using RT-PCR (RT-PCR에 의한 과채류 열매 및 종자의 바이러스 검정)

  • Tobacco mosaic tobamovirus (TMV), cucumber mosaic cucumovirus (CMV), cucumber green mottle mosaic tobamovirus (CGMMV) and zucchini yellow mosaic potyvirus (ZYMV) from individual fruits and seeds of hot pepper and cucumber were detected by the reverse transcription-polymerase chain reaction (RT-PCR). The dilution end-points for RT-PCR in curde sap from TMV. and CMV - infected hot pepper leaves and CMV - and CGMMV-infected cucumber leaves were 10-5. However, the amount of PCR product obtained from preparation of ZYMV-infected cucumber leaf was 10-fold lower than those of CMV or CGMMV-infected cucumber leaves. In hot pepper, both TMV and CMV were detected in all parts of the fruit wall tissue, but the yields of PCR products in the fruit stalk and its surrounding tissues were higher than those of the end parts of the fruit. On the other hand, in cucumber fruit infected with CMV, CGMMV or ZYMV, the fruit wall tissue and seed located in both stalk and end parts showed higher yields of PCR products than those of intermediate parts. Of five viruses that were analysed, only TMV in hot pepper seed, and CGMMV and CMV in cucumber seed were detected in testa parts.

Studies on the Soil Transmission of CGMMV and Its Control with Crop Rotation (오이녹반모자이크바이러스의 토양전염 생태 및 윤작에 의한 방제)

  • Cucumber green mottle mosaic virus (CGMMV) is one of major plant viruses infecting cucurbitaceous crops via soil or infected seeds. This study investigate ecology of infection of CGMMV in soil, and control tactics of this virus with soil hygiene and crop rotation. This virus was survival to 50% in soil without host plants for 17 months and had high vitality in debris of infected plant over 1 year. Infection rate of CGMMV was 1.0~3.6% in control soil and 12~36% in soil transplanted with wounded root of watermelon. It showed that wounded root may affect severity of soil infection. Rotation between rice and watermelon caused dramatical reduction from 76.8% in repeated cultivation to 7.3% of progeny infection by CGMMV. Therefore, it is suggested that crop rotation be effective for control of CGMMV.
