철염 응집제 중 유독성 중금속의 선택적 제거 (Selective Removal of Toxic Heavy Metals in Fe-Coagulants)

  • Among various reactions which metal sulfides can undergo in the reducing environment, the lattice exchange reaction was examined in a attempt to selectively remove heavy metal ions contained in the Fe-Coagulants acid solution. We have examined Zeta potential along with pHs to investigate surface characteristics of ${FeS}_{(s)}$. As a result of this experiment, zero point charge(ZPC) of FeS is pH 7 and zeta potential which resulted from solid solution reaction between Pb(II) and ${FeS}_{(s)}$ is similar to that of ${PbS}_{(s)}$. Solubility characteristics of ${FeS}_{(s)}$ is appeared to that dissolved Fe(II) concentration increased in less than pH 4, and also increased with increasing heavy metal concentration. Various heavy metal ions(Pb(II), Cu(II), Zn(II)) contained in Fe-coagulants acid solution were removed selectively more than ninety-five percent in the rang of pH 2.5~10 by ${FeS}_{(s)}$. From the above experiments, therefore, We could know that the products of reaction between heavy metal ions and $FeS_{(S)}$ are mental sulfide such as $PbS_{(S)}$, $CuS_{(S)}$ and $ZnS_{(S)}$.

내분비계 장애물질인 Bisphenol A의 free radical 생성을 통한 독성발현 (Cytotoxicity of Environmental Estrogenic Compound, Bisphenol A, via Generation of Free Radicals)

  • Bisphenol A shares similarities in structure, metabolism and action with DES, a known human teratogen and carcinogen. Bisphenol A, a monomer of polycarbonate and epoxy resins, has been detected in canned food and human saliva. The purpose of the this study was to evaluate the cytotoxicity, cell proliferation of bisphenol A In the presence of a rat liver S9 mix, contaning cytochrome P450 enzymes, and Cu (II). In the present study, Bisphenol A in combination with Cu (II) exhibited a enhancement in cytotoxicity which were inhibited by free radical scavengers. The content of malondialdehyde, an end product of lipid peroxidation, was also found to increase with concentration of bisphenol A. Also, we examined the change of CuZn-SOD, Mn-SOD, catalase and GPx activities in the MCF-7 cells exposed to bisphenol A. The activities of CuZn-SOD, CPx, catalase were found to decrease with bisphenol A concentration. Meanwhile, the activity of Mn-SOD was unchanged. This indicated that elevated oxidative stress caused by imbalance between the production and removal of free radicals occurred in cells.

Expression and Characterization of Recombinant Human Cu,Zn-Superoxide Dismutase in Escherichia coli

  • Expression of human Cu.Zn-superoxide dismutase (SOD) with activity comparable to human erythrocyte enzyme was achieved in E. coli B21(DE3) by using the pET-17b expression vector containing a T7 promoter. Recombinant human SOD was found in the cytosol of disrupted bacterial cells and represented > 25% of the total bacterial proteins. The protein produced by the E. coli cells was purified using a combination of ammonium sulfate precipitation, Sephacryl S-100 gel filtration and DEAE-Sephacel ion exchange chromatography. The recombinant Cu,Zn-SOD and human erythrocyte enzyme were compared using dismutation activity, SDS-PAGE and immunoblotting analysis. The mass of the subunits was determined to be 15,809 by using a electrospray mass spectrometer. The copper specific chelator. diethyldithiocarbamate (DOC) reacted with the recombinant Cu,Zn-SOD. At $50{\mu}M$ and $100{\mu}M$ concentrations of DOC, the dismutation activity was not inhibited for one hour but gradually reduced after one hour. This result suggests that the reaction of DOC with the enzyme occurred in two distinct phases (phase I and phase II). During phase I of this reaction, one DOC reacted with the copper center, with retention of the dismutation activity while the second DOC displaced the copper, with a loss of activity in phase II.

금호강 수 중의 중금속류의 장기변도 (Long-Term Change of Heavy Metal Concentration in the Kumho River Water)

  • In order to study the long-term change of heavy metal concentrations in the Kumho river water, water analysis was conducted at 13sites surrounding the Kumho river system for 18times from September 1993 to August 1999. Analytical items for the study of water quality are Cu, Zn, Cd, Cr, Fe, Mn and Pb. The six year term studied in this work was divided into Part I and Part II, which covers the period from September 1993 to August 1996 and the period from September 1996 to August 1999, respectively. The mean concentrations of Cu, Zn, Cd, Cr, Fe, Mn and Pb in the unit of ppm for the Part I period showed 0.032, 0.025, 0.006, 0.050, 0.053 and 0.019, respectively. The mean concentrations of Cu, Zn, Cd, Cr, Fe, Mn and Pb in the unit of ppm for the Part II period showed 0.001, 0.001, 0.001, 0.004, 0.020, 0.002 and 0.002, respectively. The heavy metal concentrations in the Kumho river water for te second period were found to be decreased by 1/32, 1/25, 1/6, 1/1.5, 1/2.5, 1/26.5 and 1/9.5 for Cu, Zn, Cd, Cr, Fe, Mn and Pb, respectively. The present results clearly indicate that the water quality in the Kumho river is improving in terms of heavy metal contaminations.

납-티오황산 착물생성과 구리치환에 의한 미량 납(II)의 비색분석에 관한 연구 (Spectrophotometric Determination of Trace Lead(II) After Extraction of Lead-Thiosulfate Complex into Aliquat336-CHCl$_3$ and Replacement by Cu)

  • A spectrophotometric method was developed for the acidic solution stripped after an extraction of 0.5 to 2.5 ppm of Lead(II) from 50 mL of $Na_2S_2O_3$ solution into chloroform as the ion-pairs formed between their thiosulfate complexes and alkylamine, Aliquat336. Pb(II) in the stripped solution forms an complex with DDTC in pH 7.3 buffer solution, and was developed in yellow by copper replacement. The ydlow-colored solution have the maximum absorbance at 435 nm in the measurement of absorbance by UV-Visible spectrophotometer. The interference ions such as Fe(III), Hg (II), Al(III), Co, Cu, Ni, Zn, Ca, Sn, have great effects on the extraction, but they were overcomed by the usage of adequate masking agents before an extraction. At last, a good result was obtained in applying this method to synthetic water.

(Polystyrene-divinylbenzene)-thiazolylazo phenol형 킬레이트 수지에 의한 금속이온의 분리 및 농축에 관한 연구(II) (Studies on the Separation and Preconcentration of Metal Ions by Chelating Resin containing (Polystyrene-divinylbenzene)-thiazolylazo phenol Derivatives(II))

  • 해수 중 U(VI) 등과 같은 미량 금속이온의 선택적인 분리, 농축 및 회수를 위하여 XAD-16-[2-(2-thiazolylazo)-p-cresol](TAC)형 킬레이트 수지에 대한 몇 가지 금속이온의 흡착 및 탈착 특성을 용리법으로 조사하였다. 금속이온의 흡착에 미치는 흐름속도, pH 및 완충 용액의 농도에 대한 영향을 조사하여 흡착 최적 조건을 결정하였다. 킬레이트 수지에 대한 금속이온의 총괄 흡착 용량을 조사한 결과 각각 0.41mmol U(VI)/g resin, 0.55mmol Th(IV)/g resin, 0.43mmol Cu(II)/g resin 및 0.32mmol Zr(IV) /g resin이었다. pH 5.0에서 돌파점 용량과 총괄 용량으로부터 얻은 금속이온의 용리 순서는 Th(IV)>Cu(II)>U(VI)>Zr(IV)>Pb(II)>Ni(II)>Zn(II)>Cd(II)>Mn(II)이었다. $HNO_3$, HCl, $HClO_4$, $H_2SO_4$$Na_2CO_3$ 등의 탈착제에 의한 탈착특성을 조사한 결과 Zr(IV)을 제외한 대부분의 금속이온들에 대하여 2M $HNO_3$이 탈착효율이 높게 나타났으며, 1M $H_2SO_4$ 용액을 사용하면 Zr(IV)의 탈착 및 회수가 가능하였다. 또한 킬레이트수지를 이용하여 미량의 U(VI) 이온이 함유된 인공 해수 시료를 용리시키고 회수한 결과 96% 이상의 높은 회수율을 나타내었다.

  • 두 개의 피리딘기를 포함하는 열린고리 형태의 여섯자리 질소계 리간드 1,15-bis(2-pyridyl)-2,6,10,14-tetraazapentadecane tetrahydroehloride (bpyped 4HCl)을 합성하고 원소분석, 적외선 분광법, 핵자기 공명법 및 질량 스펙트럼으로 분석하였다. 합성된 리간드의 산 해리상수($logK{^n}_H$)와 Cu(II), Ni(II), Co(II) 및 Zn(II) 에 대한 안정도 상수값을 수용액에서 전위차 적정법으로 구하였고, 이때 온도는 $25^{\circ}C$ 및 이온강도 0.10 M($KNO_3$)으로 고정시켰다. 지방족 아민의 질소 원자들 사이에 한 개 또는 두 개의 프로필기를 포함하는 bpyped와 유사한 리간드들의 지방족 아민 고리 길이 효과에 따른 산 해리 상수 및 전이금속(II)에 대한 안정도 상수 영향을 알아보았다.

  • Triton X-100이 함유된 상태에서 정색시약인 1-(2-thiazolylazo)-2-naphthol이 첨가된 물에서 구리 (II), 니켈(II)과 아연(II)의 동시 분광광도법적 정량을 위한 다변량 모델들이 개발되었다. 분광학적 간섭의 단점을 극복하기 위해서, 주성분회귀분석법(PCR)과 부분최소자승법(PLS) 다변량 분석법적 접근이 적용되었다. 다양한 시험 세트를 사용하여 본 방법의 수행이 입증되었고 그 결과들이 비교되었다. 일반적으로 PLS와 PCR 모델들 사이에 분석적 수행에서의 심각한 차이가 없었다. $Cu^{2+}$, $Ni^{2+}$ and $Zn^{2+}$ 의 세 성분들을 사용한 예측의 제곱근 평균 제곱 오차(RMSEP)들은 각각 0.018, 0.010, 0.011 ppm이었다. 또한 감도, 분석감도, 검출한계(LOD)와 같은 가치들의 측면들이 평가되었다. 본 논문에서 제안하는 과정이 화합물 혼합용액과 수돗물 속의 $Cu^{2+}$, $Ni^{2+}$ and $Zn^{2+}$의 동시 검출에 적용되었을 때에 높은 신뢰도가 성취되었다.

La이 도핑된 CuO-ZnO-Al2O3 복합 산화물의 합성공정개발 (Development and Synthesis of La Doped CuO-ZnO-Al2O3 Mixed Oxide)

  • La doped CuO-ZnO-$Al_2O_3$ powders are prepared by sol-gel method with aluminum isopropoxide and primary distilled water as precursor and solvent. In this synthesized process, the obtained metal oxides caused the precursor such as copper (II) nitrate hydrate and zinc (II) nitrate hexahydrate were added. To improve the surface areas of La doped CuO-ZnO-$Al_2O_3$ powder, sorbitan (z)-mono-9-octadecenoate (Span 80) was added. The synthesized powder was calcined at various temperatures. The dopant was found to affect the surface area and particle size of the mixed oxide, in conjunction with the calcined temperature. The structural analysis and textual properties of the synthesized powder were measured with an X-ray Diffractometer (XRD), a Field-Emission Scanning Electron Microscope (FE-SEM), Bruner-Emmett-Teller surface analysis (BET), Thermogravimetry-Differential Thermal analysis (TG/DTA), $^{27}Al$ solid state Nuclear Magnetic Resonance (NMR) and transform infrared microspectroscopy (FT-IR). An increase of surface area with Span 80 was observed on La doped CuO-ZnO-$Al_2O_3$ powders from $25m^2$/g to $41m^2$/g.

폴리히드라존계 킬레이트 고분자의 합성과 특성 (Synthesis and Characterization of Chelated Polymers of Polyhydrazones)

  • $p^-$, $m^-$, o-phthalaldehyde 및 5,5'-methylene-bis-salicyl aldehyde (PPTA, MPTA, OPTA, MBSA)와 5,5'-methylene-bis-salicylic dihydrazide, terephthalic dihydrazide 및 sebacic dihydrazide(MBSDH, TDH 및 SDH)등의 디히드라지드 화합물을 DMF-$CH_3COOH$ 혼합용액 중에서 축합반응으로 몇가지 새로운 폴리히드라존을 합성하였다. 합성한 폴리히드라존과 그의 금속킬레이트의 용해성, 흡수스펙트라 및 열적성질에 대하여 검토하였다. OPTA와 MBSDH의 반응으로 얻어진 폴리히드라존을 제외한 폴리히드라존과 그의 금속킬레이트는 일반적으로 보통 유기용매에 불응성이었다. 폴리히드라존의 중량열분석 결과 250-350$^{\circ}$C 사이에서 10%의 중량감소가 일어났고 500$^{\circ}$C고 500 폴리히드라존의 중량열분석 결과 250~350$^{\circ}$C 사이에서 10%의 중량감소가 일어났고 500$^{\circ}$C에서 32.5~63.5% 범위의 잔유중량을 나타내었다. 합성한 폴리히드라존 중에서 PPTA-MBSDH(IIa)및 MBSA-MBSDH(IIIa)가 열분해온도가 비교적 높았으며 IIa의 금속킬레이트에 있어서는 Zn(II)-IIa>Ni(II)-IIa>Co(II)-IIa>Cu(II)-IIa의 순서로 열분해온도가 저하되었다.
