  • Objective : This study was performed to evaluate the effect of immune reaction inductive substances such as phorbol-myristate-acetate(PMA), lipopolysaccharide(LPS), dermato-phagoides pteronyssus crude extract(DPE), dinitrochloro-benzene(DNCB) and Cryptotympana pustulata(CP), the Cryptotympana pustulata extracting substance at simultaneously on the translocation of nuclear factor-kappa B(NF-${\kappa}B$) towards to the nucleus and the mRNA expression patterns of various cytokine genes in Human acute monocytic leukemia cell line(THP-1 cells), monocytes of human. Experiment : To analyze cytokine genes expression patterns, the RT-PCR method was used, measuring tumor necrosis factor(TNF)-$\alpha$ that had been secreted during cell culture in the ELISA method. The morphological change in the cell observed during THP-1 cell culture was observed using a scanning electron microscope (SEM) and the quantitative distribution in the cell NF-${\kappa}B$ was analyzed through immunocytochemistry and a confocal microscopy. Result : CP showed different influences onto the mRNA expression patterns of cytokine genes with PMA, LPS. DPE and DNCB according to the types of immune inductive substances in the THP-1 cells. The expressions of inter-leukin(IL)-10, interferon(INF)-$\gamma$, TNF-$\alpha$ and monocyte chemoattractantant protein(MCP)-1 induced by PMA were suppressed by CP while the expression of transforming growth factor(TGF)-$\beta$ was promoted. Regarding the secretion pattern of TNF-$\alpha$ according to PMA processing, its secretion amount was increased by CP concurrent processing, in case of processing CP onto PMA and LPS, We discovered that the secretion amount of TNF-$\alpha$ was increased. Upon processing PMA and LPS on the THP-1 cell strain at the same time or either additionally processing CP thereon, the movement increase towards the nucleus from the NF-${\kappa}B$ cell cytoplasm, a transcription factor was able to be observed. Conclusion : In this study, Cryptotympana pustulata extracting substance was confirmed that it had an influence on expression patterns of cytokine genes according to the actions of a variety kinds of immune reaction inductive substances processed on the monocyte THP-1 cell of humans. Therefore, additional studies as for the immune adjusting function of Cryptotympana pustulata are considered to be able to offer important materials for curing immune abnormal diseases such as atopy dermatitis afterward.

Antimicrobial activity of Medicinal Plant Extracts against Streptococcus mutans

  • This study was carried out to research antimicrobial agents from medicinal plants, Lonicera japonica, Pinellia ternata, Dictamnus albus, Cryptotympana pustulata, Pinus densiflora, Bupleurum falcatum, Forsythia saxatilis, Castanea crenata, Hovenia dulcis, Prunus sargentii. The ethanol extracts of 10 medicinal plants were tested for the antimicrobial activity against Streptococcus mutans. The extracts of Pinus densiflora showed significant antimicrobial activity against Streptococcus mutans. These results suggested that the extract from Pinus densiflora could be a candidate for new antimicrobial agents against Streptococcus mutans.

  • This study was carried out to research antimicrobial agents from medicinal plants, Pueraria thunbergiana, Sophora flavescens, Lonicera japonica, Pinellia ternata, Dictamnus dasycarpus, Paeonia lactiflora, Angelica dahurica, Spirodela polyrhiza, Cryptotympana pustulata, Pinus densiflora, Cimicifuga heracleifolia, Bupleurum falcatum, Magnolia kobus, Artemisia princeps, Houttuynia cordata, Forsythia koreana, Arctium lappa, Castanea crenata, Aster tataricus, Citrus unshiu, Gardenia jasminoides, Ledebouriella seseloides, Hovenia dulcis, Prunus sargentii. The ethanol extracts of 24 medicinal plants were tested for the antimicrobial activity against Streptococcus mutans. The extracts of Sophora flavescens, Pinus densiflora, Cimicifuga heracleifolia, Gardenia jasminoides and Ledebouriella seseloides showed antimicrobial activity against Streptococcus mutans. The extracts of Pinus densiflora and Sophora flavescens among these medical plants showed significant antimicrobial activity against Streptcoccus mutans. These results suggested that the extracts from Pinus densiflora and Sophora flavescens could be the potential source of antimicrobial agents against S. mutans.

수종 곤충류 한약재 및 함유 아미노산의 알레르기 질환 치료의 유용성 연구 (The potential usefulness of several medicinal insects and their major amino acids in allergic disorders )

  • Objectives : Edible insects are the widely distributed group of animals and contain high quality proteins, fatty acids and minerals. In particular, insects are a possible interesting source of essential amino acids. Insects as traditional medicines have much potential as pharmaceuticals in modern medicines including treating infections, cancer, dissolving phlegm, relieving spasms, and inflammatory diseases. The aim of this study is to investigate the immunomodulatory effect of several kinds of insects and major amino acids. Methods : In our review we try to show the potential usefulness of insects and amino acids in searching for new therapeutic solutions for immunologic diseases. We summarized the knowledge about properties, usefulness of insect and amino acids in drug design. We hypothesized that insects and amino acids, their major ingredients, regulates airway inflammation and immunologic diseases and can be developed as therapeutic drugs for the treatment of immunologic diseases. Results : Several insects including Bombyx mori, Cryptotympana pustulata, Holotrichia diomphalia, Locusta migratoria, etc. and amino acids such as glutamine, glutamic acid, methionine and glycine may have potential protective effects against asthma and airway neutrophilia. Glutamic acid, cystine, methionine and glycine which contribute to glutathione metabolism, which are important anti-oxidant amino acids that may affect susceptibility to asthma. Conclusions : Our results provide evidence about the potential usefulness of several insects and their amino acids in allergic disorders. These findings suggest that several insects and amino acids have important roles in the way they affect the immune system and allergic responses.

The Slough of Cicadidae Periostracum Ameliorated Lichenification by Inhibiting Interleukin (IL)-22/Janus Kinase (JAK) 1/Signal Transducer and Activator of Transcription (STAT) 3 Pathway in Atopic Dermatitis

  • It is known that animal-origin medicine could be one of effective treatment to remedy atopic dermatitis (AD) by controlling the cytokines. Cicadidae Periostracum (CP), the slough of Cryptotympana pustulata, has been frequently used for treating AD and skin affliction in traditional Korean Medicine. This study is aimed at investigating the ameliorating effects of CP on AD and its potential mechanism. The dinitrochlorobenzene sensitized mice were treated with CP for 2 weeks. The various biomarkers and the dermatitis scores presented that CP treatment can induce the visual and biological improvements of AD model. Pruritus, the most serious symptom of AD, which can cause repeated scratching behaviors and finally lead to lichenification, was reduced with CP treatment by regulating the inflammatory reactions. In addition, CP treatment diminished the number of mast cells that are known for causing inflammatory reactions. Moreover, it is proven that CP can decline secretion of interleukin-22, which means CP treatment has anti-inflammatory effects. CP treatment can correct the imbalance of helper T (Th)1 and Th2, downregulating thymic stromal lymphopoietin that leads to decrease of mRNA level of inflammatory cytokines. The crucial role of CP treatment is controlling of the Janus kinase 1/signal transducer and activator of transcription 3 pathway. In addition, CP treatment has the inhibitory effects on kallikrein related peptidase (KLK) 5 and KLK7. Taken together, CP treatment can ameliorate most symptoms and problems caused by AD disease, improving the AD patients' life quality.
