정수처리 공정에서 Cryptosporidium Tracer의 제거효율 (Removal Efficiency of Cryptosporidium Tracer in Drinking Water Treatment Process)

  • 정수처리 과정에서 Cryptosporidium과 유사한 특성을 가지고 있는 C. tracer를 이용하여 공정별 제거효율을 조사하였다. PACI(Poly aluminium chloride, $Al_2O_3$(10%)) 주입량이 10 mg/L일 때 C. tracer는 97.16%로 가장 높은 제거율을 보였으며, 탁도 제거율과 SS 제거율이 높을수록 C. tracer 응집 효율이 높았다. 원수의 pH가 높을수록 C. tracer 제거율이 증가 하였으며 응집 침전 후의 유출수 탁도와 C. tracer 제거율과의 상관성이 $R^2=0.9506$로 높게 나타나 응집 침전 후의 유출수 탁도로 Cryptosporidium 제거 효율을 평가할 수 있음을 알 수 있었다. 또한 여과 실험에서는 유입 탁도에 따른 C. tracer 제거율은 $94.00{\sim}95.83%$ 범위였으며 유출수 탁도와 C. tracer 제거율과의 상관성은 $R^2=0.8704$였다. 최적 응집 조건 하에서 여과수 탁도가 양호할 경우, 예상되는 Cryptosporidium의 제거율은 응집 침전의 경우 1.55 log(97.16%), 급속모래여과의 경우 1.38 log(95.83%), 응집 침전+급속모래여과의 경우 2.31 log(99.51%)로 나타났다.

  • In order to examine the prevalence of Cryptosporidium infection in wild rodents and insectivores of South Korea and to assess their potential role as a source of human cryptosporidiosis, a total of 199 wild rodents and insectivore specimens were collected from 10 regions of South Korea and screened for Cryptosporidium infection over a period of 2 years (2012-2013). A nested-PCR amplification of Cryptosporidium oocyst wall protein (COWP) gene fragment revealed an overall prevalence of 34.2% (68/199). The sequence analysis of 18S rRNA gene locus of Cryptosporidium was performed from the fecal and cecum samples that tested positive by COWP amplification PCR. As a result, we identified 4 species/genotypes; chipmunk genotype I, cervine genotype I, C. muris, and a new genotype which is closely related to the bear genotype. The new genotype isolated from 12 Apodemus agrarius and 2 Apodemus chejuensis was not previously identified as known species or genotype, and therefore, it is supposed to be a novel genotype. In addition, the host spectrum of Cryptosporidium was extended to A. agrarius and Crosidura lasiura, which had not been reported before. In this study, we found that the Korean wild rodents and insectivores were infected with various Cryptosporidium spp. with large intra-genotypic variationa, indicating that they may function as potential reservoirs transmitting zoonotic Cryptosporidium to livestock and humans.

크립토스포리디움 활성 및 감염성 판정을 위한 direct RT-PCR, cell culture RT-PCR 및 cell culture IFA의 비교 (Comparison of Direct RT-PCR, Cell Culture RT-PCR and Cell IFA for Viability and Infectivity Assay of Cryptosporidium)

  • 크립토스포리디움의 활성 및 감염성 판정을 위해 Direct RT-PCR, 세포배양 후 RT-PCR 및 면역형광염색법을 비교한 결과는 다음과 같다. 1) 크립토스포리디움의 HSP70 gene에 대해 direct RT-PCR한 결과, 민감도가 매우 높아 저농도로 존재하는 환경시료에서의 크립토스포리디움 활성을 모니터링하는데 장점이 있을 것으로 보이나, 감염성의 판정은 알 수 없으며, 정량화가 안되는 단점이 있었다. 2) ${\beta}-tubulin$ gene에 대해 RT-nested PCR을 한 결과 크립 토스포리디움의 난포낭이 $1{\times}10^4$ 세포수정도 되어야 검출이 되는 것으로 나타나 HSP70 gene에 대한 RT-PCR결과와 비교할 때 10,000배 이상 민감도가 떨어지는 것으로 나타났다. 3) 세포배양 후 RT-PCR 또는 면역형광염색법을 이용할 경우에는 민감도가 direct RT-PCR보다 다소 떨어지는 단점이 있었으나 크립토스포리디움의 오염원이나 오염이 심한 지역의 감염성 조사에 적합할 것으로 나타났으나, 정량화가 필요한 경우에는 세포배양 후 면역형광염색법이 효과적일 것으로 나타났다.

Comparative Sensitivity of PCR Primer Sets for Detection of Cryptosporidium parvum

  • Improved methods for detection of Cryptosporidium oocysts in environmental and clinical samples are urgently needed to improve detection of cryptosporidiosis. We compared the sensitivity of 7 PCR primer sets for detection of Cryptosporidium parvum. Each target gene was amplified by PCR or nested PCR with serially diluted DNA extracted from purified C. parvum oocysts. The target genes included Cryptosporidium oocyst wall protein (COWP), small subunit ribosomal RNA (SSU rRNA), and random amplified polymorphic DNA. The detection limit of the PCR method ranged from $10^3$ to $10^4$ oocysts, and the nested PCR method was able to detect $10^0$ to $10^2$ oocysts. A second-round amplification of target genes showed that the nested primer set specific for the COWP gene proved to be the most sensitive one compared to the other primer sets tested in this study and would therefore be useful for the detection of C. parvum.

연세대학교 세브란스병원 환자에서의 Cryptosporidium 오오시스트 검출률 (Detection of Cryptosporidium oocysts from out-patients of the Severance Hospital, Korea)

  • 국내의 CUPtOSPOTidiUm 인체감염 실태를 조사하기 위하여 연세대학 세브란스 병원을 찾은 230명의 외래 환자 분변을 수거하였다. Acid-fast 염색, auramine-rhodamine 염색과 CWptosporidiwpowumoocyst에 특이적인 단클론 항체를 이용한 동정법을 이용하였다. 230명의 환자 중 48명(21%)이 AF 염색법에 의하여 50명(22%)이 AR 염색법에 의하여, 그리고 23명(10%)이 단클론 항체를 이용하는 형광현미경법으로 각각 Cryptosporidium에 감염된 것으로 조사되어 국내에서도 Cryptosporidium 인체 감염이 존재하고 있는 것으로 나타났다.

Occurrence of Cryptosporidium and Giardia and the Relationship between Protozoa and Water Quality Indicators in Swimming Pools

  • A total of 60 samples were collected from 35 swimming pools in Beijing, China, and the presence of Cryptosporidium and Giardia were investigated. The results showed that 16.7% and 15.0% of samples were positive for Cryptosporidium oocyst and Giardia cysts, respectively, with a mean concentration of 0.30 oocysts/10 L and 0.27 cysts/10 L. The oocysts and cysts were found to have higher rates of occurrence in August than in May. Genotyping confirmed the presence of Cryptosporidium hominis, C. parvum, and Giardia assemblages A and B, all of which were associated with human infections. The predominant species/assemblages were C. hominis and Giardia assemblage A. Analyses of the relationships between parasite oocysts/cysts, indicator bacteria, and physical-chemical parameters revealed that there was no correlation between 2 parasites and fecal bacterial indicators, whilst there was a significant correlation between protozoa and urea concentration, which indicates that urea concentration rather than fecal bacterial indicators might be an appropriate index for chlorine-resistant protozoa in swimming pools. This study provides useful information to improve the safety of swimming pool water and deduce the risk of protozoan infections.

  • AIDS 환자의 치명적인 2차 감염을 유발하는 Cryptosporidium parvum 의 Infective stage 인 sporozoites의 단일군 항체를 분리하였다. Oocysts를 효소처리하여 sporozoites를 excystation시킨 후 Isopycnic percoll gradients를 이용하여 sporozoites를 순수분리한 후 단일군 항체 생산을 위한 항원으로 사용하였다. 두 달된 BALB/c 쥐를 immunize한 후 splenocytes와 P3-X63-Ag8 myeloma cells를 융합시킨 후 hybridoma 기술을 이용해 Kor1(IgGl), Ea2(Ig2a) 두 clones을 분리하였으며 정제된 sporozoites를 SDS-PAGE로 분리한 후 Western blot을 이용하여 단일군 항체 Kor1과 Ea2는 20,000 daltons 크기의 항원을 인식하였다. Immunofluorescent assay에서 단일군 항체가 sporozoites 표면에 반응하는 것으로 보아 20-kDa 단백질 항원은 sporozoites 표면에 위치하는 항원으로 밝혀졌으며 C. parvum에 감염되었을 때 항체생성에 관여하는 중요 항원 중 하나일 것으로 추정되었다.

Cryptosporidium hominis Infection Diagnosed by Real-Time PCR-RFLP

  • There are approximately 20 known species of the genus Cryptosporidium, and among these, 8 infect immunocompetent or immunocompromised humans. C. hominis and C. parvum most commonly infect humans. Differentiating between them is important for evaluating potential sources of infection. We report here the development of a simple and accurate real-time PCR-based restriction fragment length polymorphism (RFLP) method to distinguish between C. parvum and C. hominis. Using the CP2 gene as the target, we found that both Cryptosporidium species yielded 224 bp products. In the subsequent RFLP method using TaqI, 2 bands (99 and 125 bp) specific to C. hominis were detected. Using this method, we detected C. hominis infection in 1 of 21 patients with diarrhea, suggesting that this method could facilitate the detection of C. hominis infections.

  • Cryptosporidium parvum causes a life-threatening diarrhea in acquired immunodeficiency syndrome (AIDS) patients. THe sporozoite stage of C. parvum has been known to be a target in treating cryptosporidiosis in AIDS patients as it is an extracellular stage. A sporozoite was ultrastructurally observed. It has a creascent shape with a rounded posterior end and a tapering body. The compact nucleus was located at the posterior end. A monoclonal antibody was produced, which recognized a 43 kDa of sporozoite antigens in a western blot analysis and showed the surface labeling in immunofluorescence.

Identification of Cryptosporidium from Dairy Cattle in Pahang, Malaysia

  • Cryptosporidium, a protozoan parasite, can cause cryptosporidiosis which is a gastrointestinal disease that can infect humans and livestock. Cattle are the most common livestock that can be infected with this protozoan. This study was carried out to determine the prevalence of Cryptosporidium infection in cattle in Kuantan, Pahang, Malaysia and to find out the association between the occurrence of infection and 3 different ages of cattle (calves less than 1 year, yearling, and adult cattle). The samples were processed by using formol-ether concentration technique and stained by modified Ziehl Neelsen. The results showed that 15.9% (24/151) of cattle were positive for Cryptosporidium oocysts. The occurrence of Cryptosporidium in calves less than 1 year was the highest with the percentage of 20.0% (11/55) followed by yearling and adult cattle, with the percentage occurrence of 15.6 % (7/45) and 11.8% (6/51), respectively. There was no significant association between the occurrence and age of cattle and presence of diarrhea. Good management practices and proper hygiene management must be taken in order to reduce the infection. It is highly important to control the infection since infected cattle may serve as potential reservoirs of the infection to other animals and humans, especially animal handlers.
