Pectin Microspheres for Oral Colon Delivery: Preparation Using Spray Drying Method and In Vitro Release of Release of Indomethacin

  • Drug delivery systems that are based on pectin have been studied for colon specific delivery using the specific activity of colon microflora. The aim of this study was to design a novel method of manufacturing pectin microspheres without oils and surfactants and to investigate the potential use of the pectin microspheres as an oral colon-specific drug carrier. The pectin microspheres were successfully formed using the spray drying method and crosslinking with calcium chloride. From the crosslinked pectin microspheres, indomethacin (IND) release was more suppressed than its release from non-crosslinked microspheres. In a low pH (pH 1.4) environment, the pectin microspheres released IND at an amount of about 18${\pm}$2% of the total loaded weight for 24 h while the release rate of IND was stimulated at neutral pH (pH 7.4). IND release from the pectin microspheres was increased by the addition of pectinase. The results clearly demonstrate that the pectin microspheres that were prepared by the spray drying and crosslinking methods are potential carriers for colon-specific drug deliveries.

키토산 가교 처리된 면직물의 천연염색에 관한 연구(II) - 오배자를 중심으로 - (Natural Dyeing of Chitosan Crossinked Cotton Fabrics (II) - Gallnut -)

  • For the purpose of standardization and practicability of natural dyeing, the mordanting and dyeing properties of gallnut was studied. In this study, the colorants of gallnut were extracted with boiling water. Chitosan crosslinked cotton fabrics were been dyed with aqueous extract of gallnut and their dyeabilities on the fabrics were studied. Additionally, the fastness to washing and light were also investigated. Cotton fabrics were treated with a crosslinking agent epichlorohydrin in the presence of chitosan to provide the cotton fabrics the dyeing properties of natural dye(gallnut) by the chemical linking of chitosan to the cellulose structure. The chitosan finishing and durable press finishing of the cotton fabrics carried out simultaneously in the mercerization bath. The dyeability(K/S), which was obtained by CCM observation, remarkably increased as the concentration of chitosan was high. Dye ability of gallnut showed higher toward chitosan treated cotton than controlled cotton fabric under condition at $60^{\circ}C$, for 20 min. The hue value indicated reddish yellow with increasing the crosslinked chitosan concentration. And the color fastness to washing and light was the almost the same.

TGA를 이용한 가교폴리에틸렌의 방사선 열화 평가 (Evaluation of Radiation Degradation or Crosslinked Polyethylene using TGA)

  • Radiation degradation of crosslinked polyethylene(XLPE) was investigated using thermogravimetric analysis(TGA), The results of TGA were compared with FT-IR, melting temperature, oxidation induction time, and elongation at break on the XLPE exposed by $\gamma$-ray. 5% decomposition temperature of $\gamma$-ray irradiated XLPE showed similar tendencies with the case of elongation at break. Both properties agreed below 1000 KGy, however, did not show any remarkable characteristics above 1000 kGy, these properties can be useful to evaluate the radiation degradation of XLPE for only low irradiated region. Above 1000 kGy, the thermal decomposition activation energy showed decreased, on the contrary, increasing below 1000 kGy. Compared with FT-IR spectrum of irradiated XLPE, it was confirmed that the oxidation reaction was still occurring below 1000 kGy. Radiation degradation of XLPE was dependent upon the irradiation doses, TGA can be a useful tool to evaluate the degradation.

  • To utilize collagen as an implantable biomateriall the mcct widely used bovine skin origin Type I collagen was investigated Pepsin treated, Type I atelocollagen was extracted and crosslinked by the ultraviolet(W) ray with wavelength of 254nm or by various concentrations of glutaraldehyde to produce collagen membranes. The crosslink rates of the specimens were observed by a polarized light microscope, a scanning electron microscope, and a Fourier transform infrared (FT-lR) spectrometer. The followings are concluded 1. The collagen membranes produced by both 2.5% glutaraldehyde solution and 254nm UV ray irra- diation demonstrated similar morphologies on polarized light microscopic and scanning electron microscopic views. 2. The chemical structures of the crosslinked membranes by glutaraldehyde over 2.5% in concentrations revealed similar intensities to that of the UV ray irradiated one in FT-lR investigation. 3. To obtain optimal croulink in bovine stalin origin Type I atelocollagen, 2.5% glutaraldehyde solution or UV ray irradiation with 254nm wavelength is acceptable.

1,3-Butadiene diepoxide에 의해 가교된 히아루론산 비드의 제조 및 특성 (Synthesis and Characteristics of Hyaluronic Acid Bead Crosslinked by 1,3-Butadiene diepoxide)

  • 인체 보형물의 생체 재료로 사용할 목적으로 히아루론산 비드를 제조하였다. 히아루론산 수용액과 콩기름을 섞어서 얻어진 현탁상태에서 히아루론산을 1,3-butadiene diepoxide로 가교시켰다. 제조된 비드의 물성을 살펴보기 위해 직경, 표면적과 팽윤도를 측정하였고 전자현미경으로 표면상태를 관찰하였다. 가교제의 농도가 $5-12\;vol\%$ 범위에서 비드가 형성되었으며 제조된 비드는 단순분산성을 보였다. 히아루론산 농도 혹은 가교제 농도가 증가할수록, 가교 온도가 감소할수록, BET표면적과 팽윤도가 감소하였다. 혼합속도의 변화에 의해 비드의 물성은 거의 변하지 않았으나 비드의 크기는 효과적으로 조절되었다.

Sulfonated Dextran/Poly(vinyl alcohol) Polymer Electrolyte Membranes for Direct Methanol Fuel Cells

  • Polymer electrolyte membranes, featuring ionic channels, were prepared from sulfonated dextran/ poly(vinyl alcohol) (sD/PVA) membranes. A stiff sulfated dextran was chosen as the route for ionic transport, since ionic sites are located along the stiff dextran main chain. The sD/PVA blend membranes were annealed and then chemically crosslinked. The characteristics of the crosslinked sD/PVA membranes were investigated to determine their suitability as proton exchange membranes. The proton conductivity was found to increase with increasing amounts of sD inside the membrane, which reached a maximum and then decreased when the sD content exceeded 30 wt%, while the methanol permeability increased with increasing sD content. The good dispersion of sD inside the membrane, which serves as an ionic channels mimic, played a significant role in proton transportation.

Preparation and Characterization of Proton Conducting Membranes by Blending PVC-g-PHEA and PVA

  • This work reports the preparation of proton conductive crosslinked polymer electrolyte membranes by blending poly(vinyl chloride)-g-poly(hydroxyl ethyl acrylate) (PVC-g-PHEA) and poly(vinyl alcohol) (PVA). The PHEA chains of the graft copolymer were crosslinked with PVA using sulfosuccinic acid (SA) via the esterification reaction between -OH of polymer matrix and -COOH of SA. The PVC-g-PHEA graft copolymer was synthesized via atom transfer radical polymerization (ATRP) using direct initiation of the secondary chlorines of PVC backbones. Ion exchange capacity (IEC) continuously increased with increasing concentrations of SA, due to the increasing portion of charged groups in the membrane. However, the water uptake increased up to 20.0 wt% of SA concentration above which it decreased monotonically. The membrane exhibited a maximum proton conductivity of 0.026 S/cm at 20.0 wt% of SA concentration, which is presumably due to competitive effect between the increase of ionic sites and the crosslinking reaction.

Electron Beam-induced Crosslinking and Characterization of Polycaprolactone Films in the Presence of Various Crosslinking Agents

  • Electron beam-induced crosslinking of polycaprolactone (PCL) films containing various crosslinking agents (CAs) was investigated in this study. PCL films containing various CAs prepared by a solution casting method were irradiated by electron beams at various absorption doses and the irradiated PCL films were investigated in terms of their crosslinking degree, thermal and mechanical properties, and biodegradability. Based on the results of the crosslinking degree measurement, triallyl isocyanurate was found to be most effective for the electron-beam induced crosslinking of PCL films. The results of the UTM, DMA, and TMA revealed that the thermal and mechanical properties of the crosslinked PCL films were greatly improved in comparison to those of the pure PCL. The results of the enzymatic degradation test revealed that the biodegradability of the crosslinked PCL films was reduced in comparison to that of the pure PCL.

LED-UV 가교에 의한 UHMWPE 미세다공막의 열 안정성 향상 (Enhancement in the Thermal Stability of Microporous UHMWPE Membrane by LED-UV Crosslinking)

  • Microporous gel films of 29 m were prepared by the gel drawing of UHMWPE/soybean oil blend (4:6, w/w) up to 800%. The stretched films containing the optimal photoinitiator concentration, depending on the film thickness, was cross-linked under 365 nm LED-UV irradiation and the oil was extracted with n-hexane, resulting in a gel fraction of 95 % or more. With the formation of the cross-linked structure, the melt-down temperature and melt-down elongation increased by 16 ℃ and by 63% respectively. Also the thermal stability of the crosslinked UHMWPE was proved by the area shrinkage under the heat treatment decreased to 3.8 % compared to 17.4 % for the pristine film, and by the reductions in the combustion heat. The enhanced thermal stability of the crosslinked UHMWPE microporous membranes can be used for various industrial applications such as filters, electric vehicles and energy storage systems.

열경화가 가능한 poly(p-phenylenevinylene)계 정공전달 물질의 합성 및 특성 (Synthesis and Characterization of Thermally Cross-linkable Hole Transporting Material Based on Poly(p-phenylenevinylene) Derivative)

  • 열경화가 가능한 PPV유도체인 poly[(2,5-dimethoxy-1,4-phenylenevinylene)-alt-(1,4-phenylenevinylene)] (Cross-PPV)를 Heck coupling 반응을 이용하여 합성하였다. Cross-PPV 박막은 $200^{\circ}C$에서 경화 시키면 일반적인 유기용매에 용해되지 않는 불용성의 고분자 박막이 된다. 열경화 전 후의 Cross-PPV의 구조는 FT-IR로 확인하였으며 구조의 차이는 크지 않았다. 경화된 Cross-PPV는 일반적인 유기용매에 대하여 내용매성이 강하다. 순환전압전류법과 흡수분광법으로 측정한 경화된 Cross-PPV의 호모 및 루모 에너지 준위는 각각 -5.11 eV와 -2.56 eV으로 ITO로 부터의 정공주입장벽(hole injection barrier)이 작아(약 0.1 eV) 정공주입층으로 효과적으로 사용 할 수 있다. 호모 및 루모 에너지 준위가 각각 -5.44 eV, -3.48 eV인 poly(1,4-phenylenevinylene-(4-dicyanomethylene-4H-pyran)-2,6-vinylene-1,4-phenylene-vinylene-2,5-bis(dodecyloxy)-1,4-phenylenevinylene) (PM-PPV)을 발광층으로 사용하여 두층의 구조(bilayer structure)를 갖는 소자(ITO/crosslinked Cross-PPV/PM-PPV/Al)를 제작, 특성을 평가한 결과 최대 효율은 0.024 cd/A, 최대 발광세기는 $45cd/m^2$으로 단층형 소자(ITO/PM-PPV/Al)(최대 효율 = 0.003 cd/A, 최대 발광세기 = $3cd/m^2$)에 비하여 매우 월등한 성능을 나타냄을 확인하였다. 또한 두층의 구조를 가지는 다층형 소자의 발광스펙트럼은 단층형 소자의 발광 스펙트럼과 동일하다. 이러한 사실들로 보아 ITO 및 Al에서 주입된 전자는 모두 발광층인 PM-PPV층에서 재결합(recombination)되어 여기자(exciton)가 형성되는 것으로 사료된다.
