Genome-Wide Comprehensive Analysis of the GASA Gene Family in Peanut (Arachis hypogaea L.)

  • The GASA protein (Gibberellic acid-stimulated Arabidopsis) are family of small cysteine-rich peptides found in plants. These GASA gene family mainly involved in biotic/abiotic stress responses and plant development. Despite being present in a wide plant species, their action and functions still remain unclear. In this study, using the in-silico analysis method we identified 41 GASA genes in peanuts (Arachis hypogaea L.). Based on the phylogenetic analysis 41 GASA genes are classified in the four major clusters and subclades. Mainly, clusters IV and III comprise the majority of GASA genes 15 and 11 genes respectively, followed by cluster I and cluster II with 9 and 6 genes respectively. Additionally, based on in-silico analysis we predicted the post-transcriptional and post-translational changes of GASA proteins under abiotic stresses such as drought and salt stress would aid our understanding of the regulatory mechanisms. Hence, a further study is planned to evaluate the expression of these GASA genes under stress in different plant tissues to elucidate the possible functional role of GASA genes in peanut plants. These findings might offer insightful data for peanut advancement.

Analysis of Molecular Variance and Population Structure of Sesame (Sesamum indicum L.) Genotypes Using Simple Sequence Repeat Markers

  • Sesame (Sesamum indicum L.) is an important oilseed crop grown in tropical and subtropical areas. The objective of this study was to investigate the genetic relationships among 129 sesame landraces and cultivars using simple sequence repeat (SSR) markers. Out of 70 SSRs, 23 were found to be informative and produced 157 alleles. The number of alleles per locus ranged from 3 - 14, whereas polymorphic information content ranged from 0.33 - 0.86. A distance-based phylogenetic analysis revealed two major and six minor clusters. The population structure analysis using a Bayesian model-based program in STRUCTURE 2.3.4 divided 129 sesame accessions into three major populations (K = 3). Based on pairwise comparison estimates, Pop1 was observed to be genetically close to Pop2 with $F_{ST}$ value of 0.15, while Pop2 and Pop3 were genetically closest with $F_{ST}$ value of 0.08. Analysis of molecular variance revealed a high percentage of variability among individuals within populations (85.84%) than among the populations (14.16%). Similarly, a high variance was observed among the individuals within the country of origins (90.45%) than between the countries of origins. The grouping of genotypes in clusters was not related to their geographic origin indicating considerable gene flow among sesame genotypes across the selected geographic regions. The SSR markers used in the present study were able to distinguish closely linked sesame genotypes, thereby showing their usefulness in assessing the potentially important source of genetic variation. These markers can be used for future sesame varietal classification, conservation, and other breeding purposes.

농작물재해보험의 농가 수입안정 및 사회적 후생 효과 분석 (Analysis of Farm Revenue Stabilization and Social Welfare Effects of Crop Yield Insurance)

  • Crop yield insurance program in Korea has rapidly grown not only in quantity but in quality for 15 years since it was introduced in 2001. Despite growth of Crop insurance, performance evaluation for crop yield insurance has not fully been evaluated at the farm, consumer and national level. The purpose of this study is to conduct the performance evaluation for crop yield insurance through estimating the effects of farm revenue stabilization and social welfare increase with three popular insurance items: apple, pear and sweet persimmon. Based on the analysis of social welfare effect, cost-benefit analysis of operating crop yield insurance was conducted at the national level. We found that crop yield insurance stabilizes farm revenue based on the estimated four risk indicators: Coefficient of Variation, Value at Risk, Certainty Equivalence, and Risk Premium. The result of cost-benefit analysis shows that crop yield insurance increases social net benefit by 44.1 billion won for the three items. As a result, crop yield insurance program has contributed remarkably on social welfare as well as farm management and its role will be more important in the future.

Comparison of Analytical Methods for Volatile Flavor Compounds in Leaf of Perilla frutescens

  • Volatile flavor compounds from perilla leaves were extracted and analyzed with different methods, head-space analysis (HS), simultaneous steam distillation and extraction (SDE) , and solvent extraction (SE), and to compare their efficiencies for quick analysis. Over 30 volatile compounds were isolated and 28 compounds were identified by GC/MSD. Major compound was perillaketone showing the compositions of which were 92% in SDE method, 86% in headspace analysis, and 62% in solvent extraction method. For quick evaluation of leaf flavor in perilla, it was desirable because the headspace analysis method had a shorter analyzing time and smaller sample amount than the other methods.

발아에 따른 현미의 아밀로펙틴 중합도와 식감관련 특성에 관한 연구 (Studies on the Degree of Polymerization of Amylopectin and Texture Analysis Test of Brown Rice After Germination)

  • 본 연구는 국내 주요 벼 품종들 중 일부 품종들에 대하여 현미와 발아현미에 대한 아밀로펙틴 중합도와 식감관련 특성에 대해 비교 분석하였다. 아밀로펙틴은 단쇄비율이 높고 장쇄비율이 낮은 품종이 밥맛이 우수한 특성을 나태는 것으로 알려져 있는데 본 연구결과 발아현미가 일반현미에 비해 유의적으로 높아지는 결과를 나타냈다. 특히 하이아미와 진보가 다른 품종들에 비하여 유의적으로 단쇄비율이 높고 장쇄비율이 낮은 것으로 나타났다. 식감측정결과 진보의 경우 현미의 경도(hardness)와 탄력성(toughness)이 각각 84.6과 51.8에서 발아과정을 통해서 50.2와 30.2로 감소하였으며 찰기(stickiness)와 부착성(adhesiveness)은 발아현미밥의 60.6와 42.9에 비하여 현미밥에서 27.8과 30.8로 나타나 발아현미밥에서 찰기와 부착성이 각각 2.18, 1.39배 높은 값을 나타내었다. 또한 식감과 아밀로펙틴 중합도 특성과의 관계를 알아보기 위하여 경도, 탄력성, 찰기, 부착성과 아밀로펙틴 중합도 특성과의 상관관계를 분석하였는데, 발아현미에서 단쇄비율이 많을수록 경도와 탄력성이 감소되고 장쇄 비율이 높을수록 경도와 탄력성, 부착성이 증가하는 경향이 나타났다. 따라서 본 연구에서는 주요 벼 품종들의 현미에 대하여 발아 전과 후의 아밀로펙틴 중합도와 식감관련 특성을 종합적으로 비교 분석한 결과 발아과정에서 현미의 조직변화가 나타나고 발아 후 식감이 부드러워지고 찰기가 증대되어 밥맛이 향상된 것을 입증할 수 있었다. 또한 식감과 아밀로펙틴 중합도 특성과의 관계는 발아현미에서 뚜렷한 경향을 나타냈으며, 본 연구가 아밀로펙틴의 길이와 식감의 상관관계를 밝히기 위한 기초자료가 될 수 있을 것으로 사료된다.
