Antioxidative Enzyme Activities and O-J-I-P Transients of Crinum asiaticum var. japonicum $B_{AK}$. Leaves under Winter Stress (겨울철 저온스트레스에 의한 문주란 잎의 항산화효소 활성과 O-J-I-P 곡선 패턴)

  • The diurnal and seasonal variations of antioxidative enzyme activity and the O-J-I-P transients were investigated from the leaves of Crinum asiaticum var. japonicum under winter stress in natural habitat, in order to diagnose quantitatively physiological states of plants under stresses. The activities of superoxide dismutase and peroxidase increased slightly in winter. Especially, peroxidase acitivity was higher at dawn and night in winter and some isoforms were detected only in early winter. In the O-J-I-P transients, the fluorescence intensity of J, I, P steps decreased significantly in winter season, contrary to its high value in summer season. Of the chlorophyll fluorescence parameters derived from the O-J-I-P transients, Fm and $\Phi_{po}$ decreased with the increase of ABS/RC depending on temperature drop in winter.

  • The chlorophyll fluorescence combined with the O-J-I-P transients were examined in the leaves of the crinum plants (Crinum asiaticum var. japonicum BAK.), in order to satisfy the demand for rapid in vivo measurement of vitality, and to apply easily to approach questions of economical interest concerning the plant vitality. The photosynthetic efficiency, Fv/Fm, of crinum plants dramatically decreased depending on temperature drop in winter. In summer, the Fv/Fm values was lower in day time than at dawn and night, suggesting that photosynthetic efficiency is chronically photoinhibited in day time. In winter, there was no prominent diurnal fluctuations of Fv/Fm values. However, based on the O-J-I-P transient, PI$\_$NO/ and SFI$\_$NO/ dramatically increased at noon in summer, and $\psi$o/(1-$\psi$o) diurnally fluctuated in winter. These results indicated that vitality indexes such as PI$\_$NO/, SFI$\_$NO/ and $\psi$o/(1-$\psi$o) can be used as the indicators for in vivo measurement of environmental stresses.

On the mutual action of boron, calcium and sucrose in pollen cell growth and pollen bursting (화분세포생장과 파열에 미치는 붕소, 석탄 그리고 자당의 상호작용에 관하여)

  • The mutual relationships of boron, Ca and sucrose were studied in relation to in vitro pollen growth and pollen bursting, by using conventient pollen from Crinum asiaticum for experiment. Crinum pollen are paticularly sensitive to Ca. Addition of very small amount of boron to cultural media was apparently synergistic to the action of sucrose and Ca in pollen germination and tube elongation. This action was extended to a higher level of boron concentraton. Combined application of boron, Ca and sucrose always gave the better results in pollen growth and protection against pollen bursting much more than when used singly. This indicated that there is a direct relationship between better growth of pollen and increased rigidity of pollen cell wall. A higher level of Ca concentration tended to increase bursting rate of pollen grains and decrease that of pollen tubes, while boron always depressed the rate of bursting. This was considered due to increased failure in pollen germination at high level of Ca that favors pollen tube elongation. The fact that Ca show an antagonistic effect on the suppressive action of high level of boron in pollen growth and shows different effect in response to pollen bursting from boron, suggested mode of Ca and boron action in the presence of sucrose is quiate different, although to increase in rigidity of pollen cell wall by them is in common nature. It was postulated therefore that Ca acts on pectins of pollen cell wall largely as "non-metabolic" and boron as "metabolic" promoter is pollen growth and protecting pollen bursting, since boron and Ca have common nature in strengthening the pollen cell wall but act differently.but act differently.

Diurnal Changes of Chlorophyll Fluorescence and Antioxidative Enzyme Activity of the Leaves from Four Subtropical Plants (아열대성 식물 4종의 엽록소형광과 항산화효소 활성의 일주기적 변화)

  • The diurnal changes of chlorophyll fluorescence and antioxidative enzyme activity were investigated in the leaves from four subtropical plant species (Crinum asiaticum var. japonicum Bak., Osmanthus insularis Koidz., Asplenium antiquum Makino and Chloranthus glaber Makino) under the natural habitats in summer and winter. The intensity of chlorophyll fluorescence was lower in O-, I-, J-, P-steps of O-J-I-P transient in winter than summer, and prominent diurnal change was not found in the fluorescence intensity of four subtropical plant species in winter. The activity and isoenzyme pattern of SOD and catalase did irregularly change seasonally and diurnally in four subtropical plant species. In contrast, the peroxidase activity and isoenzyme pattern was different depending on plant species and growth seasons; The activity increased slightly more in winter than in summer in four subtropical plant species, and several isoenzymes appeared in the leaves from C. asiaticum var japonicum, O. insularis and A. antiquum in winter.

Tropisms of Pollen-Tubes In Vitro (화분관 In Vitro 생장의 굴수성에 대하여)

  • Zephyranthes candida, Narcissus pseudonarcissus and Crinum asiaticum pollen were placed near their pistil parts respectively on agar cultural media(microslides) containing 10% sucrose and 100mg/l botic acid plus 1% agar with or without calcium and some other calciumsupporting inorganic salts. If fresh pistils (100% moisture) were used pollen grew toward their pistil parts, showing "positive" tropism. This was also true when combinations among three different species were made. Pollen tubes grew away from the pistils if they were dried (below 10% moisture), showing "negative" tropism. Pollen could not show any tropic growth and thus grew at random of all directions if the pistil parts were incompletely dried (approximately 50% moisture). The similar tropic responses of pollen-tube growth with the three species could be demonstrated with etiehr wet or dried tooth-pick segments. Calcium jons in the basic medium merely promated pollen-tube growth and so either "positive" or "negative" tropism became rather distinctive, but they were not tropically active. Pollen tubes grow toward pistil parts with more moisture content and seem to be hydrotropically sensitive. This was assumed due to the cohesive force existing in water molecules.esive force existing in water molecules.

Seasonal and Diurnal Changes of Antioxidant Enzymes in Four Subtropical Plant Species (아열대성 식물 4종의 항산화효소 활성과 Isoenzyme의 계절적.일주기적 변화)

  • 아열대성 식물 4종 (문주란, Crinum asiaticum var. japonicum; 박달목서, Osmanthus insularis; 죽절초, Chloranthus glaber; 파초일엽, Asplenium antiquum)을 대상으로 자연 환경요인의 변화에 의한 항산화 효소 (superoxide dismutase, peroxidase, catalase, ascorbate peroxidase)의 활성과 isoenzyme 패턴의 변화를 전기영동으로 조사하였다. 그 결과, peroxidase의 활성과 isoenzyme 패턴이 식물종이나 환경조건에 따라 가장 다양하게 나타났다. Peroxidase는 4종 모두에서 여름철보다 겨울철에 활성이 높았고 문주란, 박달목서, 파초일엽에서는 겨울철에 특이적으로 발현되는 isoenzyme들도 관찰할 수 있었다. Catalase는 문주란, 박달목서, 파초일엽에서 검출되었다. 문주란 잎에서는 겨울철에 비해 여름철에 다소 높은 활성을 보였으며, 박달목서와 파초일엽에서는 겨울철에 높은 활성을 나타내었다. 그리고 문주란과 박달목서에서는 겨울철에 새벽이나 밤보다 낮시간에 높은 활성을 보였는데 파초일엽에서는 낮시간의 catalase 활성이 낮았다. Superoxide dismutase는 문주란, 박달목서, 파초일엽에서 검출되었으며, 특히 박달목서에서는 겨울철에 높은 활성을 보였다. Ascorbate peroxidase는 문주란과 파초일엽에서 관찰되었으나 계절적으로 큰 차이가 없었으며, 겨울철에는 isoenzyme 패턴의 일주기적 변화가 관찰되었다. 이상의 결과, 종별로는 문주란, 파초일엽에서 4종의 항산화효소가 모두 검출되었고, 박달목서에서는 ascorbate peroxidase가, 죽절초에서는 peroxidase를 제외한 모든 항산화 효소가 검출되지 않았다. 식물종에 따라 또는 환경요인의 변화에 따라 항산화효소의 활성 또는 isoenzyme 패턴의 차이를 보이고 있지만 항산화효소의 계절적 그리고 일주기적 변화가 관찰되어, 본 연구에서 조사된 4종의 아열대성 식물이 자연환경 조건 하에서도 산화적 스트레스에 처하고 있는 것으로 보인다.

On the action of Ca in pollen growth as influenced by interaction of the different Ca concentration, acidity and temperature (화분생장에 미치는 석탄이온의 작용과 그 농도, 산성도 및 온도의 상호관계에 대하여)

  • Interaction occuring among the different Ca concentrations, pH and temperatures in the promotive effect of Ca in pollen growth was studied by using pollen from Crinum asiaticum and Cryptostegia grandiflora. Data for pollen tube elongation were found to be more indicative of representing the promotive action of Ca ion in pollen growth than those for pollen germination, and were served to evaluate the experimental results. The pollen growth increased as the concentration of Ca increase. The optimal pH range for pollen growth shifted from the lower pH to the higher as the concentration of Ca increase. The characteristic Ca effect was disappeared, and no pH effect at various ranges was observed when pollen grains were grown at the low temperature(8$^{\circ}C$). The Ca effect became quite pronounced if temperature were raised. The Ca effect became even mroe striking if the condition was in higher pH ranges (weak alkaline). Higher pH ranges were found to be more favorable for the Ca action, whereas higher temperature was required to bring about more pronounced Ca effect. Thus, the longest pollen tube was obtained with the highest pH, temperature adopted for the medium supplemented with Ca in the present experiment, and the shortest tube with the lowest temperature applied at the highest pH. Pectin synthesis in pollen tube was considered as a metabolic process, whereas Ca binding in pectin of the pollen tube wall as non-metabolic in nature. Disappearance of Ca effect at the low temperature was probably brought about by blocking the metabolic synthesis of pectin, and nonmetabolic Ca binding seems to take place more extensively with higher concentrations of Ca and at higher pH levels than the lower.

Screening of Vitality Indicators from Four Subtropical Plant Species by Chlorophyll Fluorescence Analysis (엽록소형광을 이용한 아열대성 식물 4종의 활력도 지표 검색)

  • 아열대성 식물 4종 (문주란, Crinum asiaticum var. japonicum; 박달목서, Osmanthus insularis; 죽절초, Chloranthus glaber; 파초일엽, Asplenium antiquum)을 대상으로 자연 환경요인의 변화에 따른 엽록소형광을 분석하여 이들 식물의 활력도를 검토하였다. 여름철 일변화에 있어서 양지에 자라는 문주란과 박달목서는 Fv/Fm이 새벽과 밤에는 0.80~0.83 범위의 높은 값을 보이지만, 낮시간에는 0.65~0.73으로 낮았다. 1-qN과 1-qP도 낮시간에 크게 낮아졌다. 음지에 자라는 죽절초와 파초일엽의 Fv/Fm은 높아서 0.83~0.85 범위를 유지하였으며 일변화적 특성은 관찰되지 않았다. 1-qP의 변화는 거의 관찰되지 않았으나 1-qN이 낮시간에 다소 감소하는 경향을 보여주었다. 죽절초와 파초일엽에서는 광억제가 거의 일어나지 않으나 문주란과 박달목서에서는 낮시간의 고광에 의한 광억제가 나타난다. 하지만 여기에너지의 일부를 열의 형태로 방출하여 광피해를 완화시키고 있는 것으로 보인다. 겨울철에는 Fv/Fm이 모든 종에서 0.8 이하의 값을 나타내었는데, 특히 문주란의 Fv/Fm 값은 다른 3종 보다 더 낮았다. 그리고 모든 종에서 일변화적인 특성은 관찰되지 않았다. 문주란의 1-qN은 낮시간에 다소 감소하였지만, 나머지 3종의 식물은 높은 값을 나타내었다. 그리고 l-qP는 문주란과 박달목서에서는 낮시간에 0.6 범위로 낮아졌다. 이는 종에 따라 차이가 있지만 겨울철 저온의 영향을 받은 결과로 보인다. 한편, 여름철 문주란과 박달목서의 O-J-I-P곡선은 거의 유사하며 낮에 뚜렷하게 낮았고, 죽절초와 파초일엽에서는 일변화적 특성은 두드러지지 않았다. 그리고 P $I_{NO}$ 와 SF $I_{NO}$ 가 죽절초를 제외한 3종에서 여름철 낮시간에 증가하였다. 겨울철의 O-J-I-P곡선은 모든 종에서 낮시간에 다소 낮아지지만 큰 변화는 없었다. 그리고, 문주란, 박달목서, 파초일엽에서 $\psi$o/(1-$\psi$o)가 낮시간에 다소 증가하였다. 이로부터 P $I_{NO}$ , SF $I_{NO}$ , $\psi$o/(1-$\psi$o)등의 변수는 식물의 활력도를 검정하는 지표로 활용될 가능성이 높다고 할 수 있다.

Relationship of Lycoris (Amaryllidaceae) Based on RAPD Markers (RAPD markers에 의한 상사화속 식물의 유연관계)

  • Phylogenetic relationships were examined for 17 taxa of Lycoris by RAPD analysis. The length of the amplified DNA fragments ranged from 300 bp to 1,700 bp. 57 scorable RAPD markers were observed from PCR reactions with five random oligoprimers. The analysis by UPGMA sepatated the examined taxa of Lycoris into were clusters. First group was comprised of ten taxa of L. chinensis var. sinuolata, L. sanguinea var. koreana, L. uydoensis, L. flavescens, L. radiata var. pumila, L. radiata, L. squamigera, L. chejuensis, L. aurea and L. guangxiensis, second group of L. haywardii, L. sprengeri, L. rosea, L. straminea and L. houdyshii, third group of outgroup of Narcissus tazetta var. chinensis and Crinum asiaticum var. japonicum. From the viewpoint of cytological characters such as polyploidy and karyotype, the RAPD analysis was very useful to show the relationship among the intraspecific taxa of Lycoris.
