Effects of Milk Protein levels and Casein/Whey Ratios on Organ Growth and Protein Metabolism in Early Weaned Rats (조기 이유한 흰쥐에서 유단백질의 섭취수준과 조성비가 기관성장과 단백질대사에 미치는 영향)

  • This study was conducted to investigate the effects of protein levels and casein/whey ratios on organ growth and protein metabolism in early weaned rats. Premature rats weaned by the 17th day were fed six semipurified synthetic, isocaloric and gel diets that contained three levels (low, medium and high) and two different combinations(casein/whey ; 80 : 20 or 20 : 80) of milk protein for 8 days. On the 25th day postpartum, frest weigth and DNA, RNA and milk protein contents in brain, liver, kidney and muscle were determined to ascertain organ and cellular growth. Futher, with a view to ascertain protein metabolism and renal functions, serum total protein, $\alpha$-amino N, urea N, and creatinine and creatinine and urinary urea N, creatinine and hydroxproline were determined. Total DNA contents of brain, liver and kidney, which may represent as an index of cell numbers in those organs were significantly decreased in the rats fed diets containing low level protein regardless of casein/whey ratio. However, as fat as the rats fed high protein diets were concerned, their fresh weight, protein contents and GFR of kidney were significantly increased. Furthermore, nitrogen components, $\alpha$-amino N, urea N and creatinie in serum and urine were also increassed. Another observation was that high casein/whey ratio significantly facilitated accumulation of porteins in muscle and kidney and urinary hydorxyproline excretion, not affecting the DNA content of those organs. This study showed that low(8%) or high(32%) contents of protein had less desirable effects either on protein metabolism or on organ cellular growth in prematurely weaned rats, whereas there were no effects on general growth and bone strength.

A Study on the Food Intake Sodium and Potassium Intakes and Urinary Excretion of Preschool Children in Pusan (부산지역 학령전 아동의 식품섭취와 나트륨, 칼륨의 섭취 및 소변중 배설실태에 관한 연구)

  • o assess the food intake and the sodium and potassium intakes and urinary excretion of preschool children in Pusan and to evaluate the relationship among variables dietary behaviors food and nutrient intake and 24hr urinary sodium and potassium excretion were measured with 97 subjects. The mean sodium and potassium intakes were 1890. 1mg(82,2meq) and 1479.7mg(37.8meq) respectively. The mean potassium intake(p<0.05) and density(p<0.01) were significantly low at the group who had food intake pattern absen of fruit and daily groups. The mean urinary sodium and potassium excretion were 735.3mg(32.0meq) and 418.7mg(10.7meq) respectively. The mean sodium intake(p<0.01) and urinary excretion were 735.3mg(32.0meq) and 418.7mg(10.7meq) respectively. The mean sodium intake(p<0.01) and urinary excretion (p<0.001, p<0.001) energy intake(p<0.01) sodium intake and density (p<0.001, p<0.05) the food number and intake frequency of vegetable group consumed (p<0.01, p<0.01) dietary frequency score(p<0.05) and negative correlations with the food number and frequency of dairy group consumed (p<0.05 p<0.05) The urinary potassium excretion showed positive correlations with height and weight(p<0.05, p<0.01) urine volume and urinary creatinie excretion(p<0.01 p<0.001) potassium intake(p<0.05) food number and intake frequency of dairy group consumed (p<0.05, p<0.001). Based on the results urinary sodium excretion was related to age sodium intake and food intake of vegetable and dairy group and urinary potassium excretion was related to potassium intake and food intake of dairy group. So nutritional education is needed in order to decrease sodium intake especially from food intake of vegetable group when preschool children have less food intake of diary group,.

Effects of Astragali Radix (AR) on Chronic Renal Injury in Rats (황기가 흰쥐의 만성 신손상에 미치는 영향)

  • Objective : This study was designed to investigate the possibility of AR for chronic renal injury. Methods : The author first investigated the expression levels of DNA by inducing of chronic renal injury. Then, the author investigated the effects of AR on chronic renal injury induced by combination treatment with Adriamycin and cisplatin in terms of changes in body weights and renal tissues, urine volume, BUN and creatinine levels, creatinine clearance and histopathological changes in renal tissues. Total expression levels of 546 genes were elevated or lowered by induction of chronic renal injury. Genes of which whose expression levels were elevated by induction of chronic renal injury were related to the PPAR signaling pathway and fatty acid mechanism, etc. Genes of which expression levels were lowered by induction of chronic renal injury were related to the neuroactive ligand-receptor signaling pathway. Results : Oral administration of AR restored renal mass which was reduced by induction of chronic renal injury. AR also restored creatinine clearance and lowered serum BUN level. In histopathological observation, the AR group has a tendency to prevent tissue damages as shown in the chronic renal injury group. Conclusions : AR can be used to treat patients with chronic renal injury although further study will be needed to elucidate the exact mechanisms in the efficacy of AR on chronic renal injury.

Anti-diabetic Effect of Wen-Pi-Tang-Hab-Wu-Ling-San Extract in Streptozotocin-induced Diabetic Rats (Streptozotocin으로 유도한 당뇨병 쥐에서 $WHW^{(R)}$의 항당뇨 효과에 대한 연구)

  • Objectives : This study aimed to evaluate the anti-diabetic effect of Wen-Pi-Tang-Hab-Wu-Ling-San (WHW) extract in streptozotocin(STZ)-induced type-1 diabetic rats. Methods : Experimental diabetes were induced by intraperitoneal injection of streptozotocin (60 mg/kg). Two groups of STZ-induced diabetic rats were given the following treatments for 2 weeks by oral Administrations : (1) WHW 10 mg/kg, (2) WHW 100 mg/kg. In addition, vehicle-treated diabetic and nondiabetic controls were used in the experiment. The effects of WHW extract on STZ-induced diabetes were observed by measuring the changes of body weights and the levels of fasting blood glucose, insulin, urea nitrogen (BUN) and creatinine level in sera of rats, respectively. Results : In comparison control group, WHW-treated groups (100 mg/kg) were significantly decreased fasting blood glucose levels and increased serum insulin levels in STZ-induced diabetic rats. Moreover, WHW-treated groups (100 mg/kg) were reduced s-creatinie levels in STZ-induced diabetic rats. In addition, the changes related to diabetic nephropathy with body weight were significantly lower in WHW extract-dosing groups than in the diabetic control. Conclusions : The study thus showed that WHW extract enhanced the anti-diabetic effect in STZ-induced diabetic rats by improving the hypoglycemia. It also increased pancreatic insulin content in these rats.

Effects of Ethanol-Extract of Lotus Root on the Renal Function and Blood Pressure of Fructose-Induced Hypertensive Rats (연근의 에탄올 추출물이 과당으로 유도된 고혈압 흰쥐의 혈압과 신장 기능에 미치는 효과)

  • The present study was aimed to investigate whether ethanol-extract of Nelumbo nucifera has an ameliorative effect on the renal function in high fructose-diet induced hypertensive rats. The urine osmolality (Uosmel) was decreased in rats with high fructose-diet ($60\%$) during the whole experiment period without change of the urine volume (UV). The urinary excretion of sodium and chloride were decrease significantly in rats with fructose induced hypertensive rats, wheras urinary excretion of potassium was increased. The creatinine clearance (CCr) and solute-free water reabsorption were also decreased by treatment of fructose rich diet. Among these renal functional parameters, CCr was partially restored by the administration of ethanol-extract of Nelumbo nucifera. The Uosmol was also partially restored by the administration ethanol-extract of Nelumbo nucifera at the end of the experimental period. Taken together, ethanol-extract of Nelumbo nucifera has the ameliorative effect on glomerular filtration rate in rats with high fructose-diet induced hypertension.

  • Protein malnutrition of children is one of the most serious nutritional deficiencies in developing country. Urea nitrogen excretion in ureotelic animals is the function most sensitive to dietary protein. The 24 hours excretion of creatinine in the urine of a given subject is remarkably constant from day to day. The creatinine excretion of different individuals of the same age and sex is also quite constant. Low ratios of urinary urea to creatinine are found children low protein intake. The foregiving world-wide investigations indicate that the urea nitrogen/creatinine ratios seems to be a good biochemical indicator to distinguish among group with different levels of protein intake. The purpose of this study is to evluate an indicator of protein intake on the elementary school children ranged from 6 to 8 years of age living in rural and urban areas. Each child measured for height and weight of body. weight measured by means of a plate from scale and height by a vertical measuring rod. Biochemical test were taken from a finger-tip and urine. Hemoglobin level in the blood was measured by cyanomethemoglobin method. From the urine samples, urea nitrogen and urea creatinie were determined by Folin-Wu method and: calculate the ratio. The following result were obtained: 1) Mean of the body weight and height in urban children(Seoul) was higher and heavier than rural children(Kyunggi, Kangwon). And 12% of boys, 18% of girls in Kyunggi and 25% of boys, 22% of girls in Kangwon area weight less than 80% of Korean Physical Standard weight level. 2) The mean hemoglobin values of boys and girls in Seoul are children were 13. 3g/100ml, 13.1g/100ml and the mean of hemoglobin values in Kyunggi 12.9g/100ml of boys, 12.4g/100ml of girls, and 12.4g/100ml of boys, 12.9g/100ml of girls in Kangwon children. It is found that 22% to 24% children inrural area (Kyunggi, Kangwon) had hemoglobin level less than 12g/100ml which means anemia. 3) The mean of hematocrit level of Seoul, boys and girls children were 33.5%, 34.1% and 33.4%, 33.1%, in Kyunggi area and 33.1%, 32.9% in Kangwon area. 4) Urea nitrogen/creatinine ratios in Seoul children were 9. 0, 10. 0 of boys and girls, the ratio were 8.2, 8.0 in Kyunggi boys and girls children, and 7.5 and 7.4 in Kangwon boys, girls children. Low-income rural and upper-income urban background large differences between two groups in the urea nitrogen/creatinine ratio(Seoul: Kangwon in male, female children. p<0.05, p<0.001). The urea nitrogen/creatinine ratio definetly seems to be a good indicator of the quantity of the protein intake. However, whether or not it is an indicator of the quality of the ingested protein ramains to be seen.
