도정 유무에 따른 제주산 보리의 항산화 활성 비교 (Comparison of antioxidant activities of pearled and wholegrain barley harvested in Jeju)

  • 본 연구에서는 제주산 쌀보리인 일반보리, 청보리, 흑보리를 이용하여 도정 유무에 따른 항산화 성분과 활성을 비교하였다. 제주산 쌀보리의 항산화 성분인 총 폴리페놀 함량은 도정하지 않은 쌀보리에서 139.99-149.90 ㎍ GAE/g, 도정한 쌀보리는 91.14-125.45 ㎍ GAE/g 범위로 나타나 도정하지 않은 쌀보리에서 보다 높은 총 폴리페놀 함량을 보였다. 또한 총 플라보노이드 함량도 도정하지 않은 쌀보리(0.89-1.01 ㎍ QE/g)가 도정한 쌀보리(0.50-0.54 ㎍ QE/g)보다 높은 함량을 나타내었다. HPLC로 분석한 제주산 쌀보리의 페놀성 화합물로 gallic, protocatechuic, vanillic, p-coumaric 및 ferulic acid가 주로 검출되었으며, 도정하지 않은 흑보리와 도정한 일반보리에서는 p-coumaric acid는 검출되지 않았다. Tocopherols 및 tocotrienols 분석 결과에서는 8가지 이성체 중 α-T3 함량이 높았고, 도정하지 않은 쌀보리의 α-T3는 20.68-33.09 ㎍/g으로 도정한 쌀보리 8.93-14.98 ㎍/g보다 유의적으로 높게 검출되었다. 제주산 쌀보리의 in vitro 항산화 활성 결과에서 금속 이온 제거능을 제외하고 도정하지 않은 쌀보리가 높은 항산화 활성을 나타내었고, 항산화 활성 중 SOD 소거 활성이 높게 나타내었다. In vitro 항산화 활성은 총 폴리페놀 및 총 플라보노이드 함량에 비해 총 tocopherols 및 tocotrienols 함량에 더 높은 상관관계를 보였다. 따라서 도정하지 않은 제주산 쌀보리에서 높은 항산화 성분 및 활성이 나타나 이를 천연 항산화제로 활용 가능할 것이라 생각된다.

Phosphorylation of Akt Mediates Anti-Inflammatory Activity of 1-p-Coumaroyl ${\beta}$-D-Glucoside Against Lipopolysaccharide-Induced Inflammation in RAW264.7 Cells

  • Hydroxycinnamic acids have been reported to possess numerous pharmacological activities such as antioxidant, anti-inflammatory, and anti-tumor properties. However, the biological activity of 1-p-coumaroyl ${\beta}$-D-glucoside (CG), a glucose ester derivative of p-coumaric acid, has not been clearly examined. The objective of this study is to elucidate the anti-inflammatory action of CG in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophage cells. In the present study, CG significantly suppressed LPS-induced excessive production of pro-inflammatory mediators such as nitric oxide (NO) and $PGE_2$ and the protein expression of iNOS and COX-2. CG also inhibited LPS-induced secretion of pro-inflammatory cytokines, IL-$1{\beta}$ and TNF-${\alpha}$. In addition, CG significantly suppressed LPS-induced degradation of $I{\kappa}B$. To elucidate the underlying mechanism by which CG exerts its anti-inflammatory action, involvement of various signaling pathways were examined. CG exhibited significantly increased Akt phosphorylation in a concentration-dependent manner, although MAPKs such as Erk, JNK, and p38 appeared not to be involved. Furthermore, inhibition of Akt/PI3K signaling pathway with wortmannin significantly, albeit not completely, abolished CG-induced Akt phosphorylation and anti-inflammatory actions. Taken together, the present study demonstrates that Akt signaling pathway might play a major role in CG-mediated anti-inflammatory activity in LPS-stimulated RAW264.7 macrophage cells.

  • Allelopathic compounds as naturally occurring herbicide have originally reported from local vegetation since B.C. 300. These compounds are known as secondary plant metabolites which released from plants into the environment often attract or repel, nourish or poison other organisms. In recent, many natural plant allelochemicals be used to attempt to biologically or ecologically control weed among worldwide weed scientists. Some allelochemicals have also used as fungicides, insecticides, and nematodicides, and were less than man-made agrochemicals to damage the global ecosystem. It makes efficient use of resources internal to the farm, relies on a minimum of purchased inputs. Some scientists selected for allelopathic activity when breeding weed-controlling cultivars of rice, sorghum, cucumber, surflower etc. Thus, this paper is focused on allelopathic compounds isolated from cultivated crop with the high potential of prospective herbicides. The most environmentally acceptable and sustainable approach to utilization of allelopathy for weed control is to develop plant cultivars with proven allelopathic characteristics. In rice accessions, there are 60 cultivars/lines which have known as allelopathic activity and some of these cultivars control weed more less 90% within certain radius of activity. These accessions are originated from 15 countries including Korea, Japan, USA, India, Philippines, Indonesia, Laos, Taiwan, Afghanistan, Mali, Pakistan, Colombia, Egypt, China, and Dom. Rep. From these cultivars, the most common allelopathic compounds identified in rice are p-Hydroxybenzoic, Vanillic, p-Coumaric, and Ferulic acids. In addition, allelopathic lines of the following crop have shown inhibition of weed growth : beet (Beta vulgaris), lupin(Lupinus spp.), com(Zea mays), Wheat(Triticum aestivum), oats(Avena spp.) peas(Pisum sativum), barley(Hordeum vulgare), rye(Secale cereale), and cucumber(Cucumis sativus). Thus, future allelopathy research must be designed its potentially phytotoxic propertices and the ecotoxic features of the allelochemicals from release to degradation ; its ecological sustainability, its allelopathic effect in early growth. stages, and selectivity properties in combination with chemical stages, and selectivity properties in combination with chemical concentrations. Also, research approach in allelopathy might be screened for highly allelopathic germplasm collection of crops, the idea being to ultimately transfer this agronomic character into improved cultivars by either conventional breeding or other genetic transfer techniques.

모링가 부위별 추출물의 항산화 활성 및 파이토케미컬 함량 (Antioxidant activity and phytochemical contents of the extracts from different parts of Moringa oleifera)

  • 열대 및 아열대 기후에서 자생하는 모링가가 기후변화로 인하여 국내에서도 재배가 되고 있으면서 관심이 증가하고 있다. 다양한 나라에서 식품 및 기능성제품으로 사용이 되면서 국내로 수입되는 모링가에 대한 분석이 필요하여 본 연구를 실시하게 되었다. 모링가의 부위별(잎, 가지 및 종자) 항산화 능력과 페놀 및 플라보노이드 함량은 잎과 가지에서 높았으며, 정의 상관관계를 나타내었다. 무기물 함량 역시 잎과 가지에서 풍부하였으며 특히 칼륨 이온이 가장 많았다. 또한 7종의 페놀산과 4종의 플라보노이드 성분이 검출이 되었으며 페놀산에서는 페룰릭산이, 플라보노이드에서는 쿼세틴 함량이 높게 나타났다. 이 연구결과는 향후 국내 모링가 재배농가의 비교자료로 유용하게 이용될 것으로 기대된다.

우리 나라에서 수집한 새포아풀의 분류 및 특성 (Classification and Characteristics of Annual Bluegrass(Poa Annua L.) Collected from Golf Courses in Korea)

  • This study was carried out to get better understandings about morphological, ecological, and genetical characteristics of annual bluegrass collected from different golf courses in Korea and eventually to establish a successful control strategy. Twenty five local lines of annual bluegrass collected from 20 golf courses in Korea were classified into annual or perennial type on the basis of morphological characteristics. Twelve local lines showing obvious morphological differences were selected and then genetically assessed using RAPD analysis. Classification of the 12 local lines through RAPD analysis were considerably similar to that determined by both of morphological differences and phenotype. Responses of the two types of annual blugrass to herbicides were also examined. Shoot growth of annual bluegrass was significantly suppressed by flazasulfuron and the annual type was more susceptible than perennial type, regardless of flazasulfuron concentrations used. By pendimethalin treatment, there was no clear difference in susceptibility between the two types of annual bluegrass. However, by the treatment of dithiopyr, annual type was more sensitive than perennial type in both shoot and root growth. Nine tree species were screened to detect their allelopathic potential on turfgrasses and annual bluegrass. Acacia (Robinia pseudo-acacia) leaves showed selective inhibition in the shoot and root growth as well as their seed germination when treated with 2% and 10%(v/v) of the extract. However, the other leaf extracts except acacia inhibited non-selectively the growth of three turfgrass species such as bentgrass, perennial ryegrass and zoysiagrass and annual bluegrass. The PAL activities of annual bluegrass increased at 24 h after treatment of acacia leaf extract and peaked at 36 h and then decreased till 60h. The highest PAL activity was observed at 36h after treatment of 10%. The highest activity of CA4H in annual bluegrass was observed at 2h after treatment of acacia extract and the level was 4 times greater than that of the control. The phenolic acids such as p-coumaric acid, salicylic acid and ferulic acid were increased with the treatment of acacia leaf extract. The chloroplast membrane and cell wall of annual bluegrass were destroyed by treatment of acacia leaf extract and its inner materials were released. The membranes in annual bluegrass cells might be destroyed by phytotoxic compounds from acacia leaf extract.

Anti-melanogenic effects of Hordeum vulgare L. barely sprout extract in murine B16F10 melanoma cells

  • Purpose: Barely sprout is a well-known oriental herbal medicine with a wide range of health benefits. Recent studies have provided scientific evidence of its therapeutic effects with expanded application. This study investigated anti-melanogenic effect of barley sprout water extract (BSE) in murine melanocyte B16F10. Methods: Various concentrations (0, 50, 125, and $250{\mu}g/mL$) of BSE and arbutin (150 ppm) were applied to B16F10 stimulated with or without alpha-melanocyte stimulating hormone (100 nM) for 72 hours. The whitening potency of BSE was determined altered cellular melanin contents. Activity and expression of tyrosinase and microphthalmia-associated transcription factor (MITF) were also assayed. Results: Experimental results revealed that treatment with BSE reduced cellular melanin production by approximately 40% compared to the control. Molecular findings supported that suppressed activity and expression of tyrosinase and MITF proteins by BSE were associated with declined cellular melanogenesis. Furthermore, anti-melanogenic effect of BSE ($250{\mu}g/mL$) was similar to that of arbutin, a commonly used whitening agent. Lastly, polyphenols including p-coumaric, ferulic, and vanillic acids were identified in BSE using HPLC analyses. They might be potential active ingredients showing such melanogenesis-reducing effect. Conclusion: BSE was evident to possess favorable anti-melanogenic potency in an in vitro model. As a natural food sourced material, BSE could be an effective depigmentation agent with potential application in pharmaceutical and cosmetic industries.

서양금혼초(Hypochaeris radicata L.)의 수용추출물이 사료작물에 미치는 allelopathy 효과 (The Allelopathic Effects of Aqueous Extracts of Hypochaeris radicata L. on forage Crops)

  • 서양금혼초(Hypochaeris radicata Linne)에 함유되어있는 화학물질이 사료작물의 미치는 allelopathy효과를 구명하기 위하여 본 실험을 실시하였다 서양금혼초에 함유되어 있는 allelochemicals 물질을 HPLC에 의해 성분 분석한 결과 계절적, 부위별로 차이가 있었으나 $\rho$-hydroxybenzoic acid, chlorogenic acid, catechin, caffeic acid, syringic acid, salicylic acid, $\rho$-coumaric acid, ferulic acid, naringin, hesperidin, myricetin, krans-cinnamic acid, quercetin, naringenin 등 14 종류의 phenolic compounds 성분을 확인하였고 caffeic acid, ferulic acid, naringenin 3종류는 공통으로 함유하고 있는 것으로 분석되었다. 5월에는 일, 7월에는 꽃과 뿌리에서 많은 양의 phenolic acids가 함유되어있고 특히 뿌리에 sali-cylic acid $2085.6{\mu}g/g$,와 quercetin $1522.0[\mu}g/g$,로 함량이 높았다. 서양금혼초 수용성 추출액으로 처리한 사료작물의 생장을 조사한 결과 품종에 따라 처리구와 대조구의 값이 같은 경우도 있으나 모든 조사 항목에서 대조구 보다 처리 값이 낮아 사료작물의 생장이 억제 된 것이 확인되었고 처리농도가 높을수록 강한 억제효과를 나타내었다. 수량의 변화를 조사한 결과 사료작물 품종에 따라 차이가 있으나 전체적으로 억제효과를 나타내었으며 처리농도가 높을수록 강한 억제효과를 나타내었다. 이러한 결과 서양금혼초는 사료작물에 대한 allelopathy 효과가 있음을 밝혀냈다. 특히 화본과 사료작물의 경우 italian ryegrass, 콩과 사료작물의 경우는 purple alfalfa가 생장억제효과가 높게 나타났다.

루이보스차(Asphalthus linearis)의 추출방법에 따른 페놀릭류 함량 변화연구 (Determination of Phenolic Contents in Rooibos (Asphalthus linearis) Tea Depending on the Steeping Temperature and Time)

  • 루이보스티에서 5종의 phenolic acid (gallic acid, chlorogenic acid, caffeic acid, p-coumaric acid, trans-ferulic acid)와 9종의 flavonoid (procyanidin b1, aspalathin, rutin, vitexin, hyperoside, isoquercitrin, luteolin, quercetin, chrysoeriol)를 UPLC-MSMS를 이용하여 동시 분석하였다. 14종 페놀릭류를 동시 분석하기 위하여 기기조건과 유효성을 검증하였고 확립된 분석방법을 이용하여 시중에 유통중인 루이보스티 30건을 채취하여 페놀릭류를 분석하였다. 루이보스티 1 g 혹은 1티백에 뜨거운 물 100 mL을 가하여 3분, 6분, 30분이 경과 후 그리고 차가운 물 ($25-30^{\circ}C$)에 30분 우려낸 루이보스티의 페놀릭류 함량을 구하였다. 루이보스티에서 전체 실험대상 페놀릭류 중 rutin과 aspalathin이 가장 많이 추출되어 나왔으며 각각 물질의 함량은 제품별로 달랐다. 페놀릭류 성분의 추출효율은 14종 페놀릭류의 총합 기준으로 뜨거운 물 30분 > 6분 > 3분 > 차가운 물 30분 순으로 높았다.

고압처리에 의한 발아팥의 기능성 향상 효과 (Effect of Enhancement on Functionality of Germinated Adzuki Bean (Angularis angularis var. nipponensis) with High Hydrostatic Pressure (HHP) Treatment)

  • We investigated the influence of germination and high hydrostatic pressure (HHP) treatment conditions on the conversion of functional compounds and antioxidant activity in adzuki bean. The adzuki bean germinated at $25^{\circ}C$ for three- or six-days, and was later subjected to HHP at 0.1, 50, 100, or 150 MPa for 24 h. The highest polyphenol content (5.36 mg gallic acid equivalents (GAE)/g) and flavonoid content (0.91 mg catechin equivalents (CE)/g) were observed after germination for six days and HHP treatment at 100 MPa for 24 h, respectively. The total phenolic acid contents increased with increasing applied pressure from 88.86 to $208.26{\mu}g/g$ (100MPa, 24h). Phenolic acids are divided into two categories; those that exhibit increased content upon HHP treatment, and those that exhibit decreased content. The increasing phenolic acids were gallic acid, chlorogenic acid, (+)-catechin, ${\rho}-coumaric$ acid, ferulic acid, heperidin, salicylic acid, protocatechuic acid, cinnamic acid, naringenin. The total anthocyanin content decreased with increasing applied pressure from 22.42 mg/100 g to 6.28 mg/100 g (150 MPa, 24 h). The highest ABTS radical scavenging activity (8.02 mg eq AA/g) and DPPH radical scavenging activity (1.22 eq Trolox/g) were observed after germination for six days and HHP treatment at 100MPa for 24h, respectively. These results suggested that the combination of HHP and germination can lead to improved functionality in adzuki bean.

Allopathic Effect of Black Locust (Robinia pseudoacacia L.) and Analysis for Its Allelochemicals

  • 아까시나무에 있어서 알레로파시 효과를 분석하기 위해 아까시나무를 포함하여 13개 수 종에 대한 종자발아 시험을 실시하였다. 아까시나무의 뿌리와 잎의 침출액을 실험에 사용하였다. 침출액에 대한 종자 발아실험 결과 수종에 따라 실험결과가 크게 다르게 나타났다. 아까시나무 잎 침출액에서 아까시나무 종자 발아는 30%가 억제되었다. 실험한 수종가운데 무궁화와 오리나무는 뿌리 침출액에서 억제되었으나 누운측백나무는 잎의 침출액에서 발아가 억제되었다. 아까시나무의 잎과 뿌리 침출액을 HPLC분석 한 결과 알레로파시 효과를 가지고 있는 8종류의 authentic phenolic compounds를 찾아냈다. 잎에서는 전체 페놀물질 중 40%를 차지하고 있는 gentisic acid를 뿌리에서는 syringic acid와 vanillicacid를 발견하였다. 전체적으로 볼 때 뿌리에서.보다 잎에서 7배가 많은 phenolic acids가 추출되었다 조사된 8개 phenolic acid 중에서 뿌리에서는 vanillin만이 잎에서는 coumaricacid 만이 발견 되었다. 결과적으로 13수종 중에 3개 수종에서 잎과 뿌리의 침출액에서 종자 발아가 억제되었다. 종자발아 시험결과 아까시나무의 화학물질의 상호작용은 수종에 따라 차이가 많이 있음을 밝혀냈다. 아까시나무의 phenolic compound 총량은 알레로파시 역할에 큰 영향을 미치지 않았으나 이들 물길의 상승효과가 주요한 역할을 하는 것으로 추정 하였다.
