본 연구에서는 염색폐수의 색도제거 기전을 미생물 floc에의 물리 화학적 흡착과 미생물의 대사에 의한 생물학적 제거의 두 가지로 분류하였다. 색도제거의 반응 조건, 즉 혐기/호기 조건, cosubstrate의 종류와 주입량 등을 회분식 실험에 의해 규명하고, 활성슬러지와 비활성슬러지의 biosorption 실험을 통하여 색도제거 기전을 확인하였다. 염색폐수의 색도는 호기조건과 혐기조건에서 각각 102 ${\Delta}$unit/g MLSS, 123 ${\Delta}$unit/g MLSS가 제거되어 혐기조건에서의 제거율이 높았으며, 유기물은 호기조건에서 82 $mg{\Delta}$COD/gMLSS, 혐기조건에서 75 $mg{\Delta}$COD/gMLSS 제거되어 호기조건에서 제거율이 더 높게 나타났다. Cosubstrate로서 실폐수인 가정하수와 acetate를 이용하여 주입량에 따른 염색폐수의 제거능을 분석한 결과, cosubsrate의 주입에 따라 색도 및 유기물 제거량이 증가함을 확인하였으며 가정하수보다는 acetate가 색도제거에 있어서 더 효율적인 cosubstrate임을 알 수 있었다. 활성슬러지와 멸균된 비활성슬러지를 이용한 색도제거 실험 결과. 비활성슬러지의 색도제거량은 가정하수의 주입에 따라 $20.3{\sim}37.3$${\Delta}$unit/g MLSS, 활성슬러지는 $102.0{\sim}159.0$${\Delta}$unit/g MLSS로 나타났다. 또한 반응 초기에는 물리 화학적 흡착이 우세하였으나 시간이 지나면서 생물대사 작용에 의한 제거가 증가하는 경향을 보였으며, cosubstrate의 주입에 따른 미생물의 대사에 의한 제거분율이 증가하는 것으로 분석되었고, 이는 호흡율 측정결과와도 그 경향이 일치하였다.
The concept for the decolorization in biological dye wastewater treatment systems is based on anaerobic treatment, for the reductive cleavage of the dyes' azo linkages, in combination with aerobic treatment, for the degradation of the products from azo dye cleavage, aromatic amines. Batch tests were conducted to examine the conditions and the factors affecting biological treatment of dye wastewater. From the tests, the removal efficiencies of organics and colors of dyeing wastewater were improved to $COD_{Cr}$ 27% and color 9% by injecting 10% of the domestic wastewater as a cosubstrate, and $COD_{Cr}$ 30%, color 22% with 30% injection of domestic wastewater. Therefore it was proved that decolorization efficiency is demonstrated with domestic wastewater as a cosubstrate. The analysis of aromatic amines in wastewater showed that decolorization was achieved by cometabolism while aromatic amines were produced by cleavage of azo bonds under anaerobic conditions and these products were removed in an aerobic tank subsequently.
본 연구에서는 단일 탄소원인 포도당으로부터 P PHA를 생산하는 콩시균 Pseudomonas sp. HJ를 보다 정확하게 동정하기 위하여 수치분류를 실시하 였으며, 또한 보조기질을 첨가하였을때의 PHB/HV 합성 양상을 검토하였다. Pseudomonas sp. HJ는 각 대조균주들중에서 Pseudomonas picketti와 가장 유 사도가 높은 것으로 나타났다(78.8%). 그러나 수치 분류에 서 통일 균종에 포함될 수 있는 유샤도는 85 % 정도인 것으로 얄려져 있으므로, 본 공시균 P Pseudomonas sp. HJ는 Pseudomonas 속의 다른 종 (species)이거나 새로운 종(species)일 가능성이 높 았다. 보조기 질로 hexadecane과 propionic acid를 각각 0.4%씩 첨가했을 경우, 농도증가에 비례하여 H HV 함량은 점차 증가하여 각각 60.8 mol%, 44.9 mol%까지 축적되었다. PHB/HV내의 HV 함량은 p propionic acid냐 hexadecane의 농도를 달리하여 첨가함으로써 조절할 수 있었다. 보조기질로 propl- onic acid 0.2 %를 대수기 중기 인 배양 18시간에 첨 가하였을때 균체 생육이 가장 우수하였으나, HV monomer의 비율은 대수기 말기인 배양 24시간대에 첨가하였을때 49.6 mol%로 가장 높았다.
In this research, a retrofitting process, which consists of a pretreatment system (coagulation) for dye wastewater combined with a biological nutrient system (MLE process using media), for a sewage treatment plant that has to treat dye wastewater efficiently with domestic wastewater were developed and a pilot plant was operated for verifying adoptability and performance of the developed advanced process for dye wastewater. From the results of the pilot plant operation, BOD 52.9%, $COD_{Cr}$ 55.9%, and color 71.3% were removed in pretreatment of coagulation process and the biodegradability of dye wastewater was improved to $0.32{\sim}0.59BOD/COD_{Cr}$ of the coagulated wastewater from $0.29{\sim}0.43BOD/COD_{Cr}$ of the raw dye wastewater. The final effluent concentrations were BOD of 8 mg/L, $COD_{Cr}$ of 43 mg/L, $COD_{Mn}$ of 18 mg/L, T-N of 8 mg/L, and T-P of 1.3 mg/L, respectively. Color was removed from 1655 to 468 unit by coagulation and then to 123 unit by MLE process. The HPLC analysis of aromatic amines in wastewater showed that decolorization was achieved by cometabolism while aromatic amines were produced by cleavage of azo bonds under anaerobic conditions and these products were removed in an aerobic tank subsequently. Nitrification rates of attached and suspended microorganisms were evaluated comparatively and the acclimating conditions of bacteria on media were validated by the scanning electron microscope.

Phenol, acetaminophen (AA) and salicylamide are all known to be sulfated in rats and mice. We have previously demonstrated that capacity-limited sulfation of xenobiotics in rats is due to the reduced availability of hepatic 3'-phosphoadenosine 5'-phosphosulfate (PAPS), the cosubstrate for sulfation, which in turn is limited by the availability of its precursor, inorganic sulfate. Because species differences have been reported in the extent of sulfation, this study was conducted to determine whether these xenobiotics lower hepatic PAPS and sulfate in ICR mice. All three substrates decreased serum sulfate concentrations in a dose- and time-dependent manner. However, contrary to the observations in rats, phenol markedly increased hepatic PAPS concentrations in a dose-dependent manner, 1 hr after ip injection of 0∼4 mmol/kg. Following ip injection of 4 mmol/kg phenol, hepatic PAPS concentraions were enhanced 2∼3 fold, 0.5-2 hr after dosing and returned to control values 3 hr after dosing, whereas AA and salicylamide had little effect on hepatic PAPS concentraions. In summary, these studies demonstrate that phenol markedly enhances hepatic PAPS concentrations in mice, whereas hepatic PAPS levels are not affected by AA and salicylamide. Our data suggest that 1) hepatic sulfation for high dosages of xenobiotics in ICR mice is not limited by the availability of cosubstrate and 2) there are significant species differences in the regulation of PAPS between rats and mice.
For the production of cis, cis-muconic acid via biocatalytic conversion reactions from a toxic cosubstrate, benzoic acid, a fed-batch process using computer-controlled DO-stat feeding was developed. The mutant strain of Pseudomonas putida BM014 produced cis, cis-muconic acid from benzoic acid with high conversion yield. More than 32 g/L of cis, cis-muconic acid was accumulated in 42h and a productivity of 1.4g/(L.h)was achieved.
Fermentation characteristics of recombinant Saccharomyces cerevisiae containing the xylose reductase gene from Pichia stipitis were analyzed in an attempt to convert xylose to xylitol, a natural five-carbon sugar alcohol used as a sweetener. Xylitol was produced with a maximum yield of 0.95 (g xylitol/g xylose consumed) in the presence of glucose that is used as a cosubstrate for cofactor regeneration. However addition of glucose caused inhibition of xylose transport and accumulation of ethanol. Such problems were solved by adopting glucose-limlted fed-batch fermentation. This process done with S, cerevisiae EHl3.15:pY2XR at$30\;^{\circ}C$ resulted in 105.2g/L xylitol concentration with maximum productivity of 1.69 g $L^{-1}$$hr^{-1}$.
Pseudomonas putida KCTC 2401 degrades 1,1, 2-trichloroethylene (TCE) using phenol as a cosubstrate. The initial TCE degradation rate decreased with the initial TCE concentration up to 20mg/l of TCE at $30^{\circ}C$ and pH 6.5. The initial degradation rate and total removal efficiency increased with inoculum size. The strain also degraded dichloroacetic acid, which was supposed to be a degradation by-product. Phenol monooxygenase apparently participates in the TCE degradation mechanism.
Poly(3-hydroxyalkanoate) copolyesters containing both carbon-carbon double and carbon-carbon triple bonds were produced by Pseudomonas oleovorans grown in mixtures of 10-undecynoic acid (10-UND($\equiv$)) and 10-undecenoic acid (10-UND(=)). The PHA content in the dry cells was usually 40 wt%. The bioconversion yield of ($10-UND({\equiv})$) to PHA by P. oleovorans was remarkably enhanced from 1% to over 24% as the fraction of 10-UND(=) in the carbon substrate mixtures increased from 0 to 50%. These values were higher than those obtained when P. oleovorans was grown in the same molar mixtures of ($10-UND({\equiv})$) and nonanoic acid (NA), indicating that 10-UND(=) was more efficient than NA as a cosubstrate in inducing cometabolic PHA production.
For polyhydroxyalkanoic acid (PHA) production, several microorganisms were isolated from sewage sludge. One of them, GB-77 strain, was chosen from its PHB/HV copolymer production on only fructose without cosubstrate. The isolated strain GB-77 was identified as the genus Alcaligenes. Optimal temperature and pH for cell growth were 36C and 6.8. Optimal medium composition was 10 g/l of fructose and 5 g/l of polypeptone, 1 $\times$ 10$^{-2}$M Na$^{2}$HP0$^{4}$, 1.3 $\times$ 10$^{-2}$M KH$^{2}$PO$^{4}$. To investigate the optimal condition for polyhydroxyalkanoic acid production two-stage culture technique was used; first stage for cell growth and second stage for PHA production on unbalanced growth conditions. Optimal conditions for high PHA production were C/N ratio 50, temperature 36$\circ$C and pH 6.8. To overcome fructose inhibition on cell growth, intermittent feeding fed-batch culture technique was used. Total cell concentration was 17.4 g/l with 9.1 g/l of PHA. The purified PHA was identified PHB/HV copolymer by NMR analysis.
