Urinary Cortisol Levels in Japanese Shorthorn Cattle before and after the Start of a Grazing Season

  • We conducted two experiments to assess the effect of transfer from housing to grazing on stress hormone secretion in cattle using urine samples. In a preliminary experiment, urine samples were collected following an adrenocorticotrophic hormone (ACTH) challenge, and cortisol levels in urine were compared with the levels in plasma. In a second experiment, urinary cortisol was measured before and after the start of a grazing season in 6 Japanese Shorthorn cows, all of which had experienced grazing before. In experiment 1, urinary cortisol showed a pattern of changes similar to that of plasma with a 0.5-h temporal lag time, and the peak levels were 4 to 10 times higher than the basal levels. In experiment 2, the urinary cortisol levels in cows did not change after the cows were let out to pasture, with no decreases in body weight. This study suggests that the transfer from housing to grazing did not affect physiological responses to cause high excretion of urinary cortisol in grazing-experienced cattle using a non-invasive sampling method.

  • This study was conducted to evaluate the ELISA kit for measuring the level of cortisol in the urine. The CV of within-run variation and day to day variation were 0.4~2.8 and 1.8~5.7, respectively. The minimum limitation of measurement was 1ng/ml. The cross reaction was high ($CR_{50}(%)=11.4{\sim}43.2$) in prednisolone, 11-deoxycortisol, 21-deoxycortisol and predinosone. There was low and no cross reaction in other steroid. To develop the ELISA kit we measured the cortisol level in diluted urine with PBS (procedure I), extracted urine with methylene chloride (procedure II) and extracted methylene chloride-extracted urine from thin-layer chromatography (procedure III). The CV value of procedure I, II, III was 9.4~28.3%, 7.2~8.9% and 2.5~5.7%, respectively. There was significant difference between procedure I with II, and pro-cedure I with III(p < 0.01), but no difference between procedure II with III significantly(p < 0.01). The mean UCCR of urine collected through am 8 to 10 was $9.5{\pm}7.6$(0.14~28.0) in 12-month-old dog(n = 47). In this study we can measure the cortisol level in extracted urine with methylene chloride and sequential thin-layer chromatography accurately using ELISA kit.

Correlation of Occupational Stress Index with 24-hour Urine Cortisol and Serum DHEA Sulfate among City Bus Drivers: A Cross-sectional Study

  • Objectives: The questionnaire of occupational stress index (OSI) has been popular in the workplace, and it has been tailored for bus drivers in Taiwan. Nevertheless, its outcomes for participants are based on self-evaluations, thus validation by their physiological stress biomarker is warranted and this is the main goal of this study. Methods: A cross-sectional study of sixty-three city bus drivers and fifty-four supporting staffs for comparison was conducted. Questionnaire surveys, 24-hour urine cortisol testing, and blood draws for dehydroepiandrosterone-sulfate (DHEA-S) testing were performed. The measured concentrations of these biological measures were logarithmically transformed before the statistical analysis where various scores of stressor factors, moderators, and stress effects of each OSI domain were analyzed by applying multiple linear regression models. Results: For drivers, the elevated 24-hour urine cortisol level was associated with a worker's relationship with their supervisor and any life change events in the most recent 3 months. The DHEA-S level was higher in drivers of younger age as well as drivers with more concerns relating to their salary and bonuses. Non-drivers showed no association between any stressor or satisfaction and urine cortisol and blood DHEA-S levels. Conclusion: Measurements of biomarkers may offer additional stress evaluations with OSI questionnaires for bus drivers. Increased DHEA-S and cortisol levels may result from stressors like income security. Prevention efforts towards occupational stress and life events and health promotional efforts for aged driver were important anti-stress remedies.

Cortisol, $25OHD_3$ 추출 후 원심 분리시 온도가 검사결과에 미치는 영향 (Effect of the Centrifugation Temperature on Cortisol, $25OHD_3$ Values After Extraction)

  • Purpose: We use the centrifugation of refrigeration state in separation of blood serum, Anti-ds-DNA, Vitamin $B_{12}$/Folate and GAD-Ab assay. However, Cortisol urine and 25-Hydroxyvitamin $D_3$ ($25OHD_3$) are conducted centrifuge at room temperature. This is troublesome that change centrifugation temperature into room temperature due to using of most assays at cold temperature. Therefore when using centrifuge after extraction of Cortisol urine and $25OHD_3$, we conducted researches on effect of the centrifugation temperature in assay results. Materials and Methods: In Cortisol urine, add dichloromethane 1.0 mL in urine $500\;{\mu}L$, mix for 15 minutes, and then centrifuge for 8 minutes at 2600rpm. In $25OHD_3$ add acetonitrile 0.5 mL in serum $200\;{\mu}L$, and then centrifuge for 8 minutes at 2600rpm. Those experiments were conducted centrifuge at room temperature and $4^{\circ}C$. And experiments conducted immediately after centrifugation at $4^{\circ}C$ and standing for 20 minutes after centrifugation $4^{\circ}C$. Results: In Cortisol urine, room temperature result in 1.93, 2.18, 2.43, 9.45, 14.2 (${\mu}g/dL$). Experiments of performing immediately after centrifuge at $4^{\circ}C$ result in 1.8, 2.0, 2.3, 8.1, 13.7 (${\mu}g/dL$). Experiments of performing after 20 minutes result in 2.1, 2.1, 2.7, 9.95, 14.35 (${\mu}g/dL$). On the other hand, the $25OHD_3$ tests conducted at room temperature result in 7.13, 26.6, 35.8, 48.2, 74.8 (ng/dL). Experiments were conducted immediately by pipetting after $4^{\circ}C$ centrifugation result in 7.53, 30.9, 40.3, 61.5, 89.1 (ng/dL) as results are higher than experiments at room temperature. The experiments that conducted centrifuge at $4^{\circ}C$ and then left at room temperature for 20 minutes result in 7.40, 32.4, 41.3, 51.6, 85.6 (ng/dL). Conclusions: Experiments were conducted by using centrifuge at $4^{\circ}C$ are higher or lower than room temperature. The differences between results of standing for 20 minutes after centrifuge at $4^{\circ}C$ and those of centrifuge at room temperature are less than conducting immediately. It is concerned that experiments conducted immediately after centrifuge at $4^{\circ}C$ are incorrect, because tubes become dim due to temperature differences between $4^{\circ}C$ and room temperature. Therefore, it is desirable to centrifuge at room temperature as manual and we should pipet promptly without stopping.

피부 마사지가 미숙아의 스트레스 호르몬에 미치는 효과 (The Effects of Massage on Stress Hormone in Premature Infants)

  • Purpose: This study was done to evaluate the effects of massage on the level of stress hormone in the urine in preterm infants. Method: The design was a nonequivalent control group pretest-posttest design quasi experimental study. Fifty-eight preterm infants were assigned to the experimental(31) or control group(27). The data were collected from March 2002 to August 2003. The massage stimulation was provided to infants in the experimental group for 15-minutes twice a day for 10 days. On day 1 and day 10 of the study, a 24 hour-urine sample was collected for norepinephrine, epinephrine, and cortisol assays. In data analysis, SPSSWIN 10.0 program was utilized for descriptive statistics, ANOVA and t-test. Results: General characteristics of the two groups showed no significant differences, thus the two groups were found to be homogenous. The 24 hour-urine cortisol of the massage group (t=4.61, p=.000) was significantly reduced compared to the control group after 10 days. Conclusions: The results suggest that the massage stimulation can be used to reduce 24 hour-urine cortisol in preterm infants. Therefore, massage provided in the incubator is recommended for reduction of stress in preterm infants who are hospitalized in neonatal intensive care units.

신비탕(神秘湯) 전탕액(煎湯液)이 가토(家兎)의 신장기능(腎臟機能), 혈압(血壓) 및 혈장(血漿) Cortisol농도(濃度)에 미치는 영향(影響) (Effects of Sin Bee Tang Water Extract on the Renal Function, Arterial Blood Pressure and Plasma Cortisol Concentration in the Rabbit)

  • This study was investigated to clarify the effect of Sin Bee Tang (神秘湯) on the renal function, arterial blood pressure and plasma cortisol. The results obtained were follows; 1. Urine volume and glomerular filtration rate were decreased significantly after Sin Bee Tang water extract, 0.1ml/kg, administration. 2. Glomerular filtration rate, renal plasma flow and urinary excretion of electrolytes were increased significantly after Sin Bee Tang water extract, 0.25ml/kg, administration. 3. Plasma cortisol concentration increased significantly after Sin Bee Tang water extract, 0.25ml/kg, administration. These results suggest that the therapeutic action of Sin Bee Tang for 'Su Chun (水喘)' has a relation with the increase of plasma cortisol and renal hemodynamic effect.

Urinary Catecholamine and Cortisol Responses of Japanese Shorthorn Cows to Social Isolation

  • This study was performed to investigate the use of urinary catecholamines to monitor changes in the activity of the sympathetic nervous system and to determine the relationship of urinary cortisol and catecholamines in Japanese Shorthorn cows in response to social isolation. One cow was isolated from its group, which consisted of 14 cows (457 to 756 kg BW, 2 to 12 years old), for three days. The isolated cow was in contact with the other cows visually only at meal times. This isolation was repeated for 6 cows. Spontaneously voided urine samples were collected from the experimental animals once a day, before the treatment and on days 1, 2, and 3. Urinary cortisol and adrenaline levels were significantly increased compared with pre-isolation levels on the first day, and then declined to the basal levels during the next two days. Urinary noradrenaline levels changed in the same way as cortisol and adrenaline levels, but the difference was not significant. Urinary cortisol levels tended to be correlated with those of urinary adrenaline, but not noradrenaline. This study suggests that the urinary adrenaline levels can be a non-invasive indicator of stress and that the change of urinary adrenaline is similar to that of urinary cortisol.

청각 및 전정 자극이 스트레스 호르몬에 미치는 효과 - 미숙아를 대상으로 - (The Effects of Auditory and Vestibular Stimulation on Stress Hormones in Preterm Infants)

  • Purpose: This study was done to determine whether providing auditory and vestibular stimulation to preform infants would have an effect on stress hormones. Methods: The design was a nonequivalent control group protest-posttest design in a quasi-experimental study Seventy-nine preform infants were assigned either one of two experimental groups or to a control group: 27 in the auditory stimulation group, 25 in the vestibular stimulation group and 27 in the control group. The criteria for inclusion in this study were 1) gestational age of less than 37 weeks, 2) birth weight of less than 2,500g, 3) the absence of congenital anomalies or specific diseases, 4) recovering physiological weight loss, and 5) weaned from ventilatory assistance or oxygen. The data were collected from March 2002 to May 2003. The auditory stimulation, a music audiotape, was provided 20 minutes twice a day for 10 days and the vestibular stimulation, an infant waterbed, was provided for 10 days. On day 1 and day 10 of the study, 24 hour urine sample was collected for norepinephrine, epinephrine, and cortisol assays. In the data analysis SPSSWIN 10.0 program was utilized for descriptive statistics, ANOVA and t-test. Results: General characteristics of the three groups showed no significant differences, thus three groups were found to be homogenous. The 24 hour urine cortisol for the auditory (t=3.489, p=.001) and for the vestibular (t=2.638, p=.013) stimulation group were significantly reduced compared to the control group after 10 days. Conclusions: The results suggest that auditory and vestibular stimulation can be used to reduce 24 hour urine cortisol in preform infants. Therefore, music audiotapes and waterbeds provided in incubator are be recommended for reduction of the stress in preform infants who are hospitalized in neonatal intensive care units.

감각자극이 미숙아의 체중, 스트레스호르몬 및 행동상태에 미치는 효과 (Effects of a Sensory Stimulation on Weight, Stress Hormone and Behavioral State in Premature Infants)

  • This study has been conducted on the nonequivalent control group pretest-posttest design in quasi experimental basis and newly born premature infants from intensive care unit of G Medical University Hospital in Inchon Metropolitan were selected in two groups of 21 infants each. The first group for experimental and the other for control. Data has been collected form October 30, 1997 to August 29, 1998. For the experimental group tactile and kinesthetic stimulation was applied 2 times a day for 10 days(10 : 00~11 : 00 hours in the morning and 17 : 00~18 : 00 in the afternoon). As a weight weighing instrument. electronic indicator scale(Cas Co. korea) was used. To determine urine cortisol concentration level in stress hormone, radio immune assay method was used. And high performance liquid chlomatography was used to determine urine norepinephrine, concentration level To determine behavior status, tools developed by Anderson et at(1990) and remodeled by Kim Hee-Sook(1996) were used. Collected data were analyzed with the SAS program using x$^2$-test, student t-test, repeated measures ANOVA and paired t -test. The result were as follow. 1. As for the daily weight gain. the experimental group showed first change in weight and this group also showed higher weight in the average weight than the control group. Statistically, however, there was no significant factor between the two group. 2. The cortisol concentration in urine showed decrease in the experimental group norepinephrine concentration in urine showed increase in both experimental and control groups. No statistical significance was shown between the two groups. 3. In the aspect of behavior status. the experimental group showed statistical significance by showing inactive in the state of alert and conversion to a positive state than the control group. In conclusion, the sensory stimulation in this study showed a positive aspect through there was no statistical significance in the weight gain and urine stress hormone concentration. In the behavior status, there was statistical significance in the frequency of staying inactive in the state of alert and conversion to a positive state.

위탁견에서 스트레스에 의한 혈중 Cortisol의 농도 변화 (Changes in Serum Cortisol Concentration Due to Boarding Stress in Dogs)

  • Among the dogs boarding at an animal hospital, 10 dogs each were selected as a free boarding group and a kennel boarding group. Each group was further divided into 5 puppies and 5 mature dogs to examine the changes in their serum cortisol concentrations during boarding (5 days). We collected blood at day 8 and then analyzed the blood corpuscles, ran a biochemical serum test, and an additional urine test before boarding to check their health status. After collecting the dog's blood at 9:00 am, 3:00 pm, and 9:00 pm, we analyzed the cortisol concentration by the radioimmunoassay (RIA) method. The daily serum cortisol concentration at 9:00 am, 3:00 pm, and 9:00 pm, had a circadian rhythm during both the free and kennel boarding periods in both puppies and adults. Furthermore, the average daily serum cortisol concentration was significantly increased during the 1st, 2nd, and 3rd day after free boarding in the puppy group (P < 0.01). In adults from the free boarding group, the serum cortisol concentration dramatically increased on the 1st day (P < 0.01), as well as on the 2nd and 3rd day (P < 0.05). Interestingly, the average daily serum cortisol concentration was significantly increased on the 1st day after kennel boarding as well as during the entire period of kennel boarding in the puppy group (P < 0.05). In the adult kennel boarding group, serum cortisol concentration was significantly increased during the whole kennel boarding period (P < 0.01). An interesting result was that circadian rhythmicity in the sum of the daily serum cortisol concentrations was present in the free boarding group, but not in the kennel boarding group in both puppies and adults. In summary, cortisol was released depending on the degree of stress in free and kennel boarded dogs. Taken together, these results suggest that cortisol, a stress hormone, should be maintained at physiological concentrations in a circadian rhythm when the animals are hospitalized.
