In an effort to make it possible to cultivate Coptis japonica in Korea, the environmental and ecological conditions allowing the wild growth and distribution of Coptis sp. in Japan were investigated. The environmental conditions of the culture yard and the culture methods of C. japonica were also investigated. From these studies, it was concluded that the cultivation of C. japonica requires a well-drained sandy soil, $pH\;4.5{\sim}5.5$, which is facing North or Northwest. The area suitable for the cultivation of C. japonica should have rainfall of $1,000{\sim}1,500mm$ per year, be covered with snow for $30{\sim}60$ days in the winter, and be blocked $40{\sim}70%$, of the sun shine. These findings suggest that in korea the cultivation of C. japonica may be possible in the areas ranging from Sock-cho to Kang-neung, Mt. Odae, Mt. Taebaek and Mt. Sobaek where is chill and much moisturous in summer.
Coptis rhizoma was extracted with methanol or hot water. Methanol extracts had higher berberine content than water extracts, whereas the contents of Ca, Mg, Zn and K were higher in the water extracts. Rats were fed on the purified diets rich in 0.12% cholesterol only or supplemented with 0.5% of each extracts for four weeks. The concentration of total serum cholesterol was significantly lower in the rats given the extracts, irrespective of the extractants. Although the concentration of serum HDL-cholesterol was comparable among the groups, the ratio of HDL-/total-cholesterol was higher in rats given the extracts. The concentration of serum free fatty acids and ketone body were significantly elevated in rats given the extracts. On the other hand, the extracts supplements significantly decreased the contents of liver cholesterol and triacylglycerol. The amount of fecal bile acids was markedly higher in rats given the extracts, especially in those given water extracts. The results show an effective hypocholesterolemic action of Coptis rhizoma and it seems that the lower concentration of serum and liver cholesterol is resulted from the increased fecal bile acids in rats.
In oriental medicine, Coptis japonica $M_{AKINO}$ (Ranunculaceae) has been known as bitter stomachics. This plant originally grows wild and has been cultivated in Japan but not in Korea. However, a few years ago, some workers have been trying to cultivate this plant in Korea and have made much progress. Therefore, this . study was made for the purpose of conforming possibility in cultivation of Coptis japonica $M_{AKINO}$ in Korea. The results were as follows: 1) Berberine contents of rhizomes are much more than of K.P.. Difference in contents between Eu Jeon Bu $(4.88{\pm}0.77%)$ and Bo Seong area $(3.62{\pm}0.50%)$ is significant (p<0.05). 2) Berberine contents of fibrous roots of Eu jeong Bu $(0.97{\pm}0.28%)$ area was similar to that of Gang Nam $(0.93{\pm}0.21%)$, Yang Pyung area $(0.91{\pm}0.14%)$ but only contents of Bo Seong area $(0.74{\pm}0.25%)$ is less than that of other area and it's difference is not significant. 3) It was considered that the cultivation of Coptis japonica $M_{AKINO}$ in Korea is possible considering the cultivation results and berberine contents, and also more moderate than south region.
The purpose of this study is the separation of biologically active ingredients from Coptidis Rhizoma which has been widely used as one of oriental herbal medicine for body fever. In this study, berberine-type alkaloids were tested on their biological activities in the aspect of antibacterial, antitumor, anti-herpetic and anti-HIV activity. Contents of five major alkaloids for the various origin of Coptidis Rhizoma were assayed by HPLC. As the results, the content of berberine from Coptis chinensis and Coptis japonica were 6.78% and 7.09%, respectively. The contents of coptisine, jatrorrhizine, berberastine from Coptis chinensis were higher than those of Coptis japonica. The amount of palmatine from both species were almost the same. Surprisingly for antitumor experiment, all compounds have been shown remarkable activity, especially against SNU-l(human stomach cancer) cell line. Among the compounds purified through column chromatography, palmatine, coptisine, and jatrorrhizine inhibited the growth of K-562(human chronic myelogenous leukemia) cell line whereas jatrorrhizine has been shown the effective inhibition of A-549 (human lung) cell line at the same time.
This was a study examining the fatliquoring of natural dyed cow skin with Coptis chinensis Franch. The K/S according to optimal dyeing condition, mordants and mordanting method was measured. And the surface, sections, tensile strength, elongation, stiffness and colorfastness according to post-dyeing fatliquoring were observed. The optimal conditions include 300%(o.w.f.), $50^{\circ}C$, 30 minites. Fe-simmordanting showed the highest dyeability. Coptis chinensis Franch showed a yellow color regardless of mordants and mordanting methods. Tensile strength and elongation increased in the case of simmordanting compared with untreated and the process of fatliquoring further increased. The softness significantly increased when fatliquored. Shrinkage was slightly reduced when dyed, but showed almost no change with fatliquoring. Lastly, color fastness increased overall with mordanting and partly increased with fatliquoring. The rubbing and light fastness were excellent Cu- mordanting, and drycleaning fastness also improved with Cr-mordanting.
Recently we reported that water extracts of Coptidis Rhizomas showed calcium antagonistic action and alpha-adrenoceptor inhibitory action in the vascular smooth muscle. Since ca lcium antagonistic properties are important in the treatment of various diseases including asthma. In the present study, the bronchodilatory effects of crude extract of three kinds of Coptidis Rhizoma (Coptidis chinensis, Coptis japonica and root hair of Coptis japonica) was investigated using rat isolated trachea. The result showed that all extracts relaxed carbachol-contracted tracheal smooth muscle. Concentration-dependently, in which the root hair of Coptis japonica was the least potent. The inhibitory potency expressed in terms of $IC_{50}$ against carbachol contraction was 1.8${\mu}$g/ml and 2.7${\mu}$g/ml for Coptidis chinensis and Coptis japonica, respectively. These extracts also inhibited KCI-contracted tracheal smooth muscle. But the relative potency ($IC_{50}$) was 3.5 and 4.1 folds weaker than carbachol-induced contraction for Coptidics chinenesis and Coptis japonica, respectively. Pretreatment of crude extracts also inhibited carbachol- or KCI-induced contraction, non-competitively. These findings indicate that the extracts have muscarinic blocking as well as $Ca^{2+}$ channel blocking action. When provoked intracellular stored $Ca^{2+}$ release by carbachol in $Ca^{2+}$-free conditions, initial phasic contraction due to $Ca^{2+}$ release was significantly inhibited by the extracts. As taken together, we conclude that water extracts of Coptidis Rhizoma may be beneficial in bronchospasm or other broncheal tube narrowing conditions such as asthma.
Background : Anticancer effects of herbal medicine have been reported in various types of cancer, but the systematic approaches to explain molecular mechanism(s) are not established yet. Objective : To find the anticancer-effect and mechanism(s) of Water Extract of Coptis japonica (WECJ) colon cancer cell (SNU-81). Methods : We first selected 11 herbals, and anti-cancer effects of water-extracts from those herbals have been tested in human colon cancer cell line, SNU-81. Among the tested herbals, the WECJ significantly reduced proliferation of SNU-81. To establish a basis of understanding for anti-cancer mechanism, whole proteins have been obtained from SNU-81 harvested at 48 and 96 hrs after the treatment of WECJ, protein expression has been profiled by 2DE-based proteomic approach. Results : Various changes of the protein expression have been monitored, and most frequent dysregulation was found in the molecular chaperons including heat shock protein 90-alpha (Hsp90-alpha), 14-3-3 protein epsilon, T-complex protein 1 subunit alpha, protein disulfide-isomerase A3, and calreticulin. Interestingly, proliferation-associated protein 2G4 has been up-regulated, and it suggests the possible effect of Coptis japonica on ErbB3-regulated signal transduction pathway and growth control of human colon cancer cells. Conclusion : Based upon the present findings, the further study will focus on monitoring various cancer survival factors after artificial regulation of the proteins identified, and it would be the basis for the understanding of the Coptis japonica anti-cancer effect(s) at the molecular level.

This study was to verify the skin safety of Coptis Japonica extract(CJE) -containing cosmetics by patching them on sensitive skin and checking its stimulating level. 32 subjects diagnosed with sensitive skin by the result of the lactic acid injury test, were subjected to 20 ㎕ of solubilized essence mixed with 2% CJE on the test subjects' back and closed patched for 24 hours. No stimulus reactions were found by bare eyes and scored 0 stimulus figure points after they were taken off in 30 minutes, 24 hours, and 48 hours. Coptis japonica's medical efficacies such as antioxidant, and anti-inflammatory are expectable to be used as functional cosmetics, and this research would be the foundation of further research into its cosmetical usage.
The present study was undertaken to find a protective effect of Coptis Rhizoma Extract on liver injury induced by benzo(a)pyrene in rats. This crude drug significantly suppressed the increase of several biochemical parameters such as transaminase, lactate dehydrogenase and alkaline phosphatase in serum and liver and total cholesterol. But there were little changes in elevated serum phospholipid level. The results suggested that Coptis Rhizoma prevent bezo(a)pyrene-induced hepatic injury.
The effect of the root of Coptis japonica (COPT), both dichloromethane soluble $(CH_2Cl_2)$ and insoluble $(H_2O)$ fractions, on catecholamine contents and tyrosine hydorxylase (TH) activity in PC12 cells was investigated. $(CH_2Cl_2){\;}and{\;}H_2O$ fractions showed 21 and 53% inhibitions on dopamine content, respectively, at a ocncentraction of 40 .mu.g/ml in medium : the $(H_2O)$ fraction proveided a grateer inhibitory effect. The TH activity was reduced by the treatment of COPT ($(H_2O)$ fraction). These results suggest that COPT has an inhibitory effect on the catecholamine biosynthesis by the reduction of TH activity in PC12 cells.
