Controlled Ondansetron Release Based on Hydroxyethyl Starch Hydroxyethyl Methacrylate

  • Presented study describes the synthesis of photo cross-linkable and water soluble hydroxyethyl starch hydroxyethyl methacrylate (HESHEMA) samples with different degree of substitution (DS) by functionalization of hydroxyethyl starch (HES) with hydroxyethyl methacrylate (HEMA) or hydroxyethyl methacrylate carbonylimidazole (HEMACI) in DMSO using two different routes. It was revealed that the reaction time for HESHEMA synthesis can be reduced from 5 days to 24 h by conducting the reaction at $80^{\circ}C$ instead of at room temperature. Solubility of HESHEMA was found to be dependent on DS which in turn was dependent on ratio between HES and HEMA or HEMACI. HESHEMA samples with DS > 0.24 depicted insoluble in water, whereas the samples with DS < 0.05 did not form appreciable gel. HESHEMA samples with appropriate DS were converted into hydrogels by cross-linking polymer chains under UV radiations and resulting HESHEMA hydrogels showed swelling up to 1200%. Application of HESHEMA in controlled drug delivery was investigated by diffusion based encapsulation of Ondansetron, a serotonin 5-$HT_3$ receptor antagonist drug, mainly used for nausea and vomiting treatment.

Impact of Thickness of Sand Capping on Phosphorus Release Rate from Sediment (모래 캡핑 두께가 퇴적물 인 용출에 미치는 영향)

  • This research was carried out to understand the impacts of thickness of sand capping to control phosphorus release from sediment into overlying water. As capping effectively retards release flux, phosphorus concentration in water body can be maintained if phosphorus release rate was kept under microbial degradation rate. With capping thickness increases, deaeration rate become less, while reaeration coefficient become higher. Phosphorus release rate and capping thickness were linearly correlated. The results of regression analysis indicated that phosphorus release can be controlled effectively by sand capping of least 20${\sim}$40 mm thickness.

  • To explore the potential of shell crosslinked micelle (SCM) as a drug carrier, the drug release behavior of poly($\varepsilon$-caprolactone)-b-poly(acrylic acid) (PCL-b-PAA) SCMs was investigated. PCL-b-PAA was synthesized by ring opening polymerization of $\varepsilon$-caprolactone and atom transfer radical polymerization of tert-butyl acrylate, followed by selective hydrolysis of tert-butyl ester groups to acrylic acid groups. The resulting amphiphilic polymer was used to prepare SCMs by crosslinking of PAA corona via amidation chemistry. The drug release behavior of the SCMs was studied, using pyrene as a model drug, and was compared with that of non-crosslinked micelles, especially below the critical micelle concentration (CMC). When the shell layers were crosslinked, the drug release behavior of the SCMs was successfully modulated at a controlled rate compared with that of the non-crosslinked micelles, which showed a burst release of drug within a short time.

  • Purpose. To prepare PVP-based solid dispersions containing lovastatin (LOS) and solubilizers (sodium lauryl sulfate. Tween80. oleic acid) to enhance dissolution of practically insoluble LOS. Methods. Solid dispersions containing LOS were prepared by dissolving two different organic solvent systems (acetone/ethanol or acetonitrile/ethanol). Results. The stickiness and flowability of solid dispersion powders were dependent on the composition and ratio of the solubilizers. (omitted)

  • In case of oral application of quinupramine, antidepressants, it may cause adverse effects such as diarrhea, nausea due to transient high blood concentration. Ethylene vinyl acetate (EVA) which is heat-processible, flexible, inexpensive material was used for transdermal drug delivery. The purpose of this study was to develop the new transdermal delivery system of quinupramine using EVA polymer matrix that can provide sustained release and avoid the side effects. The EVA matrix containing quinupramine was prepared by solvent-evaporation method. (omitted)

Effects of Bovine Somatotropin (bST) Administration Combined with Controlled Internal Drug Release (CIDR) on Embryo Quality and Pregnancy of Hanwoo (Korean Native Beef Cattle) during Commercial Embryo Transfer Program

  • Effects of recombinant bovine somatotropin (bST) on plasma hormonal concentration, embryo quality, and pregnancy rate were examined during the superovulation and synchronization treatment in donor and recipient cows. Hanwoos (Korean native beef cattle) were treated with controlled internal drug release (CIDR) combined with bST (CIDR+bST) or without bST (CIDR) as donor cows. The embryos recovered from donors were transferred into Holstein recipient heifers treated with bST (CIDR+bST) or without bST (CIDR) for synchronization. The correlation between IGF-I and P4 showed a positive pattern in the CIDR+bST group (r=0.44, p<0.01), but a negative pattern was shown in the CIDR group (r = -0.59, p<0.02) at day 7 of estrous cycles. Although the number of recovered, transferable, and degenerated embryos was not different, quantities of grade 1 (excellent) embryos in CIDR+bST group were significantly higher than those of the CIDR group (p<0.01). The pregnancy rate was higher in the CIDR+bST recipient group compared to CIDR group (p<0.05), when the embryos were recovered from the donors treated with CIDR. However, the pregnancy was maintained highly in both recipient groups, when the embryos were produced by CIDR+bST treated donors. It can be concluded that bST administration combined with CIDR is an effective method for superovulation and synchronization treatment to stabilize plasma hormonal levels, to obtain excellent quality of embryos, and to get higher pregnancy rate.

Preparation of Dexamethasone-21-palmitate Incorporated Lipid Nanosphere: Physical Properties by Varying Components and Ratio of Lipid (팔미틴산덱사메타손이 봉입된 지질나노입자의 제조: 지질종류와 함량에 따른 물리적 특성)

  • Intraarticular corticosteroid injections for therapy of rheumatic arthritis are administered with the aim of optimal local anti-inflammatory effect at the injection site. Since the side effects of corticosteroidal drug, dexamethasone(DEX), administered at hish dose limited the therapeutic efficacy, there was a need to design a new drug delivery system for controlled release of dexamethasone. As a prodrug for continuous therapeutic efficacy, dexamethasone-21-palmitate(DEX-PAL) was prepared via esterification of palmitoyl chloride and dexamethasone. DEX-PAL was identified by NMR and MASS analysis. DEX-PAL or DEX was entrapped in lipid nanosphere which could be prepared by using a self emulsification-solvent evaporation method. Physicochemical characteristics such as mean particle diameter, zeta potential and drug loading efficiency of the lipid nanospheres were investigated with variation of either the kind of lipid or the lipid composition. The lipid nanospheres had a mean diameter $83{\sim}95$ nm and DEX-PAL loading efficiency of up to 95%. The drug loading efficiency increased with the increase of aliphatic chain length attached to the phospholipid. The incorporation of cationic lipid was very efficient for both reducing particle size of lipid nanospheres and enhancing drug loading efficiency. The lipid nanospheres containing DEX-PAL may be a promising novel drug carrier for the controlled release of the poorly water-soluble drugs.

  • A Gompertz modeling, sigmoid in shape, is a widely used application for social science, natural science, engineering, and medical research to allow confident approximation and accurate analysis and has been applied to estimate an elderly population on aging of population. Due to the high toxicity of currently available drug delivery vehicles, various efforts have been made to reduce side-effects in clinical fields, but its application to preclinical and clinical studies is limited and there are some difficulties to optimize the parameters of Gompertz modeling applicable to preclinical studies. Therefore, in this study, we demonstrated the ability of sickle red blood cells loaded by hypotonic dialysis then photosensitized and light-activated ex vivo for controlled release and simultaneously optimized Gompertz function to evaluate controlled drug release properties of photosensitized sickle red blood cells to reduce pain-related treatments in cancer patients.

Leaching, soil residue, and volatilization of dicamba from controlled release granular formulations (방출조절형 dicamba 입제의 용탈, 토양잔류 및 휘산)

  • In order to minimize harmful side effects, extend weed control performance, and control the releasing rate of the active ingredient, dicamba controlled release granular formulations were prepared with starch as matrix. Leaching, soil residue, and volatilization of the granules were compared with dicamba soluble concentrate formulation (SL). Leaching of dicamba through the soil applied with the granules could be reduced more than 50% as compared with that of dicamba SL. The half life of dicamba in loamy soil treated with the granules was 50 to 51 days, while that in sand loamy soil was 50 to 58 days. Whereas, the half life of dicamba in loamy and sand loamy soil applied with dicamba SL was 24 and 22 days, respectively. Volatilization of dicamba from the granules was 10 times less than that of dicamba SL.
