Development of ArcGIS-based Model to Estimate Monthly Potential Soil Loss (월단위 토양유실가능성 추정을 위한 ArcGIS 기반의 모형 개발)

  • Soil erosion has been issued in many countries since it causes negative impacts on ecosystem at the receiving water bodies. Therefore best management practices to resolve the problem in a watershed have been developed and implemented. As a prior process, there is a need to define soil erosion level and to identify the area of concern regarding soil erosion so that the practices are effective as they are designed. Universal Soil Loss Equation (USLE) were developed to estimate potential soil erosion and many Geographic Information System (GIS) models employ USLE to estimate soil erosion. Sediment Assessment Tool for Effective Erosion Control (SATEEC) is one of the models, the model provided several opportunities to consider various watershed peculiarities such as breaking of slope length, monthly variation of rainfall, crop growth at agricultural fields, etc. SATEEC is useful to estimate soil erosion, however the model can be implemented with ArcView software that is no longer used or hard to use currently. Therefore SATEEC based on ArcView was rebuild for the ArcGIS software with all modules provided at the previous version. The rebuilt SATEEC, ArcSATEEC, was programmed in ArcPy and works as ArcGIS Toolset and allows considering monthly variations of rainfall and crop growth at any watershed in South-Korea. ArcSATEEC was applied in Daecheong-dam watershed in this study, monthly soil erosion was estimated with monthly rainfall and crop growth variation. Annual soil erosion was computed by summing monthly soil erosion and was compared to the conventional approach to estimate annual soil erosion. The annual soil erosion estimated by the conventional approach and by summing monthly approach did not display much differences, however, ArcSATEEC was capable to provide monthly variation of soil erosion.

Effect of Active Master Packaging System on Preservation of Fresh Shiitake Mushrooms in Supply Chain (유통과정에서 생표고버섯에 대한 Active 마스터 포장 시스템의 적용 효과)

  • Master packaging system consists of an inner individual package and secondary outer package. During the stages of chilled transport and distribution, the combination of primary individual package and secondary package was used to maintain a modified atmosphere for shiitake mushrooms. During the retail stage at higher temperature ($25^{\circ}C$), the primary individual package was exposed to display conditions after dismantling of the secondary packaging. The master packaging system was constructed to contain eight individual $30-{\mu}m$ thick polypropylene film bags of 500 g shiitake mushrooms inside a $40-{\mu}m$ low-density polyethylene bag. Carbon dioxide absorbent material [$Ca(OH)_2$] and/or moisture absorbent (superabsorbent polymer) were designed in their required amounts based on respiration characteristics and then applied to the outer secondary packaging in sachet form. Gas concentration of the packaging, temperature, and humidity were monitored throughout transport and storage. The quality of shiitake mushrooms was measured at the retail stage to determine the packaging effect. During the distribution stage of 108 h, $O_2$ and $CO_2$ concentrations in the master packaging system were maintained at 9~11% and 1~4% in the inner packaging, respectively, which are good for quality preservation. Compared to the control, the master packaging with $CO_2$ and/or moisture absorbents improved mushroom preservation and particularly reduced decay.

Analysis of Bovine Interferon-tau Gene subtypes Expression in the Trophoblast and Non-trophoblast cells

  • Interferon-tau (IFNT) is known as a major conceptus protein that signals the process of maternal recognition of pregnancy in ruminants. Also, multiple interferon genes exist in cattle, However, molecular mechanisms of these bovine IFNT (bIFNT) genes whose expressions are limited have not been characterized. We and others have observed that expression levels of bovine subtype IFNT genes in the tissues of ruminants; thus, bIFNT1 and other new type I (bIFNTc1/c2/c3) gene co-exist during the early stages of conceptus development and non-trophoblast cells. Its genes transcription could be regulated through CDX2 and ETS2 and JUN and/or cAMP-response element binding protein (CREB)-binding protein (CREBBP) expression, a transcription factor implicated in the control of cell differentiation in the trophectoderm. Bovine ear-derived fibroblast cells, were co-transfected with luciferase reporter constructs carrying upstream (positions -1000 to +51) regions of bIFNT1 and other new type I gene and various transcription factor expression plasmids. Compared to each - 1kb-bIFNT1/c1/c2/c3-Luc increased when this constructs were co-transfected with CDX2, ETS2, JUN and/or CREBBP. Also, Its genes was had very effect on activity by CDX2, either alone or with the other transcription factors, markedly increased luciferase activity. However, the degree of transcriptional activation of the bIFNTc1 gene was not similar to that bIFNT1/c2/c3 gene by expression plasmid. Furthermore, Sequence analyses also revealed that the expression levels of bIFNT1/c2/c3 gene mRNAs expression were highest on day 17, 20 and 22 trophoblast and, Madin-Darby bovine kidney (MDBK), Bovine ear-derived fibroblast (EF), and endometrium (Endo) non-trophoblast cells. But, bIFNTc1 mRNA had not same expression level, bIFNTc1 lowest levels than those of IFNT1/c2/c3 gene in both trophoblast and non-trophoblast cells. These results demonstrate that bovine subtype bIFNT genes display differential, in the trophoblast and non-trophoblast cells.

  • One of the major problems with the livestock industry is that in the summer, livestock will die from heat waves or infectious diseases. Under these circumstances, what livestock need is a proper indoor temperature and a regular sterilization system. Therefore, in this study, we developed a system that automatically controls the power of the fan according to the temperature inside the shaft, and a function that provides feed and sterilization on a regular basis, so that we could manage the toast efficiently. It also proposed the ability to automatically control the power of the fan, to display the temperature inside the shaft to mobile applications, and to provide food and sterilization. First, the function of controlling the power of the fan automatically turns the fan on when the temperature inside the shaft rises above a certain level. Conversely, if the temperature inside the shaft falls below a certain level, turn off the fan. Second, the function of the mobile application is to check the temperature inside the shaft. The third feeding function is periodically fed using a servo motor, and the feeding provided is delivered to livestock through a conveyor belt. Finally, the sterilization function is a function to sterilize livestock periodically using DC motor pump. The intelligent congratulatory functions proposed in this study may contribute to the health of livestock.

A Microcomputer-Based Data Acquisition System (Microcomputer를 이용(利用)한 Data Acquisition System에 관(關)한 연구(硏究))

  • A low cost and versatile data acquisition system for the field and laboratory use was developed by using a single board microcomputer. Data acquisition system based on a Z80 microprocessor was built, tested and modified to obtain the present functional system. The microcomputer developed consists of 6 kB ROM, 5 kB RAM, 6-seven segment LED display, 16-Hex. key and 8 command key board. And it interfaces with an 8 channel, 12 bits A/D converter, a microprinter, EPROM programmer for 2716, and RS232C interface to transfer data between the system and HP3000 mini-computer manufactured by Hewlett Packard Co., A software package was also developed, tested, and modified for the system. This package included drivers for the AID converter, LED display, key board, microprinter, EPROM programmer, and RS232c interface. All of these programs were written in 280 assembler language and converted to machine codes using a cross assembler by HP3000 computer to the system during modifying stage by data transferring unit of this system, then the machine language wrote to the EPROM by this EPROM programmer. The results are summarized as follows: 1. Measuring program developed was able to control the measuring intervals, No. of channels used, and No. of data, where the maximum measuring speed was 58.8 microsec. 2. Calibration of the system was performed with triangle wave generated by a function generator. The results of calibration agreed well to the test results. 3. The measured data was able to be written into EPROM, then the EPROM data was compared with original data. It took only 75 sec. for the developed program to write the data of 2 kB the EPROM. 4. For the slow speed measurements, microprinter instead of EPROM programmer proved to be useful. It took about 15 min. for microprinter to write the data of 2 kB. 5. Modified data transferring unit was very effective in communicating between the system and HP3000 computer. The required time for data transferring was only 1~2 min. 6. By using DC/DC converting devices such as 78-series, 79-series. and TL497 IC, this system was modified to convert the only one input power sources to the various powers. The available power sources of the system was DC 7~25 V and 1.8 A.

The Experimental Study for Myocardial Preservation Effect of Ischemic Preconditioning (허혈성 전조건화 유발이 심근보호에 미치는 영향에 관한 실험적 연구)

  • Decrease in cardiac function after open heart surgery is due to an ischemia induced myocardial damage during surgery, and ischemic preconditioning, a condition in which the myocardial damage does not accumulate after repeated episodes of ischemia but protects itself from damage after prolonged ischemia due to myocytes tolerating the ischemia, is known to diminish myocardial damage, which also helps the recovery of myocardium after reperfusion, and decreases incidences of arrythmia. Our study is performed to display the ischemic preconditioning and show the myocardial protective effect by applying cardioplegic solution to the heart removed from rat. Material and Method: Sprague-Dawley male rats were used, They were fixed on a modified isolated working heart model after cannulation. The reperfusion process was according to non-working and working heart methods and the working method was executed for 20 minutes in which the heart rate, aortic pressure, aortic flow and coronary flow were measured and recorded. The control group is the group which the extracted heart was fixed on the isolated working heart model, recovered by reperfusion 60 minutes after infusion and preserved in the cardioplegic solution 20 minutes after the working heart perfusion and aortic cross clamp, The thesis groups were divided into group I, which ischemic hearts that were hypoxia induced were perfused by cardioplegic solution and preserved for 60 minutes; group II, the cardioplegic solution was infused 45 seconds (II-1), 1 minutes (II-2), 3 minutes (II-3), after the ischemia induction, 20 minutes after working heart perfusion and aortic cross clamp; and group III, hearts were executed on working heart perfusion for 20 minutes and aortic cross clamp was performed for 45 seconds (III-1), 1minute (III-2), 3 minutes (III-3), reperfused for 2 minutes to recover the heart, and then aortic cross clamping was repeated for reperfusion, all the groups were compared based on hemodynamic performance after reperfusion of the heart after preservation for 60 minutes. Result: The recovery time until spontaneous heart beat was longer in groups I, II-3, III-2 and III-3 to control group (p<0.01). Group III-1 (p<0.05) had better results in terms of recovery in number of heart rates compared to control group, and recovered better compared to II-1 (p<0.05). The recovery of aortic blood pressure favored group III-1 (p<0.05) and had better outcomes compared with II-1 (p<0.01). Group III-1 also showed best results in terms of cardiac output (p<0.05) and group III-2 was better compared to II-2 (p<0.05). Group I (p<0.01) and II-3 (p<0.05) showed more cardiac edema than control group. Conclusion: When the effects of other organs are dismissed, protecting the heart by infusion of cardioplegic solution after enforcing ischemia for a short period of time before the onset of abnormal heart beats for preconditioning has a better recovery effect in the cardioplegic group with preconditioning compared to the cardioplegic solution itself. we believe that further study is needed to find a more effective method of preconditioning.

Identification of Differentially Expressed Genes Related to Intramuscular Fat Development in the Early and Late Fattening Stages of Hanwoo Steers

  • Marbling of cattle meat is dependent on the coordinated expression of multiple genes. Cattle dramatically increase their intramuscular fat content in the longissimus dorsi muscle between 12 and 27 months of age. We used the annealing control primer (ACP)-differential display RT-PCR method to identify differentially expressed genes (DEGs) that may participate in the development of intramuscular fat between early (12 months old) and late fattening stages (27 months old). Using 20 arbitrary ACP primers, we identified and sequenced 14 DEGs. BLAST searches revealed that expression of the MDH, PI4-K, ferritin, ICER, NID-2, WDNMI, telethonin, filamin, and desmin (DES) genes increased while that of GAPD, COP VII, ACTA1, CamK II, and nebulin decreased during the late fattening stage. The results of functional categorization using the Gene Ontology database for 14 known genes indicated that MDH, GAPD, and COP VII are involved in metabolic pathways such as glycolysis and the TCA cycle, whereas telethonin, filamin, nebulin, desmin, and ACTA1 contribute to the muscle contractile apparatus, and PI4-K, CamK II, and ICER have roles in signal transduction pathways regulated by growth factor or hormones. The final three genes, NID-2, WDNMI, and ferritin, are involved in iron transport and extracellular protein inhibition. The expression patterns were confirmed for seven genes (MDH, PI4-K, ferritin, ICER, nebulin, WDNMI, and telethonin) using real-time PCR. We found that the novel transcription repressor ICER gene was highly expressed in the late fattening stage and during bovine preadipocyte differentiation. This information may be helpful in selecting candidate genes that participate in intramuscular fat development in cattle.

A Study on the Yogurt Manufacture Suitability and Antimicrobial Activity of Lactobacillus plantarum LHB55 Isolated from Kimchi (김치에서 분리한 Lactobacillus plantarum LHB55의 항균성과 요구르트 제조 적합성 연구)

  • The aim of this study was to develop a new starter for fermented milk. The approach started with 103 acid-producing isolates from Kimchi, a type of spiced, fermented cabbage and then PCR screening was used to identify 72 Lactobacillus strains. The ability to inhibit the growth of food-borne human pathogens (Escherichia coli, Salmonella enteritidis, Staphylococcus aureus) of these strains were measured, using the paper disk method. Among them, one bacterium (LHB55) that showed a strong antibacterial activity against food-borne human pathogens was identified and further characterized, using 16S rDNA sequencing and API 50CHL system. Because this isolate was identified as L. plantarum, it was named as L. plantarum LHB55. The yogurt produced using commercial LAB with L. plantarum LHB55 did not display properties that are microbially or physico-chemically different from the control group, which suggests that L. plantarum LHB55 can be used as a useful starter for yogurt containing high antibacterial activity. We think that identifying effective starter strains enabling further development of fermented milk that can deliver better health benefits such as antimicrobial properties is of high significance, and thus our effort in this type of approach will continue.

A Study on the Natural Mapping between Burner and Switch of Gas Range by Color coding (가스레인지에 있어서 칼라코딩을 통한 버너와 스위치의 자연적 대응에 관한 연구)

  • People store information not only in their Drain but also material things. Norman called it knowledge in the world. The general way to store the information is to paste labels. 4 burner gas range force user to make conceptual model between burner and switch to see labels. but those are cognitive stress. Norman suggested spatial analogies for natural mapping between display and control. However the way of his methods in spatial analogies was not compatible with kitchen atmosphere. To solve those problems 1 suggested color coding . This study hypothesized that the mapping between burner and switch is realized by color coding. To testy the hypothesis 1 compared A group using general gas range with B group using color coded gas range. The result showed difference between A and B in accuracy ( F (1, 38) = 17.892, p < 0.01) and response time ( F (1, 38) = 6.726 p < 0.05). The result of this test is to certify that color coding affect peoples by presenting the difference accuracy and response time. As result this study presents that color coding can be compatible the product having importance to certify in the design process.

A study on the RDF(Refuse Derived Fuel) making process of Livestock manure sludge by oil-drying method (유중건조를 이용한 축산분뇨슬러지의 고형연료화 공정 연구)

  • In this study, we found the optimal manufacturing conditions of livestock manure sludge RDF with the oil-drying method. We performed oil evaporation, oil drying and pelletizing of the sludge to evaluate the value of the product (sludge RDF), and measured the performance of the product using calorimeter and PXRF equipment. Also, we conducted the calorie comparison test between sludge RDF manufactured in this study and wood RDF generally used in the field. Experimental results showed that 30g of the sludge treated by vegetable oil at $130^{\circ}C$ for 25 minutes were the optimal conditions to make the sludge RDF (considering the aspects of eco-friendly and mass production). The caloric value of the sludge RDF manufactured in this study was 5211kcal/kg which is higher than that of wood RDF used widely in the market. Finally, PXRF results showed sludge RDF contains no heavy metals with the exception of sulfur. Therefore, we recommend more study about the sulfur control process for future development of the industrial manufacturing process.
