Thermotolerance Inhibits Various Stress-induced Apoptosis in NIH3T3 Cells

  • When NIH3T3 cells were exposed to mild heat and recovered at $37^{\circ}C$ for various time intervals, they were thermotolerant and resistant to subsequent stresses including heat, oxidative stresses, and antitumor drug methotrexate which are apoptotic inducers. The induction kinetics of apoptosis by stresses were determined by DNA fragmentation and protein synthesis using $[35^S]$methionine pulse labeling. We investigated the hypothesis that thermotolerant cells were resistant to apoptotic cell death compared to control cells when both cells were exposed to various stresses inducing apoptosis. The cellular changes in thermotolerant cells were examined to determine which components are involved in this resistance. At first, the degree of resistance correlates with the extent of heat shock protein synthesis which were varied depending on the heating times at $45^{\circ}C$ and recovery times at $37^{\circ}C$after heat shock. Secondly, membrane permeability change was observed in thermotolerant cells. When cells prelabeled with $[^{3}H]$thymidine were exposed to various amounts of heat and recovered at $37^{\circ}C$ for 1/2 to 24 h, the permeability of cytosolic $[^{3}H]$thymidine in thermotolerant cells was 4 fold higher than that in control cells. Thirdly, the protein synthesis rates in thermotolerant and control cells were measured after exposing the cells to the same extent of stress. It turned out that thermotolerant cells were less damaged to same amount of stress than control cells, although the recovery rates are very similar to each other. These results demonstrate that an increase of heat shock proteins and membrane changes in thermotolerant cells may protect the cells from the stresses and increase the resistance to apoptotic cell death, even though the exact mechanism should be further studied.

뇌 해마가 제거된 흰쥐의 전기충격에 대한 내력(耐力) (Tolerance by Electric Shock in Hippocampectomized Rats)

  • 뇌 해마가 전기충격에 의한 내력(tolerance)에 미치는 영향을 구명하기 위하여, 두정 후두 경계 부위의 대뇌피질을 양측성으로 둥글게 떼어내고, 이를 거쳐 해마를 평균 97%가량 떼어버린 흰쥐(해마군) 14마리와 해마를 덮는 대뇌피질 부분만을 떼어버린 흰쥐(대뇌피질 대조군) 17마리 및 두개골을 노출하는데 그치고 뇌에 손상을 주지 않은 흰쥐(정상 대조군) 17마리를 사용하여 다음과 같은 실험을 실시하였다. 24시간 동안 먹이 없이 물만으로 지나게 한 다음 구금판에 세 무리를 구금하고 24시간 동안 매회 1.5초 동안씩 계속되는 4mA의 교류전류에 의한 충격을 1분 간격으로 반복하여 폐사하는 동물의 수효를 세어 다음과 같은 결과를 얻었다. 1. 전기충격을 가하는 동안 폐사한 동물의 수효에서 해마군의 값은 두 대조군의 값에 비하여 유의하게 적었으나, 두 대조군 사이에는 이렇다 할 차가 없었다. 2. 전기충격으로 인하여 폐사한 예에서 전기충격의 시작부터 폐사할 때까지의 시간은 해마군의 값이 두 대조군의 값에 비하여 연장된 경향을 보여 주었다. 이상의 결과로 미루어 뇌 해마는 전기충격에 대한 내력(耐力)에 억제적 영향을 끼친다고 추리된다.

Effect of Prophylactic Supplementation of Vitamin E and Se on Antioxidant Enzymes during Endotoxic Shock in Buffalo Calves

  • This study was conducted to determine the effects of prophylactic supplementation of vitamin E and Se on oxidative damage and antioxidant status. Fifteen healthy male buffalo (Bubalus bubalis) calves between the age of 6 to12 months were divided into three groups of five animals each: Group I-control, group II-endotoxic shock group infused with lyophilized E coli endotoxin @ 5 ${\mu}g$/kg body wt, and group III-supplemented with vitamin E @ 250 mg and Se @ 7.5 mg, one month prior to induction of endotoxic shock. All the animals in group II and group III exhibited signs of endotoxic shock. When the endotoxic shock was induced, there was significant (p<0.05) increase in the circulating levels of malonyl dialdehyde MDA (an indicator of lipid peroxidation). In the supplemented group III the magnitude of formation of MDA was also less as compared to group II at every stage of study. There was significant (p<0.05) decrease in circulating levels of SOD, GSH-Px, Catalase and G-6-PD activity from the normal (0 h) value with passage of time. As a result of endotoxic shock, these values reached a lowest value, and then showed a tendency towards the 0 h value. Prophylactic supplementation with vitamin E and Se was successful in reducing the quantum of oxidative damage due to formation of free radicals because of endotoxic shock.

스로틀 전자제어 방식에서 제어 특성이 차량의 가/감속 시 운전성에 미치는 영향 (The Effect of the ETC(Electronic Throttle Control) Characteristics on the Vehicle Driveability at Tip-in/out)

  • The passenger car drivers want in general to feel good driveability, but they sometimes feel uncomfortable by shock and jerk phenomenon when they push or release acceleration pedal with clutch engaged. In this paper, the shock and jerk characteristics are studied on the vehicles of the throttle-by-wire system. With this system, the throttle is not directly controlled by drivers but via a microprocessor. So the control logic of the throttle is very important on tip-in and tip-out driveability. Experiments were carried out on two vehicles which show different control characteristics. The torque control logics were analyzed by measuring cylinder pressures. The results show that special torque control logic is needed at tip-in/out state for good driveability.

A Speed Control for the Reduction of the Shift Shocks in Electric Vehicles with a Two-Speed AMT

  • In the present paper, a speed control algorithm with fast response characteristics is proposed to reduce the shift shock of medium/large-sized electric vehicles equipped with a two-speed AMT. Shift shocks, which are closely related with to the vehicles' ride comfort, occur due to the difference between the speed of the motor shaft and the load shaft when the gear is engaged. The proposed speed control method for shift shock reduction can quickly synchronize speeds occurring due to differences in the gear ratios during speed shifts in AMT systems by speed command feed-forward compensation and a state feedback controller. As a result, efficient shift results without any shift shock can be obtained. The proposed speed control method was applied to a 9 m- long medium- sized electric bus to demonstrate the validity through a simulated analysis and experiments.

  • The present study describes an experimental work to reduce supersonic jet noise using a control wire device that is placed into the supersonic jet stream. The jet pressure ratio is varied to obtain the supersonic jets which are operated in a wide range of over-expanded to moderately under-expanded conditions. The wire device is composed of long cylinders with a very small diameter. X-type wire device is applied to control the supersonic jet noise, and its location is varied to investigate the effect of the control wire device on supersonic jet noise. A high-quality Schlieren optical system is used to visualize the flow field of supersonic jet with and without the control wire device. Acoustic measurement is performed to obtain the overall sound pressure level and noise spectra. The results obtained show that the present wire device destroys the shock-cell structures, reduces the shock strength, and consequently leading to a substantial suppression of supersonic jet noise.

  • This paper presents optimal design of controllable magnetorheological (MR) shock absorbers for passenger vehicle. In order to achieve this goal, two MR shock absorbers (one for front suspension; one for rear suspension) are designed using an optimization methodology based on design specifications for a commercial passenger vehicle. The optimization problem is to find optimal geometric dimensions of the magnetic circuits for the front and rear MR shock absorbers in order to improve the performance such as damping force as an objective function. The first order optimization method using commercial finite element method (FEM) software is adopted for the constrained optimization algorithm. After manufacturing the MR shock absorbers with optimally obtained design parameters, their field-dependent damping forces are experimentally evaluated and compared with those of conventional shock absorbers. In addition, vibration control performances of the full-vehicle installed with the proposed MR shock absorbers are evaluated under bump road condition and obstacle avoidance test.

산성 pH 충격 스트레스에 의한 스트랩토마이세스 시에리컬러 M511의 프로디지닌 생산 증대 (Enhanced Prodiginines Production in Streptomyces coelicolor M511 by Stress of Acidic pH Shock)

  • 운데실프로지닌과 스트래토루빈 B는 S. coelicolor가 생산하는 붉은색 항생물질이다. 이번 연구에서 이들 붉은색 항생제의 생산성과 pH shock 스트레스와의 상관관계를 연구 하였다. 운데실프로디지닌과 스트랩토루빈 B의 생합성은 고체 R2YE 배지에서 산성 pH shock에 의해 증가되었다. 최적 pH shock은 pH 4로 비교군과 비교하여 각각 1.6배 및 2배 운데실프로디진과 스트랩토루빈 B의 생산성이 증가되었다. 게다가, 산성 pH 4의 세포 추출물은 T. mentagrophytes 에 대한 주목할만한 저항 활성을 나타내었다. 그러나, 중성 및 염기성 pH shock에서는 이들 항생제의 생산성뿐만 아니라 항진균 활성 증가가 일어나지 않았다. 그러므로, 비록 산성 pH shock이 간단하고 쉬운 방법이지만, 이들 붉은색 항생물질과 다른 이차대사산물의 생산성 향상에는 매우 효과적인 접근방법일 것이다.

충격파 개념에 기반한 유체 추력벡터제어에 관한 연구 (Fluidic Thrust Vector Control Using Shock Wave Concept)

  • 충격파 개념을 이용하는 유체 추력벡터 제어는 빠른 벡터링 응답, 간단한 구조 및 낮은 무게로 인하여 큰 벡터링 성능을 달성하는데 많은 이점을 제공한다. 본 논문에서는 전산유체역학 기법을 사용하여 슬롯 인젝터를 가진 3차원 직사각형 초음속 노즐에 대하여 연구를 수행하였다. 계산 방법론을 검증하기 위하여 수치 결과를 실험 데이터로 비교하였다. 대칭 평면에서의 상부 및 하부 노즐벽을 따르는 압력분포는 시험 결과와 잘 일치하였다. $k-{\omega}$ SST 난류모델을 기반으로 한 수치해석을 통하여, 운동량 플럭스 비율의 영향을 철저히 조사하여 추력의 성능 변화를 명확하게 나타내었다.
