Dose-Dependent Cytotoxic Effects of Menthol on Human Malignant Melanoma A-375 Cells: Correlation with TRPM8 Transcript Expression

  • Background: Transient receptor potential melastatin 8 (TRPM8), a principle membrane receptor involved in calcium ion influx and cell signal transduction, has been found to be up-regulated in some cancer types, including melanomas. Efficiency of menthol, an agonist of TRPM8, in killing melanoma cancer cells has been reported previously, but the mechanisms remain unclear. We here determined whether in vitro cytotoxic effects of menthol on A-375 human malignant melanoma cells might be related to TRPM8 transcript expression. Materials and Methods: The $PrestoBlue^{(R)}$ cell viability assay was used to assess the in vitro cytotoxic effect of menthol after 24h of treatment. RT-PCR was used to quantify TRPM8 transcript expression levels in normal and menthol-treated cells. Cell morphology was observed under inverted phase contrast light microscopy. Results: TRPM8 transcript expression was found at low levels in A-375 cells and down-regulated in a potentially dose-dependent manner by menthol. Menthol exerted in vitro cytotoxic effects on A-375 cells with an $IC_{50}$ value of 11.8 ${\mu}M$, which was at least as effective as 5-fluorouracil ($IC_{50}=120{\mu}M$), a commonly applied chemotherapeutic drug. Menthol showed no dose-dependent cytotoxicity on HeLa cells, a TRPM8 non-expressing cell line. Conclusions: The cytotoxic effects on A-375 cells caused by menthol might be related to reduction of the TRPM8 transcript level. This suggests that menthol might activate TRPM8 to increase cytosolic $Ca^{2+}$ levels, which leads to cytosolic $Ca^{2+}$ imbalance and triggers cell death.

  • Goniothalamin, a natural compound extracted from Goniothalamus sp. belonging to the Annonacae family, possesses anticancer properties towards several tumor cell lines. This study focused on apoptosis induction by goniothalamin (GTN) in the Hela cervical cancer cell line. Cell growth inhibition was measured by MTT assay and the $IC_{50}$ value of goniothalamin was $3.2{\pm}0.72{\mu}g/ml$. Morphological changes and biochemical processes associated with apoptosis were evident on phase contrast microscopy and fluorescence microscopy. DNA fragmentation, DNA damage, caspase-9 activation and a large increase in the sub-G1 and S cell cycle phases confirmed the occurrence of apoptosis in a time-dependent manner. It could be concluded that goniothalamin show a promising cytotoxicity effect against cervical cancer cells (Hela) and the cell death mode induced by goniothalamin was apoptosis.

The effect of the cytotoxicity of sodium lauryl sulfate containing toothpaste on HaCaT and NIH-3T3 cells (구강세치제에 함유된 SLS(Sodium lauryl Sulfate)가 HaCaT 세포와 NIH-3T3 세포에 미치는 독성 효과)

  • Objectives: The purpose of this study was to determine the toxic effects of sodium lauryl sulfate(SLS) in human keratinocyte HaCaT cells and mouse fibroblast NIH-3T3 cells. Methods: The effect of sodium lauryl sulfate(SLS) cell viability and proliferation were determined by WST-1 assay and changes shape of nucleus were evaluated by Hoechst staining under fluorescence microscopy. Additionally, observation of cell morphological changes under light microscopy. Results: SLS induced cytotoxicity and a marked apoptosis in both HaCaT and NIH-3T3 cell lines. With the result of the WST-1 assay, SLS induced the cytotoxicity of 0.005% and 0.0075%, 0.01% SLS for 24 h after HaCaT and NIH-3T3 cells in time and dose-dependent manner(p<0.005). SLS inhibited cell growth and caused apoptosis as evidenced by nuclear fragmentation and condensation. Thus, determination of the morphological changes to define apoptosis was visualized using inverted phase contrast microscopy. Conclusions: SLS had toxicity of the human keratinocyte cells and mouse fibroblast cells and this study will provide the basic data for the development of proper SLS concentration in dentifrice.

Morphological aspect of the attached bacteria by the sterilization method of the ultrasonic scaling tip (소독멸균 방법에 따른 초음파 스케일링 팁에 부착된 세균의 양상)

  • Objectives: The purpose of the study was to investigate the bacterial morphology attached on ultrasonic scaler tips using no cleansing solution, alcohol cotton, liquid chemical disinfecting agent, and autoclave method. Methods: Scaling tip was applied to the mouth and the ultrasonic scaler tips were assigned to four groups. Group 1 was control group with no cleansing solution. Group 2 was treated with alcohol cotton. Group 3 was treated with 2% green Y-Na solution in liquid chemical disinfecting agent, and Group 4 was sterilized by autoclave method. Live bacteria were observed by phase contrast microscopy. The scanning electron microscopy(SEM) revealed the morphological characteristics of scaler surface. The type of attached bacteria were analyzed using SPSS 21.0 program. The data were analyzed by one-way analysis of variance(ANOVA) and Tukey's post-hoc test. Results: The types of sterilization methods had influences on the bacterial viability. The numbers of cocci, bacilli, spiral form bacteria, and filamentous bacteria was observed in $89.00{\pm}3.60%$, $29.67{\pm}3.51%$, $3.33{\pm}0.57%$ and $1.67{\pm}0.57%$ in control group, $31.67{\pm}3.51%$, $63.33{\pm}4.04%$, $2.00{\pm}1.00%$ and $1.67{\pm}0.57%$ in alcohol cotton group, $69.67{\pm}4.50%$, $12.33{\pm}2.51%$, 0% and 0% in liquid chemical disinfecting agent group, and 0.0%, 0.0%, 0.0% and 0.0% in autoclave method group. The clean surface of ultrasonic scaler tip was shown on SEM by autoclave method. Conclusions: The most effective sterilization method of ultrasonic scaler tip was the autoclave method. Autoclave method is the most effective sterilization method and can reduce the cross-infection in the dental clinic.

Viscoelastic behavior of aqueous surfactant micellar solutions

  • A cationic surfactant, cetyltrimethylammonium $\rho$-toluenesufonate (CTA$\rho$TS), forms long threadlike micelles in aqueous solution. The threadlike micelles make concentrated entanglement networks, so that the solution shows pronounced viscoelastic behavior as concentrated polymer systems do. However, a mechanism for a process responsible for the longest relaxation time of the threadlike micellar system is different from that of semi-dilute to concentrated polymer systems. The threadlike micellar system exhibits unique viscoelasticity described by a Maxwell model. The longest relaxation time of the threadlike micellar system is not a function of the concentration of CTA$\rho$TS, but changes with that of $\rho$-toluenesufonate ($\rho$$TS^{-}$) ions in the bulk aqueous phase supplied by adding sodium $\rho$-toluenesulfonate (NapTS). The rates of molecular motions in the threadlike micelles are not influenced by the concentration of $\rho$$TS^{-}$ anions, therefore, molecular motions in the threadlike micelles (micro-dynamics) are independent of the longest relaxation mechanism (macro-dynamics). A nonionic surfactant, oleyldimethylamineoxide (ODAO), forms long threadlike micelles in aqueous solution without any additives. The aqueous threadlike micellar system of ODAO also shows Maxwell type viscoelastic behavior. However, the relaxation mechanism for the longest relaxation process in the system should be different from that in the threadlike micellar systems of CTA$\rho$TS, since the system of ODAO does not contain additive anions. Because increase in the average degree of protonation of head groups of ODAO molecules in micelles due to adding hydrogen bromide causes the relaxation time remarkably longer, changes in micro-structure and micro-dynamics in the threadlike micelle are closely related to macro-dynamics in contrast with the threadlike micellar system of CTA$\rho$TS.

Quality Control of Asbestos Fiber Counting for Airborne Samples Using Phase Contrast Microscopy (위상차 현미경법을 이용한 공기중 석면농도 분석에서의 정도관리)

  • This study was conducted to evaluate the accuracy and the precision of asbestos counting data produced by the Division of Industrial Health, School of Public Health, Seoul National Universitys (SNU). The study was performed from July 18 to October 4, 1991, and the results are summarized as follows. 1. Intracounter Relative Standard Deviations (Sr) in the category of 5-50.5 fibers as total fibers counted ranged from 0.27 to 0.37, which were greater than 0.10-0.17 which were reported by the NIOSH. The reasons are supposed to be as follows. First, inexperience of counters in asbestos fiber counting was considered to be a main reason. Second, poor quality of samples due to sampling and mounting error increased variation of counting. Third, fiber density of many samples were less than $100fibers/mm^2$. But Intracounter Relative Standard Deviations (Sr) in samples with >50.5 fibers ranged from 0.l6 to 0.20, approaching the value 01 NIOSH. 2. Intralaboratory Relative Standard Deviations (Sr) in categories of 5-20.5, >20.5-50.5 and >50.5 fibers were 0.54, 0.37 and 0.26, respectively. Intralaboratory Sr in samples with fiber density greater than $100fibers/mm^2$ was 0.26. This was similar to the values reported by other foreign experienced laboratories. 3. Comparing results of three counters, Counter C, a beginner, overestimated asbestos fiber concentrations. 4. Since our SNU laboratory has participated in two quality control programs, IOMA-F.R.I.C.A., U.K. and NIOSH PAT Program, U.S.A., this laboratory has been evaluated as " Rating 1" and "Proficient" laboratory, by IOM and NIOSH, respectively.

Characterization of Recombinant Baculovirus Ewpressing Polyhedrin Gene of Bombyx mori and Autographa californica Nuclear Polyhedrois Virus (두 종류의 다각체 단백질 유전자를 발현하는 유전자 재조합 핵다각체병 바이러스의 특성)

  • Recombinant baculoviruses having expanded host range were selected by coinfection of Autographa california NPV and Bombyx mori NPV into Sf-9 and BmN-4 insect cell lines. In order to determine the polyhedra morhplogy of RecS-A6, one of a recombinant baculovirus, polyhedra of RecS-A6 produced in insect cells were observed by phase contrast microscope and scanning electron microscope. The results revealed that the recombinant baculovirus had a various polyhedra morphology which was different from its parental viruses, suggesting that the various morhpology of recombinant baculovirus with an expanded host range was due to the genetic recombination of viral genome. To analyze the genomic recombinantion of the recombinant baculoviruses, genomic DNAs of two parent viruses and RecS-A6 were digested with restriction endonuclease and subjected to agarose gel electrophoresis. Southern blot analysis revealed that RecS-A6 has two polyhedrin gene of AcNPV and BmNPV in a viral genome. Polyhedral protein of recombinant baculovirus was analysed by SDS-PAGE. The result showed that molecular weight of polyhedral protein of RecS-A6 containing two polyhedrin gene of AcNPV and BmNPV was as the 31 kDa band of AcNPV and 30 kDa band of BmNPV.

Articulatory modification of /m/ in the coda and the onset as a function of prosodic boundary strength and focus in Korean

  • An articulatory study (using an Electromagnetic Articulography, EMA) was conducted to explore effects of prosodic boundary strength (Intonational Phrase/IP versus Word/Wd), and focus (Focused/accented, Neutral, Unfocused/unaccented) on the kinematic realization of /m/ in the coda (${\ldots}$am#i${\ldots}$) and the onset (${\ldots}$a#mi${\ldots}$) conditions in Korean. (Here # refers to a prosodic boundary such as an IP or a Wd boundary). Several important points have emerged. First, the boundary effect on /m/s was most robustly observed in the temporal dimension in both the coda (IP-final) and the onset (IP-initial) conditions, generally in line with cross-linguistically observable boundary-related lengthening patterns. Crucially, however, in contrast with boundary-related slowing-down effects that have been observed in English, both the IP-final and IP-initial temporal expansions of Korean /m/s were not accompanied by an articulatory slowing down. They were, if anything, associated with a faster movement in the lip opening (release) phase (into the vowel). This suggests that the mechanisms underlying boundary-related temporal expansions may differ between languages. Second, observed boundary-induced strengthening effects (both spatial and temporal expansions, especially on the IP-initial /m/s) were remarkably similar to prominence (focus)-induced strengthening effects, which is again counter to phrase-initial strengthening patterns observed in English in which boundary effects are dissociated from prominent effects. This suggests that initial syllables in Korean may be a common focus for both boundary and prominence marking. These results, taken together, imply that the boundary-induced strengthening in Korean is different in nature from that in English, each being modulated by the individual language's prosodic system. Third, the coda and the onset /m/s were found to be produced in a subtly but significantly different way even in a Wd boundary condition, a potentially neutralizing (resyllabification) context. This suggests that although the coda may be phonologically 'resyllabified' into the following syllable in a phrase-medial position, its underlying syllable affiliation is kinematically distinguished from the onset.

Development of Equipment to Measure Insulation Resistance and Evaluate the Lifetime of High-voltage Cable in Operation (운전 중인 고전압 케이블의 절연저항 측정 및 수명평가장치의 개발)

  • In this paper, we find out the lifetime index in order to determine the time-dependent trend of deteriorating performance of 6.6kV high-voltage power cable in operation at a power station. The cable systems used in our study have been in operation for 13 years. With measurements for the 13 years, we analyzed the insulation resistances. By developing measuring equipment (comprized mainly of transformer, temperature sensor, and LPF) operating by the three-phase electric power, we analyzed the changing characteristics of insulation resistance of power cable. In contrast to 22kV cables, 6.6 kV cables have thicker insulation. Therefore the characteristics of 6.6kV cables are different from that of 22kV cables. The study found that as time passes, the insulation resistance does not decrease continuously; it decreases to a certain value, then does not decrease any more and shows properties of oscillation. We could not detect the process of deterioration in the preceding twelve years. The cable system showed great stability so that deterioration was not apparent. In this case, it is not possible to measure the future life indices of power cables because the lifetime indices are not predictable

Microstructure and CO Gas Sensing Properties of Ag-CuO-SnO2 Thin Films Prepared by Co-Evaporation and Thermal Oxidation (공증발과 열산화로 제조한 Ag-CuO-SnO2 박막에서 미세조직과 CO 가스 감지특성)

  • In this study, we investigated microstructure and the CO gas sensing properties of Ag-CuO-$SnO_2$ thin films prepared by co-evaporation and subsequently thermal oxidation at air atmosphere. The sensitivity of a Cu-Sn films, thermally oxidized at $600^{\circ}C$, is strongly affected by the amount of Cu. At Cu:7 wt%-Sn:93 wt%, the film exhibited a maximum sensitivity of ${\sim}2.3$ to CO gas of 1000 ppm at $300^{\circ}C$. In contrast, the sensitivity of a Sn-Ag film did not change significantly with the amount of Ag. An enhanced sensitivity of ${\sim}3.7$ was observed in the film with a composition of Ag:3 wt%-Cu:4 wt%-Sn:93 wt%, when thermally oxidized at $600^{\circ}C$. In addition, this thin film shows a response time of ${\sim}80$ sec and a recovery time of ${\sim}450$ sec to 1000 ppm CO gas. The results demonstrate that the CO sensitivity of the Ag-CuO-$SnO_2$ thin films may be closely associated with coexistence of $SnO_2$ and SnO phase, decrease in average particle size, and a porous microstructure. We also suggest that co-evaporation and followed by thermal oxidation is a very simple and effective method to prepare oxide gas sensor thin films.
