Improved Large-Scale Refolding Techniques for Inclusion Body Proteins (내포체 단백질의 개선된 대규모 재접힘 기술)

  • Techniques for protein refolding from inclusion body are discussed in view of its engineering application to large scale protein purification. Among the techniques, dilution and dialysis are mainly utilized due to simple operation. Membrane reactor, gel filtration chromatography, and continuous tank operation are emerging tools for their process-scale possibility in refolding. Reaction engineering approaches could be used to analyze the kinetic behaviour in the process scale refolding reactor. The kinetic analysis is helpful in the optimization of refolding yield in the refolding reactor.

  • Lymphangioleiomyomatosis(LAM) is defined as an abnormal proliferation of smooth muscles in the lung tissue throughout lymphatics, vascular and bronchial structure. A 52-year-old postmenopausal woman was admitted to our hospital for recurrent pneumothorax. She was treated for medroxyprogesterone by LAM 1 month ago. We performed operation of pulmonary partial resection and pleurodesis. The patient is receiving continuous medroxyprogesterone and Leuplin administration, and currently, 6 months after the operation, is still showing good results.

  • There are many results on the extended operations of two fuzzy numbers based on the Zadeh's extension principle. For the calculation, we have to use existing operations between two ${\alpha}$-cuts. In this paper, we define parametric operations between two ${\alpha}$-cuts which are different from the existing operations. But we have the same results as the extended operations of Zadeh's principle.

  • Permeabilization of Zymomonas mobilis with CTAB(Cetyltrimethylammoniumbromide) was investigated in order to obtain stable process for sorbitol production in the immobilized system. The optimum conditions for sorbitol formation were obtained in the case of using cells treated with 0.2% CTAB at$ 4^{\circ}C$ for 10 min. Permeabilized cells were treated with glutaraldehyde to cross-link the internal enzyme for the improvement of the enzyme stability. In this way, no significant loss of enzyme activity was apparent during 30-day operation in a continuous process. The productivity of the continuous process at dilution rate 0.2h-1 was 6.51g/1/h for sorbitol. The CTAB permeabilized cells could be used to produce sorbitol in the long term continuous process.

Continuous Conditional Random Field Model for Predicting the Electrical Load of a Combined Cycle Power Plant

  • Existing power plants may consume significant amounts of fuel and require high operating costs, partly because of poor electrical power output estimates. This paper suggests a continuous conditional random field (C-CRF) model to predict more precisely the full-load electrical power output of a base load operated combined cycle power plant. We introduce three feature functions to model association potential and one feature function to model interaction potential. Together, these functions compose the C-CRF model, and the model is transformed into a multivariate Gaussian distribution with which the operation parameters can be modeled more efficiently. The performance of our model in estimating power output was evaluated by means of a real dataset and our model outperformed existing methods. Moreover, our model can be used to estimate confidence intervals of the predicted output and calculate several probabilities.

  • Using a flocculating Saccharomyes cerevisiae CA-1, an air-lift reactor equipped with a modified settler was used for ethanol fermentation. The effects of conditions such as aeration rate, initial glucose concentration, and dilution rate were studied using the air-lift reactor. In batch fermentation, optimum aeration rate was 0.5 vvm. In continuous fermentation, aeration rate and initial pH were fixed 0.5 vvm and 4.5, substrate concentration and dillution rate were changed 10-15% and 0.1-1.3. The maximum ethanol productivity was shown to be 20.4 g/l$\cdot $h in 10% glucose and 0.7 h$^{-1}$ dilution rate., and optimum operation condition considering the ethanol productivity and glucose utilization ratio was 0.5 h$^{-1}$ dilution rate in 10% glucose concentration.

  • As the modern industrial processes become more complex, it is getting more difficult to model and control the processes. Naturally, an advanced type of DCS(Distributed Control System) with higher level functions is being sought. Advanced DCS is a DCS with advanced functions such as fault diagnosis, GPC(Generalized Predictive Control), NN(Neural Network), and Fuzzy Control. In this thesis, we have studied a fuzzy control algorithm for realizing an advanced DCS. Its algorithm is implemented in a form of function code which is a process control language, being used by the industrial engineers. To verify the realized function code of the fuzzy control, the function code is applied to a continuous casting process of the Pohang Iron & Steel Works in Kwangyang. The rules of the fuzzy control were collected via interviews of the field operators and their operation documents. Finally under a real-time operating system environment, usability of the function code of the fuzzy control is shown via simulation for the continuous casting process.

Continuous Production of Sorbitol with Permeabilized Zymomonas mobilis Immobilized in Alginate and Chitin (알저네이트 및 카이틴 고정화 Zymomonas mobilis 에 의한 쏠비톨의 연속생산)

  • This study describes the sorbitol production with permeabilized cells of Zymomonas mobilis immobilized in Ca-alginate. Toluene treated cells lose glucose-fructose oxidoreductase activity due to leaking of enzyme from the cells. In order to prevent this leakage, the permeabilized cells were immobilized in alginate and chitin. No significant loss of enzyme activity was apparent during 210h operation in a continuous process. The productivity of the continuous process was estimated to be about 3.5g/l -h for sorbitol at dilution rate $0.2h^{-1}$.

  • This paper proposes a tension control to compensate friction loss using on-line friction torque observer for a continuous strip processing line. Friction loss of roller results in significant deviation of strip tension, accordingly it has an influence on the operation of other adjacent rolls. To avoid tension variation of the strip, a friction torque observer is designed in adjacent roll, which operates in speed control mode. The observed torque is added to the torque limit reference of the pay-off reel for on-line compensation of both friction loss and acceleration/deceleration torque at the same time. The simulation and experimental results show improvement of tension control performance by the proposed friction compensation method.

  • Porphyrin-catalyzed reduction of TNT to triaminotoluene was performed in both batch reactions and a continuous process. Packed-bed reactors were used to study porphyrin-catalyzed reduction in a continuous process. A reactor was packed with DEAD(diethylaminoethyl)-substituted glass beads on which $Co^{+3}$_centered deuteroporpgyrin Ⅸ, -2, 4-disulfonic acid was immobilized, and another containing only DEAE glass beads was used as a control. The porphyrin exhibited catalytic activity in its immobilized state up to 100 hours of operation. Based on the successful abatement of nitrobodies by porphyrin-catalyzed reduction in both batch and continuous process, this process is recommended to be used as a pretreatment for biological treatment or carbon adsorption treatment of TNT wastes.
