  • For commercial application of the MCFC, the life time of the MCFC should exceed 40,000 hours, But the life time of the state-of-the-art MCFC was limited to 15,000 hours. The main reasons of the life time limit can be classified as the deficiency of the electrolyte and cathode dissolution. It has been found that the electrolyte deficiency is caused by the continuous evaporation of the electrolyte. However a recent reaserch shows that an electrolyte pumping phenomenon, which implies, the migration of the electrolyte through the gasket material of the external gas manifold, is also the reason of the electrolyte deficiency. Due to the electrolyte pumping phenomenon, positive end cell of the stack suffers the electrolyte deficiency and negative end cell of stack is flooded with electrolyte. Therefore, the cell performance is degraded. The author invented a new manifoldcasing, which prevents the contact between the wet seal and the gasket of the manifold, and gives a complete elimination of an electrolyte pumping effect.

정관절제가 흰쥐의 정자두부의 미세구조에 미치는 영향 (Effect of the Vasectomy on the Fine Structure of the Sperm-Acrosome in Rats)

  • Vesectomy has been increased as a popular method of birth control because it is simpler than other methods for men. But the vasectomy results in several problems such as relation to effect changes on the structure and function of the reproductive organ. The fate of non-ejaculated spermatozoa is postulated by some authors that those are disappeared by a progress of dissolution and reabsorption in the epididymis, and we have attempted to prove the true state of sperm-acrosome on the fine structure in vasectomized rats. The results were as follows: 1. Apical segments of the acrosome were swollen similar to the shape of club in many spermatozoa. 2. Discontinuities of the outer and inner acrosomal membranes were occasionally noted and there were complete losses of acrosomes in the certain place. 3. There was no evidence of significant changes in the nuclear structure, nor dilatation of the subacrosomal space. 4. Vasectomy might effect destructive changes in the acrosomes of the non-ejaculated spermatozoa in situ.

Structural and Morphological Alterations of Candida albicans Cells after Treatment with Atratoxin $B_1$ from Holothuria atra (Jaeger)

  • Atratoxin $B_1$ which was found to inhibit the growth of Candida albicans caused structural and morphological alteration of the cells. Increased accumulation of vesicles and membranous bodies in the cytoplasm, and alterations of the cell membrane and cell wall were most obvious. Sequential lytic events of the cells eventually resulted in complete disintegration of the cytoplasmic structures. These results suggested that atratoxin $B_1$ functioned by either blocking the biosynthetic step during cell wall synthesis, altering cell wall metabolism or dissolution of the cell organelles. These changes caused a progressive destruction of the cell wall leading to cell lysis.

7075 Al합금의 RRA처리에서 저온 퇴화처리의 효과 (Effects of Low Temperature Retrogression on RRA Treatment of 7075 AI Alloy)

  • 가공열처리한 7075Al합금을 $170^{\circ}C$에서 퇴화처리하여도 GP대는 완전히 분해되지 않았으며 초기의경도저하는 부분적인 GP대의 분해에 기인하고 $\mu '$의 생성으로 강도는 증가하다가 $\mu '$의 성장 및 $\mu$의 생성으로 강도는 다시 저하한다. 퇴화처리시 경도 곡선의 극소점이나 2차 피크점(극대점)에서 RRA처리한 시편의 응력부식 특성은 T6재에 비하여 상당히 개선되며 강도의 저하를 수반하지 않는다. 또한 저온인 $170^{\circ}C$에서 퇴화처리시 기지조직의 변화가 크지 않아 퇴화처리시 경도곡도의 극소점과 2차 피크점에서의 특성차이는 거의 없었다.

Preparation and Characterization of Simvastatin Solid Dispersion using Aqueous Solvent

  • Simvastatin is an inhibitor of 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase, which catalyzes the conversion of HMG-CoA to mevalonate, an early and rate-limiting step in the biosynthesis of cholesterol. Simvastatin has good permeability, but it also has low solubility (BCS class II), which reduces its bioavailability. To overcome this problem, a solid dispersion is formed using a spray-dryer with polymeric material carrier to potentially enhance the dissolution rate and extend drug absorption. As carriers for solid dispersion, Gelucire$^{(R)}$44/14 and Gelucire$^{(R)}$ 50/13 are semisolid excipients that greatly improve the bioavailability of poorly-soluble drugs. To avoid any toxic effects of an organic solvent, we used aqueous medium to melt Tween$^{(R)}$ 80 and distilled water. The structural behaviors of the raw materials and the solid dispersion were analyzed by differential scanning calorimetry (DSC), powder X-ray diffraction (PXRD) and scanning electron microscopy (SEM). The DSC and PXRD data indicated that the crystalline structure of simvastatin was transformed to an amorphous structure through solid dispersion. Then, solid dispersion-based tablets containing 20 mg simvastatin were prepared with excipients. Dissolution tests were performed in distilled water and artificial intestinal fluid using the USP paddle II method. Compared with that of the commercial tablet (Zocor$^{(R)}$ 20 mg), the release of simvastatin from solid dispersion based-tablet was more efficient. Although the stability study is not complete, this solid dispersion system is expected to deliver poorly water-soluble drugs with enhanced bioavailability and less toxicity.

신부전 치료제 WHW 엑스의 pH 특이적인 방출 제형 연구 (A pH-specific Released Formulation Study of WHW Extract for Therapeutics of Renal Failures)

  • Effective therapeutics for renal failures have not yet been developed until now. Recently, there was a report showing that Wen-pi-tang-Hab-Wu-ling-san (WHW) prescriptions had the potential to prevent renal failures through the increased expression of HSP-27 and HSP-72 after ischemia/reperfusion. Therefore, formulation studies by pH-specific released systems were carried out to exhibit the optimal activity of WHW prescriptions in this study. WHW prescriptions were separately extracted using water into two parts of stomach-released (SR) and intestine-released (IR) extracts. Subsequently, the double-layered tablet was prepared using the SR extracts and pharmaceutical additives and enteric-coated IR tablet. Dissolution studies were carried out to figure out the release of cinnamic acid and icarrin from SR tablet, IR tablet and double-layered tablet, respectively. The complete release of cinnamic acid from SR tablet showed 90min after dissolution in pH 1.2 and insignificant drug released from IR tablet. As well as, icarrin from IR tablet completely released in pH 6.8 and 7.4 as enteric-coating film dissolved.

시멘트 페이스트 및 지오폴리머 내의 PLA 섬유의 용해성 평가 (Evaluation of PLA Fiber Dissolution in Cement Paste and Geopolymer)

  • PLA(Poly-Lactic Acid)섬유는 친환경소재이며 자연분해가 되므로 건설재료에 사용할 경우 내부 공극연결구조를 가진 다공성 재료의 제조가 가능하다. 본 연구에서는 국내에서 생산된 PLA 섬유(직경 0.5mm, 1.0mm, 길이 10mm)를 대상으로 강알칼리와 고온에서의 용해실험을 수행하였으며, 이를 이용하여 FA기반 지오폴리머에 활용하였다. 고온양생과 강알칼리 용액을 통하여 강도는 확보하였으나, 완전한 PLA 섬유의 용해을 확보하지 못하였다. 기존의 연구인 0.003mm의 직경에서는 완전히 용해하였으나 0.5mm는 약 42.5%, 1.0mm는 약 33.3%의 용해율을 가지고 있는 것으로 평가되었다. 또한 섬유의 체적이 커짐에 따라 양생시 부유하는 섬유가 발생하여 작업성과 용해에 부정적인 영향을 나타내었다. PLA 특성은 생산하는 원재료와 생산조건에 따라 달라질 수 있으나, 다공성 건설 자재로서 사용하기 위해서는 0.1mm 내외의 PLA 섬유의 사용이 바람직하다고 판단된다.

중금속 제거용 흡착제로서의 철광사 적용 (Application of Iron Sand as Adsorbent for the Removal of Heavy Metal)

  • 본 연구에서는 철이 주성분 이면서 다양한 금속산화물의 조성을 갖는 채광물질인 철광사(Iron Sand)를 구리 혹은 납을 함유한 합성폐수 처리에 대한 적용성을 조사하였다. 먼저 철광사의 내산성을 조사하기 위해 pH $2{\sim}4.5$ 범위에서 철 및 알루미늄의 용출특성을 조사였을 때, 가장 많은 조성을 갖는 철의 용출량은 강산성인 pH 2에서도 수질오염 배출기준 이하로 나타났다. 알루미늄의 경우 pH 2에서는 상당량의 용출현상이 나타났지만 pH 3 이상에서는 용출농도가 급격히 감소하였다. 이러한 내산성 실험결과는 철광사를 pH 3 이상에서의 폐수처리에 적용하는 데는 큰 문제가 없는 것을 제시한다. 철광사의 중금속 흡착능력을 조사하기 위해 구리 및 납을 흡착질로 사용하고 실험실 규모의 회분식 흡착실험과 칼럼 여과실험을 실시하였다. pH 5에서 철광사에 의한 구리이온의 제거는 반응초기 시간대에 빠르게 이루어졌으며 2시간이 경과되었을 때 구리 주입량의 50%가 제거되었으며 24시간 경과 후에 거의 완전한 흡착평형에 도달하였다. 철광사에 대한 Cu(II) 및 Pb(II)의 흡착은 pH 2 이상에서 증가되기 시작하였으며 pH 6 이상에서는 거의 모든 흡착질들이 제거되는 전형적인 양이온형 흡착경향을 보였다. 철광사에 대한 구리 및 납 흡착질의 농도비 변화에 따른 흡착등은 실험결과, 구리 및 납의 최대 흡착량은 각각 2,170 mg/kg 및 3,450 mg/kg으로 나타났다.

  • 코발트, 니켈, 구리 그리고 귀금속분말에 적용할 수 있는 polyol법은 균질한 크기와 형상을 갖는 금속분말합성에 매우 효과적인 공정이다 이 때 polyol은 용매, 환원제 그리고 보호제의 역학을 한다 $AgNO_3$글 촉매제로 첨가하여 비균질계 핵생성 반응을 야기할 경우 서브마이크폰 크기(0.5$\mu$m)의 코발트 분말을 한성학 수 있었다. 또한 촉매제인 Ag 핵의 수출 변화시키므로써 코발트 분말의 입도를 제어할 수 있음을 확인하였다.

Bacillus thuringiensis subsp. sotto의 내독소 결정체 용해 과정 및 활성기작과 항원 발현 양상 (In Vitro Dissolution and Proteolytic Activation of $\delta$-endotoxin and Antigenic Expression Pattern of Bacillus thuringiensis subsp, sotto)

  • The solubilization and proteolytic process of $\delta $-endotoxin was analvsed to compare the biochemical property of the toxin isolated from B. thuringiensis subsp. sotto. The purified crystals were dissolved in 50 mM carbonate buffer containing 10 mM dithiothreitol at pH 10 for various times. The electrophoretic pattern showed that a rapid disappearance of 138 kDa protein band. This disappearance of protein with high molecular weight was accompanied by the appearance of new protein fragment with 104 kDa, 60 kDa, and 25 kDa. For proteolvtic processing, the soluble crystals were digested with trypsin for various times. The soluble crystal protein of 104 kDa was completely disappeared. However, the protein fragment of 60 kDa and 25 kDa still remained after complete proteolysis. The comparative immunoblot analysis showed that the antiserum against intact crystals showed strong immunoreactivity to the homologous inclusion protein of 138 kDa, 104 kDa, and 25 kDa, and to the intact spores of 221 kDa and 138 kDa, but not to the vegetative cell homogenate. The sera against crystals and spores had no immunoreactivity to the vegetative cell homogenate.
