A Simple Method for Generation of Homologous Internal Standards for Competitive PCR

  • In competitive PCR, which is used to quantify target DNA an internal standard is needed. Here we present a simple method to construct homologous competitive standards. The method, which is based upon deletion of a portion of the target DNA, does not require any additional primers. This is the simplest method developed thus far to construct a competitive standard. The whole procedure. from construction of a competitive standard to quantitation by PCR, can be completed within a single day.

  • The forecasting of container cargo volumes should be estimated correctly because it has a key roles on the establishment of port development planning, and the decision of port operating system. Container cargo volumes have a dynamic characteristics which was changed by effect of competitive ports. Accordingly forecasting was needed overall approach about competitive port's development, alternation and information. But, until now, traffic forecasting was not executed according to competitive situation, and that was accomplished at the point of unit port. Generally, considering the competition situation, simulation method was desirable at forecasting because system's scale was increased, and the influence power was intensified. In this paper, considering this situation, the objectives can be outlined as follows. 1) Structural model constructs by System dynamics method. 2) Structural simulation model develops according to modelling of competitive situation by expended SD method which included HEP(Hierarchical Fuzzy Process) And actually, effectiveness was verified according to proposed model to major port in northeast asia.

  • This introduces a improved method of the forward kinematics analysis, which finds the 6DOF motions and velocities from the given six cylinder lengths in the Steward platform. The numerical method(Newton Raphson Mehotd)of the forward kinematics analysises has the disadvantage of the long calculated time. To overcome this, we propose the competitive method that determine a proper initial value. Through the competitive method, we can select a proper initial value so that the calculate time is reduced. therefore we can give the property of the real time process to the forward kinematics analysis. We show the result comparing between general Newton-Raphson method and proposed one. From the result we verify the performance of the proposed method.

  • In this paper, we introduce MTS(Mahalanobis-Taguchi System) Method which is suggested Dr. Taguchi in later 1990. We apply MTS Method for Quality Competitive Appraisal System(QCAS) Model which is executived from 1997 by Agency for Technology and Standards, Ministry of Commerce, Industry and Energy(MOCIE). We can measure company Quality competitive by using MTS. MTS Measure can not be compared statistical sum by calculated QCAS. MTS can be possible distinct subtle which can not distinct using statistical sum. Also MTS Method can seek more strong effect factor among of many factor. If A Company use MTS Method, can find vital factor and level of destination.

A Study on the Structural Analysis of the Port Competition Power by FSM Method (FSM법에 의한 항만경쟁력의 구조분석에 관한 연구)

  • Although the ports are actually competing with various strategies, the definition and structural understanding of port competitive power are not known very much. Therefore this study has launched from this fact, and has the objective of obtaining the structural model of the competitive power, and understanding the components of the port competitive power. The following are the results of the study. First, the process began by abstracting the components that composed the port competitive power through recent research, and grouping it by the most core components using the KJ method. Also, by using the FSM(Fuzzy Structural Modeling) method to understand the structure of the grouped components, and the structural model of the port competitive power was able to obtain as the result. Second, when analyzing the obtained structural model, port expenses, main trunk location, port congestion and port facility came out to be the most important component groups, and especially port expenses was the most effective component that effected all the other components overall. Third, the component groups that were relatively less important, effected by most of the other components, and located on the top level of the structure model were the hinterland accessibility, port ownership, customs duties speed, and large ship port entrance possibility etc. Fourth, the results of this study will be able to be used when establishing competing strategies for our country's ports by proposing the relatively important components with the port competitive rower considered.

  • Portage kinetic constants of peptide transport can be measured by competitive spectrophotometry. The kinetic constants of L-Glu-L-Glu transport in Escherichia coli were ascertained using L-Phe-L-3-thia-Phe (PSP) as a detector. Since the production of thiophenol upon intracellular hydrolysis of PSP was competitively inhibited by L-Glu-L-Glu, it was able to compute the kinetic constants of L-Glu-L-Glu using this method. The resulted data were in agreement with the values obtained by the method of Michaelis-Menten kinetics. The potential of this method was examined against dipeptide transport systems in various microorganisms. These results strongly suggest that the overall properties of individual systems for dipeptide transports can be easily characterized by competitive spectrophotometry.

  • With the shift of the electric power industry from a regulated monopoly structure to a competitive market environment, the focus of the transmission expansion planning has been moving from reliability-driven transmission expansion to market-based transmission expansion. In market-based transmission expansion, however, a growing demand for electricity, an increasing number of transmission bottlenecks, and the falling levels of transmission investment have created the need for an incentive to motivate investors. The expectation of profit serves as a motivational factor for market participants to invest in transmission expansion in a competitive market. To promote investment in transmission expansion, there is an increasing need for a systematic method to examine transmission expansion for investment incentives from multiple perspectives. In this paper, the transmission expansion problem in a competitive market environment is formulated from ISO and investors' perspectives. The proposed method uses parametric analysis to analyze benefits for investors to identify the most profitable location and amount for transmission addition. Numerical results are presented to demonstrate the effectiveness of the proposed method.

  • If a system such as a port has a large boundary and complexity, and the system's substance is considered as a black box, forecast accuracy will be very low. Futhermore various components in a port exert significant influence on each other. To copy with these problem the form of structure models were introduced by using SD method. The Competitive Ports Model had several sub-systems consisting of each Unit Port models, and each Unit Port model was made by quantitative, qualitative factors and their feedback loops. The fact that all components of one port have influence on the components of the other ports should be taken into account to construct Competitive Port Models. However, with the current approach that is impossible, and in this paper therefore, models were simplified by HFP adapted to integrate level variables of unit port models. Although many studies on modelling of port competitive situation have been conducted, both theoretical frame and methodology are still very weak. In this study, a new algorithm called ESD(Extensional System Dynamics) for the evaluation of port competition was presented, and applied to simulate port systems in northeast asia.

Application of Competitive ELISA Method for Estimation of Urinary Aflatoxin M1 Level (ELISA 방법을 이용한 요중 아플라톡신 M1 측정)

  • We compared the efficacy of the competitive ELISA method for measuring the level of urinary aflatoxin M1 (AFM1) with that of the HPLC-fluorescence detector (HPLC-FLD) method. The recovery rate of AFM1 with the ELISA method was 105% (73-124%), and the coefficient of variation of the analysis was 6.85%. The ELISA method showed a 0.20 pg/ml and 0.62 pg/ml limit of detection and limit of quantitation, respectively. In correlation analysis, the two methods showed a very strong and statistically significant correlation (R=0.96, p<0.01). However, in spite of the strong correlation, the ELISA method tended to overestimate the urinary AFM1 concentration compared to the HPLC-FLD method. These results suggest that the competitive ELISA method may be a useful technique for measuring the AFM1 level in high-throughput urine samples, but it needs to be corrected with a regression equation from regression analysis with the HPLC-FLD method.
