Effect of temperature on pharmacokinetics of nalidixic acid, piromidic acid and oxolinic acid in olive flounder Paralichthys olivaceus following oral administration (넙치, Paralichthys olivaceus에 nalidixic acid, piromidic acid, oxolinic acid의 경구투여 약물동태에 미치는 수온의 영향)

  • Effects of temperature ($13{\pm}1.5^{\circ}C$, $23{\pm}1.5^{\circ}C$) on the pharmacokinetic properties of nalidixic acid (NA), piromidic acid (PA) and oxolinic acid (OA) were studied after oral administration to cultured olive flounder, Paralichthys olivaceus. Serum concentrations of these antimicrobials were determined after oral administration of a single dosage of 60 mg/kg body weight (average 700 g). At $23{\pm}1.5^{\circ}C$, the peak serum concentrations of NA, PA and OA, which attained at 10 h, 24 h and 30 h post-dose, were 11.55, 3.79 and $1.12{\mu}g/m\ell$, respectively. At $13{\pm}1.5^{\circ}C$, the peak serum concentrations of NA, PA and OA, which attained at 10 h, 15 h and 30 h post-dose, were 6.36, 1.4 and $1.01{\mu}g/m\ell$, respectively. Better absorption of NA and PA was noted at $23{\pm}1.5^{\circ}C$ compared to $23{\pm}13^{\circ}C$. The elimination of NA from serum of olive flounder was considerably faster at $23{\pm}1.5^{\circ}C$ than at $13{\pm}1.5^{\circ}C$. However, both absorption and elimination of OA were not affected significantly by temperature. The kinetic profile of absorption, distribution and elimination of these antimicrobials in serum were analyzed by fitting to a one- and two compartment model, with WinNonlin program. In the one compartment model for NA, AUC, Tmax and Cmax at $23{\pm}1.5^{\circ}C$ were $258.26{\mu}g{\cdot}h/m\ell$, 10.67 h and $8.91{\mu}g/m\ell$, respectively. The AUC, $T_{max}$ and $C_{max}$ at $13{\pm}1.5^{\circ}C$ were $341.45 {\mu}g{\cdot}h/m\ell$, 7.72 h and $6.23{\mu}g/m\ell$, respectively. In the one compartment model for PA, AUC, $T_{max}$ and $C_{max}$ at $23{\pm}1.5^{\circ}C$ were $248.12{\mu}g{\cdot}h/m\ell$, 21.15 h and $3.09{\mu}g/m\ell$, respectively. The AUC, $T_{max}$ and $C_{max}$ at $13{\pm}1.5^{\circ}C$ were $103.89{\mu}g{\cdot}h/m\ell$, 12.89 h and $1.22{\mu}g/m\ell$, respectively. In the two compartment model for OA, AUC, $T_{max}$ and $C_{max}$ at $23{\pm}1.5^{\circ}C$ were $138.20{\mu}g{\cdot}h/m\ell$, 23.95 h and $1.06{\mu}g/m\ell$, respectively. The AUC, $T_{max}$ and $T_{max}$ at $13{\pm}1.5^{\circ}C$ were $159.10{\mu}g{\cdot}h/m\ell$, 28.03 h and $1.02{\mu}g/m\ell$, respectively.

Numerical Analysis on the Initial Cool-down Performance Inside an Automobile for the Evaluation of Passenger's Thermal Comfort (차량 내부 탑승자의 쾌적성 평가를 위한 초기 냉방운전 성능에 대한 수치해석적 연구)

  • Cool-down performance after soaking is important because it affects passenger's thermal comfort. The cooling capacity of HVAC system determines initial cool down performance in most cases, the performance is also affected by location, and shape of panel vent, indoor seat arrangement. Therefore, optimal indoor designs are required in developing a new car. In this paper, initial cool down performance is predicted by CFD(computational fluid dynamics) analysis. Experimental time-averaging temperature data are used as inlet boundary condition. For more reliable analysis, real vehicle model and human FE model are used in grid generation procedure. Thermal and aerodynamic characteristics on re-circulation cool vent mode are investigated using CFX 12.0. Thermal comfort represented by PMV(predicted mean vote) is evaluated using acquired numerical data. Temperature and velocity fields show that flow in passenger's compartment after soaking is considerably unstable at the view point of thermodynamics. Volume-averaged temperature is decreased exponentially during overall cool down process. However, temperature monitored at different 16 spots in CFX-Solver shows local variation in head, chest, knee, foot. The cooling speed at the head and chest nearby panel vent are relatively faster than at the knee and foot. Horizontal temperature contour shows asymmetric distribution because of the location of exhaust vent. By evaluating the passenger's thermal comfort, slowest cooling region is found at the driver's seat.

  • The objective of the present study is to predict the characteristics of the fire and smoke propagations in a clean room. Numerical calculations have been performed by using the finite volume method to obtain temperature and velocity distributions in the clean room. In odor to account for the turbulent flow characteristics, the standard $k-{\varepsilon}$ model is used. From this study, it was found that the fire propagation could be fully developed only after 150 seconds when the ventilation system in the clean room was off. And the smoke mass fraction showed a similar distribution as the gas temperature. Since the simulated fire was proceeded up to $20{\sim}30%$ of the room within 60 seconds. it could be recommended that the occupants should be evacuated from the room within 30 seconds.

Effects of Temperature on the Uptake and Retention of Cesium-137 by the Clam Cyclina sinensis (가무락조개에 의한 세슘-137 의 농축(濃縮)과 잔류(殘留)에 미치는 온도(溫度)의 영향)

  • The effects of temperature on the uptake of $^{137}Cs$ from seawater and on the retention after its uptake by the clam Cycling sinensis was investigated under laboratory conditions. The clams exhibited a greater bioaccumulation of $^{137}Cs$ in $25^{\circ}C$-acclimated animals than those acclimated at $15^{\circ}C$. The viscera of the clams reached the highest bioconcentration factor after 14 days uptake from seawater, but the tissue distribution pattern of $^{137}Cs$ was little influenced, if any, by the uptake temperature. The uptake rate slightly decreased with an increase of temperature in order of $10^{\circ}C$. The radionuclide accumulated in clams was released again in a radionuclide-free seawater according to a two-exponential compartment model. A temperature increase of $10^{\circ}C$ reduced the biological half-life of the long-lived component with a factor of about two, whereas it caused no change in the short-lived component.

At $23{\pm}1.5^{\circ}C$, the peak serum concentrations of NA and PA, which attained at 24 h post-dose, were 5.87 and $0.43\;{\mu}g/ml$, respectively. At $13{\pm}1.5^{\circ}C$, the peak serum concentrations of NA and PA, which attained at 10 h post-dose, were 6.22 and $1.57\;{\mu}g/ml$, respectively. Better absorption of PA was noted at $13{\pm}1.5^{\circ}C$ compared to $23{\pm}1.5^{\circ}C$. However, absorption of NA was not affected significantly by temperature. The elimination of NA and PA from serum of black rockfish was considerably faster at $23{\pm}1.5^{\circ}C$ than at $13{\pm}1.5^{\circ}C$. The AUC, $T_{1/2}$, $T_{max}$ and $C_{max}$, respectively, were: $161.25\;{\mu}g{\cdot}h/ml$, 0.15 h, 12.29 h and $8.91\;{\mu}g/ml$ at $23{\pm}1.5^{\circ}C$, and $134.12{\mu}g{\cdot}h/ml$, 0.18 h, 8.79 h and $5.00\;{\mu}g/ml$ at $13{\pm}1.5^{\circ}C$ with NA; $41.57\;{\mu}g{\cdot}h/ml$, 0.58 h, 8.24 h and $0.21\;{\mu}g/ml$ at $23{\pm}1.5^{\circ}C$, and $40.36\;{\mu}g{\cdot}h/ml$, 0.59 h, 5.04 h and $1.20\;{\mu}g/ml$ at $13{\pm}1.5^{\circ}C$ with PA.

Separation of Soybean Protein by Free-flow Electrophoresis (자유유동 전기이동법에 의한 대두단백질 분리)

  • The effect of operating conditions on separation of soybean proteins in a home-made free-flow electrophoresis apparatus was investigated. Measurement of the pH, conductivity, and UV-absorbance(280 nm) were carried out at each run and the purity of the sample was tested with SDS-PAGE analysis. The soybean extract pretreated with Tris and boric acid was mixed with the amino acids composed of glutamic acid, histidine, arginine, glycine(1 mM each) with glycyl-glycine(2mM) and KCl(1mM). When the cellulose acetate was used as a compartment between the electrode and the buffer solution in the cell, pH distribution in the separation cell varied from 3.0 at the anodic side to 8.0 at the cathodic side and had two inflection point. The applied voltage was from 300V to 1000V and the separation was better at a higher voltage but the voltage was limited by the capability of the cooling system due to Joule heat. The proteins focused near the middle of the channel. From the change of pH and conductivity it was found that the ions in the channel moved out to the electrodes through the membrane. In the case when the concentration of the buffer solution was increased 5 times, proteins were focused at 300V. We could not increase up to the ten times of the concentration since the temperature difference between inlet and outlet was more than $25^{\circ}C$ and denaturation of proteins was expected. When ion-exchange membranes were used U-type pH distribution was set up due to the ionic polarization near the membrane. The commercial ampholytes, instead of the mixed amino acids showed not much improvements in purity of the separated sample.

  • The heating, ventilating, air conditioning (HVAC) system is a very important part of an automotive vehicle: it controls the microclimate inside the passenger's compartment and removes the frost or mist that is produced in cold/rainy weather. In this study, the numerical analysis of the defrost duct in an HVAC system and the de-icing pattern is carried out using commercial CFX-code. The mass flow distribution and flow structure at the outlet of the defrost duct satisfied the duct design specification. For analyzing the de-icing pattern, additional grid generation of solid domain of ice and glass is pre-defined for conductive heat transfer. The flow structure near the windshield, streakline, and temperature fields clearly indicate that the de-icing capacity of the given defrost duct configuration is excellent and that it can be operated in a stable manner. In this paper, the unsteady changes in temperature, water volume fraction, and static enthalpy at four monitoring points are discussed.
