$17{\alpha}-ethinylestradiol$ (EE2), a synthetic estrogen which interfere the endocrine and reproductive function in living organisms, has been found extensively to be deposited into municipal wastewater treatment plants and the environment via human excretion. EE2 has long been known to be efficiently cometabolized by ammonia-oxidizing bacteria (AOB) during ammonia ($NH_3$) oxidation. Current study aims to investigate the effect of culture history on the biotransformation of EE2 by nitrifying sludge which was enriched under different ammonia loading rates in continuous flow reactors. Result showed that past growth condition largely affected not only the metabolic rate of $NH_3$ oxidation but also EE2 cometabolism. Sludge previously acclimated with higher $NH_3$ loads as well as sludge dominated with AOB belong to high growth cluster (Nitrosomonas europaea-Nitrosococcus mobilis) showed higher rate of EE2 biotransformation than those one being acclimated with lower $NH_3$ loads because of its ability to provide more reducing power from $NH_3$ oxidation. Moreover, the correlation between the degradation rates of $NH_3$ and EE2 was higher in sludge being acclimated with higher load of $NH_3$ in comparison with other sludge. Implication of the findings emphasized the role of volumetric $NH_3$ loading rate in determining EE2 removal in wastewater treatment system.

Single-well-gas-sparging tests were developed and evaluated for assessing the feasibility of in-situ aerobic cometabolism of trichloroethylene (TCE), using propane as a growth substrate. To evaluate transport characteristics of dissolved solutes [sulfur hexafluoride (SF6) or bromide (non-reactive tracers), propane (a growth substrate), ethylene, propylene (nontoxic surrogates to probe for CAH transformation activity), and DO], push-pull transport tests were performed. Mass balance showed about 90% of the injected bromide and about 80% of the injected SF6 were recovered, and the recoveries of other solutes were comparable with bromide and slightly higher than SF6. A series of Gas-Sparging Biostimulation tests were performed by sparging propane/oxygen/argon/SF6 gas mixtures, and temporal ground water samples were obtained from the injection well under natural gradient 'drift' conditions. The decreased time for propane depletion and the longer time to deplete SF6 as a conservative tracer indicate the progress of biostimulation. Gas-Sparging Activity tests were performed. .Propane utilization, DO consumption, and ethylene and propylene cometabolism were well demonstrated. The stimulated propane-utilizers cometabolized ethylene and propylene to produce ethylene oxide and propylene oxide, as cometabolic by-products, respectively. Gas-Sparging Acetylene Blocking tests were performed by sparging gas mixtures including acetylene to demonstrate the involvement of monooxygenase enzymes. Gas substrate degradation was essentially completely Inhibited in the presence of acetylene, and no production of the corresponding oxides was also observed. The Gas-Sparging tests supports the evidences that the successive stimulation of propane-oxidizing microorganisms, cometabolic transformation of ethylene and propylene by the enzyme responsible for methane and propane degradation.
생촉매 및 생물막 반응기를 이용한 TCE 생분해는 TCE를 무해한 최종산물로 처리할 수 있는 환경친화적 방법이며, 초기 시설비와 운영비도 낮아 경제성도 우수한 기술로 평가할 수 있다. 그러나, TCE 및 독성 분해산물로 인하여 생촉매 불활성화가 일어나서 장기간 안정된 반응기 운전이 어렵고, TCE와 성장기질사이의 경쟁적 저해로 인하여 처리효율이 저하된다는 단점이 있다. 이러한 문제점을 극복하기 위하여 TCE 처리 단계와 생촉매 재활성화 단계를 구분시킨 2단계 CSTR/TBF 시스템에 대하여 TCE 연속처리용 시스템으로써의 실규모 적용 가능성을 평가해 보았다. B. cepacia 및 M. trichosporium을 생촉매로 사용한 2단계 CSTR/TBF 시스템은 고농도 유입 TCE와 다양한 운전조건에서도 28∼525mg TCE/1$.$day수준의 높은 TCE 처리효율을 안정되게 유지할 수 있어 산업폐가스 처리를 위한 실규모 처리 시스템으로 적용 가능성이 높다고 평가할 수 있었다.
Cometabolism technology was employed to degrade lignin wastewater in Sequencing Batch Biofilm Reactor. Cometabolic system (with glucose and lignin in inflow) and the control group (only lignin in inflow) were established to do a comparative study. In contrast with the control group, the average removal rates of lignin increased by 14.7% and total oarganic carbon increased by 32% in the cometabolic system with glucose as growth substrate, under the condition of 5 mg/L DO, $0.2kgCOD/(m^3{\cdot}d)$ lignin and glucose $1.0kgCOD/(m^3{\cdot}d)$. Functional groups of lignin are degraded effectively in cometabolic system proved by fourier transform infrared spectroscopy and Gas Chromatography-Mass Spectrometer, and the degradation products were amides (mainly including acetamide, N-ethylacetamide and N, N-diethylacetamide), alcohols (mainly including glycerol and ethylene glycol) and acids. Meanwhile, results of Polymerase Chain Reaction-Denaturing Gradient Gel Electrophoresis showed great differences in microbial population richness between cometabolic system and the control group. The Margalef's richness index and Shannon-Wiener's diversity index of microorganism in cometabolic system were 3.075 and 2.61, respectively. The results showed that extra addition of glucose, with a concentration of 943 mg/L, was beneficial to lignin biodegradation in cometabolic system.
공업용 유기용매나 그리스 제거제, 전자기판 세척제로 널리 사용되어 온 TCE는 적절한 처리 방법이 개발되지 못하여 환경오염을 일으키고 있으며, 유전자변이 및 암을 유발하는 것으로도 알려져 효율적인 처리기술 개발이 요구된다. 미생물막을 이용한 처리 방법은 TCE를 무해한 최종산물로 분해시킬 수 있으며, 운전비 및 설비비도 낮아 경쟁력 있는 처리 기술로 평가되고 있다. TCE 자체는 미생물의 성장기질로 사용되지 못하므로 공동대사 과정을 거쳐 분해된다. TCE 및 TCE 분해산물의 높은 독성으로 인하여 TCE 분해효소 및 세포 분해산물의 높은 독성으로 인하여 TCE 분해효소 및 세포 불활성화가 일어나서 장기간 안정된 반응기 운전이 어렵다는 단점과 성장기질을 공급하는 경우 유발되는 경쟁저해로 인한 분해효율 감소를 막을 수 있는 여러 종류의 생물반응기 시스템들이 개발되었다. 이러한 생물반응기 시스템의 개발과 우수한 분해능을 가지는 균주 선별 및 개량 등을 통해 실제 현장에 적용시킬 수 있는 생물학적 TCE 처리 기술의 실용화가 가능할 것이다.
Sing]e-well-push-pull tests were developed for use in assessing the feasibility of in-situ aerobic cometabolism of chlorinated aliphatic hydrocarbons (CAHs). The series includes Transport tests, Biostimulation tests, and Activity tests. Transport tests are conducted to evaluate the mobility of solutes used in subsequent tests. These included bromide or chloride (conservative tracers), propane (growth substrate), ethylene, propylene (CAH surrogates), dissolved oxygen (electron acceptor) and nitrate (a minor nutrient). Tests were conducted at an experimental well field of Oregon State University. At this site, extraction phase breakthrough curves for all solutes were similar, indicating apparent conservative transport of the dissolved gases and nitrate prior to biostimulation. Biostimulation tests were conducted to stimulate propane-utilizing activity of indigenous microorganisms and consisted of sequential injections of site groundwater containing dissolved propane and oxygen. Biostimulation was detected by the increase in rates of propane and oxygen utilization after each injection. Activity tests were conducted to quantify rates of substrate utilization and to confirm that CAH-transforming activity had been stimulated. In particular, the transformation of injected CAH surrogates ethylene and propylene to the cometabolic byproducts ethylene oxide and propylene oxide provided evidence that activity of the monooxygenase enzyme system, responsible for aerobic cometabolic transformations of CAHs had been stimulated. Estimated zero-order transformation rates decreased in the order propane > ethylene > propylene. The series of push-pu3l tests developed and field tested in this study should prove useful for conducting rapid, low-cost feasibility assessments for in situ aerobic cometabolism of CAHs.
본 연구에서는 유입수의 변동이 심하고 전문가가 부재한 환경인 선박에서 발생하는 오수의 효과적인 처리를 위하여 RCM공법을 선박오수처리장치에 적용하는 실험실 규모의 실험을 수행하였다. 질소 인의 고도처리 효율과 선박이라는 특수환경과의 접목성을 검토한 결과 RCM공정에 유효미생물을 주입하는 방법은 선박환경에 적합한 것으로 평가되었다. 또한 RCM공정은 활성슬러지 공정에서 배출되는 슬러지는 배출시키지 않고 슬러지액화분해조(SDC)에서 재분해하여 순환함으로써, 최근 해양투기가 금지됨으로 인해 문제가 되고 있는 슬러지의 발생량을 최소한으로 하여 친환경적인 수처리가 가능하다. 복합미생물제제 주입 후 미생물 관찰결과 고도처리에 유리한 미생물종의 출현을 확인하였으며 이들의 상호기작으로 질소 인의 처리에 도움을 주어 처리효율이 높은것이라 판단된다. 유기물 제거효율 실험결과 $BOD_5$, CODcr T-N, T-P의 처리효율이 각각 96, 97, 78, 81.68 %로 나타나 Membrane이나 Filter없이도 강화되어가는 해양오염기준을 충족시킬 수 있는 공정으로 판단된다.
본 논문에서는 순수균주인 ENV425와 유류오염토양에서 butane을 탄소원 및 에너지원으로 이용하여 분리한 혼합균주를 대상으로 MTBE와 기타 가솔린 산화제 분해특성을 조사했다. ENV425와 혼합균주의 butane monooxygenase (BMO)에 의해 butane 분해시 1-butanol이 주요 부산물로 축적되었다. 또한 monooxygenase의 방해자로 알려진 acetylene의 첨가시에는 butane의 분해 및 1-butane떠 축적이 일어나지 않아 butane monooxygenase에 의한 분해임을 알 수 있다. 본 연구결과에서, propane, pentane, butane을 포함한 alkane류는 MTBE 공대사에 우수한 성장기질이었고, BTEX 화합물 역시 MTBE 공대사에 가능성 있는 기질임이 관찰되었다. 또한 균주농도 역시 MTBE분해에 영향을 미치는 것으로 나타났는데, 균주 농도 증가에 따라 MTBE 분해량은 증가하나 transformation capacity는 상대적으로 감소하는 경향을 보였다. 그리고 대표적인 가솔린 산화제인 MTBE 외에 ETBE, TAME도 부탄분해균에 의해 효과적으로 분해가 이루어짐이 관찰되었다.
