The Effect of Dietary Fat Inclusion on Nutrient Intake and Reproductive Performance in Postpartum Awassi Ewes

  • The objective of this study was to evaluate the effect of dietary fat inclusion on nutrient intake, body weight, milk production, return to estrus, pregnancy and lambing of winter-lambing, postpartum Awassi ewes. Thirty multiparous, winter-lambing Awassi ewes (body weight=51${\pm}$7.0 kg) were randomly assigned to three dietary treatments (n=10) for 62 days using a completely randomized design. Experimental diets were isonitrogenous, and were formulated to contain 0 (CON), 2.5 (MF), and 5% (HF) added fat, and 33% of the dietary crude protein (CP) as undegradable intake protein (UIP). On day 26 postpartum (day 0=parturition), ewes and their lambs were housed in individual pens for 28 days. Feed offered and refused was recorded daily. At the end of this period, ewes and their lambs within each treatment were combined into one group and fed their respective diet ad libitum. One fertile Awassi ram fitted with a marking harness was allowed with each group for 34 days. No significant (p>0.05) differences in dry matter intake, organic matter intake, and crude protein intake were observed for ewes fed the three experimental diets. No difference was observed in metabolizable energy intake (MEI) for ewes fed the CON and the MF diets (average 8.3 Mcal/d) diet. However, ewes fed the HF diet had greater(p<0.05) MEI compared with the rest of the treatments. Ewe body weights increased throughout the study, unaffected by the experimental diets. No significant differences in milk production were found among ewes fed the three experimental diets. No significant differences were observed in pregnancy rate (6/10, 5/10, 6/10 for CON, MF and HF diets, respectively), lambing rate and the number of lambs per ewe among the three treatments. postpartum reproductive performance of well-fed, winter-lambing Awassi ewes.

Maternal selenium-supplementation at various stages of periconception period: influence on murine blastocyst morphology and implantation status

  • Background: Selenium is one of the trace minerals whose deficiency is known to lead to complications of female reproduction. The identified gaps in researches regarding selenium and pregnancy include optimizing the dosage of selenium supplementation, timing of supplementation, finding the best form and type of selenium, and selenium administration combined with other antioxidants. Hence, this study was conceptualized to address one of the identified gaps, that is, to find out the best timing of selenium administration around the time of pregnancy. Specifically, this study aimed to assess the effects of maternal Selenium-supplementation, administered at various stages of periconception period, on murine blastocyst morphology, percent occurrence of good quality blastocysts, and implantation status. Methods: ICR female mice were randomly assigned into the unsupplemented group (Group I) receiving basal diet without selenium, and treatment groups given with $3.0{\mu}g$ selenium-supplement per day during pregestation only (Group II), pregestation-throughout-gestation (Group III) and gestation only (Group IV). Both blastocyst morphology and implantation status were assessed. Results: The morphometric measurements of blastocysts appeared to be unaffected by selenium-supplementation at different stages of periconception. Selenium-supplementation at pregestation only (Group II) and gestation only (Group IV) produced higher percent occurrence of good quality blastocysts and lower percent pre-implantation loss than Group III. Among all the treatment groups, Group III (Selenium-supplementation during pregestation-to-gestation) yielded the lowest quality blastocysts and highest percent pre-implantation loss. Conclusion: Maternal selenium-supplementation during pregestation and gestation stages of the periconception period yielded a high percent occurrence of good quality blastocysts and pre-implantation success.

  • Although anejaculation is a relatively uncommon occurrence in the general population, over 12,000 new cases are reported annually. Anejaculation may result from spinal cord injury, retroperitoneal lymph node dissection, diabetes mellitus, transverse myelitis, multiple sclerosis, or psychogenic disorders. At least 30% of men with this problem are or will be married and many will seek help to remedy their infertile state. The evolution of technique and instrumentation over the last 30 years has made electroejaculation an accessible and acceptable form of therapy. Recent successes in inducing ejaculation by means of rectal probe electrostimulation or vibratory stimulation combined with assisted reproductive techniques, such as zygote intrafallopian transfer (ZIFT), gamete intrafallopian transfer (GIFT), and in vitro fertilization (IVF), have provided these men means of producing their own biologic offspring. We have experienced a successful pregnancy with electroejaculation and in vitro fertilization in a infertile patient whose husband had an ejaculatory disturbance due to a spinal cord injury. So we report this case with a brief review of literatures.

Studies on Development of Breeding Technique to Increase Hanwoo (Bos taurus coreanae) III. Hormonal Treatment of Reproductive Disorders and Effect of Intraovarian $\textrm{PGF}_{2a}$ Administration in Hanwoo (한우의 신속한 증식을 위한 번식기술 개발에 관한 연구 - III. 한우에서 번식장애 처치 및 $\textrm{PGF}_{2a}$의 난소실질내 투여효과에 관한 연구)

  • In order to develop the breeding techniques to increase Hanwoo (Bos taurus coreanae) population, the present study was performed 1) to establish the treatment protocol on reproductive disorders with GnRH or PGF/sub 2$\alpha$/, 2) to improve intraovarian PG $F_{2}$$\alpha$/ administration for reducing open period. Among total of 43 diagnosed, high percentage of cows (41.9%, 18 cows) were diagnosed as silent heat, followed by inactive ovaries (32.6%, 14 cows), ovarian cysts (9.3%, 4 cows), persistent corpus luteum (7.0%, 3 cows), endometriosis (4.7%, 2 cows), pyometra (2.3%, 1 cow) and luteal cysts (2.3%, 1 cow). To treat silent heat, 18 cows were administrated with 25 mg PGF/sub 2$\alpha$/, heat-detected, artificially inseminated and monitored pregnancy. All treated cows were heat-detected and 16 cows (88%) were successfully pregnant. With 200 $\mu\textrm{g}$ GnRH treatment, 7 cows (70%) with inactive ovaries and 3 cows (75%) with ovarian cysts were successfully pregnant. Administration with 25mg PGF/sub 2$\alpha$/, successfully treated 3 cows (100%) with persistent corpus luteum and 1 cow (100%) with luteal cysts, followed by 100% pregnancy rate. With the combined treatment of 25 mg PG $F_{2}$$\alpha$/and antibiotics, 2 cows (100%) with endometriosis were effectively treated and got pregnant after. artificial insemination (AI). In order to reduce open period, 5 mg PGF/sub 2$\alpha$/ was administrated intraovarian to 20 days cows after delivery, heat-detected, artificially inseminated and monitored pregnancy. In the first experiment, in order to recover uterus, 5mg PGF/sub 2$\alpha$/were administrated, followed by administration of 5mg PGF/sub 2$\alpha$/ at the interval of 14 days. As results, 74% (17/23 cows) of pregnancy rate after AI. In order to further reduce the open period, 5 mg PGF/sub 2$\alpha$/was administrated at the interval of 11 days without the period of uterus recovery, resulted in 94% (16/17 cows) pregnancy rate. In conclusion, these results showed that PGF/sub 2$\alpha$/ and GnRH treatment were effective hormonal treatment resume in Hanwoo with various reproductive disorders. In addition, modified protocol of intraovarian PGF/sub 2$\alpha$/ administration could be the effective method for reducing the open period.

Effect of Progesterone Implant and Follicular Rupture on Estrus Induction and Fertility in Anestrus Cows (무발정우에서 Progesterone Implant와 Follicular Rupture에 따른 발정유도 및 임신율)

  • The objective of this study was to compare the effect of four different estrus induction methods in anestrus cows on the estrus induction and pregnancy following artificial insemination (AI). Sixty-five cows (3∼4 years old) were selected and divided into four different estrus induction treatment groups. Group 1, 12 cows were treated by Ovsynch program combined with GnRH and PGF$_2$a. Group 2, 12 cows were treated by "Tow plus Two" program with GnRH and PGF$_2$a. Group 3, 20 cows were treated by "Tow plus Two" program following intravaginal progesterone implantation (CIDR). Twenty one cows in Group 4 were treated by "Tow plus Two" program following follicular rupture and intravaginal progesterone implantation. Cows were then observed estrus induction and inseminated artificially at 12 h and 24 h after standing estrus. The rates of estrus induction in Group 4 (18/21, 86%) was significantly (P<0.05) higher than those in groups 1, 2 and 3 (8/12, 67%; 9/12, 75%; 14/20, 70%). In the mean time of onset of estrus after final administration of GnRH in different hormone-treated cows, the cows in Group 3 (24.2$\pm$2.2) and Group 4 (23.4$\pm$2.0) were significantly (P<0.05) shorter than that in Group 1 (28.5$\pm$4.6) and Group 2 (26.4$\pm$3.3). The rates of pregnancy diagnosed on Day 28 were significantly different between treatment groups. Significantly (P<0.05) higher rate of pregnancy was observed in Group 4 (17/20, 85.0%) than those in Groups 1, 2 and 3 (7/11, 63.6%; 8/12, 66.7%; 15/20, 75.0%, respectetively). The rate of abortion diagnosed on 49 days of gestation was significantly (P<0/05) lower in Group 4 (1/17, 5.9%) than those in Groups 1, 2 and 3 (2/7, 28.7%; 2/8, 25% and 3/15, 20%, respectively). In conclusion, combined treatments with GnRH and PGF$_2$a following follicular rupture and progesterone implant in anestrus cows was considered to be most effective in estrus induction and maintenance of pregnancy. Further studies are needed to verify the functional mechanisms of residual follicles in anestrus ovaries on retarding the response of hormonal treatments.sponse of hormonal treatments.

Presence of Transcription Factor OCT4 Limits Interferon-tau Expression during the Pre-attachment Period in Sheep

  • Interferon-tau (IFNT) is thought to be the conceptus protein that signals maternal recognition of pregnancy in ruminants. We and others have observed that OCT4 expression persists in the trophectoderm of ruminants; thus, both CDX2 and OCT4 coexist during the early stages of conceptus development. The aim of this study was to examine the effect of CDX2 and OCT4 on IFNT gene transcription when evaluated with other transcription factors. Human choriocarcinoma JEG-3 cells were cotransfected with an ovine IFNT (-654-bp)-luciferase reporter (-654-IFNT-Luc) construct and several transcription factor expression plasmids. Cotransfection of the reporter construct with Cdx2, Ets2 and Jun increased transcription of -654-IFNT-Luc by about 12-fold compared with transfection of the construct alone. When cells were initially transfected with Oct4 (0 h) followed by transfection with Cdx2, Ets2 and/or Jun 24 h later, the expression of -654-IFNT-Luc was reduced to control levels. OCT4 also inhibited the stimulatory activity of CDX2 alone, but not when CDX2 was combined with JUN and/or ETS2. Thus, when combined with the other transcription factors, OCT4 exhibited little inhibitory activity towards CDX2. An inhibitor of the transcriptional coactivator CREB binding protein (CREBBP), 12S E1A, reduced CDX2/ETS2/JUN stimulated -654-IFNT-Luc expression by about 40%, indicating that the formation of an appropriate transcription factor complex is required for maximum expression. In conclusion, the presence of OCT4 may initially minimize IFNT expression; however, as elongation proceeds, the increasing expression of CDX2 and formation of the transcription complex leads to greatly increased IFNT expression, resulting in pregnancy establishment in ruminants.

Optimization of Trichomonas vaginalis Diagnosis during Pregnancy at a University Hospital, Argentina

  • The aim of this study was to evaluate different methods for Trichomonas vaginalis diagnosis during pregnancy in order to prevent maternal and perinatal complications. A total of 386 vaginal exudates from pregnant women were analyzed. T. vaginalis was investigated by 3 types of microscopic examinations direct wet mount with physiologic saline solution, prolonged May-Grunwald Giemsa (MGG) staining, and wet mount with sodium-acetate-formalin (SAF)/methylene blue method. PCR for 18S rRNA gene as well as culture in liquid medium were performed. The sensitivity and specificity of the microscopic examinations were evaluated considering the culture media positivity or the PCR techniques as gold standard. The frequency of T. vaginalis infection was 6.2% by culture and/or PCR, 5.2% by PCR, 4.7% by culture, 3.1% by SAF/methylene blue method and 2.8% by direct wet smear and prolonged MGG staining. The sensitivities were 83.3%, 75.0%, 50.0%, and 45.8% for PCR, culture, SAF/methylene blue method, and direct wet smear-prolonged MGG staining, respectively. The specificity was 100% for all the assessed methods. Microscopic examinations showed low sensitivity, mainly in asymptomatic pregnant patients. It is necessary to improve the detection of T. vaginalis using combined methods providing higher sensitivity, such as culture and PCR, mainly in asymptomatic pregnant patients, in order to prevent maternal and perinatal complications.

Effect of choline chloride supplementation on milk production and milk composition of Etawah grade goats

  • Background: The effect of choline chloride supplementation through forced drinking combined with concentrate diets containing Ca-fish oil on milk production and milk composition of Etawah Grade goats was evaluated. Choline chloride is an essential component in ruminant diets as it is required for fat metabolism. Method: The experiment was conducted in a completely randomized block design with three types of treatments and eight replications. The trial had two successive experimental periods; the first, during the eight weeks of late pregnancy, and the second, during the first 12 weeks of lactation. Twenty-four Etawah Grade does in the second gestation period were divided into three treatment groups. Commercial choline chloride 60 % in corncobs-based powder was used as a source of choline chloride. The treatments were no supplementation (control) and supplemented with either 4 g or 8 g/2days of choline chloride. Choline chloride was given to the animals through a forced drinking technique, after dissolving it in 60 ml drinking water. The initial body weight of does was $38.81{\pm}3.66kg$. The does were penned individually, and were given fresh chopped King Grass ad libitum and 700 g/day of concentrate diets containing Ca-fish oil, starting eight weeks prior to expecting kidding and continuing for 12 weeks of parturition. Results: All nutrient intakes were not significantly different (p > 0.05) among the treatments during the late pregnancy and the lactation periods. Supplementation did not affect (p > 0.05) the average daily gains and feed conversion ratio during pregnancy but gave effects (p < 0.05) on the average daily gains, feed conversion ratio and income over feed cost during lactation. The highest average daily milk yields and 4 % fat corrected milk yields were found in goats supplemented with 4 g/2days of choline chloride and increased by 17.00 % and 24.67 %, respectively, compared to the control. Moreover, milk composition percentage and milk constituent yields improved significantly (p < 0.05) in those supplemented with 4 g/2days of choline chloride. Conclusion: The supplementation of 4 g/2days of choline chloride through forced drinking increased milk yields, the 4 % fat corrected milk yields, milk composition, milk constituent yields, and improved feed conversion ratio and income over feed cost of Etawah Grade goats.

  • Objective: The aim of this study was to compare the clinical efficacy of clomiphene citrate (CC) and letrozole combined with gonadotropins for controlled ovarian stimulation (COS) in patients with CC-induced thin endometrium Methods: Fifty-one intrauterine insemination cycles performed in patients who previously had a thin endometrium (<8 mm) to ovulation induction using CC were included in this study. A CC 100 mg/day (CC+gonadotropin group, n=26) or letrozole 2.5 or 5 mg/day (letrozole+gonadotropin group, n=25) was administered on day 3~7 of the menstrual cycle, combined with gonadotropins at dose 75~150 IU every other day starting on day 5~7. We compared total dose of gonadotropin used, endometrial thickness, endometrial pattern, number of follicles ${\geq}14\;mm$ on hCG day, pregnancy rate and multiple pregnancy rate between the two groups, which were statistically analyzed using Mann-Whitney U test or Fisher's exact test, where appropriate. Results: There were no significant differences in clinical characteristics such as age, duration of infertility, number of previous IUI cycles, basal serum hormone levels and cause of infertility between the two groups. In both groups, the endometrium was significantly thicker than that of previous ovulation induction cycles using CC. No significant differences were found in the total dose of gonadotropin used, day of hCG administration, the rate of triple endometrium and pregnancy rate. The number of follicles ${\geq}14\;mm$ was significantly lower ($3.7{\pm}1.7$ vs. $2.8{\pm}1.7$, p=0.03) and the endometrium on hCG day was significantly thicker ($7.7{\pm}1.5$ vs. $9.1{\pm}1.7$, p=0.001) in letrozole+gonadotropin group compared to CC+gonadotropin group. Conclusion: The clomiphene citrate and letrozole combined with gonadotropins appear to avoid the undesirable effects on the endometrium frequently seen with CC for ovulation induction. However, in terms of adequate endometrial development or optimal follicular growth, letrozole may be more beneficial than CC for gonadotropin-combined COS in patients with CC-induced thin endometrium. Further prospective randomized controlled studies in a larger scale will be necessary to confirm our findings.

Estrous Response and Fertility in Postpartum Suckled Female Yaks (Bos grunniens) Treated with an Intravaginal Device Containing Progesterone(CIDR), Pregnant Mares' Serum Gonadotrophin and Prostaglandin Analogue

  • The efficiency of a short-term treatment with an intravaginal device containing progesterone (CIDR) combined with pregnant mares' serum gonadotrophin (PMSG) and prostaglandin analogue ($PGF_{2\alpha}$) was evaluated for the induction of estrus, initiation of cyclic activity, and fertility in postpartum suckled yak cows. Seventy-five postpartum suckled yak cows were assigned to three treatments: (1) insertion of a CIDR intravaginal progesterone (1.9 g) (day 0), an administration of $PGF_{2\alpha}$ (0.2 mg i.m.) on day 6 and PMSG (1,000 IU i.m.) at the time of CIDR withdrawal on day 7 (CPP group, n=28); (2) an administration of $PGF_{2\alpha}$ (0.2 mg i.m.) on day 6 and PMSG (1,000 IU i.m.) on day 7 (PP group, n=21); (3) untreated animals served as the control (CG group, n=26). Seven yak bulls were placed in pastures with the cows for natural mating. Estrus rate in the CPP group (28/28) was higher (p<0.01) than in the PP group (6/21) and in the CG group (0/26) within 96 h after the end of treatment. The first service conception rate in the CPP group (21/28) was higher (p<0.01) compared with in the PP group (2/9) as judged by serum $P_4$ concentration $\geq$2.35 ng/ml on day 21 after breeding. It is concluded that a short-term progesterone treatment combined with PMSG and prostaglandin increased the proportion of yak cows that exhibited behavioral estrus with more synchronized estrus response and satisfactory conception rate in postpartum suckled yak cows.
