Role of Ser-33 and Asp-112 Residues in In vivo Folding of E, coli Tryptophan Synthase $\alpha$ Subunit (트립토판 중합료소 $\alpha$ 소단위체의 대장균내 구조형성과정에서의 Ser-33과 Asp-112 잔기의 역할)

  • In the present report, a p[ossibility of the interaction fo Ser-33 and Asp-112 residues in folding of tryptophan synthase $\alpha$ subunit was explored by examining the effect of single or double substitution of these residues on folding of $\alpha$ subunit in E. coli. $\alpha$ subunit of which Ser-33 was substituted with Leu (SL33) was accumulated as insoluble aggregate form, when overproduced in E. coli, whereas $\alpha$ subunit of which Asp-112 was replaced by Asn (DN112) or Gly (DG112) was accumulated as soluble form to the similar extent as wild type $\alpha$ subunit was. When these alterations were combined into one protein, the synergistic effect of residues 33 and 112 on the amount of aggregate form was shown. The amount of doubly altered SL33/DG112 $\alpha$ subunit as aggregate form was increased 5-13 fold that of SL33 $\alpha$ subunit, and the amount of SL33/DG112 $\alpha$ subunit as aggregate form was decreased 3-4 fold that of SL33 $\alpha$ subunit. Aggregates are derived from the specific association of partially folded or unassembled subunits in the folding process. Therefore, this result suggests that residues 33 and 112 of $\alpha$ subunit may unteract during the folding of this enzyme in E. coli.

  • In this study, earlier analyzed tetrapod, one of an original group of offshore constructions studied for manufacturability of the concrete using the properties of steel making slag aggregate. steel making slag aggregate assessment, RCS and Blast Furnace Slag : the 20 mm air-cooled slag and combinations by 50 %, aggregate properties on the most appropriate for the properties of recycled aggregate concrete optimal mix, and assessing it. Properties of concrete used to be derived are judged as to bury the studies show that the hollowing-out of the RCS, plastic sole use is in the workability of the aggregate, plastic in the 20 mm slag also assessed to be a slight disadvantage, but RCS by mixing air-cooled coarse and 50 percent to 20 percent 50 mm. Thus, steel making slag marine structures using recycled aggregate, in rapid chilled slag or air-cooled slag. The sole use of the aggregate them than to combine the aggregate of concrete. After they satisfy the quality standard quality shall be used will aggregate steel making slag who meet the criteria concrete manufacturing in general or par with the aggregate of concrete. Performance was assessed as to develop a more than that.

Antioxidative Activity and Flavonol Glycosides Analysis in Callus Derived from Leaf Tissue of Ginkgo biloba L. (은행(Ginkgo biloba L.)의 잎 유래 캘러스의 항산화능력 및 플로보놀 배당체 검정)

  • This study was carried out to establish an in vitro culture method of callus having a high antioxidant activity from Ginkgo biloba L. Leaf explants were cultured on Murashige and Skoog's medium supplemented with various growth regulators. The explants were incubated in the dark or 3,000 lux cool-white light. Methanol extracts from incubated callus were evaluated for scavenging activity of the free radicals using DPPH. The best callus growth rate was achieved in MS medium combined with 10 ${\mu}M$ NAA and 5 ${\mu}M$ kinetin in the light condition. Total antioxidant activity of cell aggregates in suspension culture [MS medium supplemented with 10 ${\mu}M$ NAA in the light] was up to 80% of ascorbic acid. By means of HPLC analysis, quantification of the quercetin dehydrate and keamperol profiles from suspension callus was compared. Contents of quercetin dehydrate and keamperol from leaf extracts were 0.07 and 2.24 ${\mu}g/20{\mu}l$, and those from callus 0.56 and 0.18 ${\mu}g/20{\mu}l$, respectively.
