In this study, we evaluated the stability of amoxicillin (AMX) and clavulanic acid (CLA) in aqueous solution, and compared the stability of AMX and CLA in commercial combination products. In aqueous solutions, the degradation of AMX ($t_{90}=8.88\;day\;at\;10^{circ}C$) and CLA ($t_{90}=3.53\;day\;at\;10^{circ}C$) occurred rapidly. From the pH-rate profile, AMX and CLA were the most stable at the range of pH 5.5 and 6.0. After reconstitution of commercial dry syrups, the contents of AMX and CLA in suspensions were gradually decreased for 7 days. However, AMX and CLA in dispersible tablet were not changed at all. The contents of CLA in the dispersible tablet ($87.92\%$) and dry syrups (2.16 and $3.91\%$) were remained in the accelerated stability test ($75\%\;RH,\;at\;40^{circ}C$) after 10 hours. And the colors of the dry syrups were rapidly changed from white to yellow. From these results, we concluded that the dispersible tablet could overcome the stability problems of dry syrups.
본 연구의 목적은 배뇨장애 치료를 위한 실로도신과 솔리페나신 숙신산염을 함유한 새로운 복합 정제를 개발하는 것이다. 이러한 목표를 달성하기 위하여 실로도신과 솔리페나신 숙신산염의 동시 정량법을 확립하였다. 두 약물은 다양한 완충액에서 1 mg/ml 이상의 수용해도 값을 나타내었으며, 실로도신과 솔리페나신 숙신산염이 함유된 시판제품의 용출은 다양한 용출조건에서 30분 이내에 완료되었다. 시차 주사 열량계를 사용하여 부형제와 약물 간 상호 반응성을 확인하여 선정된, 약물과 반응성이 없는 부형제를 사용하여 습식 과립화 방법을 사용하여 결합제와 붕해제의 사용에 따른 다양한 처방을 제조한 후 용출시험을 진행하였다. 제조한 정제의 처방 중 실로도신, 솔리페나신 숙신산염, 유당, 미결정셀룰로오스 PH101, 소듐라우릴설페이트, 포비돈 K-30, 크로스포비돈 및 스테아린산마그네슘이 8/10/56/112/2/6/6/2(w/w)의 비율로 제조된 정제는 트루패스정(실로도신 시판 제품) 및 베시케어정(솔리페나신 숙신산염 시판 제품)과의 비교용출 시험시 동등성을 나타내는 것을 확인하였다. 따라서, 본 연구를 통해 얻어진 복합 정제는 각 약물의 시판 제품과 동등한 생체이용률을 나타낼 수 있을 것으로 판단되며, 향후 배뇨 장애 치료에 복약순응도가 우수한 의약품으로 활용될 수 있을 것으로 사료된다.
Objective: This study was conducted to compare the adherence, clinical and economical utility of fixed-dose combination tablets of sitagliptin/metformin with concomitant administration of sitagliptin and metformin in patients with type 2 diabetes mellitus. Methods: Adherence was measured as the medication possession ratio (MPR) of ${\geq}80%$, and MPR was calculated as the number of total prescription days divided by the total treatment period. Hemoglobin $A_{1C}$ ($HbA_{1c}$) differences between baseline and predetermined periods were analyzed. Proportions of patients who achieved $HbA_{1c}$ less than 6.5% for three or more consecutive times were compared. To evaluate cost-effectiveness, prices of sitagliptin, metformin and sitagliptin/metformin tablets were investigated. Results: More than 90% of patients showed adherence in both groups (92.0% in fixed-dose combination group vs 95.9% in concomitant administration group), and there was no statistically significant difference (P = 0.113). Proportion of patients with HbA1c less than 6.5% for three or more consecutive times tended to be somewhat higher in fixed dose combination group than in concomitant administration group without a statistically significant difference (32.6% vs. 28.0%, P = 0.344). Total price of metformin and sitagliptin was cheaper up to 222 KRW in the case of fixed-dose combination tablets compared to the case of concomitant administration. Conclusion: The sitagliptin/metformin fixed-dose combination tablet had a similar patient adherence and was not significantly different in efficacy to the concomitant administration of each component. In terms of drug prices, fixed-dose combination tablets were cheaper than concomitant administration of each tablet.
A novel polymeric tablet of tinidazole (TD) was formulated to treat Helicobacter pylori and Giardia lambria more efficiently with reduced hepatotoxicity by controlling the release of TD after oral administration. TD tablets containing various concentrations of either xanthan gum (XG, viscosity enhancer) and/or polycarbophil (PC, mucoadhesive) were prepared by the wet granulation method. In vitro release of TD into pH 2.0 and pH 5.0 buffer solutions was observed at 37°C by using an USP dissolution tester and an UV (313 nm) spectrophotometer. In vivo absorption of TD tablets was investigated in rabbits by measuring the blood concentration of TD after oral administration using a HPLC. Compared to a commercial TD tablet, in vitro release of TD in both pH 2.0 and pH 5.0 buffer solutions significantly decreased as the concentration: of XG or PC in the tablet increased up to 30%. However, when XG and PC was added in combination, TD was completely released in a pH 5.0 buffer solution within 8 hours, whereas the release of TD in pH 2.0 buffer solution significantly decreased. TD in a commercial tablet was rapidly absorbed after oral administration in rabbits. After oral administration of the polymeric tablets that contain both XG and PC, plasma concentration of TD dramatically decreased. Since the oral absorption of TD significantly decreased by the addition of XG and PC in the tablets while TD completely released in a pH 5.0 buffer solution, it was speculated that more TD was retained in the gastrointestinal tract. Thus, it was possible to control the release of TD by changing the content of XG and/or PC in the tablet, thereby manipulating the release rate and the gastrointestinal retention of TD after oral administration in rabbits.
본 연구에서는 VDT(Visual Display Terminals) 환경에서 배경색과 가독성의 상관관계를 알아보았다. 본 실험에 앞서 예비실험으로 색 온도가 다른 White 지각도를 알아보았으며, 실험 자극물은 태블릿 PC를 활용하였다. 실험 환경은 주광과 야외광이 함께 공존할 수 있는 창가에서 실험 자극물을 제시하였다. White 지각도의 인식과 선호 결과 중 단파장계열의 Nearly Whites 색 에서 백색에 대한 높은 선호도와 인식도를 보였다. 이러한 예비실험 결과를 바탕으로 본 연구에서는 동일한 실험 환경과 실험 기기를 가지고 빨간색, 초록색, 파란색, 노란색의 총 4가지 Nearly Whites와 한 가지 백색을 배경색으로 사용하여 배색 평가, 가독성, 피로도를 조사하였다. 또한, 글자는 검은색으로 제한하였다. 그 결과, 초록색의 Nearly White 배경색에서 가독성과 배색 평가가 높게 나타났으며, 가독 시 느끼는 피로도는 가장 낮은 것으로 나타났다. 이와 반대로 빨간색의 Nearly White 배경색에서는 가독성과 배색 평가가 가장 낮게 나타났으며, 가독 시 느끼는 피로도는 가장 높은 것으로 나타났다. 이러한 연구 결과를 통해서 VDT 환경에서의 가독성을 향상시키며, 눈의 피로도를 낮출 수 있는 배경색을 제안해 보고자한다.
The implementation of NIR and chemometrics in the Pharmaceutical industries is still in strong progress, both regarding qualitative and quantitative applications and beneficial results are seen. Looking at the development so far, NIR will change the pharmaceutical industry even more in the future. This presentation will address the experiences and progress achieved regarding the application and implementation of quantitative methods for determination of content uniformity and assay of tablets with less than 10% w/w of active, using Near Infrared transmittance spectroscopy in combination with PLS. Also qualitative methods for identification of the same tablets by Near Infrared reflectance spectroscopy will be discussed. Four commercial tablet strengths are formulated and produced from two different compositions by direct compression. Three different strengths are dose proportional, i.e. fixed concentration by varying in size. The aim was to replace the conventional primary methods for analysing content uniformity, assay and identification by NIR. Studies were performed on comparing transmittance versus reflectance spectroscopy for both applications on the dose proportional tablets. The model for determination of content uniformity and assay was developed to cover both coated and uncoated tablets, whereas the qualitative model was developed to identify coated tablets only. The impact of the tablet formulation, tablet size and coating, resulted in individual models far each composition The best calibration was achieved using diffuse reflectance for the identification purposes and diffuse transmittance for the quantitative determination of the active content within the tablets. As NIR in combination with other techniques opens up the possibility of total quality management within the production, the transfer of the above-mentioned models from a laboratory based approach to an at-line approach at H.Lundbeck will be addressed too.
An oral adhesive tablet of acyclorir [9-(2-hydroxyethoxymethyl) guanine] for herpetic stomatitis was prepared and its physical properties were evaluated. 300 mg weighed tablets containing 30 mg of acyclovir were prepared with six kinds of polymers from direct compression, and the stickiness, fracture resistance and dissolution in pH 6.8 buffer solution were tested. HPMC and MC showed good stickiness and fracture resistance, and their dissolution rates were significantly different from each other. Three factors-HPMC:MC ratio, acyclovir content, compression force-were chosen as an important factor of manufacture and factorial analyses for these three factors were carried out. Eight kinds of formulations from different combination of three factors were prepared and tested in stickiness, fracture resistance and dissolution in pH 6.8 buffer solution. Dissolution rate was significantly affected by polymer ratio, fracture resistance was affected by compression force, and stickiness was not significantly affected by acyclovir content and polymer ratio.
비 스테로이드성 항염증제에 대한 개별적 독성증상을 예방하기 위해 로박스 플라티넘(Robax Platinum), 로박스아세트(Robaxacet), 나프록센(Naproxen) 그리고 재활치료를 선택적으로 처방하여 비 스테로이드 진통소염제(Non-Steroidal Anti-Inflammatory Drugs)에 대한 독성증상을 예방을 위한 효과 비교 연구를 진행하였다. 이 연구에서는 로박스 플라티넘(Robax platinum), 로박스아세트(Robaxacet), 나프록센(Naproxen)을 주한미군 처방전(Annex A-Over-The-Counter Prescription)을 기준으로 하여 처방하였고, 앨리스 리치의 통증 척도(Alice Rich's Pain Scale)를 사용하여 통증 경과를 비교하고 그 결과를 IBM SPSS statics 24 버전을 사용하여 데이를 계산했다. 결합된 메토카바몰(Methocarbamol, 500 mg), 아세트아미노펜(Acetaminophen, 325 mg), 이부프로펜(Ibuprofen, 200 mg) 정제는 결합된 메토카바몰(Methocarbamol, 500 mg) 정제와 함께 사용할 수 있다. 아세트아미노펜 (Acetaminophen, 325 mg)의 알약이 통증을 조절하는 방법으로 사용되었다. 메도카바몰 500mg, 아세트아미토펜 325 mg 및 이부프로펜 200 mg 정제의 약물 조합은 메도카바몰 500 mg 및 아세트아미노펜 325 mg 정제가 전반적인 통증 조절과 관련하여 물리적인 스트레칭 운동과 쌍을 이룬 것과 유사한 통증 완화효과를 나타내었다.
Advances in the Internet and network technology and the rapidly growing number of personal devices result in the fast growth of E-Commerce, M-Commerce. Nowadays, location-based services are widely used in E-commerce. A location-based service (LBS) is a service based on the geographical positions of mobile handheld devices such as smart phones or tablet computers. In this paper I will view the combination of the E-commerce and LBS.
본 연구는 carvedilol과 ivabradine의 이층정 제형 개발에 있어 carvedilol 속방층의 제형조건을 최적화하기 위해 수행되었다. Full-factorial design of experiments를 사용하여 carvedilol 속방층(wet granulation part)의 첨가제(microcrystalline cellulose, citric acid, crospovidone) 및 타정 시 공정변수(main compression)를 확인하였고 각 독립변수에 대한 7가지 종속변수(함량, 함량 균일성, 경도, 마손도, 붕해, 용출(pH 1.2 및 6.8)를 design expert software를 사용하여 평가하였다. Design of experiments (DoE) 실험에서 도출된 Analysis of Variance (ANOVA) 결과를 통해 main compression은 경도, 마손도, 붕해 시간에 유의한 영향을 주었고, microcrystalline cellulose는 마손도, 용출에 주요한 영향을 주었음을 확인하였다. 또한, citric acid는 마손도, crospovidone은 마손도, 용출에 주요한 영향을 주었음을 확인하였다. DoE실험 결과를 종합하여 최종적으로 도출된 설계공간에 따라 microcrystalline cellulose (약 18.0-32.0 mg), citric acid (약 0.5-12.0 mg), main compression (약 615-837 kgf)의 최적의 범위 내에서 초기 위험평가에서 평가된 모든 위험요인이 제거된 carvedilol 속방층의 최적 제조 범위를 확인하였다.
