This experiment was carried out to investigate the effects of gamma-radiation on the rooting, growth, and color mutation in poinsettia. Using 10 poinsettia varieties ('Lollipop', 'Little Peace', 'Happy Day', 'Early Bird', 'Pixy Red', 'Happy Time', 'Heidi', 'Red Bell', 'Clara', and 'Scarlet') bred by National Institute of Horticultural and Herbal Science, 100 Gy of gamma ray was irradiated at the stage of callused cuttings. Four weeks after sticking cuttings in the rooting media, 8 cultivars showed 100% of root formation, but 'Early Bird' rooted 24.4% and even died off during the cutting propagation. After planting rooted cuttings, survival rate until flowering time varied among irradiated cultivars. While 'Pixy Red' and 'Heidi' survived about 98%, 'Clara', 'Happy Day', and 'Early Bird' survived lesser than 30%. All irradiated plants showed remarkably shorter plant height, lesser branch numbers than non-irradiated control plants. Thirty color mutants were obtained among 281 plants survived until flowering time. Nine mutants were complete color mutated branches, whereas 21 mutants were partially color mutated bracts and transitional leaves. Color patterns mutated by 100 Gy of gamma ray were divided into pink, hot pink, light red and spotted (pink spots with red main color). Pink mutants were commonly obtained. Complete color mutants were discovered from 4 plants of 'Pixy Red', 2 plants of 'Red Bell' and 3 plants of Lollipop.
A full-length cDNA and genomic DNA of a $leucoanthocyanidin$$dioxygenase$ ($DgLDOX$) gene was isolated from the petals of chrysanthemum 'Argus', and comparative features of the gene among three flower color mutants derived from a gamma-ray mutagenesis were characterized. The cDNA coding region of the gene was 1068 bp and was translated into 356 amino acids accordingly. The genomic DNA size was 1346 bp for 'Argus', while three mutants revealed ranges of 1363 to 1374 bp. A single intron between two coding exons for the $DgLDOX$ gene was found, of which size was 112 bp for 'Argus', but 128 or 137 bp for three flower color mutants, indicating that a genomic insertion in the intron occurred during the gamma-ray mutagenesis. DNA blot analysis revealed the $DgLDOX$ gene presenting as a single copy in the chrysanthemum genome. The $DgLDOX$ gene was expressed in both 'Argus' of light-pink color and two purple color mutants (AM1 and AM3) but had very weak expression in only white color mutant (AM2). The results demonstrated that variations in the flower color of the mutants might be associated with changes in the amino acid moieties in the coding exons or fragment insertions in the intron of the $DgLDOX$ gene, which potentially resulted in less expression of the gene in the white colored mutant.
The white-colored and the dark gray-colored mutants were frequently happened in cultivated areas of Pleurotus ostreatus (Wonhyeong-neutari). These caused conflicts between farmers and spawn companies. Our studies were conducted to elucidate the mechanism of mutagenesis. The results from the studies would provide valuable informations that could be used to prevent the color-related mutation, and also will be applied in breeding programs of P. ostreatus. Oyster mushroom variety, Wonhyeong-neutari, is somatic hybrid of Pleurotus and has genetic makers for arginine, ornithine, proline, riboflavine. Genetic markers analysis of monospore isolates derived from color mutants show identical tendency with that of Wonhyeong-neutari, these results indicate that color mutants were derived from Wonhyeongneutari. Twenty-one and four homokaryons were selected from the white-colored mutant MGL 2205 and gray-colored ASI 2029. All 34 F1 hybrids derived from the white-colored mutant MGL 2205 produce white-color fruiting bodies, indicating that the white color trait is heritable. In the first generation hybrids between the white-colored MGL 2205 and the gray-colored ASI 2029, all 16 hybrids produced pigmented fruiting bodies. Homokaryons isolated from the hybrid MGL 2205 X ASI 2029 were mated with homokaryon tester strains derived from MGL 2205. By these result, we could assumed that white color trait is a heritable character which is controlled by more than one recessive gene.
This study was carried out to compare the pattern of mutant variation and to evaluate the characteristics of mutants obtained by gamma irradiation in rose 'Kardinal'. Forty four rooted cuttings of 'Kardinal' were irradiated at 70 Gy gamma-ray dose from a $^{60}Co$ source to induce mutants in 2002. The irradiated plants were planted in field, and observed spotting of petal color mutants from 2002 to 2004. Four different kinds of mutant twigs with each different color flower were obtained from the irradiated 'Kardinal' with red petal. After being identified to be a stable mutant from 2004 to 2008, each mutant line propagated by cutting was hydroponic-cultured to evaluate the characteristics in the greenhouse from 2008 to 2009. Four mutant lines obtained from 'Kardinal' with red petal (Red group, 44A, 45B) include KA1 with light pink petal (Red group, 55B-55D), KA2 with pink petal (Red group, 63A-63B), KA3 with deep pink (Red purple, N57A-N57C), and KA4 with orange red (Red group, 43A-43B). Diameters of each flower in four mutant lines were different from 'Kardinal'. The line KA1 was 9.5 cm wide, and it showed the smallest diameter when compared to other mutants. While the line KA2 was the largest one with 12.5 cm 'Kardinal'. Petal number per flower was also variable among the mutants. The line KA2 had 39.8 petals being the largest number among the mutants, while the line KA1 was the lowest one compared to 35.5 petals of 'Kardinal'. Petal color was measured by using colorimeter. Brightness (L) measured at each petal of four mutants increased more than 'Kardinal'. CIE Lab values, a and b decreased more than 'Kardinal' at the petal color of three mutants except the line KA4. Characteristics of shoot, leaf, etc. from four mutants were also different from the ones of 'Kardinal'. The line KA1 was shortest in shoot, node and peduncle length, and lowest in prickle number. The reverse side of leaves was reddish green color in 'Kardinal' as well as the line KA4, but green color in the line KA1, KA2, and KA3.

Although innate color preference of motile organisms may provide clues to behavioral biases, it has remained a longstanding question. In this study, we investigated innate color preference of zebrafish larvae. A cross maze with different color sleeves around each arm was used for the color preference test (R; red, G; green, B; blue, Y; yellow). The findings showed that 5 dpf zebrafish larvae preferred blue over other colors (B > R > G > Y). To study innate color recognition further, tyrosinase mutants were generated using CRISPR/Cas9 system. As a model for oculocutaneous albinism (OCA) and color vision impairment, tyrosinase mutants demonstrated diminished color sensation, indicated mainly by hypopigmentation of the retinal pigment epithelium (RPE). Due to its relative simplicity and ease, color preference screening using zebrafish larvae is suitable for high-throughput screening applications. This system may potentially be applied to the analysis of drug effects on larval behavior or the detection of sensory deficits in neurological disorder models, such as autism-related disorders, using mutant larvae generated by the CRISPR/Cas9 technique.
The objectives of this study were to isolate and the sequence of novel $F3'H$ gene related to an anthocyanin pathway, and to confirm the expression patterns of the gene involved in the flower color variations of chrysanthemum mutants. In this study, we isolated the full-length cDNAs and the genomic DNAs of an $F3'H$ gene from a wild type (WT) chrysanthemum (cv. Argus) and its three color mutants. The sequence analysis revealed a putative open reading frame of 1,527 bp that encodes a polypeptide of 509 amino acids. Sequence homology ranged from 97% to 99% between 'Argus' and its three color mutants. The sequence analysis from the genomic DNA revealed that the chrysanthemum $DgF3'H$ gene consisted of three exons and two introns spanning a 3,830 bp length. The sizes of the gene for three mutants ranged from a shorter size of 3,828 bp to a longer size of 3,838 bp when compared to the size of WT. The total size of the two introns was 2,157 bp for WT, but those for three color mutants ranged from 2,154 bp to 2,159 bp. A result of an RT-PCR analysis indicated that the color variations of the mutants AM1 and AM2 can be partly explained by the structural modification derived from the sequencial changes in the gene caused by gamma ray. A Southern blot analysis revealed that the $DgF3'H$ gene existing as multiple copies in the chrysanthemum genome. A systemic study will be further needed to provide a genetic mechanism responsible for the color mutation and to uncover any involvement of genetic elements for the expression of the $DgF3'H$ gene for the color variation in chrysanthemum.

Phenotypes of panicle, hull and seed of mutant rice (Oryza sativa L.) were characterized. Panicle mutants were classified in 4 groups with their internode length of main rachis, primary rachis, secondary rachis and pedicel. Hull and seed mutants were grouped into 12 based on their mutant characters in shape, size and color of seeds. These natural and spontaneous mutant collections showed distinct phenotypes to wild type rice. This might be useful for the identification of the functions of genetic factors in the Mendelian inheritance.
To identify genes involved in the decolorization of brilliant green, we isolated random mutants generated by transposon insertion in brilliant green-decolorizing bacterium, Citrobacter sp. The resulting mutant bank yielded 19 mutants with a complete defect in terms of the brilliant green color removing ability. Southern hybridization with a Tn5 fragment as a probe showed a single hybridized band in 7 mutants and these mutants appeared to have insertions at different sites of the chromosome. Tn5-inserted genes were isolated and the DNA sequence flanking Tn5 was determined. By comparing these with a sequence database, putative protein products encoded by bg genes were identified as follows: bg 3 as a LysR-type regulatory protein; bg 11 as a MalG protein in the maltose transport system; bg 14 as an oxidoreductase; and bg 17 as an ABC transporter. The sequences deduced from the three bg genes, bg 2, bg 7 and bg 16, showed no significant similarity to any protein with a known function, suggesting that these three bg genes may encode unidentified proteins responsible for the decolorization of brilliant green.
This study was carried out to establish a system for mutation breeding by irradiation of gamma-ray in $Rosa$$hybrida$ Hort. The rooted cuttings of two roses, 'Spidella' and 'Cabernet' were irradiated with different gamma-ray doses (0, 30, 50, 70, 90, 110, 130, 150 and 170 Gy) from a $^{60}Co$ source to reveal an optimal dose for induction of mutants. The irradiated plants were planted in a greenhouse, and investigated on the appearance of petal color mutants and shoot growth by gamma ray dose. The 50% lethal doses ($LD_{50}$) of plant were 110 Gy for 'Spidella' and 150 Gy for 'Cabernet', respectively. The 50% decrease dose of shoot length was observed at 70-90 Gy dose for 'Spidella', and 110 Gy dose for 'Cabernet'. Solid, chimeric and mosaic petal mutants with various colors were induced from pink petal of 'Spidella' and red petal of 'Cabernet' when 30-170 Gy dose was irradiated. The mutants obtained from 'Spidella' had white, ivory, pinky ivory, light pink and deep pink petal colors. The mutants obtained from 'Cabernet' had pink, deep pink, purple red (magenta), orange red and purple petal colors. It was suitable to irradiate 70-90 Gy dose for 'Spidella' and 90-110 Gy dose for 'Cabernet' for the induction of various mutants considering plant survival rate, shoot growth and mutant occurrence rate.
This study was aimed to select useful mutants of rapeseed (Brassica napus L.) and leaf mustard (Brassica juncea L.), the seeds of three lines S-14, S-27, and S-28 were treated with gamma-ray and EMS. The optimum ranges of gamma-ray dose and EMS concentration to enlarge the characteristic morphological variations were also separately investigated. The survival rates of S-28 only linearly decreased with increasing the gamma-ray dose. The overall growth parameters decreased of gamma-ray dose in all three lines of S-14, S-27, and S-28. The reduction dosage 50 of gamma-ray was identified as 1,200 Gy for S-14 leaf mustard, while those of S-27 and S-28 rapeseed lines were appeared as same 1,000 Gy. The emergence rates of S-14 and S-27 showed no significant differences by EMS treatment, while the growth of all three lines were significantly decreased. The reduction concentration 50 in S-14 could not be determined, demonstrating that this leaf mustard line is presumably insensitive to mutagenic EMS, while those in S-27 and S-28 were identified as 3.0 and 2.5%, respectively, showing that these rapeseed lines possess higher sensitivity to EMS than S-14. Various morphological characteristics of $M_1$ generation obtained from mutagen treatment were elaborately investigated for further maintenance of $M_2$ generation. In $M_2$ generation variants showing short stem, yellow color in seed coat, chlorophyll deficiencies in leaf or pod, abnormal flower color were selected as potentially useful mutants for breeding.
