A Study on Airborne Microbes Contamination that Influences on Nosocomial Infection (병원내 감염에 영향을 미치는 공기오염 상태에 대한 조사 연구)

  • Nosocomial infection is important in the management of the nursing care and has been found to be related with the airborne microbes contamination. The purpose of this study is to identify the differences of the airborne microbes between hospitals, nursing units, days and times and to identify the difference in the types of microorganisms between hospitals, nursing units, days and times. This study was conducted from May 25, to July 10, 1981, according to R. Koch's methods for quantative samplings of airborne microbes and the results of this study were reviewed in a statistical method. The following conclusions were obtained: 1. There was a significant difference in the types of airborne microbes between 8 hospitals (F=5.0491, p<0.01) 2. There was a significant difference in the types of airborne microbes between surgical, medical, nursery, 1.C.U., operating theatre and outpatient nursing unit. (F=2.1764, p<0.05) 3. There was not a significant difference in the types of airborne microbes between Monday, Thursday and Saturday (F= 1.6365, p>0.05) 4. There was a significant difference in the types of airborne microbes between AM 7 : 00, AM 11:00 and PM 3 : 00. (F=7.2951, p<0.01) 5. 4061 colonies were divided into more 48 types and the classification was as follows. gram positive cocci (2024 colonies) 49.3% gram positive bacillus (1211 colonies) 29.8% gram negative bacillus (577 colonies) 14.2% fungus(200colonies) 4.9% gram negative cocci (41 colonies) 1.0% other's(8 colonies) 0.3% 6. There was a significant difference of airborne microbes between 8 hospitals. (F=7.7943, P<0.01) 7. There was not a significant difference of airborne microbes between 6 nursing units. (F= 1.8461, P>0.05) outpatient nursing unit : 12.6 colonies surgical nursing unit. 10.4 colonies Medical nursing unit : 9.8 colonies nursery nursing unit : 9.4 colonies operating theatre nursing unit:8.2 colonies Intensive care nursing unit : 7.8 colonies 8. There was not a significant difference of airborne microbes between Monday, Thursday and Saturday. (F=1.4371, p>0.05). Saturday : 11.0 colonies Monday : 9.1 colonies Thursday : 9.1 colonies 9. There was a significant difference of airborne microbes between A. M. 7 ; 00, A. M. 11 : 00 and P .M. 3:00 (F=7.6658, p<0.05) P. M. 3 : 00 : 12.2 colonies A. M. 11 : 00 : 10.0 colonies A. M. 7 : 00 : 7.1 colonies.

Studies on Nosema Disease of Honey Bees 1. Isolation of Nosema apis and a Survey of Nosema Disease in Honey Bees (꿀벌의 Nosema 병(病)에 관한 연구(硏究) 1. 원인체분리(原因體分離) 및 감염실태조사(感染實態調査))

  • The experiments were conducted to isolate the etiogical agent and to survey the distribution of Nosema disease in honey bees. The results obtained were as follows: 1. The etiological agent of the so-called "crawling disease" in honey bees characterized by the symptoms of crawling, diarrhea, and enteritis etc. was first isolated and identified with Nosema apis (Zander 1909) in Korea. 2. 455 colonies were randomely sampled and surveyed in 4,766 bee colonies out of 56 apiaries and 51 colonies (11.2%) out of 455 bee colonies were infected with N, apis. 3. Infection rates according to the period of honeyflow as follows: Brassica napus (Apr.): 25/130 colonies (18.4%) Rohinia pseudoacacia (May) : 8/55 colonies (14.%) Trifolium repels(Jun.): 15/99 colonies (13.6%) Castanea crenate (Jul.): 3/46 colonies (6.5%) Lespedeza bicolor(Aug.): 0/60 colonies (-) Fagopyrumesculentum(Sept.) & Perilla frutescens(Oct.) 0/65 colonies (-) 4. The typical clinical signs of Nosema disease were appeared on loth day after N. apis was orally administered with the level of $16{\times}10^4$ spores/ml to the healthy adult bees. Spores could be harvested with the level of $121{\sim}236{\times}10^4$ spores/ml on 10th day and $392{\sim}429{\times}10^4$ spores/ml on 15 days after infection. 5. In adult honey bees infected with N. apis artificially the 50% lethal day of life-span was 9 to 10 days and 100% lethal day was 16 to 19 days. However, in the control 50% lethal day was 19 to 23 days and 100% lethal day was 31 to 33 days.

  • 1. Of the 35 various grains, 446 colonies of fungi were isolated. Among the 446 colonies, 423 were possible to identify into 17 genera. 2. Of the 10 bean-pastes, fungi were isolated 193 colonies. It can be identified 11 genera. 3. It will be noted that Aspergillus sp., Penicillium sp., Mucor sp., Cladosporium sp. were predominated genera. 4. A total of 155 colonies of Aspergillus sp. isolated from various grains were obtained from the samples. Asp. versicolor was the frequent species and 47 colonies isolated. 5. Penicillium where was isolated from various grains, obtained 115 colonies. Pen. camemberti was the most frequent species. 6. Penicillium palitans was most excellent amylase producing fungi and which was 1ml per 6.300 unit.

Image Analysis of Typhoon Impacts on Soft Coral Community at Munseom in Jeju, Korea (제주도 문섬 조하대에 서식하는 연산호군락의 태풍에 의한 영향 분석)

  • Impacts of Typhoon Maemi on a soft-coral community located on subtidal cliff at Munseom, Jeju were investigated in this study using underwater photography. Typhoon Maemi hit Jeju Island in late September 2003 and its impact was strong enough to destruct most shallow water sessile benthos including soft corals. To estimate numbers and size of soft-coral colonies, a line transect was installed on the cliff at depth from 3 to 9 m and photographs were taken serially by every 1m. From each $1{\times}1m$ underwater photograph, species and size of soft-coral colony was determined. Number of soft-coral colony and its Percent coverage (PC) in each $1m^2$ quadrat was calculated. Soft corals Scleronephthya gracillium, Dendronephthya gigantea, D. spinulosa and D. castanea were identified from the photographs. Dendronephthya sp. was mainly distributed at 3-6m while S. gracillimum was mostly occurred at $6{sim}9m$. A survey conducted before the typhoon showed that number of the soft-coral colonies at $3{\sim}4m,\;4{\sim}5m,\;5{\sim}6m,\;6{\sim}7m,\;7{\sim}8m\;and\;8{\sim}9m$ was 17, 24, 20, 23, 18 and 30 $colonies/m^2$ or 21, 48, 36, 28, 24 and 43%, respectively. After the typhoon, number of soft-coral colonies in the transect increased, 31, 35, 21, 10, 21 and 50 $colonies/m^2$ while PC was remarkably decreased as 21, 23, 21, 5, 9 and 13%, respectively. Our data suggested that the impact was limited in larger colonies; larger soft coral colonies were selectively destroyed and removed while the small colonies underneath the larger colonies remained undestroyed.

On Phylogenetic Relationships Among Native Goat Populations Along the Middle and Lower Yellow River Valley

  • This paper is based on the 9 goat colonies along the middle and lower Yellow River valley and 7 local goat colonies in the Northeast, Tibet and the Yangtze valley. After collecting the same data about the 22 goat colonies in China and other countries, it establishes and composes the matrix of fuzzy similarity relation describing the genetic similarities of different colonies. It also clusters 38 colonies according to their phylogenetic relationship. The establishment of the matrix and the cluster are effected in terms of the frequency of 18 loci and 43 allelomorphs in blood enzyme and other protein variations. The study proves that the middle Yellow River valley is one of the taming and disseminating centers of domestic goats in the South and East of Central Asia. Compared with other goat populations in this vast area, the native goat populations in the west of Mongolian Plateau, the Qinghai-Tibet Plateau and the middle Yellow River valley share the same origin. The colonies in the lower Yellow River valley and those in the middle valley, however, are relatively remote in their phylogenetic relationship. The native goat colonies in the southeast of Central Asia can be classified into two genetic groups: "East Asia" and "South Asia" and the colonies in Southeast Asia belong to either group.

Trait-based algal community assembly associated with Pectinatella magnifica (Bryozoa, Phylactolaemata)

  • Habitat-forming species increase spatial complexity and alter local environmental conditions, often facilitating the assembly of plants and animals. We conducted a trait-based approach to algal assemblages associated with the freshwater bryozoan, Pectinatella magnifica. Association with algae leads to the inner bodies of the bryozoans being colored green; this is frequently observed in the large rivers of South Korea. We collected the green-colored gelatinous matrices and phytoplankton from waterbodies of the two main rivers in South Korea. Algal assemblages within the colonies and in the waterbodies were compared using the three diversity indices (richness, diversity, and dominance), and the composition of functional groups (FGs) and morphologically based functional groups (MBFGs) between the colonies within and outside of P. magnifica colonies. The most dominant and common species within the colonies were Oscillatoria kawamurae and Pseudanabaena catenata, both of which were assigned to the same FG (codon S1). Of the algal assemblages within the colonies, the dominance was higher, while the richness and diversity were lower, than those in the waterbodies. There was variation in the compositions of FGs and MBFGs in the waterbodies outside the colonies. Total nitrogen and orthophosphate led to dominance, and were significant factors for the variation in FGs in the waterbodies, whereas there were no such significant factors within the colonies. This trait-based approach to the community structure of associated algae provides the status and habitat gradient of these communities, which are stable, isolated, and consistent with the overgrowth of shade-adapted tychoplanktonic cyanobacteria.

Indoor-wintering for the honeybee colonies of Aips mellifera in Yeongju area (영주지방에 있어서 봉군의 실내월동 시험)

  • The studies was conducted to establish the indoor overwintering methods for productive honeybee colonies in the wintering chamber which provides satisfactory imide temperature ranging $2{\sim}9^{\circ}C$ and ventilation for the wintering hooeybee colonies in Korea. Mortality of the indoor wintered hooeybee colonies was 6.3~7.1% during the winter season of 1997~1998 and 5~10% during the winter season of 1998~1999. The decreased rates of colony weights dwing the wintering season were 10.6~10.7% in the strong colonies and 10.2~11.7% in the weak colonies. The increased rates of colony population overwintered in the chamber were 136.1~142.3% in the strong colonies and 128~136.5% in the weak colonies.

Effects of Kimchi Extract on the Development of Multicellular Structures from Rat Mammary Organoids Cultured in Matrigel

  • The effect of methanol soluble fraction(MSF) of kimchi on the proliferating and differentiating activity of normal rat mammary epithelial cells or organoids in culture were studied. Reconstituted basement membrane, Matrigel, supported the growth and development several different multicellular structures from mammary organoids. The five type colonies of multicellular structures, stellate, ductal, webbed, squamous, and lobulo-ductal colonies, were observed in Matrigel culture. In methanol extract groups, webbed colonies were more and squamous colonies were less than control group. and the lobulo-ductal colonies which is known that it formed in well differentiated mammary epoithelial cells were developed more in MSF treated group than control group. These results showed that methanol extract of kimchi affected on the proliferation and differentiation of normal rat mammary epithelial cells cultured in serum free medium condition.

Effects of Natural Selection, Mutagenesis, and Protoplast Formation and Cell Wall Regeneration on the Production of Aminoglycoside Antibiotics

  • High producers or blocked mutants of aminoglycoside antibiotic-producing Streptomyces spp. were selected by application of an agar plug method and by culturing individual colonies in broth. The productivities of aminoglycoside antibiotic producing organisms were increased by selection of a high producer from colonies obtained by spreading spores of wild strain, or survived from treatment of a mutagen or from the colonies regenerated from protoplast-formation and cell-wall regenerations. Some mutagen treated colonies lost the ability to produce antibiotics (5-8%). Some A-factor negative and deostreptamine or streptidine negative mutants were obtained by N-methyl-N'-nitro-N-nitrosomethylguanidine (MNNG) treatment. Many of the survivors from the MNNG treatment lost the ability to produce antibiotics. Major colonies produced less amount of antibiotics ; only few survived colonies produced more antibiotics than the parent. Resistance of Streptomyces spp. against the antibiotics produced by itself was also markedly affected by mutagen treatment.

Development of an Approach for Analysing Vegetation Community Mosaic Using Landscape Metrics (경관지수를 활용한 식생군락 모자이크화 분석법)

  • Whereas the demand for development of forested areas covering more than 60% of Korean territory, permission on the forest development has been still given from the perspective of effective land utilization rather than conservation. As the assessment of large forested areas usually focuses more on forest structure, it has its limitation of observing and analyzing the interior change in forest in this way. This study was aimed at computing landscape metrics using a presence vegetation map and FRAGTSTATS 4.2 and analyzing vegetation mosaics. Colonies in native vegetation were classified into a series of major groups and sub-groups based on the native species within the colonies. The colonies were investigated by analyzing a suite of landscape metrics - Core Area, Percentage of Landscape, Number of Patches, Patch Density, Largest Patch Index, Total Edge, Edge Density, Landscape Shape Index, Mean Patch Area, Euclidean Nearest Neighbor. In the Chungnam province major groups and sub-groups of colonies classified based on the proportion of pine and oak species, and pine species was the principal one in terms of distribution area. As for the competition between pines and oaks, while the coverage of pine-centered colonies were three times larger than those of oak-centered ones, pine colonies showed the greater number of patches and therefore higher fragmentation than oaks at the major group level. For the sub-groups, the largest coverage colonies were not only indicated by Pinus densiflora-Quesrcus mongolica colonies among P. densiflora-centered colonies, Q. accutissima colonies among Q. accutissima-centered ones, Q. accutissima-P. densiflora colonies among Q. accutissima-centered ones, Q. mongolica colonies among Q. mongolica-centered ones, P. thumbergii colonies among P. thumbergii-centered ones, and Q. serrata-Q. acutissima colonies among Q. serrata-centered ones, but also revealed more severely mosaicked than other smaller colonies. The overall mosaicking degree estimated by landscape metrics was considered useful for monitoring and investigating vegetation. However, in order to develop management strategy based on analyzing the reason for the mosaicking process and anticipating a trend in vegetation succession, it is essential to further study about ecological characteristics of each colony in the vegetation.
