The effect of Picrorrhiza Rhizoma (PR) aqueous extracts were evaluated on 2,4-dinitrofluorobenzene (DNFB)-induced contact dermatitis, type I allergic model. Contact dermatitis was induced by sensitization with dinitrophenyl-derivatized ovalbumin (DNP-OVA) and DNFB challenge as antigen. Three different concentrations of PR extracts (300,150 and 75mg/kg) were orally administered to DNP-OVA sensitization mice once a day for 7 days with reference materials; dexamethasone (15mg/kg, intraperitoneal treatment). End of 7 days oral administration of PR extracts or intraperitoneal treatment of dexamethasone, the changes on the edematous changes and scratching behavior were measured. Immediate after DNFB challenge on ear or paw of DNP-OVA sensitized mice, increases of ear and paw thicknesses and weights were detected with anterior ear skin (dermis to epidermis) thickness and paw scratching behavior increases. However, these DNFB-induced increases on ear and paw thicknesses, weights and scratching behaviors were decreased by treatment of all three different dosages of PR extracts and dexamethasone, respectively. In addition, the increases of anterior skin thicknesses were also dramatically inhibited by treatment of all three different dosages of PR extracts and dexamethasone at histopathological observations. The results obtained in this study suggest that oral treatment of PR extracts also has relatively favorable effects on allergic dermatitis.
To evaluate the antitumor activity of Paridis Rhizoma(PR), studies were done experimentally. The results were obtained as follows: 1. In cytotoxicity against MCF-7, SK-OV-3, HCT15, concentration inhibiting cell growth up to below 50% of control was recognized at $100-200{\mu}g/m{\ell}$ of PR and also against A549, XF498 was recognized at $50-100{\mu}g/m{\ell}$. 2. In Inhibitory effect on activity of DNA topoisomerase I, the $IC_{50}$ was shown $50-100{\mu}g/m{\ell}$ of PR. 3. The concentration inhibiting adhesion of A549 and SK-OV-3 to complex extracellular matrix up to below 50% of control was recognized at $10-100{\mu}g/m{\ell}$ of PR. 4. The T/C% was 137.9 in PR-treated group in S-180 bearing ICR mice. From above results it was concluded that PR could be usefully applied for the prevention and treatment of cancer.
This study examined Korean Public Relations trend and social acceptance in 1960s. It centers on the acceptance conditions of PR concept, corporate PR and governmental PR. It analyzed PR recognition and practical activities of the general corporations, financial institutions and public corporations in dealing with corporate PR. It also analyzed Korean governmental PR through examining the structure of governmental PR system, PR activities in rural communities and the police. In those days, PR concept had been misused to no small degree and not been accepted correctly that the necessity to let people know PR in its proper meaning was raised. Laying emphasis upon the technical side, corporate PR activities had centered on publicity. Especially, at that time, the banks, the insurance companies and the public corporations had perceived the importance of PR and implemented a set of PR activities. It can be said that a special feature in government PR in 1960s lies in the expansion of the public information system and reinforcement of PR policies. Also, PR in rural communities had been doing to give information relating to agriculture technique and to improve the living conditions of the farm area. The importance of PR in police had been on the side of promoting relations with people and improving treatment of the people by policeman.
Objectives: The aim of this study was to estimate the single oral toxicity of Peucedani Radix (PR) ethanol extracts. PR is one of the important herbs for removal of phlegm, the viscous turbid pathological product that can accumulate in the body and cause a variety of diseases. However, research on the pharmacologic toxicity of PR is lacking. Methods: In this study, PR was orally administered to 5-week-old male ICR mice at an oral dose of 2,000, 3,000, or 5,000 mg/kg. After a single-dose administration, the mortality and behavioral changes were observed daily and body weights were measured every two days. After 14 days, the organ weight, organ index, macroscopy, hematological analysis, and serum biochemistry analysis were determined. Results: No mortality, body weight changes, abnormal behavioral changes, or anatomical signs of toxicity were found. The organ weight, organ index, hematological analysis, and serum biochemistry analysis were also within the normal ranges. Conclusions: These results suggest that the 50% lethal dose of PR is more than 5,000 mg/kg. This could indicate that PR is a safe drug without acute toxicity and side effects.
A normal prion protein (PrPc) is converted to a protease resistant isoform (PrPsc) by an apparent self-propagating activity in bovine spongiform encephalopathies (BSE), which is a neurodegenerative disease. The cDNA encoding bovine PrP open reading frame (ORP) in Korean cattle was cloned by polymerase chain reaction (PCR). The cloned cDNA had a length of 795 base pairs which coded for a protein of 264 amino acid residues with a calculated molecular mass of 28.6 kDa. Identities of 90, 90, 79 and 78% on nucleotide and 94, 94, 84, and 84% on amino acid sequence were shown to PrP genes from sheep, goat, human, and mouse, respectively. The cloned DNA was ligated into the pQE30 expression vector and transformed into E. coli M15. The PrP was expressed by induction with isopropyl-$\beta$-D-thiogalactoside (IPTG) and purified on the Ni-NTA affinity column. High specific activities of the recombinant PrP were observed in the fraction of pH 5.8 eluate and showed a molecular mass of-29 kDa on SDS-PAGE and Western blot analysis.
Objectives : Polygalae Radix (PR) from Polygalae tenuifolia (Polygalaceae) has been clinically used as a sedative, anti-inflammatory, and anti-bacterial agent. To extend pharmacological effects of PR in the central nervous system (CNS) on the basis of its CNS protective effect, the present study was conducted to identify the effect of PR, whether it shows the neuroprotective action against excitatory neurotoxicity. Methods : To identify the protective effect of PR to excitatory neuro-toxic agent, the present study was focused on the PR effect on cell death, that was caused by applying NMDA to nerve cell, elevation of $(Ca^{2+})_i$, releasement of glutamate, and ROS generation. Result : 1. PR methanol extract, at the concentration range of 0.05 to 5 g/ml, significantly inhibited NMDA (1 mM)-induced neuronal cell death as well as MK-801 (non competitive NMDA antagonist). 2. PR methanol extract $(0.5\;{\mu}g/ml)$ inhibited NMDA (1 mM)-induced elevation of cytosolic calcium concentration $[Ca^{2+}]_i$. NMDA application in the presence of MK-801 $(10\;{\mu}M)$ failed to produce the increase of $[Ca^{2+}]_i$ through all the measurement time. 3. PR methanol extract $(0.5\;{\mu}g/ml)$ inhibited the NMDA-induced elevation of glutamate release. Also, MK-801 showed similar protective effects. 4. PR methanol extract $(0.5\;{\mu}g/ml)$ inhibited the NMDA-induced elevation of ROS generation. Also, MK-801 showed similar protective effects. Conclusion : The present study provides the availability of PR to exert its protective effect on the neuronal cell death in various neurodegenerative pathophysiological conditions.
In this note, we mainly discuss the Gorenstein $Pr{\ddot{u}}fer$ domains. It is shown that a domain is a Gorenstein $Pr{\ddot{u}}fer$ domain if and only if every finitely generated ideal is Gorenstein projective. It is also shown that a domain is a PID (resp., Dedekind domain, $B{\acute{e}}zout$ domain) if and only if it is a Gorenstein $Pr{\ddot{u}}fer$ UFD (resp., Krull domain, GCD domain).
In this research, the genotoxic effect of Picrorrhiza Rhizoma (PR) aqueous extract was evaluated using the mouse micronucleus test. PR extract was administered once a day for 2 continuous days by oral gavage to male ICR mice at doses of 2000, 1000 and 500 mg/kg. Cyclophosphamide was used as a known genotoxic agent in a positive control. The appearance of a micronucleus (MN) in polychromatic erythrocyte (PCE) is used as an index for genotoxic potential, and PCE ratio is used as an index of cytotoxicity. Although significant (p < 0.01) increase of the number of PCE with one or more nuclei (MNPCE) was detected in cyclophosphamide treated groups, no significant increases of MNPCE numbers were observed in all three different dosages of PR extracts treated mice with over 0.39 of the individual polychromatic erythrocyte ratio in all mice used in this study. The results obtained indicated that PR extract shows no genotoxicity effects up to 2000 mg/kg dosing levels.
Objectives: The purpose of this study was to examine the efficacy of Picrorrhizae Rhizoma (PR) aqueous extracts on the formalin-induced paw chronic inflammation in mice. Methods: PR extracts (500, 250 and 125mg/kg) or distilled water (DW) were orally administered once a day for 10 days to formalin-injected chronic inflammatory mice. The paw thicknesses and volumes were measured daily and the paw wet-weight and histological profiles were conducted at termination with paw tumor necrosis factor (TNF)-${\alpha}$ contents measurement. The anti-inflammatory effects of PR extracts were compared with dexamethasone. Results: In DW treated control group, the paw thickness, paw wet-weights and paw TNF-${\alpha}$ contents were markedly increased. Severe chronic inflammation signs such as severe fibrosis, the formation of necrotic debris, and infiltration of inflammatory cells were detected in histopathological observations. However, these formalin-induced changes were dramatically decreased by treatment of dexamethasone and all three different dosages of PR extracts. The anti-inflammatory effects of PR at highest dose were slighter than that of dexamethasone, but it did not show any harmful effects on the body weight contrary to dexamethasone. Conclusion: These results suggest that PR extracts have safe and favorable effects on formalin-induced chronic inflammation.

We have isolated and artificially expressed three cDNA clones of Capsicum annuum PR5 genes for elucidating the antifungal activity against Phytophthora capsici which contracted a hot pepper root rot in field condition. Three divergent PR5 proteins from hot pepper were designated as CAPR5-1 and CAPR5-2 from susceptible cultivar (Subicho) as well as CAPR5-3 from resistant cultivar (CM331) in response to P. capsici. The cDNA similarity was found over 80% of identity among the three CAPR5s, and deduced amino acid sequence was characterized that all of CAPR5s contained 16 cysteine residues which possibly had a significant role in the structural formation. The result of genomic DNA blot showed that CAPR5-1 and CAPR5-2 existed as single copy in the Subicho genome. Three recombinant CPARs in E. coli were identified by SDS-PACE, and each expressed protein was treated on the PDA medium which contained cultured pathogens. Although three CAPR5 proteins did not affected the hyphal growth of Glomerella glycines and Colletotrichum fagenarium, CAPR5-1, CAPR5-2, and CAPR5-3 showed a specific antifungal activities against P. capsici.
