Inhibitory Effects of Type IV Collagenase by Disulfiram (Disulfiram에 의한 type IV collagenase 저해효과)

  • The cancer cells, characterized by local invasion and distant metastasis, are very dependant on extracellular matrix. The expression of matrix metalloproteinases (MMPs) has been implicated in the invasion and metastasis of cancer cells. Among the human MMPs, matirx metalloproteinase-2 (MMP-2) and matrix metalloproteinse-9 (MMP-9) are key enzymes that degrade type IV collagen of the matrix. Here, we studied the effect of disulfiram, an anti-tumor compound, on the suppression of the tumor invasion and the activity of MMP-2, MMP-9 in human osteosarcoma cells (U2OS). Disulfiram had the type IV collagenase inhibitory activity, the effect of inhibition of gene and protein expression, and these inhibitions were responsible for blocking invasion through cell mediated and non-cell mediated pathways. In conclusion, disulfiram inhibited expression of MMP-2 and MMP-9, and regulated the invasion of U2OS, Caki-1 and Caski. These observations raise the possibility of clinical therapeutic applications for disulfiram used as a potential inhibitor of cancer invasion.

Elevation of anti-oxidative activity and inhibitory activities against tyrosinase, elastase, collagenase and hyaluronidase of Oplismenus undulatifolius by elicitor treatment (Elicitor 처리한 주름조개풀(Oplismenus undulatifolius)의 항산화, tyrosinase, elastase, collagenase 및 hyaluronidase 억제 활성 증대)

  • This study was designed to assess the change of the antioxidative and biological enzyme activities [tyrosinase, elastase, collagenase, and hyaluronidase (HAase)] of extracts from elicitor-treated Oplismenus undulatifolius. Elicitor-treated O. undulatifolius showed higher total phenolic content than the non-treated extract. As a result of comparing the anti-oxidant activity of elicitor-treated O. undulatifolius extract and non-treated extract, the elicitor-treated group showed high activity. Elicitor-treated O. undulatifolius showed higher elastase and collagenase inhibitory activities as anti-wrinkle effect, tyrosinase inhibitory activity as whitening effect, and anti-inflammation effect to induced as HAase inhibition than the non-treated extract. Therefore, elicitor-treatment during O. undulatifolius cultivation in outdoors will elevate total phenolics content in the plant and elevate of various bioactivities, which will yield high quality for industrialization.

Study on Biological Effect of Euiiin-tang(y$\grave{i}$y$\check{i}$r$\acute{e}$n-t$\bar{a}$ng) Extracts by Extraction Methods (의이인탕 추출방법에 따른 생리활성 연구)

  • Objectives: The purpose of this study is to investigate biological effects of Euiiin-tang(Y$\grave{i}$y$\check{i}$r$\acute{e}$n-t$\bar{a}$ng) extracts by extraction methods. Methods: To investigate the effects of anti-oxidative and anti-inflammatory activity, we measured DPPH radical scavenging assay, collagenase inhibition assay and COX-2 inhibition assay. Results: In case of measuring anti-oxidative and anti-inflammatory activities of Euiiin-tang(Y$\grave{i}$y$\check{i}$r$\acute{e}$n-t$\bar{a}$ng) by collagenase and COX-2 inhibition activity, it came out that Euiiin-tang(Y$\grave{i}$y$\check{i}$r$\acute{e}$n-t$\bar{a}$ng) was unable to inhibit collagenase, but was effective to inhibit COX-2. Cytotoxicity analysis of Euiiin-tang(Y$\grave{i}$y$\check{i}$r$\acute{e}$n-t$\bar{a}$ng) extracts by MTT assay principles, there was no cytotoxicity in aqueous and 50% ethanol extracts in any level of concentration. Conclusions: This results suggest that Euiiin-tang(Y$\grave{i}$y$\check{i}$r$\acute{e}$n-t$\bar{a}$ng) extracts have anti-oxidative and anti-inflammatory activities.

Study on the Physiological Activity of Chrysanthemum indicum Flower Extracts Using the Hydrothermal Water Extraction Method (감국꽃 열수 추출물의 생리활성에 관한 연구)

  • In this study, the hydrothermal extract of Chrysanthemum indicum Flowers are analyzed for the identification of its functional material value. Various aromatic compounds were detected as a result of GC-MS analysis, and antioxidant analysis confirmed the high antioxidant activity of the extract. High antibacterial activity was confirmed in the TIME KILL TEST using the 70% ethanol extract, which showed the highest activity, and the inhibition of intracellular melanogenesis was confirmed to be 115.77 ± 6.67%, 112.42 ± 1.64%, and 67.12 ± 3.89% compared to the α-MSH treatment group at concentrations of 1%, 1.2%, and 1.5%, respectively, when the extract was treated with 1%, 1.2%, and 1.5%. Collagenage activity inhibition assay showed collagenase inhibition of 56.57 ± 2.28%, 58.64 ± 2.99 and 29.53 ± 1.03% when treated at concentrations of 1.5%, 3% and 5%. The cytotoxicity test of the extract showed no cytotoxicity at concentrations of 1.5%, 3% and 5%. Therefore, this study predicts that the hydrothermal extract of Chrysanthemum indicum Flowers will be useful as a functional cosmetic material.

Whitening and Anti-wrinkling Effects of Fractions from Prunus persica Flos (도화(Prunus persica Flos) 분획물의 미백 및 주름개선 효과)

  • In this study, the compounds of Prunus persica Flos were extracted with 70.0% acetone and were purified using a sephadex-LH-20 column chromatography. As a result, eight fractions were isolated. For whitening effects, the tyrosinase inhibitory activity was determined to be 92.2% in Fr.-8 isolated from P. persica Flos at 1.000 ppm. The melanoma cell-originated tyrosinase inhibitory effect of Fr.-8 from P. persica Flos was approximately 63.4% at 100 ppm. The inhibitory activity on melanin synthesis by Fr.-8 isolated from P. persica Flos was about 71.7% at 100 ppm concentration. For anti-wrinkling effects, the elastase inhibition activities by Fr.-5, 7 isolated from P. persica Flos were around 71.4 and 74.5% respectively at 1,000 ppm. The collagenase inhibition activity and collagen synthesis by Fr.-8 isolated from P. persica Flos was about 80.0% at 100 ppm. All these findings suggested that the fractions of P. persica Flos have great potential as cosmeceutical ingredients with whitening and anti-wrinkling effects.

Antiaging and antioxidant activity of Hypsizygus marmoreus extracts (흰만가닥버섯(Hypsizygus marmoreus)추출물의 항노화 및 항산화 활성)

  • This study was carried out to investigate the effects of the extracts of 80% EtOH and EtOAc, BuOH, and D.W in Korean native Hypsizygus marmoreus. Antioxidation and antioxidant analysis to provide the possibility of a multifunctional cosmetic raw material. As a result of analyzing the cytotoxicity of the sample using the animal cell line HaCaT cell, it was confirmed that the toxicity of the sample hardly affected the cells. The inhibition rate of elastase was 30.41% at 10% of sample concentration and inhibition activity of collagenase was 11.65% at 1% of sample concentration. Hypsizygus marmoreus showed better antioxidant activity than similar mushroom. The total phenol contents of the fractions were 778.4 mg and 2.59 mg per 1 g of white mushroom, confirming its potential as a natural antioxidant. As a result, the extract of Hypsizygus marmoreus is considered to be highly valuable as a natural cosmetic material having antioxidant and antioxidant functions.

Analysis on Antioxidant Activity and Agronomic Characteristics of Extract from Smilacis Chinae Radix

  • The Smilacis chinae Radix refers to the root of Smilax chinae L distributed in mountain and filed of Korea, and it is a vine shrub in the Lilaceae family, called Berchemia berchemiaefolia, and is referred to as Smilacis chinae Radix in it's a natural medicine name. Antibacterial, inflammatory, and antioxidant activity were studied in Smilacis chinae Radix. In this study, biological activities such as antioxidant (DPPH, ABTs, TPC), cytotoxicity, wrinkle improvement, and whitening improvement to increase the utilization value of Smilacis chinae Radix and identify the botanical value. Therefore, we tried to explore the applicability of Smilacis chinae Radix as a functional cosmetic material. Smilacis chinae Radix (SCR) was dried and extracted with ethanol. In order to measure the biological activity of the SCR, antioxidant activity, inhibition activities of collagenase, tyrosinase and cell viability were measured. The DPPH (1,1-diphenyl-2-picryl hydrazyl) radical scavenging activity in the extract with a concentration of 400㎍/mL is 91.22% ± 0.41%%. ABTs (2,2'-azinobis-3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity in the extract with a concentration of 400㎍/mL is 99.60% ± 0.03%. Total polyphenol contents (TPC) are 0.203 ± 0.05 mg GAE/mg Ext when SCR was lmg/mL. And the Cell viability for HaCaT derived human keratinocyte and Raw264.7, a mouse-derived macrophage was determined using the MTT assay. When cell was treated with 100㎍/mL of SCR, HaCaT cell showed cell viability of 78.09 ± 0.1% and Raw264.7 cell showed cell viability of 91.88 ± 0.42%. From the above results, we have shown the possibility that the CSR have antioxidant ability, inhibition activity of collagenase and tyrosinase and cell safety ability which can be useful in a functional cosmetic material.

Effects of Aloe and Violae herba Extract on the Anti-oxidant, Anti-inflammatory, Anti-wrinkle and Whitening (노회(蘆薈)(알로에), 자화지정(紫花地丁)의 항산화, 항염증, 주름, 미백에 미치는 영향)

  • Objective : This study was performed to assess the effects of Aloe and Violae herba extracts on skin disease and skin beauty. Methods : Anti-oxidant effects were measured by the scavenging for DPPH radical, xanthine oxidase activity. Anti-inflammatory effects were examined by relations in NO synthesis, IL-$1{\beta}$, IL-6, TNF-$\alpha$, NF-kB, COX-2, MAP kinase. The skin wrinkle formation effects were measured by collagenase and elastase activities. The whitening effects were examined by tyrosinase activities, melanin synthesis in MNT-1 cell. Results : 1. In an anti-oxidant test, Aloe and Violae herba extracts showed high radical scavenging activity. 2. In an anti-inflammatory test, Aloe and Violae herba extracts strongly inhibited the lipopolysaccharide(LPS)-induced nitric oxide(NO) release from the RAW 246.7 macrophage cells. Aloe and Violae herba extracts also inhibited the LPS-induced IL-$1{\beta}$ and COX-2 expressions. The inhibitory effects of Aloe and Violae herba extracts on macrophage activation were via the inhibition of NF-kB, evidenced by transient transfection assay. Furthermore, Aloe and Violae herba extracts weakly inhibited the activation of Jun-N-terminal kinase(JNK) but they did not have any effects on p38 MAP kinase in RAW 264.7 cells. 3. In the skin wrinkle formation assay, Aloe extract strongly inhibited collagenase and elastase, whose activity are tightly related with the wrinkle formation. 4. In the skin whitening assay, Aloe and Viloae herba extracts weakly inhibited tyrosinase activity, however, it was not statistically significant. Besides they did not have any effects on melanin synthesis, indicating that they could not be applicable for skin whitening. Conclusion : This study show that Aloe and Violae herba extracts may play a significant role in skin disease and skin beauty.

Effect of ethanol extract from mixture including Angelicae Dahuricae Radix on Dermal Anti-aging and Whitening (백지를 포함하는 한약재 복합 에탄올 추출물이 피부 항노화 및 미백에 미치는 영향)

  • Objective : Herbal medicinal mixture (JMB) are consisted of Caryophylli Flos, Aucklandiae Radix, and Angelicae Dahuricae Radix. Each of these herbal medicines has studied on anti-aging effect in vitro. So this study was conducted to investigate efficacy and potency of JMB extract on dermal anti-aging and whitening. Methods : The JMB was extracted at room temperature by 80% ethanol. Collagenase and elastase inhibition activity in JMB ethanol extract were determined at 10, 50, 100, 500, 1000 mg/ml concentrations by colorimetric method. The toxic range of JMB ethanol extract was evaluated using MTT assay. Also, The inhibitory effect of JMB ethanol extract on tyrosinase activity and melanin contents in mouse melanoma cell line (B16F10 cell) was identified at 50, 100, 200 ㎍/㎖ levels by spectrometric assay. In each analysis, EGCG (epigallocatechin gallate) and Kojic acid were used as positive controls, respectively. Results : The elastase and collagenase inhibitory activity of JMB ethanol extract increased dose dependently. Also, The MTT assay showed that JMB, up to 400 ㎍/㎖ concentration, exhibited no toxic effect to the B16F10 cell. And following the JMB ethanol extract treat, cellular melanin contents and tyrosinase activity were dose-dependently decreased compared to those of control. Conclusion : These results suggest that JMB ethanol extract has effects to inhibitory activity on dermal wrinkle enzyme and melanogenesis. Therefore, JMB has applicable benefits for development of materials or products to have whitening and anti-aging functions on skin.

Establishment of Optimum Extraction Conditions and Wrinkle Improvement Evaluation of Glycosaminoglycans in Styela plicata (오만둥이(Styela plicata)에서 글리코스아미노글리칸의 최적 추출조건 설정 및 주름개선 효능)

  • Styela plicata are naturally-occurring marine resources easily found along the coastlines that have established their niche as functional food and nutraceuticals ingredient along with their increasing consumer demand. Ascidian contain a large amount of dietary fiber but only the meat has been utilized and consumed while the rest of its parts are discarded. Also, various studies have been conducted on the meat of ascidians while studies on the functionality of the ascidian tunics, which were mostly undervalued, were scarce. In this study, we investigated and explored the glycosaminoglycans (GAGs) contents in the tunics of S. plicata, and their potential use as functional ingredient in pharmaceutical and nutraceutical applications. Sulfated GAGs and uronic acids contents were 8.9-10.7 g/100 g and 9.4-11.3 g/100 g, respectively. Highest GAGs content was extracted with optimum Brix at 7-9. Extraction efficiency using hot water at 121℃ was 4.22% while enzyme extraction using Protamex was more efficient at 5.91%. GAGs extracted from S. plicata tunics exhibited collagenase inhibitory activity of 75.2% at 100 ㎍/mL and procollagen synthesis activity of 80.1% at 100 ㎍/mL.
